An optimized extraction and cleanup method for the analysis of chlortetracycline (CTC), doxycycline (DC), oxytetracycline (OTC) and tetracycline (TC) in soil is presented. Soil extraction in a pressurized liquid extraction system, followed by extract clean up using solid-phase extraction (SPE) and tetracycline determination by liquid chromatography tandem mass spectrometry (LC-MS/MS) provided appropriate efficiency and reproducibility. Different dispersing agents and solvents for soil extraction and several SPE cartridges for cleanup were compared. The best extraction results were obtained using ethylenediamine tetraacetic acid-treated sand as dispersing agent, and water at 70 °C. The most effective cleanup was obtained using Strata-XTM sorbent in combination with a strong anion exchange cartridge. Recoveries ranged from 71% to 96% and precision, as indicated by the relative standard deviations, was within the range of 8–15%. The limits of quantification (LOQs) by using LC-MS/MS, based on signal-to-noise ratio (S/N) of 10, ranged from 1 μg kg−1 for TC to 5 μg kg−1 for CTC. These results pointed out that this technique is appropriate to determine tetracyclines in soils. Analysis of 100 samples taken in the Valencian Community revealed that, in soil, up to 5 μg kg−1 CTC, 15 μg kg−1 OTC, 18 μg kg−1 TC, and 12 μg kg−1 DC could be detected. Detection of the analytes in several samples, which typify great part of the Spanish agricultural soils, should be outlined as most important result of this study. An efficient and selective automated analytical method for the determination and quantification of a selected group of 12 organochlorine and organophosphorous pesticides in milk-based infant formulas has been developed. The samples were extracted by pressurized liquid extraction (PLE) and analysed using GC-MS/MS. The use of alumina as the fat retainer in the PLE extraction cell, together with the application of an injector temperature program during the GC injection process, avoided typical matrix interferences without the application of additional cleanup steps. Mean recoveries of between 70 and 110% were achieved for most of the compounds, except for chlorpyrifos methyl (50%), vinclozoline (48%), fenitrothion (56%) and procymidone (53%), with relative standard deviations ranging from 9 to 17%. Low limits of quantification were obtained for the studied compounds, from 0.01 to 2.6 μg kg−1, thus guaranteeing their accurate determination within the rigorous requirements established for baby food. The validated method was applied to a pilot monitoring study in Spain. Twenty five samples of different brands of powdered infant formulas were obtained from supermarkets. Positive findings of endosulfan I, endosulfan II, fenitrothion, chlorpyrifos ethyl and bifenthrin were detected at concentrations ranging from 0.03 to 5.03 μg kg−1.  相似文献

3-Chloropropane-1,2-diol (3-MCPD) and 1,3-dichloro-2-propanol (1,3-DCP) were determined for the first time in bakery foods using pressurized liquid extraction (PLE) combined with in situ derivatization and GC-MS analysis. This one-step protocol uses N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) as silylation reagent. Initially, screening experimental design was applied to evaluate the effects of the variables potentially affecting the extraction process, namely extraction time (min) and temperature (°C), number of cycles, dispersant reagent (diatomaceous earth in powder form and as particulate matter with high pore volume Extrelut NT) and percent of flush ethyl acetate volume (%). To reduce the time of analysis and improve the sensitivity, derivatization of the compounds was performed in the cell extraction. Conditions, such as the volume of BSTFA, temperature and time for the in situ derivatization of analytes using PLE, were optimized by a screening design followed to a Doehlert response surface design. The effect of the in-cell dispersants/adsorbents with diatomaceous earth, Florisil and sodium sulfate anhydrous was investigated using a Box-Behnken design. Using the final best conditions, 1 g of sample dispersed with 0.1 g of sodium sulfate anhydrous and 2.5 g diatomaceous earth was extracted with ethyl acetate. 1 g of Florisil, as clean-up adsorbent, and 70 μL of BSTFA were used for 3 min at 70°C. Under the optimum conditions, the calibration curves showed good linearity (R(2)>0.9994) and precision (relative standard deviation, RSD≤2.4%) within the tested ranges. The limits of quantification for 1,3-DCP and 3-MCDP, 1.6 and 1.7 μg kg(-1), respectively, are far below the established limits in the European and American legislations. The accuracy, precision, linearity, and limits of quantification provided make this analytical method suitable for routine control. The method was applied to the analysis of several toasted bread, snacks, cookies and cereal samples, none of which contained chloropropanols at concentrations above the legislation levels.  相似文献

