将已建立的 7 cm 柱长的磷酸基团强阳离子交换富集整体柱与85 cm柱长的C12烷基反相整体柱结合的在线二维分离平台应用于软骨提取蛋白的蛋白质组分析。对20 μg软骨提取蛋白的酶解产物进行14个盐梯度的分级,然后对14个馏分进行反相色谱梯度分离及串联质谱鉴定,成功地鉴定得到了7434个独立肽段对应的1901个非冗余蛋白质。对所鉴定到的蛋白质进行定位分类,结果表明鉴定到的大部分蛋白质是来自于软骨细胞内部的低丰度蛋白质,这对于许多关节类疾病的研究有重要意义。  相似文献

以4-乙烯基苯硼酸(VPBA)为功能单体,季戊四醇三丙烯酸酯(PETA)为交联剂,乙二醇和二甘醇为二元致孔剂,通过热引发自由基聚合反应,在毛细管内原位聚合制备得到一种poly(VPBA-co-PETA)毛细管整体柱。所制备的整体柱具有典型的硼酸亲和特性,可以特异性地捕获含有顺二羟基的化合物。在前期实验的基础上,将其用于两种中草药 蒲公英和刺果卫矛中含顺二羟基的小分子活性物质的富集。这两种中药提取物直接用高效液相色谱进行检测时,其活性成分绿原酸等因含量太低而使检测困难或无法检测;经过硼酸亲和整体柱富集后收集洗脱液进行检测,色谱峰响应则显著提高。该结果表明所制备的poly(VPBA-co-PETA)整体柱可以作为富集中药提取物中含顺二羟基活性物质的有效手段。  相似文献

采用甲基丙烯酸月桂酯为基础功能单体,乙二醇二甲基丙烯酸酯为交联剂,正十二醇、1,4-丁二醇及二甲基亚砜为致孔剂,在内径为75 μm的石英毛细管内制备了具有良好机械性能及化学稳定性的反相毛细管整体柱。考察了致孔剂的种类、比例以及交联剂在单体混合物中的比例对柱压和分离效果的影响;以单体15%、交联剂15%、致孔剂70%(均为质量分数)作为优化配方,在70 ℃条件下反应24 h;并对所合成的毛细管整体柱进行了电镜表征,测试了流速、柱长与柱压的关系。结果表明,毛细管整体柱的通透性良好,可通过延长柱长的方法提高分离效果。将所制备的毛细管整体柱装于纳升级高效液相色谱仪上进行牛血清白蛋白及血浆样本的胰蛋白酶酶切液的分离,获得了比较理想的分离效果。  相似文献

An optimized high‐performance liquid chromatography (HPLC) method is used to show that, as myoblasts differentiate into multinucleated muscle fibers, there is a shift to a more oxidized cell redox state. The HPLC method incorporated derivatization with monobromobimane for the determination of the reduced (GSH) and oxidized (GSSG) forms of glutathione and the reduced (Cys) and oxidized (CysSS) forms of cysteine. The derivatization was optimized to improve the sensitivity of the approach; the limits of detection for glutathione and cysteine were 3 × 10?8 and 5 × 10?8 M , respectively. 在线单柱二维液相色谱法快速纯化牛胰腺中细胞色素C   总被引:1,自引:0,他引:1  
用二维(弱阳,疏水)色谱柱首次完成了在线单柱二维液相色谱法快速纯化牛胰腺中的细胞色素C.在将牛胰腺粗提液进样到该二维色谱柱后,在弱阳离子交换模式下,以梯度洗脱方式进行一维色谱分离,并将分离得到的细胞色素C样品液收集到色谱仪的附加样品储液管内.然后将储液管中样品液全部排出,并二次进样到同一根二维色谱柱中,与此同时也完成了对该样品液的缓冲溶液交换,按疏水色谱(HIC)分离模式进行分离.最终对细胞色素C完成了第二维的HIC纯化.上述全部操作均为在线,在一具有正压的封闭体系中进行并可在52分钟内完成.细胞色素C的最终产品纯度高达94.7%(RSD=1.91%),质量回收率为80.5%(RSD=2.20%).预计此在线单柱二维液相色谱法也可能用于牛胰腺中其他功能蛋白的快速纯化,并可能将其放大到制备和生产规模.  相似文献

