In this work, a novel method is described for the determination of bromhexine in biological fluids using molecularly imprinted solid-phase extraction as the sample cleanup technique combined with high performance liquid chromatography (HPLC). The water-compatible molecularly imprinted polymers (MIPs) were prepared using methacrylic acid as functional monomer, ethylene glycol dimethacrylate as cross-linker, chloroform as porogen and bromhexine as the template molecule. The novel imprinted polymer was used as a solid-phase extraction sorbent for the extraction of bromhexine from human serum and urine. Various parameters affecting the extraction efficiency of the polymer have been evaluated. The optimal conditions for molecularly imprinted solid-phase extraction (MISPE) consisted of conditioning 1 mL methanol and 1 mL of deionized water at neutral pH, loading of 5 mL of the water sample (25 μg L−1) at pH 6.0, washing using 2 mL acetonitrile/acetone (1/4, v/v) and elution with 3× 1 mL methanol/acetic acid (10/1, v/v). The MIP selectivity was evaluated by checking several substances with similar molecular structures to that of bromhexine. Results from the HPLC analyses showed that the calibration curve of bromhexine using MIP from human serum and urine is linear in the ranges of 0.5-100 and 1.5-100 μg L−1 with good precisions (3.3% and 2.8% for 5.0 μg L−1), respectively. The recoveries for serum and urine samples were higher than 92%.  相似文献

In this paper, a highly selective molecularly imprinted polymer (MIP) for tramadol hydrochloride, a drug used to treat moderate to severe pain, was prepared and its use as solid-phase extraction (SPE) sorbent was demonstrated. The molecularly imprinted solid-phase extraction procedure followed by high performance liquid chromatography with ultraviolet detector (MISPE-HPLC) was developed for selective extraction and determination of tramadol in human plasma and urine. The optimal conditions for molecularly imprinted solid-phase extraction (MISPE) consisted of conditioning with 1 mL methanol and 1 mL of deionized water at neutral pH, loading of tramadol sample (50 μg L−1) at pH 7.5, washing using 1 mL acetone and elution with 3 × 1 mL of 10% (v/v) acetic acid in methanol. The MIP selectivity was evaluated by checking several substances with similar molecular structures to that of tramadol. Results from the HPLC analyses showed that the calibration curve of tramadol (using MIP from human plasma and urine) is linear in the ranges of 6–100 and 3–120 μg L−1 with good precisions (1.9% and 2.9% for 5.0 μg L−1), respectively. An automated system has been developed for the determination of trace tetracycline antibiotics (TCs) in egg samples, based on online molecularly imprinted solid-phase extraction (MISPE) coupling with high-performance liquid chromatography (HPLC). Oxytetracycline and chlortetracycline were chosen as mixed templates to synthesize highly selective molecularly imprinted polymers for online extraction. Under the optimal online MISPE-HPLC condition, 10 mL egg samples were injected into the MISPE column and then the matrix was washed out. By rotating the switching valve, TCs were transferred to the analytical column and then separated by HPLC. Because sample pretreatment and chromatographic separation were carried out simultaneously, the whole analytical time (18 min) was significantly shortened compared with conventional offline techniques. The detection limits ranged from 0.8 to 1.3 ng/g. The enhancement factors were in the range of 159-410. The spiked recoveries of TCs in real egg samples ranged from 91.6 to 107.6% and the relative standard deviations (RSDs) were not higher than 4.0%.  相似文献

A novel molecularly imprinted polymers based on multiwalled carbon nanotubes synthesized by precipitate polymerization was applied as a selective sorbent for separation and determination of rhein (4,5-dihydroxyanthraquinone-2-carboxylic acid) from the root of kiwi fruit samples coupled with high performance liquid chromatography (HPLC). The molecularly imprinted polymers were prepared with methacrylic acid and 4-vinylpyridine as bifunctional monomers. The chemical structure of the molecularly imprinted polymers was characterized by Fourier transform infrared spectrometer. The equilibrium rebinding experiment and competitive adsorption experiment showed that these imprinted polymers exhibited good adsorption ability toward rhein. The Langmuir adsorption equilibrium constant, K(m) , and theoretical maximum adsorption capacity, Q(m) , were estimated to be 0.43 and 6.77 mg g(-1) , respectively. Compared with molecularly imprinted polymers prepared with methacrylic acid or 4-vinylpyridine solely, the molecularly imprinted polymers synthesized with bifunctional monomers showed enhanced molecular imprinting effect and higher adsorption capacity for the template rhein. The performances of the molecularly imprinted polymers utilized as solid phase extraction sorbent were investigated in detail. The molecularly imprinted polymers prepared by the method proposed in this work could successfully apply to extraction and determination of rhein from the root of kiwi fruit samples coupled with HPLC.  相似文献