The effect that a high amount of mineral filler might have on the extraction process of antioxidants from polyethylene and polypropylene was investigated. Extraction of Irganox 1010, Irganox 1076 and Irgafos 168, along with its oxidation product 2,4-di-tert-butylphenol, from freeze ground polyethylene–based (PE–based) and polypropylene–based (PP–based) mineral concentrates of 85 w/w calcium carbonate (CaCO3) and 75 w/w talc was carried out by pressurized fluid extraction (PLE) prior to high performance liquid chromatography (HPLC). Results indicate that 85 w/w CaCO3 did not affect the extraction process from PE or PP. For talc concentrates, additive recovery from PE and PP was considerably lower. The relation of recovery yield and mixing time was investigated for the talc concentrates and it was concluded that the presence of talc seemed to accelerate the rate of antioxidant consumption during sample processing, thus, less antioxidant was left to be extracted from the polyolefin; rather than talc has limited the extraction process. The method developed in this work has been applied to determine these compounds in several commercial samples.  相似文献

An analytical methodology including pressurized liquid extraction (PLE) as sample treatment to isolate retinyl acetate and tocopherols from infant formulas has been developed. The milk extracts were kept at −18 °C for 30 min and after filtration could be injected directly into the chromatographic system. Thus, a rapid and simple routine control method of these products is possible.

The parameters affecting both the extraction process and the liquid chromatography (LC) system were optimized. PLE was performed using one cycle of extraction during a static time of 5 min. Methanol was chosen as the extraction solvent for a temperature of 50 °C. Chromatographic separation was accomplished using a RP-18 column; the mobile phase used was methanol–water (94:6, v/v) containing 2.5 mM acetic acid/sodium acetate buffer. Electrochemical detection in amperometric mode with a glassy carbon electrode at +1100 mV was applied. The proposed methodology was successfully used for the determination of retinyl acetate, δ-tocopherol, (β + γ)-tocopherol and -tocopherol in different infant formulas. The analytes were evaluated in the same chemical form present in the samples. Recoveries were between 92 and 106%. A certified reference material of milk powder was also analyzed.  相似文献

A multiresidue method for the analysis, in egg matrices, of residues of nine quinolones used in veterinary medicine, has been developed and validated according to the provisions of Council Decision 2002/657. Compounds were extracted by a pressurized liquid extraction (PLE) technique using a 1:1 mixture of acetonitrile and a phosphoric acid buffer (pH 3.0) at 70 °C. The obtained extract was clear enough, so that no further clean-up was necessary. Analytes were determined by liquid chromatography (LC) with fluorescence detection (FL). Two chromatographic columns were compared: a high-purity silica Inertsil C8 column and a newly developed Kinetex C18 core-shell technology column. Validation was carried out at four concentration levels, using both chromatographic columns. Precision in terms of reproducibility standard deviation was between 7% and 23%, and good recoveries were obtained. Decision limit (CCα) and detection capability (CCβ) values obtained with the Inertsil and Kinetex columns were in the 0.2-19.8 μg kg−1 and 0.4-33.5 μg kg−1 concentration ranges, respectively. The proposed method allows residues of quinolones banned for use with laying hens to be detected and quantified in eggs. About twenty-four samples per day can be processed.  相似文献

