Chemiluminescence-HPLC for the assay of guanidino compounds   总被引:1,自引:0,他引:1  
A chemiluminescence-HPLC system for the assay of serum guanidino compounds was established. In this study, we found that lucigenin added to the reaction mixture containing guanidino compounds and alkaline-ninhydrin reagent resulted in strong chemiluminescence. The lucigenin chemiluminescence was not derived from fluorescent derivatives of guanidino compounds or ninhydrin. The interaction of guanidino compounds and ninhydrin under alkaline conditions produced reactive oxygen species. We applied this phenomenon to the microanalysis of guanidino compounds.  相似文献

High-performance liquid chromatographic microanalyses for guanidino compounds in human physiological fluids have been accomplished by means of a pre-column fluorescence derivatization method using benzoin. The guanidino compounds in urine or deproteinized serum after ultrafiltration are converted to the fluorescent derivatives with benzoin in an alkaline medium, and the derivatives are separated simultaneously within 25 min on a reversed-phase column (mu Bondapak Phenyl) with a linear gradient elution of methanol in aqueous mobile phase (pH 8.5). The method permits the quantitative determination of guanidinosuccinic acid, methylguanidine, taurocyamine and guanidinobutyric acid at concentrations of as low as 8-78 pmol/ml in human urine and serum.  相似文献

Derivatives of 1,3-naphthoxazines containing from five to seven-membered spiro ring – under Vilsmeier–Haack reaction conditions are rearranged into novel xanthene-type compounds. All synthesized aminochromene derivatives fluoresce in organic solvents with extra large Stokes shifts (100–133?nm). It was found that compounds containing five-membered annulated aliphatic rings in methanol solution have the best spectral characteristics. These compounds have moderate quantum yields of 28.36–28.94%, and extra large Stokes shifts 106 and 115?nm.  相似文献

A new and convenient one-pot methodology for the reaction of N-methyl isatin, alkynes and phenacyl bromides to selectively afford spiro dihydrofuran oxindole derivatives catalyzed by indium bromide under ambient temperature conditions has been documented. The method has been applied for the synthesis of a range of compounds with variable functionalities in good to excellent yields (76–92%). The significant advantages of this protocol are highlighted by excellent yields, cleaner reaction profiles and avoidance of expensive catalysts.  相似文献

Manganese(III)-mediated oxidative annulation of vinylidenecyclopropanes with 1,3-dicarbonyl compounds in acetonitrile/acetic acid produces the corresponding functionalized dihydrofuran derivatives in moderate to good yields under mild conditions. A simple and efficient protocol has been developed for the synthesis of 3‐phenylnaphtho[2,3‐b]furan‐4,9‐diones by domino reaction of α‐bromonitroalkenes to 2‐hydroxynaphthalene‐1,4‐dione. With the optimal reaction conditions [NaOAc (120 mol%), water, 70°C, 7 h], the scope of the domino reaction was explored and the green approach provided the desired products in moderate to good yields at elevated temperature under aqueous‐mediated conditions. A mechanistic rationalization for this reaction is also provided. The absorption characteristics of the compounds were examined by UV‐Vis spectra and fluorescence spectroscopy. All compounds were fluorescent in solution emitting at blue light (432–433 nm), green light (512–536 nm), or yellow light (591 nm).  相似文献

利用新型荧光试剂1,2-苯并-3,4-二氢咔唑-9-乙基肼基甲酸酯(BCEC)作柱前衍生化试剂,在HypersilBDSC18(4.6mm×200mm,5μm)色谱柱上,采用梯度洗脱对皮质醇、皮质酮、睾酮、孕酮4种类固醇荧光衍生物进行了优化分离。65℃下在乙腈溶剂中以三氯乙酸作催化剂,衍生反应2h后获得稳定的荧光产物。激发和发射波长分别为333nm和390nm。采用大气压化学电离源(APCI)正离子模式,实现了黑线姬鼠粪便中4种类固醇化合物的定性及定量测定。线性回归系数均在0.9999以上,检出限为47.3~71.2fmol。  相似文献

A simple and efficient synthesis of novel spirooxindole–furan derivatives has been investigated by a modified version of the“interrupted” Feist–Bénary (IFB) reaction of isatin derivatives, 1,3-dicarbonyl compounds and N-phenacyl pyridinium salts in the presence of triethylamine. The reaction has been carried out under mild conditions in ethanol, and the products were obtained in good to moderate yields with a simple work-up procedure.  相似文献

The mechanisms that trigger the phototoxic response to 2-chlorophenothiazine derivatives are still unknown. To better understand the relationship between the molecular structure of halogenated phenothiazines and their phototoxic activity, their photophysics and photochemistry were studied in several alcohols. The photodestruction quantum yields were determined under anaerobic conditions using monochromatic light (313 nm). Absorption- and emission-spectroscopy, 1H- and 13C-NMR and GC-MS were used to characterize the photoproducts and reference compounds. An electron transfer mechanism had been previously proposed by Bunce et al . ( J. Med. Chem . 22 , 202–204) to explain the large difference between the photodestruction quantum yield of 2-chlorpromazine (φ = 0.46) and 2-chlorphenothiazine (φ = 0.20). According to these authors, the alkylamino chain transfers an electron to the phenothiazine moiety. Our results demonstrate that this mechanism is incorrect, because the photodestruction quantum yields of all chlorinated derivatives of this study are the same under the same conditions of solvent and irradiation wavelength. The quantum yield has no dependence on the 10-substituent, but it depends on the solvent. The percentage of each photoproduct, on the other hand, strongly depends on that substituent, but not very much on the solvent. Finally, it is demonstrated that the phototoxic effect of chlorinated phenothiazines is not related to the photodechlorination, although both processes share the same transient.  相似文献