Primary creatine deficiency syndromes (CDS) are a new group of disorders caused by guanidinoacetate methyltransferase (GAMT) deficiency, which affects endogenous creatine biosynthesis with depletion of body creatine. A deficiency in creatine can be corrected by treatment with oral creatine supplementation and this necessitates a simple and sensitive screening method for early detection of creatine in dilute physiologic fluids. In this work an artificial receptor, molecularly imprinted polymer (MIP), for creatine was used both as a material for solid-phase extraction (SPE) and as a sensing element in a voltammetric sensor. Using the combination of molecularly imprinted solid-phase extraction (MISPE) with a complementary MIP sensor, the minimum detectable amount was found to be 0.0015 ng mL(-1) (RSD = 1.3%, S/N = 3). The MISPE-MIP sensor combination provided up to 60-fold preconcentration, which was more than sufficient for achieving the required quantification limit 50 ng mL(-1) (or 0.0025 ng mL(-1) after 2 x 10(4)-fold dilution) for creatine in human blood serum.  相似文献

Surface molecularly imprinted polymers for mezlocillin were used for the selective solid-phase extraction of mezlocillin from eggs. The molecularly imprinted polymers were prepared using mezlocillin as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker, silica particle as the support and were characterized by infrared spectroscopy, scanning electron microscopy, and thermogravimetric analysis. The adsorption properties of polymers were investigated in detail. The obtained polymers provided high imprinting efficiency to mezlocillin with an imprinting factor of 3.72 and were used as selective sorbents for mezlocillin. Molecularly imprinted solid-phase extraction followed by high-performance liquid chromatography was used for the determination of mezlocillin in eggs and the conditions were evaluated. The average recovery of mezlocillin in fortified eggs was between 69.8 and 82.3% with a relative standard deviation less than 6.9%. The results demonstrate the application of molecularly imprinted polymers for the extraction of mezlocillin from biological samples.  相似文献

A molecularly imprinted polymer solid phase extraction (MISPE) method combined with flow-injection chemiluminescence (FI-CL) for the determination of residual tetracycline (TC) in fish samples is presented. The molecularly imprinted polymer (MIP) of TC was synthesized and particles of this MIP were packed into a polytetrafluoroethylene (PTFE) tube, which was connected into the sampling loop of an eight-way injection valve and served as the MISPE column for on-line selective adsorption of TC. The eluent (CH3CN : HNO3 (0.01 mol L(-1)) = 4 ratio 1, v ratio v) was used for extracting the adsorbed TC, which could be detected by its good enhancing effect on the CL reaction between Ce(iv) and rhodamine B. The CL intensity is linear to TC concentration in the range from 4 x 10(-9) to 4 x 10(-7) g mL(-1). The detection limit is 1 x 10(-9) g mL(-1) (3 sigma) and the relative standard deviation is 2.4% (n = 9). The conditions of preconcentration, extraction and CL reaction were carefully studied. The selectivity experiment shows that the selectivity and sensitivity of the CL method could be improved greatly when MIP was used as a recognition material in SPE. However, the MISPE column interacted indiscriminately with oxytetracycline (OTC) with a 49 +/- 2% binding. An intermediate differential pulsed elution (DPE) step using 3% acetic acid as eluent was employed to remove OTC and other interfering substances. The proposed MISPE-CL method has been applied successfully to the determination of TC in fish samples. At the same time, the binding characteristics of the polymer to tetracycline were evaluated by batch and dynamic methods.  相似文献

In this work, the preparation and evaluation of water-compatible molecularly imprinted polymers for triazines using 2-hydroxyethyl methacrylate and methacrylic acid as comonomers is described. Four sets of molecularly imprinted and non-imprinted polymers for propazine were prepared at varying monomer molar ratios (from 4:0 to 1:3), and evaluated for the recognition of several triazines directly in aqueous media. The evaluation was performed by loading 1 mL of an aqueous solution containing 500 ng of each selected triazine, washing with 500 μL of acetonitrile, and eluting with 500 μL of methanol followed by 2 × 500 μL of a solution of methanol containing 10% of acetic acid. Final determinations were performed by high-performance liquid chromatography-ultraviolet detection. Improvement in molecular recognition of triazines in water was obtained on those molecularly imprinted polymers incorporating 2-hydroxyethyl methacrylate in 3:1 or 2:2 molar ratios, being the former selected as optimum providing recoveries for propazine up to 80%. A molecularly imprinted solid-phase extraction protocol was developed to ensure that triazines-selective recognition takes place inside selective binding sites in pure water media. Finally, the developed method was successfully applied to the determination of the selected triazines in environmental waters providing limits of detection from 0.16 and the 0.5 μg/L concentration range.  相似文献