A method to determine organochlorine pesticides in horticultural samples (lettuce, tomato, spinach, potato, turnip leaf and green bean) using pressurized liquid extraction (PLE) is described and compared with microwave assisted extraction (MAE). Significant parameters affecting PLE procedure such as temperature, static extraction time and extraction solvent were optimised and discussed. Clean-up of extracts was performed by solid phase extraction (SPE) using a carbon cartridge as adsorbent. Pesticides were determined by gas chromatography and electron capture detection (GC-ECD). Analytical recoveries obtained were ca. 100% and the relative standard deviations were lower than 15% for most of the studied pesticides with the proposed methods in each analysed matrix.  相似文献

A simple and sensitive method for simultaneous analysis of 43 pharmaceutical compounds in sewage sludge and sediment samples was developed and validated. The target compounds were extracted using pressurized liquid extraction (PLE) and then purified and pre-concentrated by solid phase extraction (SPE) using a hydrophilic-lipophilic balanced polymer. PLE extraction was performed on temperature of 100 °C, with methanol/water mixture (1/2, v/v) as extraction solvent. The quantitative analysis was performed by liquid chromatography tandem mass spectrometry using a hybrid triple quadrupole-linear ion trap mass spectrometer (LC-QqLIT-MS). Data acquisition was carried out in selected reaction monitoring (SRM) mode, monitoring two SRM transitions to ensure an accurate identification of target compounds in the samples. Additional identification and confirmation of target compounds were performed using the Information Dependent Acquisition (IDA) function. The method was validated through the estimation of the linearity, sensitivity, repeatability, reproducibility and matrix effects. The internal standard approach was used for quantification because it efficiently corrected matrix effects. Despite the strong matrix interferences, the recoveries were generally higher of 50% in both matrixes and the detection and quantification limits were very low. Beside the very good sensitivity provided by LC-QqLIT-MS, an important characteristic of the method is that all the target compounds can be simultaneously extracted, treated and analysed. Hence, it can be used for routine analysis of pharmaceuticals providing large amount of data. The method was applied for the analysis of pharmaceuticals in river sediment and wastewater sludge from three treatment plants with different treatment properties (i.e. capacity, secondary treatment, quality of influent waters). The analysis showed a widespread occurrence of pharmaceuticals in the sludge matrices.  相似文献

A method combining HPLC and pressurized liquid extraction was developed for simultaneous quantification of nine components, including eight triterpenes (ganoderic acid A, ganoderic acid Y, ganoderic acid DM, ganoderol A, ganoderol B, ganoderal A, methyl ganoderate D and ganoderate G) and a sterol (ergosterol), in Ganoderma. The determination was achieved by using a Zorbax ODS C18 analytical column (4.6 x 250 mm id, 5 microm) and gradient elution with diode-array detection. All calibration curves showed good linearity (r2 > 0.9997) within the test ranges. The developed method showed good repeatability for the quantification of the nine investigated components in Ganoderma with intra- and inter-day variations of less than 2.4% and 4.1%, respectively. The validated method was successfully applied to quantify the nine components in two species of Ganoderma, i.e. G. lucidum and G. sinense, used as Lingzhi in China. Furthermore, hierarchical clustering analysis based on the nine components in HPLC profiles from the tested 11 samples showed that chemical characteristics were significantly different between G. lucidum and G. sinense, which suggested that clinical investigation should be performed so as to ensure the safety and efficacy of medication.  相似文献

