The diastereospecific chemical syntheses of uridine-2',3',4',5',5' '-(2)H(5) (21a), adenosine-2',3',4',5',5' '-(2)H(5) (21b), cytidine-2',3',4',5',5' '-(2)H(5)(2)H(5) (21c), and guanosine-2',3',4',5',5' '-(2)H(5) (21d) (>97 atom % (2)H at C2', C3', C4', and C5'/C5' ') have been achieved for their use in the solution NMR structure determination of oligo-RNA by the Uppsala "NMR-window" concept (refs 4a-c, 5a, 6), in which a small (1)H segment is NMR-visible, while the rest is made NMR-invisible by incorporation of the deuterated blocks 21a-d. The deuterated ribonucleosides 21a-d have been prepared by the condensation of appropriately protected aglycone with 1-O-acetyl-2,3,5-tri-O-(4-toluoyl)-alpha/beta-D-ribofuranose-2,3,4,5,5'-(2)H(5) (19), which has been obtained via diastereospecific deuterium incorporation at the C2 center of appropriate D-ribose-(2)H(4) derivatives either through an oxidation-reduction-inversion sequence or a one-step deuterium-proton exchange in high overall yield (44% and 24%, respectively).  相似文献

Hydrolytic reactions of guanosyl-(3',3')-uridine and guanosyl-(3',3')-(2',5'-di-O-methyluridine) have been followed by RP HPLC over a wide pH range at 363.2 K in order to elucidate the role of the 2'-hydroxyl group as a hydrogen-bond donor upon departure of the 3'-uridine moiety. Under neutral and basic conditions, guanosyl-(3',3')-uridine undergoes hydroxide ion-catalyzed cleavage (first order in [OH(-)]) of the P-O3' bonds, giving uridine and guanosine 2',3'-cyclic monophosphates, which are subsequently hydrolyzed to a mixture of 2'- and 3'-monophosphates. This bond rupture is 23 times as fast as the corresponding cleavage of the P-O3' bond of guanosyl-(3',3')-(2',5'-di-O-methyluridine) to yield 2',5'-O-dimethyluridine and guanosine 2',3'-cyclic phosphate. Under acidic conditions, where the reactivity differences are smaller, depurination and isomerization compete with the cleavage. The effect of Zn(2+) on the cleavage of the P-O3' bonds of guanosyl-(3',3')-uridine is modest: about 6-fold acceleration was observed at [Zn(2+)] = 5 mmol L(-)(1) and pH 5.6. With guanosyl-(3',3')-(2',5'-di-O-methyluridine) the rate-acceleration effect is greater: a 37-fold acceleration was observed. The mechanisms of the partial reactions, in particular the effects of the 2'-hydroxyl group on the departure of the 3'-linked nucleoside, are discussed.  相似文献

A series of new square-planar 4'-(5'-R-pyrimidyl)-2,2':6',2'-terpyridyl platinum(II) phenylacetylide complexes ( 1a- 5a) bearing different substituents (R = H, OEt, Ph, Cl, CN) on the pyrimidyl ring have been synthesized and characterized. The electronic absorption, photoluminescence, and triplet transient difference absorption spectra were investigated. All of the complexes exhibit broad, moderately strong absorption between 400 and 500 nm that can be tentatively assigned to the metal-to-ligand charge transfer ( (1)MLCT) transition, possibly mixed with some ligand-to-ligand charge transfer ( (1)LLCT) character. Photoluminescence arising from the (3)MLCT state was observed both in fluid solutions at room temperature and in a rigid matrix at 77 K. The (1)MLCT/ (1)LLCT absorption bands and the (3)MLCT emission bands for 1a- 5a red-shift in comparison to those of the corresponding 4'-toly-2,2':6',2'-terpyridyl platinum(II) phenylacetylide complex. In addition, the energies of the (1)MLCT/ (1)LLCT absorption and the (3)MLCT emission bands exhibit a linear correlation with the Hammett constant (sigma p) of the 5'-substituent on the pyrimidyl ring. The lifetime of the (3)MLCT emission at room temperature is governed by the energy gap law. The triplet transient difference absorption spectra of 1a- 5a exhibit a broad absorption band from 500 to 800 nm, and a bleaching band between 420 and 500 nm. Complex 5a, which contains the -CN substituent, exhibits a lower-energy triplet absorption band at 785 nm and a shorter lifetime (130 ns) in CH 3CN than 2a, which has the -OEt substituent, does (lambda T1-Tn (max) = 720 nm, tau T = 660 ns). The triplet excited-state absorption coefficients at the band maxima for 1a- 5a vary from 36 600 L.mol (-1).cm (-1) to 115 090 L.mol (-1).cm (-1), and the quantum yields of the triplet excited-state formation range from 0.19 to 0.66. All complexes exhibit a moderate nonlinear transmission for nanosecond laser pulses at 532 nm. Moreover, these complexes can generate singlet oxygen efficiently in air-saturated CH 3CN solutions, with the singlet oxygen generation quantum yield (Phi Delta) varying from 0.24 to 0.46.  相似文献