A unique stir bar coated with dummy molecularly imprinted polymers for bisphenol A was prepared by sol-gel technique. The scanning electron microscopic image of the coating presented a homogeneous surface with a thickness of about 57 ± 2.5 μm. The Fourier transform infrared spectrum of the coating proved the incorporating of dummy molecularly imprinted polymers with the sol-gel network. When used to extract bisphenol A from aqueous solution containing bisphenol A and its three analogs (4-tert-butylphenol, 4,4'-dihydroxybiphenyl, and 3,3',5,5'-tetrabromo-bisphenol A). Dummy molecularly imprinted polymers-coated stir bar showed better selectivity than the bars coated with polydimethylsiloxane or non-imprinted polymers. The extraction conditions including stirring speed, pH, and extraction time were optimized. After back extraction with methanol, the extracts were analyzed by high-performance liquid chromatography-fluorescence detection. The linear range was 0.0228-0.456 ng/mL with correlation coefficient of 0.9994 and the detection limit was about 5.70 × 10(-3) ng/mL based on three times ratio of signal-to-noise. The method was applied to the determination of trace bisphenol A in tap water.  相似文献

The isolation and preconcentration of estrogens from new types of biological samples (acellular and protein-free simulated body fluid) by molecularly imprinted solid-phase extraction has been described. In this technique, supramolecular receptors, namely molecularly imprinted polymers (MIPs) are used as a sorbent material. The recognition sites of MIPs were prepared by non-covalent multiple interactions and formed with the target 17β-estradiol as a template molecule. High-performance liquid chromatography with spectroscopic UV, selective, and a sensitive electrochemical CoulArray detector was used for the determination of 17β-estradiol, estrone, and estriol in simulated body fluid which mimicked human plasma.  相似文献

A method was developed for the determination of ractopamine in pig urine using molecularly imprinted solid-phase extraction (MISPE) as the sample clean-up technique combined with high-performance liquid chromatography. The molecularly imprinted polymer (MIP) was synthesized in acetonitrile-triethylamine system using ractopamine (RAC) as the template and acrylamide as the monomer. The binding capacity of the polymer toward RAC was found to be about 2.57 mg of ractopamine/g of polymer. The optimal procedures for MISPE consisted of conditioning with 3 mL methanol, equilibrating with 3 mL of water, loading volume of <10 mL of aqueous sample (pH 7), washing with 3 mL water and 3 mL methanol, and eluting with 5 mL of 5% ammonia in methanol. In the four spiked samples with the levels of 0.01, 0.1, 1.0 and 5.0 μg/mL, the mean recoveries of analyte on the MIP were higher than 90% with relative standard deviation <10%, and significant differences between imprinted and non-imprinted materials were observed. The MIP selectivity was evaluated by checking 11 drugs with similar and different molecular structures to that of RAC. The characteristics of three-dimensional cavities and hydrogen bond interaction were regarded as the main factors that dominated the retention of RAC on the MISPE cartridge.  相似文献

