Derivatives of sym-Triazines. 6. Some Special Features of the Addition of Substituted Acetylenes to Triazine Monoazides

The special features of the interaction of 2-azido-4-R-6-R'-sym-triazines with some acetylenic compounds have been investigated. A series of new derivatives of sym-triazine linked at position 2 with a 1,2,3-triazole ring has been synthesized.     

Derivatives of <Emphasis Type="Italic">sym</Emphasis>-Triazine. 1. Synthesis and Conversions of Quaternary Methylammonium Salts of 2-Chloro-4,6-disubstituted 1,3,5-Triazines in Nucleophilic Substitution Reactions

New quaternary ammonium salts of monochloro-4,6-disubstituted 1,3,5-triazines have been obtained. These compounds are convenient synthons for the synthesis of new cyano, alkoxy, and other derivatives of sym-triazine. Some conversions of the synthesized salts have been studied. An elimination-isomerization reaction of the sym-triazine ring into a 2-oxo-1,2-dihydro-sym-triazine ring was discovered.     

sym-Triazine Derivatives. 2. Synthesis, Properties, and Structure of 2-Oxo-1,2-dihydro-sym-triazines

Previously unreported 2-oxo-1,2-dihydro-sym-triazines have been prepared and their alkylation reactions have been studied. It was found that, independently of the structure and nature of the substituent in positions 4 and 6 of the triazine ring or the structure of the alkyl halide used, the reaction occurs regioselectively at the oxygen atom. The stereostructure of the 2-oxo-1,2-dihydro-sym-triazines was investigated by X-ray analysis and it was shown that they exist as dimers in the crystalline state.     

Synthesis and Properties of sym-Triazine Derivatives. 18. Synthesis of N-Substituted 2,4-Diamino-6-(benzothiazolyl-2-thiomethyl)-sym-triazines

The condensation of methyl benzothiazolyl-2-thioacetate with N-substituted biguanides in the presence of sodium methylate gave 2-amino-4-(RR'-amino)-6-(benzothiazolyl-2-thiomethyl)-sym-triazines. In a number of cases the 1,1-disubstituted 5-(benzothiazolyl-2-thioacetyl)biguanides could also be separated and these could be cyclized by refluxing in DMF to the corresponding substituted sym-triazines described. The latter type of compound was also synthesized by treating 2-amino-4-trichloromethyl-6-(benzothiazolyl-2-thiomethyl)-sym-triazine with primary and secondary aliphatic and heterocyclic amines or by the reaction of N-substituted 2,4-diamino-6-chloromethyl-sym-triazines with 2-mercaptobenzothiazole.     

sym-triazines. 7. Hydrolysis and cyclization of 1,3,5-triazine series mononitriles

The routes of base and acid hydrolysis of sym-triazine mononitriles has been studied for aqueous or aqueous-alcohol media. Depending on the pH of the medium, the concentration of reagents, and the temperature it was found that the hydrolysis led to the formation of amides, oxo or alkoxy 1,3,5-triazines, or to an unstable sym-triazine carboxylic acid, the latter leading to formation of substituted sym-triazines. Synthesis and structures of 4,6-disubstituted 2-(5-methyl-1,2,4-oxadiazol-3-yl)-1,3,5-triazines

New 1,2,4-oxadiazolyl-1,3,5-triazines were synthesized from amidoximes derived from sym-triazine mononitriles. The structure of one of the resulting compounds was studied in detail by X-ray diffraction. __________ Published in Russian in Izvestiya Akademii Nauk. Di-4(3H)-quinazolinon-2-yl Derivatives from the Diacid Chlorides of Pinic and sym-Homopinic Acids

The corresponding diamides have been synthesized by the interaction of the diacid chlorides of cis-2,2-dimethyl-3-carboxycyclobutaneacetic acid (pinic acid) and cis-2,2-dimethylcyclobutane-1,3-diacetic acid (sym-homopinic acid) with two equivalents of anthranilic acid. Treatment of the diamides with formamide gave 2,2-dimethyl-1-[4(3H)-quinazolinon-2-yl]methyl-3-[4(3H)-quinazolinon-2-yl]cyclobutane and 2,2-dimethyl-1,3-di[4(3H)-quinazolinon-2-ylmethyl]cyclobutane respectively.     

Reactions of Amidines with 5-Methylene-1,3-dioxolan-2-ones

The composition and yields of the products from the reaction of 4,4-dialkyl-5-methylene-1,3-dioxolan-2-ones with amidines depend on the structure of the initial amidine and on the reaction conditions. 2-Aminopyridines lead to 3-substituted 4-hydroxy-4-methyloxazolidin-2-ones and 4-methylene-oxazolidin-2-ones and also to sym-carbamides. 2-Amino-4,6-dimethylpyrimidine leads to the corresponding 4-methyleneoxazolidin-2-one. 3-Aminothiazoles give linear oxourethanes and sym-carbamides.     