The cooperative action of multiple Cu(II) nuclear centers is shown to be effective and selective in the hydrolysis of 2'-5' and 3'-5' ribonucleotides. Reported herein is the specific catalysis by two trinuclear Cu(II) complexes of L3A and L3B. Pseudo first-order kinetic studies reveal that the L3A trinuclear Cu(II) complex effects hydrolysis of Up(2'-5')U with a rate constant of 28 x 10(-)(4) min(-)(1) and Up(3'-5')U with a rate constant of 0.5 x 10(-)(4) min(-)(1). The hydrolyses of Ap(3'-5')A and Ap(2'-5')A proceed with rate constants of 24 x 10(-)(4) min(-)(1) and 0.5 x 10(-)(4) min(-)(1) respectively. The L3A trinuclear Cu(II) complex demonstrates high specificity for Up(2'-5')U and Ap(3'-5')A. Similar studies with the more rigid L3B trinuclear Cu(II) complex shows no selectivity and yields lower rate constants for hydrolysis. The selectivity observed with the L3A ligand is attributed to the geometry of the ligand-bound diribonucleotide which ultimately dictates the proximity of the attacking hydroxyl and the phosphoester to a Cu(II) center for activation and subsequent hydrolysis.  相似文献

Peroxynitrite reacts with 2',3',5'-tri-O-acetyl-guanosine to yield a novel compound identified as 1-(2,3,5-tri-O-acetyl-beta-D-erythro-pentofuranosyl)-5-guanidino-4-nitroimidazole (6). This characterization was achieved using a combination of UV/vis spectroscopy and ESI-MS. Additionally, 1-(beta-D-erythro-pentofuranosyl)-5-guanidino-4-nitroimidazole (6a) was synthesized by an independent route, characterized by UV/vis spectroscopy, ESI-MS, and (1)H- and (13)C NMR, and shown to be identical to deacetylated 6. This product is extremely stable in aqueous solution at both pH extremes and is formed in significant yields. These characteristics suggest that this lesion may be useful as a specific biomarker of peroxynitrite-induced DNA damage. We also observed formation of 2',3',5'-tri-O-acetyl-8-nitroguanosine (2',3',5'-tri-O-acetyl-8-NO(2)()Guo), 2-amino-5-[(2,3,5-tri-O-acetyl-beta-D-erythro-pentofuranosyl)amino]-4H-imidazol-4-one (2',3',5'-tri-O-acetyl-Iz), and the peroxynitrite-induced oxidation products of 2',3',5'-tri-O-acetyl-8-oxoGuo. The formation of 6 and 2',3',5'-tri-O-acetyl-8-NO(2)()Guo was rationalized by a mechanism invoking formation of the guanine radical.  相似文献

A general procedure for the first synthesis of a 3',5'-cyclic boranomonophosphate was established. Specifically, adenosine 3',5'-cyclic boranomonophosphosphate (cyclic AMPB, 4c), a P-borane (BH(3)) analogue of adenosine 3',5'-cyclic monophosphate (cAMP), was synthesized via a phosphite approach in good yield. The method is also applicable for syntheses of natural cAMP and its phosphorothioate analogue. The two diasteromers of cyclic AMPB 4c were separated, and their chemical structures were established via spectroscopic methods.  相似文献