Summary The retention behaviour of nucleobases and nucleosides on unmodified silica with dichloromethane-methanol-water mixtures has been systematically investigated. The degree and order of retention can be varied over a wide range by changing the pH, the type and concentration of the acidic additives and by the methanol and water content of the mobile phase. The retention process cannot be considered as simple adsorption but rather as a very complex mixed distribution process of adsorption and absorption including the involvement of ion-pair formation. Further data on the effect of the type of silica (source of supply) on retention behaviour, column efficiency and column reproducibility are reported. The practical application of unmodified silica for the separation of nucleobases and nucleosides is demonstrated by the analysis of a hydrolysate of calf thymus DNA.  相似文献

In this work a chiral stationary phase was prepared by dynamically coating a monolithic reversed-phase HPLC column with a vancomycin-derivative as chiral selector. A hydrophobic alkyl-chain was attached to the vancomycin molecule, providing the immobilization of the chiral selector on the reversed-phase material. Dansyl amino acids were chosen as model analytes for testing the separation power of the dynamically coated phase. All investigated compounds were separated into their enantiomers. Compared with a conventionally packed vancomycin-CSP, a reversal of the enantiomer elution order was obtained.  相似文献

This work reports a fast and simple liquid chromatographic method for the simultaneous determination of five banned fat-soluble synthetic colorants, namely Sudan I-IV and Para-Red, in spice samples. The analytes were successfully separated isocratically in less than 5 min on the new narrow bore monolithic column, FastGradient® Chromolith (50 mm × 2.0 mm i.d.) using a mobile phase of 0.1% (v/v) HCOOH/acetonitrile (35/65%, v/v) at a flow rate of 1.5 mL min−1. All colorants were detected at 506 nm. The main parameters (mobile phase composition, flow rate, injection volume) affecting the separation were studied. The proposed method was thoroughly validated in terms of linearity, LODs, precision and accuracy. The method was applied to the determination of the studied azo-dyes in various spices (paprika, chilli and mixed spice powders) after ultrasound-assisted extraction. Satisfactory recoveries, ranging from 92% to 109% were obtained.  相似文献

建立了整体柱离子对色谱-间接紫外检测和填充柱离子对色谱-间接紫外检测分析四乙基铵根离子的两种方法。用反相整体柱和反相填充柱,以咪唑离子液体-离子对试剂-有机溶剂为流动相,研究了背景紫外吸收试剂、检测波长、离子对试剂、有机溶剂、柱温和流速对测定四乙基铵根离子的影响,比较了两种色谱柱的差异,并讨论了保留规律。在优化的实验条件下,两种方法测定四乙基铵根离子的保留时间分别是2.40和3.02 min;检出限分别是0.04和0.07 mg/L;峰面积的相对标准偏差分别是0.16%和0.11%;保留时间的相对标准偏差分别是0.02%和0.01%。将这两种方法用于分析实验室合成的溴化四乙基铵离子液体,加标回收率分别为98.2%和99.1%。两种方法均能满足四乙基铵根离子测定的需要。  相似文献

With the analysis of Chinese liquor Moutai as an example, the effect of different column combinations was studied on two-dimensional separation in comprehensive two-dimensional gas chromatography (GC × GC). A method to optimize column combinations was developed for achieving maximum orthogonality. Using a geometric approach to factor analysis, the degree of separation orthogonality was quantitatively estimated. The parameters evaluated include peak spreading angle, retention correlation, and practical peak capacity. When using the “reversed-type” column combinations (a polar column as the first dimension and a non- or less polar one as the second dimension), correlation coefficient was lower than or equal to 0.221, the spreading angle was higher than or equal to 77°, and more than 92% of the theoretical peak capacity was reasonably used. For Moutai liquor mainly consisting of some polar compounds, the HP-Innowax + DB1701 column combination was optimal. In addition, through the test of Grob mixture and McReynolds constant, the mechanism of solute-stationary phase interactions was disclosed in details, which validated the estimation of GC × GC orthogonality in a molecular level.  相似文献