Synthesis of substituted 2-amino-4,6-bis(nonafluoro-tert-butyl)-1,3,5-triazines

A procedure was developed for the synthesis of substituted 2-amino-4,6-bis(nonafluoro-tert-butyl)-1,3,5-triazines from 2-chloro-4,6-bis(nonafluoro-tert-butyl)-1,3,5-triazine and aliphatic and aromatic amines. The data of¹⁹F NMR spectroscopy are indicative of the presence of a barrier to internal rotation. Translated fromIzvestiya Akademii Nauk. The investigations in 2,3′-biquinoline series. 25*. Synthesis of 4-(2-quinolinyl)-pyrrolo[1,2-<Emphasis Type="Italic">a</Emphasis>]quinolines and 4-(2-quinolyl)-imidazo[1,2-<Emphasis Type="Italic">a</Emphasis>]quinolines

A method has been developed for the synthesis of 4-(2-quinolinyl)-1,2,3,3a-tetrahydropyrrolo-[1,2-a]quinolines based on the 1,3-dipolar cycloaddition reaction of α,β-unsaturated carbonyl compounds to 2,3′-biquinolinium salts. Oxidation in benzene using MnO₂ gave 4-(2-quinolinyl)-pyrrolo[1,2-a]quinolines. It was found that a side product of the 1,3-dipolar cycloaddition is 7,14-di-benzoyl-6,13-di(2-quinolyl)-6a,7,13a,14-tetrahydro-7a,14a-diazadibenzo[a,h]anthracene. Reaction of 1′-phenacyl-2,3′-biquinolinium salts with hydroxylamine in acetic acid gave 4-(2-quinolyl)imidazo-[1,2-a]quinolines. *For Communication 24 see [1]. Synthesis of isomeric 2-oxazolidinones from (1<Emphasis Type="Italic">R</Emphasis>,2<Emphasis Type="Italic">R</Emphasis>)-and (1<Emphasis Type="Italic">S</Emphasis>,2<Emphasis Type="Italic">S</Emphasis>)-2-amino-1-(4-nitrophenyl)-1,3-propanediols

A synthesis is reported for (4R,5R)-and (4S,5S)-4-hydroxymethyl-5-(4-nitrophenyl)oxazolidin-2-ones and (1′R,4R)-and (1′S,4S)-4-[hydroxy(4-nitrophenyl)methyl]oxazolidin-2-ones from (1R,2R)-and (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediols. Reaction of 4-Amino-3-methylthio-5-oxo-6-R-4,5-dihydro-1,2,4-triazines with Sulfonylacetic Acid Nitriles

The reactions of 4-amino-3-methylthio-5-oxo-6-R-4,5-dihydro-1,2,4-triazines with arylsulfonylacetonitriles and with dinitriles of sulfonyldiacetic acid and methylenebis(sulfonylacetic) acid have been studied. 7-Amino-3-R-8-(R'-sulfonyl)-1,4-dihydropyrazolo[5,1-c][1,2,4]triazin-4-ones have been synthesized and these are the first representatives of geminal sulfones in which the heterocyclic ring is bound directly to the sulfur atoms of the sulfonyl groups.     

Synthesis of 6-Amino-5-R2-7-(6-R1-4-oxo-3,4-dihydro-2-quinazolyl)-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles

It has been found that malonodinitrile and 2-(6-R¹-oxo-3,4-dihydro-2-quinazolyl)acetonitrile in the presence of triethylamine undergo hetarylation by 5,6-dichloro-2,3-pyrazinedicarbonitrile at the active methylene group to give the triethylammonium salt of 2-(3-chloro-5,6-dicyano-2-pyrazinyl)malononitrile or 5-chloro-6-cyano(6-R¹-4-oxo-1,2,3,4-tetrahydro-2-quinazolylidene)methyl-2,3-pyrazinedicarbonitriles. Reaction of these with primary amines leads to annelation of the pyrrole ring at the pyrazine [b] edge to give 6-amino-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3,7-tricarbonitriles and 6-amino-5-R²-7-(6-R¹-4-oxo-3,4-dihydro-2-quinazolyl)-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles respectively.     

