Separation of Peptides by Pressurized Capillary Electrochromatography   总被引:2,自引:0,他引:2  
A novel gradient pressurized capillary electrochromatography (pCEC) instrument was developed to separate peptides.Two gradient elution modes,hydrophobic and hydrophilic interaction mode in pCEC,were performed on this instrument.Baseline separation of six peptides was obtained on two gradient modes with C18 column and strong cationic exchange column respectively.The effects of mixer volume and total frow rate of pumps on resolution were also discussed.  相似文献

In recent years, capillary electrophoretic immunoassay (CEIA) has become a primary analytical tool in clinical diagnostics. The primary detection methods in CEIA are UV absorbance detection and laser induced fluorescence detection (LIF)1. A new method for the quantitative determination of the enantiomers of salsolinol, an endogenous neurotoxin, was reported. Enantiomeric separation of salsolinol was obtained by capillary zone electrophoresis using hydroxypropyl-β-cyclodextrin (HP-β-CD) as chiral selector. Coupled with UV detection, this separation was applied to the determination of R,S-salsolinol in dried banana chips, and both R- and S-salsolinol were found at 40 ktg/g level.  相似文献

A micellar electrokinetic capillary chromatography (MECC) was developed for the determination of aniline and 6 substituted anilines. The seven components were separated within 25min in the buffer solution of 40mmol/L sodium borate and 100mmol/L SDS. It was found that the separation was dependent on operating voltage, pH value, borate and SDS concentrations.The analytical performance was examined in terms of linear response and reproducibility.Wastewater was determined by the established method.  相似文献

The mixtures of two polymers, poly (N,N-dimethylacrylamide) (PDMA) and polyvinylpyrrolidone (PVP) were synthesized and used as the separation medium for double-stranded and single-stranded DNA fragments by capillary electrophoresis with UV detector. A vanillin imprinted capillary monolithic column was synthesized by in situ polymerization reaction using ethylene-glycol dimethacrylate as cross-linking monomer and methacrylic acid as functional monomer. Under the optimum conditions of capillary electrochromatography, this molecularly imprinted polymer (MIP)-based column showed high selectivity and could recognize not only template molecule vanillin but also positional isomer o-vanillin from their structural analogues.  相似文献

Quasi-interpenetrating network of polyacrylamide (PAA) and polyvinylpyrrolidone (PVP) had been successfully used for single-base resolution of double-stranded DNA (0.76 for 123 bp/124 bp) and single-stranded DNA fragments (0.97 for 123 b/124 b) with UV detection. This quasi-IPN (interpenetrating network) sieving matrix showed low viscosity (23.5 mPa·s at 25 ℃) and decreased with increasing temperature. This polymer also exhibited dynamically coating capacity and could be used in the uncoated capillary. The effects of temperature and electric field strength on the DNA separation of quasi-IPN matrix were also investigated and found that the temperature and electric field strength could markedly affected the mobility behavior of DNA fragments. This polymer matrix has also applied to separate the bigger DNA fragments by capillary electrophoresis with UV detection. Under the denaturing conditions, this matrix separated the samples with last fragment of 1353 base in 40 rain, in which the doublet of 309/310 base was partial separated and the resolution was 0.88.  相似文献

A capillary electrophoresis (CE) method has been firstly used for the separation of the therapeutically important xanthones from Securidaca inappendiculata. The separation of the nine xanthones was systematically optimized with respect to pH, concentration of running buffers,addition of sulfated β-CD,applied voltage and column temperature.Baseline separation was achieved for the nine xanthones in less than 15 minutes using a background electrolyte consisting of 200 mmol/L borate (pH9.5) and 10 mmol/L sulfated β-CD.  相似文献