亲水作用色谱(HILIC)作为一种分离极性化合物的液相色谱模式,近年来越来越受到关注和重视。一方面是因为强极性化合物的分离问题引起了各个研究领域的重视,如药物分析、代谢组学、蛋白质组学等研究领域都不同程度地涉及强极性化合物的分离问题;另一方面是由于HILIC具有流动相组成简单、分离效率较高、与质谱兼容以及反压较低等优势。固定相是HILIC发展和应用的基础,本文主要从固定相分子结构的角度对HILIC固定相的结构特征、保留特性以及应用概况等进行了综述。对传统正相色谱固定相用于HILIC以及专门设计的HILIC固定相进行了介绍,评述了各自的优缺点和应用概况;对近年来HILIC固定相在中药分离中的应用进行了介绍;并对HILIC固定相的发展进行了展望。  相似文献

A hybrid monolithic column with sulfonate functionality was successfully prepared for the simultaneous separation of common inorganic cations in ion‐exchange chromatographic mode through a simple and easy single‐step preparation method. The strong cation‐exchange moieties were provided directly from allylsulfonate, which worked as an organic monomer in the single‐step reaction. Inorganic cations (Li+, Na+, K+, NH4+, Cs+, Rb+, Mg2+, Ca2+, and Sr2+) were separated satisfactorily by using CuSO4 as the eluent with indirect UV detection. The allysulfonate hybrid monolith showed a better performance in terms of speed and pressure drop than the capillary packed column. The number of theoretical plates achieved was 19 017 plates/m (in the case of NH4+ as the analyte). The relative standard deviations (n = 6) of both retention time and peak height were less than 1.96% for all the analyte cations. The allysulfonate hybrid monolithic column was successfully applied for the rapid and simultaneous separation of inorganic cations in groundwater and the effluent of onsite domestic wastewater treatment system.  相似文献

An analytical method that combined high‐performance liquid chromatography with inductively coupled plasma mass spectrometry has been developed for the determination of hexavalent chromium in traditional Chinese medicines. Hexavalent chromium was extracted using the alkaline solution. The parameters such as the concentration of alkaline and the extraction temperature have been optimized to minimize the interconversion between trivalent chromium and hexavalent chromium. The extracted hexavalent chromium was separated on a weak anion exchange column in isocratic mode, followed by inductively coupled plasma mass spectrometry determination. To obtain a better chromatographic resolution and sensitivity, 75 mM NH4NO3 at pH 7 was selected as the mobile phase. The linearity of the proposed method was investigated in the range of 0.2–5.0 μg L?1 (r2 = 0.9999) for hexavalent chromium. The limits of detection and quantitation are 0.1 and 0.3 μg L?1, respectively. The developed method was successfully applied to the determination of hexavalent chromium in Chloriti lapis and Lumbricus with satisfactory recoveries of 95.8–112.8%.  相似文献

Fatty acid methyl esters from various fats and oils were separated by comprehensive two-dimensional supercritical fluid chromatography with conventional packed columns and FID detection. The first dimension was a silica gel column and the second dimension was an ODS column. This combination was largely orthogonal for the separation of fatty acid methyl esters. The first dimension separations were primarily based on the number of double bonds while the second dimension separations were based on the chain length. The highly-ordered chromatograms and improved resolution allowed the easy detection and identification of minor components. Although the first dimension separations were performed under isobaric conditions where the peak width increased in proportion to the retention, the programming of the sampling duration allowed us to maintain the optimum re-injection frequency (3–4 times) per peak into the second dimension and so to minimize the total analysis time without deteriorating the resolution.  相似文献

A rapid, sensitive and reproducible HPLC method was developed and validated for the analysis of haloperidol and its three main metabolites in human plasma. The analysis was carried out on a monolithic silica column (Chromolith Performance RP-18e, 100 x 4.6 mm). The mobile phase consisted of sodium phosphate (0.1 m, pH 3.5)-acetonitrile (80:20, v/v) at a flow rate of 2.0 mL/min. UV detection at 230 nm was used, with the detection limits of these compounds ranging from 2 to 5 ng. The separation factors of all studied compounds were in the range 2.30-16.32, while the resolution factors were from 1.00 to 5.37.  相似文献