Mechanisms and kinetics of the elementotropic rearrangements of tetrahydro-4,4,8,8-tetramethyl-4,8-disila-sym-indacene

The dynamic stereochemistry of silatropic rearrangements of tetrahydro-4,4,8,8-tetra-methyl-4,8-disila-sym-indacene (dimer of 6,6-dimethyl-6-silafulvene), which exists in solution as an equilibrium mixture of two interconverting isomers, was studied. The mechanisms and complete kinetic scheme of rearrangements were established using¹H,¹³C, and²⁹Si 2D quantitative EXSY NMR spectroscopy. It was found that the interconversion and degenerate rearrangements of the observed isomers proceedvia two concurrent pathways due to the formation of different intermediates. The activation parameters of the rearrangements were determined by means of total lineshape analysis of dynamic NMR (DNMR) spectra. Translated fromIzvestiya Akademii Nauk. Derivatives of imidazo-<Emphasis Type="Italic">sym</Emphasis>-triazine

2-Dimethylamino-4,6,7,8-tetrahydroimidazo[1,2-a]-sym-triazin-4-one synthesized by us previously is converted by the action of aryl isocyanates and arenesulfonyl chlorides into the corresponding derivatives of urea and N-arenesulfonyl-substituted imidazo-sym-triazinones. It was shown that 6-(2-chloroethoxy)-2-dimethylamino-4-(methoxycarbonylmethylamino)-sym-triazine is converted by thermolysis into 2-dimethylamino-8-methoxycarbonylmethyl-4,6,7,8-tetrahydroimidazo[1,2-a]-1,3,5-triazin-4-one, which is also obtained by the action of methyl bromoacetate on the imidazo-sym-triazinone. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 8, pp. 1230–1235, August, 2008.     

4-(3-Cyanopyridin-2-ylthio)acetoacetates in synthesis of heterocycles

Substituted 2-amino-4-aryl-3-cyano-5-oxo-5,6-dihydro-4H-pyrano[2,3-d]pyrido[3",2":4,5]thieno[3,2-b]pyridines were synthesized by the reactions of 4-hydroxy-1H-thieno[2,3-b;4,5-b]dipyridin-2-ones with arylidenemalononitriles or by the three-component reactions of hydroxythienodipyridinones with aldehydes and malononitrile in DMF in the presence of triethylamine. Methods for syntheses of substituted 3-alkoxycarbonyl-6-amino-4-aryl-2-(3-cyanopyridin-2-ylthiomethyl)-4H-pyrans were developed on the basis of the reactions of 4-(3-cyanopyridin-2-ylthio)acetoacetates and arylidenemalononitriles or aldehydes and malononitrile. Ethyl 4-(3-cyanopyridin-2-ylthio)acetoacetate and 4-methoxybenzylidenecyanothioacetamide were used for the synthesis of 6-(pyridin-2-ylthiomethyl)-3-cyanopyridine-2(1H)-thione.     

Synthesis of substituted benzoxazinylthieno[2,3-<Emphasis Type="Italic">b</Emphasis>]pyridines by the reaction of (3-cyanopyridin-2-ylthio)acetic acids or their amides with <Emphasis Type="Italic">o</Emphasis>-aminophenyl(diphenyl)carbinol

A general method for the synthesis of 3-amino-2-(4,4-diphenyl-4H-3,1-benzoxazin-2-yl)thieno[2,3-b]pyridines was proposed. The method involves reactions of (3-cyanopyridin-2-ylthio)acetic acids or their amides with o-aminophenyl(diphenyl)carbinol in nitromethane in the presence of perchloric acid followed by neutralization of the resulting salts.__________Published in Russian in Izvestiya Akademii Nauk. Synthesis and Alkylation of Piperidinium 4,5-trans-3-Cyano-6-hydroxy-4-(2-iodophenyl)-6-methyl-5-(2-methylphenyl)carbamoyl-1,4,5,6-tetrahydropyridine-2-thiolate. Molecular and Crystal Structure of 3-(2-Iodophenyl)-2-(4-phenyl-2-thiazolyl)acrylonitrile

The reaction of o-iodobenzaldehyde, cyanothioacetamide, and N-acetoacetyl-o-toluidine in the presence of piperidine gave piperidinium 4,5-trans-3-cyano-6-hydroxy-4-(2-iodophenyl)-6-methyl-5-(2-methylphenyl)carbamoyl-1,4,5,6-tetrahydropyridine-2-thiolate used in the synthesis of substituted 4,5-trans-2-alkylthiotetrahydropyridines and 2-(2-thiazolyl)acrylonitriles. The structure of 3-(2-iodophenyl)-2-(4-phenyl-2-thiazolyl)acrylonitrile was studied using X-ray diffraction structural analysis.     

Sonochemical synthesis of Biginelli compounds

The ultrasound effect accelerates the Biginelli reaction 40 and more times. A sonochemical method for the synthesis of ethyl 4-R-6-methyl-2-oxo- and 4-R-6-methyl-2-thioxo-1,2,3,4-tetrahydropyrimidine-5-carboxylates was developed. The target products were obtained within 2 to 5 min in 90—95% yields.