Hydrolytic reactions of 2',3'-O-methyleneadenos-5'-yl bis(2',5'-di-O-methylurid-3'-yl) phosphate (1), a sugar O-alkylated trinucleoside 3',3',5'-monophosphate, have been followed by RP HPLC over a wide pH range. Under neutral and mildly acidic conditions, the only reaction observed was a pH-independent cleavage of the O-C5' bond of the 5'-linked nucleoside. Under more alkaline conditions nucleophilic attack by hydroxide ion starts to compete. The reaction is first order in [OH(-)] and becomes predominant at pH 10. Each of the 3'-linked nucleosides is displaced 2.9 times as readily as the 5'-linked one. To determine the beta(lg) value for the hydroxide ion catalyzed hydrolysis of 1, two diesters (2a,b) having 2',3'-O-methyleneadenosine (7) and 2',5'-di-O-methyluridine (4) as leaving groups were hydrolyzed under alkaline conditions. Since the beta(lg) value for this reaction is known, DeltapK(a) between 4 and 7 could be calculated. The beta(lg) for the hydrolysis of 1 was estimated to be -0.5 with use of this information. The mechanisms of the partial reactions and the role of leaving group properties in ribozyme reactions of large ribozymes are discussed.  相似文献

Ammonium [1-(3',5',6'-trideoxy-beta-D-erythro-hexofuranosyl)thymine]-6'- phosphonate (1), ammonium 3',5'-dideoxycytidine-5'-C-methylphosphonate (2) and 3',5'-dideoxyadenosine-5'-C-methyl phosphonic acid (3) have been synthesized and tested for anti-HIV activity. The key steps involved an Arbuzov reaction between triethyl phosphite and 3,5,6-trideoxy-6-iodo-1,2-O-isopropylidene-alpha-D-erythro- hexofuranose (7), followed by condensation with the appropriate nucleoside bases. The substances 1, 2 and 3 have been tested in vitro against HIV.  相似文献

Diindeno[1,2,3,4-defg;1',2',3',4'-mnop]chrysene (1), the smallest possible alkene-centered C60 substructure with a curved pi-system, is obtained in 25-35% yield by flash vacuum pyrolysis of the twisted 1,1'-dibromobifluorenylidene (2) on a 100 mg scale at 1050 degrees C. At 1200 degrees C, the bowl-shaped hydrocarbon 1 rearranges to the planar isomer diindeno[5,6,7,1-defg;5',6',7',1'-lmnop]chrysene (14) by a double 5/6 ring-expansion/ring-contraction. X-ray crystallography establishes that the central carbon atoms of 1 are nearly 80% as pyramidalized as the carbon atoms of C60 (POAV angles = 9.0 degrees and 11.6 degrees for 1 and C60, respectively). A four-step synthesis has been developed to prepare the pyrolysis precursor (2) as a mixture of (E)- and (Z)-isomers in 39% overall yield from commercially available 9-fluorenone-1-carboxylic acid (10).  相似文献

The synthesis and characterization of Ru(II) terpyridine complexes derived from 4'-functionalized 2,2':6',2'-terpyridine ligands by a multi step procedure have been described. The complexes are redox-active, showing both metal-centred (oxidation) and ligand-centred (reduction) processes. The antibacterial and antifungal activity of the synthesized ruthenium(II) complexes [Ru(attpy)2](PF6)2 (attpy = 4'-(4-acryloyloxymethylphenyl)-2,2':6',2'-terpyridine); [Ru(mttpy)2](PF6)2 (mttpy = 4'-(4-methacryloyloxymethylphenyl)-2,2':6',2'- terpyridine); [Ru(mttpy)(MeOPhttpy)](PF6)2 (MeOPhttpy = 4'-(4-methoxyphenyl)-2,2':6',2'-terpyridine); and [Ru(mttpy)(ttpy)](PF6)2 (ttpy = 4'-(4-methylphenyl)-2,2':6',2'-terpyridine) were tested against four human pathogens (Proteus vulgaris, Proteus mirabilis, Pseudomonas aeruginosa and Escherichia coli) and five plant pathogens (Curvularia lunata, Fusarium oxysporum, Fusarium udum, Macrophomina phaseolina and Rhizoctonia solani) by the well diffusion method and MIC values of the complexes are reported. A biological study of the complexes indicated that the complexes [Ru(mttpy)2](PF6)2 and [Ru(mttpy)(MeOPhttpy)](PF6)2 exhibit very good activity against most of the test pathogens and their activity is better than those of some of the commercially available antibiotics like tetracycline and the fungicide carbendazim.  相似文献