This work presents an effective sample preparation method for the determination of eight UV filter compounds, belonging to different chemical classes, in freeze-dried sludge samples. Pressurized liquid extraction (PLE) and gas chromatography-mass spectrometry (GC-MS) were selected as extraction and determination techniques, respectively. Normal-phase, reversed-phase and anionic exchange materials were tested as clean-up sorbents to reduce the complexity of raw PLE extracts. Under final working conditions, graphitized carbon (0.5 g) was used as in-cell purification sorbent for the retention of co-extracted pigments. Thereafter, a solid-phase extraction cartridge, containing 0.5 g of primary secondary amine (PSA) bonded silica, was employed for off-line removal of other interferences, mainly fatty acids, overlapping the chromatographic peaks of some UV filters. Extractions were performed with a n-hexane:dichloromethane (80:20, v:v) solution at 75°C, using a single extraction cycle of 5 min at 1500 psi. Flush volume and purge time were set at 100% and 2 min, respectively. Considering 0.5 g of sample and 1 mL as the final volume of the purified extract, the developed method provided recoveries between 73% and 112%, with limits of quantification (LOQs) from 17 to 61 ng g(-1) and a linear response range up to 10 μg g(-1). Total solvent consumption remained around 30 mL per sample. The analysis of non-spiked samples confirmed the sorption of significant amounts of several UV filters in sludge with average concentrations above 0.6 μg g(-1) for 3-(4-methylbenzylidene) camphor (4-MBC), 2-ethylhexyl-p-methoxycinnamate (EHMC) and octocrylene (OC).  相似文献

A new method based on solid phase dispersion-pressurized liquid extraction (PLE) followed by gas chromatography-mass spectrometry (GC-MS) has been developed for the determination of 26 suspected fragrance allergens (all the regulated in the EU Cosmetics Directive amenable by GC, as well as pinene and methyleugenol) in cosmetic samples. The effects of the temperature, extraction time and solvent, and dispersing sorbent, affecting the whole proposed procedure, have been evaluated using a multifactor strategy. The optima conditions after the analysis of main and second order effects entailed the extraction at 120°C for 15 min, using hexane/acetone as solvent, and florisil as dispersing sorbent. The method performance has been studied, showing good linearity (R≥0.996) as well as good precision (RSD≤10%). Detection limits (S/N=3) ranged from 0.000001 to 0.0002% (w/w), values far below the established restrictions as regard labelling in the European Cosmetics Regulation. Reliability was demonstrated through the quantitative recoveries of all the studied compounds. The absence of matrix effects allowed quantification of the compounds by calibration with standard solutions. The analysis of 10 samples (several moisturizing and anti-wrinkle creams and lotions, hand creams, and sunscreen and after-sun creams), covering very different matrices, showed the presence of suspected allergens in all the analyzed samples; in fact, half of the samples contained an elevated number of them. Although the ubiquity of these compounds was demonstrated, labelling was in all cases in consonance with the European Cosmetics Regulation.  相似文献

Rhubarb, a well-known Chinese herbal medicine, is also used in Europe and other places of the world. Anthraquinones derivatives are thought to be the major active components. A pressurized liquid extraction (PLE) and capillary zone electrophoresis (CZE) separation were developed for simultaneous determination of five anthraquinones including aloe-emodin, emodin, chrysophanol, physcion, and rhein in Rhubarb. The effects of the experimental variables on PLE and CZE have been optimized. The optimum conditions of PLE were: solvent, methanol; temperature, 140 degrees C; particle size, 0.13-0.2 mm; static extraction time, 5 min; pressure, 1500 psi; and one extraction. The best separation of the five anthraquinones could be obtained using 50 mM borate buffer (pH 8.2) containing 25% isopropyl alcohol and 25% acetontrile as modifier, while the separation voltage was 25 kV and the temperature was at 20 degrees C. The method developed is accurate, simple, and reproducible, and could be used for quality control of Rhubarb and its medical preparations.  相似文献

The scope of this work is the development of a rapid, reliable and sensitive method for the analysis of organochlorine pesticides from soils by pressurized liquid extraction (PLE). The effect of four parameters (temperature, pressure, static time and cell volume) on the extraction efficiency was studied. The great extracting power of the PLE causes the extraction of numerous interfering substances, so a more efficient purification of this extract was necessary. In this work several sorbents have also been assayed to carry out the purification of soil samples: Florisil, silica, alumina, carbon, as well as combinations of them. Finally, the proposed analytical method was validated using a certified reference soil material (CRM804-050) and the results were compared with those obtained by other extraction techniques (Soxhlet and microwave-assisted extraction).  相似文献