Cat's claw is a large woody vine with hook‐like thorns, and has been traditionally used to treat inflammatory disorders in South and Central America. In this study, a rapid, validated high‐performance liquid chromatographic (HPLC) method using a silica monolithic column was developed for the simultaneous determination of oxindole alkaloids, namely rhynchophylline, pteropodine, isomitraphylline and isopteropodine, in cat's claw. The ionic liquid‐based microwave‐assisted extraction (ILMAE), considered as an environmentally friendly and powerful tool, was first applied in the extraction of oxindole alkaloids. To optimize the HPLC method, the stationary phases, pH values of mobile phase and flow rates were investigated. The validated HPLC method using a Monolithic RP18e column (100 × 4.6 mm) enables these analytes to be separated almost twice as fast as with a conventional particulate column (~16 vs ~30 min) with limits of quantification and detection of 0.5 and 0.15 μg/mL, respectively. The ILMAE conditions were optimized by the Taguchi orthogonal array design. In comparison with conventional water boiling extraction, ILMAE offers almost four times higher yields within an extremely short extraction time. The developed HPLC coupled with ILMAE method could be efficient and practical for rapid determination of oxindole alkaloids in cat's claw.  相似文献

A comprehensive two-dimensional liquid chromatographic (2D-LC) separation system based on the combination of a CN column and a Merck Chromolith Flash reversed-phase column was developed for the separation of components in Adinandra nitida, one type of traditional Chinese medicine (TCM). The two dimensions were connected by a ten-port, dual-position valve controlled automatically by software written in-house. The effluents were detected by both ultraviolet and atmospheric pressure chemical ionization source ion trap tandem mass spectrometry (MS). The calculated peak capacity of the 2D-LC–MS/MS system was above 1240. More than 57 components were resolved in the methanol extract from Adinandra nitida leaves, and five of these were identified based on their relative retention times, molecular weights and MS/MS spectra.  相似文献

A separation method for a mixture of eight sulfur-containing peptides and proteins characterized by a wide molar mass (1-18.4 kDa) and pI range (4.5-10.7) was developed onto a monolithic phenyl phase. Based on the first optimization steps that revealed an increase of the acetonitrile content to 45 vol.% as sufficient for the elution of all biomolecules and the addition of the ion pairing reagent trichloroacetic acid (TCA) as preferable over the eluent additives formic acid or ammonium acetate buffer, the critical variables TCA concentration, gradient time, and eluent flow rate were optimized using a Box-Behnken experimental design. To achieve optimum values for separation factors of all peak pairs, a TCA content of 0.025% (m/v), a gradient time of 10 min, and a flow rate of 3.5 mL min(-1) were selected. Arsenic binding studies were undertaken under conditions optimized with respect to the crucial separation factor of the nonapeptides vasotocin (Vtc) and vasopressin (Vpr) in a shortened gradient time of 7.5 min. A complete separation of phenylarsenic-substituted and unmodified forms of these peptides allowed the calculation of both consumptions and apparent equilibrium constants K from HPLC-UV peak areas. The nonapeptide consumptions by the reaction with phenylarsine oxide (PAO) increased from 7% up to 100% in dependence on the molar ratio of the reaction components. Due to an enhanced UV absorption of the phenylarsenic-substituted biomolecules, the calculation of apparent equilibrium constants led to increasing K values with rising PAO molarities from 9.6×10(5) to 1.2×10(8) in case of Vtc and from 2.2×10(6) to 1.4×10(9) in case of Vpr. For α-lactalbumin, a consumption of 59.2±6.1% by the reaction with molar excesses of PAO varying from 1.4 to 21 can be derived from the chromatograms. The quantitative evaluation of the reaction of the small protein aprotinin with PAO was hindered by a pronounced peak broadening that occurred after reduction of the disulfide bridges.  相似文献