Black dye (BD), isomer 1 ([Ru(II)(H3-tctpy)(NCS)3](-1), where H3-tctpy = 4,4',4' '-tricarboxy-2,2':6,2' '-terpyridine) is known to be an excellent sensitizer for dye-sensitized solar cells and exhibits a very good near-IR photo response, compared to other ruthenium dyes. Because isothiocyanate is a linear ambidentate ligand, BD has three other linkage isomers, [Ru(H3-tctpy)(NCS)2(SCN)](-1), isomer 2 and 2', and [Ru(H3-tctpy))(SCN)3](-1), isomer 3. In this study, we have calculated the geometry of BD and its isomers by DFT. Further, we have analyzed the bonding in these isomers using NBO methods. TDDFT calculations combined with scalar relativistic zero-order regular approximations (SR-ZORA) have been carried out to simulate the absorption spectra. Calculations have been performed for the isomers both in vacuo and in solvent (ethanol). The inclusion of the solvent is found to be important to obtain spectra in good agreement with the experiment. The first absorption bands are dominated by the metal-to-ligand charge transfer (MLCT) and ligand-to-ligand charge transfer (LLCT).  相似文献

(E)-4-(2',4',5'-trimethoxyphenyl)but-1,3-diene (4) and (E)-4-(2',4',5'-trimethoxyphenyl)but-1-ene (6), bioactive phenylbutanoids of Zingiber cassumunar, were synthesized exclusively with trans geometry. Treatment of methylmagnesium iodide with (E)-2',4',5'-trimethoxycinnamaldehyde (2), an oxidized product of abundantly available toxic (Z)-phenylpropanoid (1) of Acorus calamus, gave (E)-4-(2',4',5'-trimethoxyphenyl)but-3-en-2-ol (3) which upon dehydration with copper sulphate/silica gel under microwave irradiation for 3 min afforded 4 in 58% yield. Further, catalytic hydrogenation of 4 with 10% Pd/C afforded 4-(2',4',5'-trimethoxyphenyl)butane (5) which upon dehydrogenation with DDQ/SiO2 afforded hypolipidemic 6 in 54% yield.  相似文献

[reaction in text] A novel, short, and efficient synthesis of (S)-5,5,5,5',5',5'-hexafluoroleucine (6) in greater than 99% ee starting from the protected oxazolidine aldehyde 1 is described. The enantiomeric excess of the product was calculated from an NMR analysis of a dipeptide formed by reaction with a protected L-serine derivative. Furthermore, a racemic sample of N-acylated hexafluoroleucine was enzymatically resolved by treatment with porcine kidney acylase I and was found to have the same optical rotation as a synthetic sample of 6.  相似文献

An electroactive luminescent switch has been synthesized that comprises a hydroquinone-functionalized 2,2':6',2'-terpyridine ligand coordinated to a ruthenium(II) (4'-phenylethynyl-2,2':6',2'-terpyridine) fragment. The assembly is sufficiently rigid that the hydroquinone-chromophore distance is fixed. Excitation of the complex via the characteristic metal-to-ligand charge-transfer (MLCT) absorption band produces an excited triplet state in which the promoted electron is localized on the terpyridine ligand bearing the acetylenic group. The triplet lifetime in butyronitrile solution at room temperature is 46 +/- 3 ns but increases markedly at lower temperature. Oxidation of the hydroquinone to the corresponding benzoquinone switches on an electron-transfer process whereby the MLCT triplet donates an electron to the quinone. This reaction reduces the triplet lifetime to 190 +/- 12 ps and essentially extinguishes emission. The rate of electron transfer depends on temperature in line with classical Marcus theory, allowing calculation of the electronic coupling matrix element and the reorganization energy as being 22 cm(-1) and 0.84 eV, respectively. The switching behavior can be monitored using luminescence spectroelectrochemistry. The on/off level is set by temperature and increases as the temperature is lowered.  相似文献