A fast method for simultaneous determination of five compounds (one coumarin, two alkaloids and two bibenzyls) in Dendrobium spp. using pressurized liquid extraction and ultra‐performance liquid chromatography coupled with photo diode array detection was developed. The separation was performed on an Acquity UPLC BEH C18 column (50 mm×2.1 mm id, 1.7 μm) with gradient elution of phosphate buffer (pH=7.1, 20 mM) and acetonitrile within 6 min. The method was validated for linearity, LOD and LOQ, precision and accuracy. All calibration curves showed good linearity (R2≥0.9999) within test ranges. The overall intra‐ and inter‐day variations (RSDs) of five analytes were less than 2.5%, and the detection recoveries were between 95.6 and 102.3%. The developed method was successfully applied to quantitative analysis of five investigated compounds in different species of Dendrobium. The results showed that there were great variations of their contents, though all these materials were officially used as Chinese herb, Shihu. In addition, the chromatographic fingerprints of Dendrobium spp. were also investigated.  相似文献

建立了一个检测动物源性食品中6种氟喹诺酮类药物残留的高效液相色谱方法。不同基质的样品前处理对检测影响较大:鱼、肉及肝脏等样品需经过乙腈-0.1 mol/L KH2PO4缓冲液提取,乙腈饱和的正己烷洗涤去除油脂;蛋及乳制品样品用正己烷饱和的乙腈提取,乙腈饱和的正己烷去脂。目标化合物采用高效液相色谱-荧光检测器检测,外标法定量。对市售鸡肉、猪肉、鸡蛋进行检测,添加10、20、50、100μg/kg浓度水平时,回收率在82%~105%之间,相对标准偏差在4%~12%之间,方法的检出限诺氟沙星、环丙沙星、沙拉沙星及单诺沙星为5.0μg/Kg,恩诺沙星、达氟沙星为3.0μg/Kg。  相似文献

Tocopherols and tocotrienols have been simultaneously determined in food samples using a rapid and simple analytical method including pressurized liquid extraction (PLE) and LC with electrochemical detection. Separation was carried out on a Phenomenex Synergi 4 μm Hydro‐RP 80A column, using a solution of 2.5 mM acetic acid/sodium acetate in methanol/water (99:1, v/v) as mobile phase at a flow rate of 1.0 mL/min. Column temperature was maintained at 30°C. Detection was performed by coulometric detection at 500 mV except for (β+γ)‐tocotrienol, in wheat and rye samples, which was at +350 mV. A palm oil containing a relatively large amount of γ‐tocotrienol and lower concentrations of α‐ and δ‐tocotrienols and α‐ and γ‐tocopherols was used to provide reference retention times for the tocotrienols. Analyte quantification was performed using the external standard method. The calibration equations of tocopherols were used to quantify both tocopherols and their corresponding tocotrienols. The extraction recoveries obtained using the optimized PLE conditions were in the 80–114% range, with RSDs lower than 15%. The method was successfully applied to the determination of tocotrienols and tocopherols in cereal (wheat, rye, barley, maize and oat) and palm oil samples.  相似文献

A dispersive liquid–liquid microextraction procedure was developed to extract nine fluoroquinolones in porcine blood, six of which were quantified using a univariate calibration method. Extraction parameters including type and volume of extraction and dispersive solvent and pH, were optimized using a full factorial and a central composite designs. The optimum extraction parameters were a mixture of 250 μL dichloromethane (extract solvent) and 1250 μL ACN (dispersive solvent) in 500 μL of porcine blood reached to pH 6.80. After shaking and centrifugation, the upper phase was transferred in a glass tube and evaporated under N2 steam. The residue was resuspended into 50 μL of water–ACN (70:30, v/v) and determined by CE method with DAD, under optimum separation conditions. Consequently, a tenfold enrichment factor can potentially be reached with the pretreatment, taking into account the relationship between initial sample volume and final extract volume. Optimum separation conditions were as follows: BGE solution containing equal amounts of sodium borate (Na2B4O7) and di‐sodium hydrogen phosphate (Na2HPO4) with a final concentration of 23 mmol/L containing 0.2% of poly (diallyldimethylammonium chloride) and adjusted to pH 7.80. Separation was performed applying a negative potential of 25 kV, the cartridge was maintained at 25.0°C and the electropherograms were recorded at 275 nm during 4 min. The hydrodynamic injection was performed in the cathode by applying a pressure of 50 mbar for 10 s.  相似文献