A high-performance liquid chromatographic procedure has been developed for the separation and fluorometric detection of guanidino compounds in physiologic fluids. All guanidino compounds were separated on a 17 X 0.46 cm cation-exchange column using a stepwise pH gradient. The chromatographic system was designed to enable the use of the specific reagent 9,10-phenanthrenequinone as a means of monitoring the guanidino compounds of physiologic fluids. This new analytical method is so sensitive that it enables the analysis at the picomole level. Our automatic guanidino-compound analyzer was successfully applied to the quantitative determination of all guanidino compounds in physiologic fluids from normal controls and uremic patients.  相似文献

A simple and straight forward solvent free method for the synthesis of a series of highly functionalized tetrahydropyridine derivatives has been achieved via multicomponent reaction of aromatic aldehydes, various amines, and β‐keto esters at room temperature using readily available (±)‐camphor‐10‐sulfonic acid as an organocatalyst. The current protocol offers an atom economic and environmentally benign method for the synthesis of the title compounds under solvent free conditions in good yields with high substrate scope and avoids column chromatographic purification.  相似文献

A series of derivatives of meso-tetraphenylporphine, with neutral, acidic and basic functional groups, has been prepared. Several of these compounds were synthesized directly via the Rothemund reaction, under a variety of conditions to obtain optimal yields; others were prepared by interconversions of various functional groups. Drastic reaction conditions employed for hydrolysis, alcoholysis, or lithium aluminum hydride reduction did not affect the porphine ring system. The two amino derivatives showed anomalous spectra in the visible range.  相似文献

A simple and convenient one‐pot method has been used for the synthesis of new polycyclic compounds articulated around 3‐cyanopyridine derivatives from 2‐chloroquinolin‐3‐carbaldehydes, acetophenone derivatives, active methylene compounds, and ammonium acetate as a source of ammonia in the presence of catalytic amounts of PPh3 at room temperature. The salient features of the protocol used are mild reaction conditions, good yields, short reaction time, safety, and high atom economy. No column chromatographic was needed, and pure products were obtained by simple filtration and washing with Et2O.  相似文献

Ag support on silica has been used as an effective heterogeneous catalyst for the facile synthesis of chloro‐8‐substituted‐9H‐purine derivatives via the one‐pot reaction of 6‐chloro‐pyrimidines and substituted acids. The title compounds were formed as excellent yields with short reaction time under eco‐friendly conditions. The prepared catalyst (Ag/SiO2) can be reused for a number of times with insignificant loss in its activity. This route has the advantage of being a cost‐effective, readily available, easy workup procedure.  相似文献

 一种新的衍生试剂9,10 蒽醌 2 磺酰氯(ASC)首次用于酚类衍生。几种不同极性的酚被用于评价该试剂。为便于考察ASC对酚衍生的机理及优化衍生条件,制备了不同酚的标准衍生物并对它们进行了结构确证。衍生过程涉及去质子酚与特丁基铵阴离子形成离子对后被有机溶剂提取。衍生反应可以在室温下3min内在两相界面上定量完成。衍生产物很稳定,可以分别被正相和反相分离(相应地在320nm或256nm波长处检测),其浓度和响应在0 2μmol/L~200μmol/L内存在很好的线性关系。  相似文献

A sensitive fluorimetric method for the determination of monosubstituted guanidino compounds is based on their reaction in potassium hydroxide solution with benzoin in the presence of dimethylformamide. The fluorescence produced is stabilized by β-mercaptoethanol and shows excitation and emission maxima around 325 and 435 nm, respectively. The method is simple, selective for monosubstituted guanidino compounds, including polypeptides with one or two arginyl residues, and sensitive; compounds can be determined at concentrations as low as 0.08—0.22 nmol ml-1.  相似文献

The synthesis of organophosphorus compounds is of great importance to industrial, agricultural, and pharmaceutical chemistry. In this paper, we have reported the synthesis of highly substituted pyrazole derivatives through P–C bond formation under catalyst-free conditions. On the other hand, the first catalytic asymmetric version of this reaction has also been developed under catalysis by an isosteviol derived thiourea organocatalyst. The optically active phosphorus-containing compounds have been obtained in good chemical yields with moderate enantioselectivities.  相似文献

A variety of carbonyl compounds have been successfully converted to the corresponding thioacetal derivatives in good to excellent yields on reaction of carbonyl compounds with 1,2-ethanedithiole, 1,3-propanedithiol, and ethylthiol in the presence of a catalytic amount of benzyltriphenylphosphonium tribromide (BTPTB) under solvent-free conditions. Some of the major advantages of this method are mild reaction conditions, high efficiency, and the compatibility with other reported methods. In addition, no bromination occurs at the double bond or α to the keto position or even in the aromatic ring under these experimental conditions.  相似文献

A simple method for the synthesis of thioamide derivatives in DMSO at room temperature and at 120 °C has been developed. Total 27 compounds were prepared under both conditions via a one-pot, three component reaction between substituted aromatic aldehydes, elemental sulfur powder, and cyclic secondary amines. By optimizing the mole ratio of sulfur powder and amines, we have successfully carried out Willgerodt-Kindler reaction of aromatic aldehydes at room temperature. At 120 °C, it is catalyst free reaction with lower reaction time whereas at room temperature, due to the additional amine molecule, Willgerodt-Kindler reaction of aromatic aldehydes is successfully carried out at room temperature. On gram-scale, the reaction is successfully attempted under both conditions with good yields.  相似文献