A rapid, specific, and sensitive method has been developed using molecularly imprinted polymers (MIPs) as solid-phase extraction sorbents for extraction of trace tetracycline antibiotics (TCs) in foodstuffs. MIPs were prepared by precipitation polymerization using tetracycline as the template. Under the optimal condition, the imprinting factors for MIPs were 4.1 (oxytetracycline), 7.0 (tetracycline), 7.4 (chlortetracycline), 7.7 (doxycycline), respectively. Furthermore, the performance of MIPs as solid-phase extraction sorbents was evaluated and high extraction efficiency of molecularly imprinted solid-phase extraction (MISPE) procedure was demonstrated. Compared with commercial sorbents, MISPE gave a better cleanup efficiency than C18 cartridge and a higher recovery than Oasis HLB cartridge. Finally, the method of liquid chromatography–tandem mass spectrometry coupled with molecular-imprinted solid-phase extraction was validated in real samples including lobster, duck, honey, and egg. The spiked recoveries of TCs ranged from 94.51% to 103.0%. The limits of detection were in the range of 0.1–0.3 μg kg−1. In this work, a novel magnetic nanomaterial functionalized with a molecularly imprinted polymer was prepared for the extraction of protoberberine alkaloids. Molecularly imprinted polymers were made on the surface of Fe3O4 nanoparticles by using berberine as template, acetonitrile/water as porogen, acrylamide as functional monomer and ethylene glycol dimethacrylate as cross‐linker. The optimized molar ratio of template/functional monomer was 1:7. The polymeric magnetic nanoparticles were characterized by transmission electron microscopy and Fourier transform infrared spectroscopy. The stability and adsorption capacity of the molecularly imprinted polymers were investigated. The molecularly imprinted polymers were used as a selective sorbent for the magnetic molecularly imprinted solid‐phase extraction and determination of jatrorrhizine, palmatine, and berberine. Extraction parameters were studied including loading pH, sample volume, stirring speed, and extraction time. Finally, a magnetic molecularly imprinted solid‐phase extraction coupled to high‐performance liquid chromatography method was developed. Under the optimized conditions, the method showed good linear range of 0.1–150 ng/mL for berberine and 0.1–100 ng/mL for jatrorrhizine and palmatine. The limit of detection was 0.01 ng/mL for berberine and 0.02 ng/mL for jatrorrhizine and palmatine. The proposed method has been applied to determine protoberberine alkaloids in Cortex phellodendri and rat plasma samples. The recoveries ranged from 87.33–102.43%, with relative standard deviation less than 4.54% in Cortex phellodendri and from 102.22–111.15% with relative standard deviation less than 4.59% in plasma.  相似文献

A molecularly imprinted polymer solid-phase extraction method is used to extract esculetin from the ash bark of Chinese traditional medicine. Ratio of ethanol and water as washing solution were investigated. Data of accumulative adsorption on molecularly imprinted polymers from the continuous loading experiment suggests that there are two different kinds of recognition sites in molecularly imprinted polymers. By selecting the washing and eluting solution a scheme was designed to separate esculetin and its analogues including esculin, coumarin, 7-methoxylcoumarin and daphnetin. Finally, by applying the revised scheme esculetin was extracted from the ash bark of Chinese traditional medicine that was purchased from two big drugstores, respectively, with both molecularly imprinted polymers and non-molecularly imprinted polymers.  相似文献

Therapeutic drug monitoring of captopril, which is a commonly used antihypertensive agent in clinical practice, is necessary. However, matrix effect-induced pretreatment is the bottleneck for determination. Metal-mediated molecularly imprinted polymers, an essential branch of molecularly imprinted polymers with better specificity and selectivity, have been used to separate/enrich analytes from complex matrices. In this work, Cu2+ was introduced to dynamically establish the binding sites of metal-mediated molecularly imprinted polymer towards captopril. All evidence demonstrated that the metal-mediated molecularly imprinted polymer based on Cu2+ coordination obtained a higher adsorption capacity (81.23 mg/g), faster adsorption rate (adsorption equilibrium within 50 min), and better selectivity (with the unrecognized analog). Subsequently, the Cu2+-mediated molecularly imprinted polymer was used as dispersive molecularly imprinted solid-phase extraction to successfully establish an analytical platform for the determination of trace captopril in rat plasma. The enrichment factor was up to 20, the detection limit was as low as 0.16 μg/ml, and the average recovery was in the range of 87.51%–98.28% with a relative standard deviation of less than 3.29%. This study provides a promising reference for the preparation of selective adsorbents to improve pretreatment.  相似文献

The computer‐assisted design and synthesis of molecularly imprinted polymers for the simultaneous capture of six carbamate pesticides from environmental water are reported in this work. The quantum mechanical computational approach was employed to design the molecularly imprinted polymers with carbofuran as template. The interaction energies between the template molecule and different functional monomers in various solvents were calculated to assist in the selection of the functional monomer and porogen. The optimised molecularly imprinted polymer was subsequently used as a class‐selective sorbent in solid‐phase extraction for pre‐concentration and determination of carbamates from environmental water. The parameters influencing the extraction efficiency of the molecularly imprinted solid‐phase extraction procedure were systematically investigated to facilitate the class‐selective extraction. For the proposed method, linearity was observed over the range of 2–500 ng/mL with the correlation coefficient ranging from 0.9760 to 1.000. The limits of detection ranged from 0.2 to 1.2 ng/mL, and the limit of quantification was 4 ng/mL. These results confirm that computer‐assisted design is an effective evaluation tool for molecularly imprinted polymers synthesis, and that molecularly imprinted solid‐phase extraction can be applied to the simultaneous analysis of carbamates in environmental water.  相似文献