A pressurized liquid extraction (PLE) method has been optimized for the determination of polycyclic aromatic hydrocarbons (PAHs) in soil samples and it was compared with ultrasonic extraction. The extraction step was followed by gas chromatography-triple quadrupole mass spectrometry (GC-QqQ-MS/MS) analysis. Parameters such as type of solvent, extraction time, extraction temperature and number of extractions were optimized. There were no significant differences among the two extraction methods although better extraction efficiencies were obtained when PLE was used, minimizing extraction time and solvent consumption. PLE procedure was validated, obtaining limits of detection (LODs) ranging from 0.02 to 0.75 μg kg−1 and limits of quantification (LOQs) ranging from 0.07 to 2.50 μg kg−1 for the selected PAHs. Recoveries were in the range of 59-110%, except for naphthalene, which was the most volatile PAH. Finally, the method was applied to real soil samples from Southeast of Spain. PAHs concentrations were low, and phenanthrene, pyrene, fluorene, benzo[a]pyrene and chrysene were the most frequently detected analytes in the samples.  相似文献

A new HPLC coupled with evaporative light scattering detection (ELSD) method was developed for simultaneous determination of 11 major triterpene saponins, namely notoginsenoside R1 (1), ginsenosides Rg1 (2), Re (3), Rf(4), Rb1 (5), Rg2 (6), Rc (7), Rb2 (8), Rb3 (9), Rd (10), and Rg3 (11) in Panax notoginseng, a commonly used traditional Chinese medicine (TCM). Pressurized liquid extraction (PLE) was employed for sample preparation, and the analysis was achieved using a Zorbax ODS C18 column eluted with gradient water-ACN in 60 min. The drift tube temperature of ELSD was set at 60 degrees C, and nitrogen flowrate was at 1.4 L/min. The method provided good repeatability and sensitivity for quantification of 11 saponins with overall precision (including intra- and interday) and LOD of less than 2.9% (RSD) and 98 ng, respectively. The validated method was successfully applied to quantify 11 saponins in 28 samples of P. notoginseng collected in different places, which is helpful to control the quality of P. notoginseng and its related products.  相似文献

Organophosphate esters have been extensively used as flame retardants and plasticizers. The analysis of organophosphate esters in the environment is a hot topic because many of them are toxic and persistent. We developed a novel procedure for determining organophosphate esters in sediment. In this work, pressurized liquid extraction and solid‐phase microextraction are used for sample preparation to extract and concentrate the analytes, which are then analyzed by gas chromatography with flame photometric detection. The extraction parameters of pressurized liquid extraction were investigated and optimized by orthogonal design and then evaluated by range analysis and analysis of variance. Under the optimal conditions, the proposed procedure showed wide linear ranges (0.90–100 ng/g) with correlation coefficients ranging from 0.9921 to 0.9990. The detection limits of the method were in the range of 0.009–0.280 ng/g with standard deviations ranging from 2.2 to 9.5%. Recoveries of the proposed method ranged from 82.3 to 108.9% with relative standard deviations <8.4%. The obtained method was applied successfully to the determination of organophosphate esters in real sediments with recoveries varying from 79.8 to 107.3%. The proposed method was proved to be simple, easy, and sensitive for analyzing organophosphate esters in sediment samples.  相似文献