A series of novel 5-phenyl-3-(2,2':5',2″-terthien-5-yl)-4,5-dihydro-lH-pyrazolines were synthesized in this report. Their photoactivated cytotoxicities on the 5podoptera litura (SL) cell line were evaluated using the MTT method. It was noticed that the inhibitory activities of all the conjugates was enhanced when irradiated with UV-A light. Compounds 4, 6 and 8 were found to be most effective with inhibition rates of 88.1%, 88.0%, and 90,5%, respectively, For compound B, the inhibition rate value was only slightly enhanced under the irradiation treatment (78.3%) compared to the dark treatment (74.8%). The relationship analysis between structure and activity showed that the middle thiophene ring played an important role on the inhibitory activities. It was shown that these compounds could be the potential candidates for new photoactivated pesticides.  相似文献

Electrochemical dimerization reactivity has been studied for 5-substituted uracils (5XU) including thymine (1a: X = Me) and 5-halouracil derivatives (1b: X = F; 1c: X = Cl; 1d: X = Br; 1e: X = I). Upon galvanostatic electrolysis of Ar-saturated aqueous solution 1a underwent anodic oxidation to produce N(1)-C(5')- and N(1)-C(6')-linked dimer hydrates, 1-(6'-hydroxy-5',6'-dihydrothymin-5'-yl)thymine (5a) and 1-(5'-hydroxy-5',6'-dihydrothymin-6'-yl)thymine (6a), as the major products. These N-C-linked dimerizations were accompanied by the formation of novel stereoisomeric C(5)-C(5')-linked dimers (meso isomer: 13a[meso]; racemic isomer: 13a[rac]) with a condensed tetrahydrofuran ring skeleton. Similar electrolyses of 5-fluorouracil (1b) and 5-chlorouracil (1c) also afforded the corresponding N(1)-C(5')-linked dimer hydrates, 1-(5'-fluoro-6'-hydroxy-5',6'-dihydrouracil-5'-yl)-5-fluorouracil (5b) and 1-(5'-chloro-6'-hydroxy-5',6'-dihydrouracil-5'-yl)-5-chlorouracil (5c), respectively, while resulting in neither N(1)-C(6')-linked dimer analogues nor C(5)-C(5')-linked dimers, unlike the reactivity of 1a. In contrast to 1a-c, no dimeric products were obtained from 5-bromouracil (1d) and 5-iodouracil (1e). The present electrochemical method was applicable to the cross-dimerization into N(1)-C(5')-linked heterodimer hydrates composed of binary 5-substituted uracils that occurred in competition with the formation of homodimer hydrates. A mechanism of the N(1)-C(5')-linked dimerization of 1a-c has been proposed, by which allyl-type radical intermediates with limiting mesomeric forms of N(1)-centered and C(5)-centered pyrimidine radicals (2a-c [N(1)]/2a-c [C(5)]) are generated via anodic one-electron oxidation and subsequent deprotonation at N(1) and undergo a head-to-tail coupling.  相似文献

In vitro selection was used to identify deoxyribozymes that ligate two RNA substrates. In the ligation reaction, a 2'-5' RNA phosphodiester linkage is created from a 2',3'-cyclic phosphate and a 5'-hydroxyl group. The new Mg(2+)-dependent deoxyribozymes provide 50-60% yield of ligated RNA in overnight incubations at pH 7.5 and 37 degrees C, and they afford 40-50% yield in 1 h at pH 9.0 and 37 degrees C. Various RNA substrate sequences may be joined by simple Watson-Crick covaration of the DNA binding arms that interact with the two RNA substrates. The current deoxyribozymes have some RNA substrate sequence requirements at the nucleotides immediately surrounding the ligation junction (either UAUA GGAA or UAUN GGAA, where the arrow denotes the ligation site and N equals any nucleotide). One of the new deoxyribozymes was used to prepare by ligation the Tetrahymena group I intron RNA P4-P6 domain, a representative structured RNA. Nondenaturing gel electrophoresis revealed that a 2'-5' linkage between nucleotides A233 and G234 of P4-P6 does not disrupt its Mg(2+)-dependent folding (DeltaDeltaG degrees ' < 0.2 kcal/mol). This demonstrates that a 2'-5' linkage does not necessarily interfere with structure in a folded RNA. Therefore, these non-native linkages may be acceptable in modified RNAs when structure/function relationships are investigated. Deoxyribozymes that ligate RNA should be particularly useful for preparing site-specifically modified RNAs for studies of RNA structure, folding, and catalysis.  相似文献
