The effects of ultraviolet (UV) radiation on chlorophyll content and accumulation of the anti-inflammatory monoterpene-indole alkaloid brachycerine in plants and calli of Psychotria brachyceras (Rubiaceae) were investigated. In this study, we also investigated a protective role for brachycerine against stress conditions. Calli and tip cuttings incubated in nutrient media were daily supplemented with 4 or 16 h of UV. High-performance liquid chromatography analyses of methanolic extracts showed only traces of brachycerine in irradiated aseptic cultures, with no alkaloid being observed in control calli. In cuttings, a 10-fold increase in brachycerine content was seen after exposure for 16 h to UV-C, whereas a 4 h daily supplementation doubled the amount of the alkaloid in leaves. Exposure to a UV-B source also doubled the alkaloid yield. In vitro brachycerine was able to quench singlet oxygen. The data indicate a potential protective role for brachycerine against UV radiation, acting as a UV filter (absorption peaks are within the UV range) and a reactive oxygen species scavenger. In addition, UV radiation may be used to increase yields of this compound of pharmaceutical interest.  相似文献

The effects of UVA (365 nm) radiation on the cellular distribution of F-actin and formation of binucleated cells have been studied using 3T3 Swiss albino mouse fibroblasts and V79 Chinese hamster fibroblasts. Ultraviolet A at biologically relevant fluences was found to disintegrate the actin filaments in the cells shortly (5 min) after irradiation, concomitant with the formation of cells with two nuclei. In 76-100% of the bi- and multinucleated cells the distribution of F-actin was clearly altered. Cells in GI phase of the cell cycle were most probably involved in the formation of binucleated cells. The disintegration of F-actin was presumably not due to depolymerization of F-actin to G-actin, as the amount of F-actin in the cells was unaltered after UVA exposure but rather due to direct breakage of the actin filaments. Ultraviolet B (297/302 nm) had no effect on the cellular distribution of microfilaments, not even at highly lethal fluences.  相似文献

UVA radiation penetrates deeply into the skin reaching both the epidermis and the dermis. We thus investigated the effects of naturally occurring doses of UVA radiation on mitogen-activated protein kinase (MAPK) activities in human dermal fibroblasts. We demonstrated that UVA selectively activates p38 MAPK with no effect on extracellular-regulated kinases (ERK1-ERK2) or JNK-SAPK (cJun NH2-terminal kinase-stress-activated protein kinase) activities. We then investigated the signaling pathway used by UVA to activate p38 MAPK. L-Histidine and sodium azide had an inhibitory effect on UVA activation of p38 MAPK, pointing to a role of singlet oxygen in transduction of the UVA effect. Afterward, using prolonged cell treatments with growth factors to desensitize their signaling pathways or suramin to block growth factor receptors, we demonstrated that UVA signaling pathways shared elements with growth factor signaling pathways. In addition, using emetine (a translation inhibitor altering ribosome functioning) we detected the involvement of ribotoxic stress in p38 MAPK activation by UVA. Our observations suggest that p38 activation by UVA in dermal fibroblasts involves singlet oxygen-dependent activation of ligand-receptor signaling pathways or ribotoxic stress mechanism (or both). Despite the activation of these two distinct signaling mechanisms, the selective activation of p38 MAPK suggests a critical role of this kinase in the effects of UVA radiation.  相似文献

Exposure to ultraviolet (UV) radiation may induce erythema, DNA damage and suppression of immune responses. Melanin pigmentation offers protection against the first two of these effects, but immunosuppression seems to occur irrespective of the subject's pigmentation. Cis-urocanic acid (cis-UCA), produced by isomerization of trans-UCA in the stratum corneum on UV exposure, initiates some of the immunomodulatory effects of UV radiation. In the present study the relationship between skin pigmentation and UCA isomerization has been examined in 28 healthy individuals of skin types I-IV. Pigmentation is measured in five areas of not recently exposed back skin before irradiation with 0, 0.45, 0.9, 1.8 and 3.6 standard erythema dose (SED) of filtered broadband UV-B (1 SED = 10 mJ cm-2 at 298 nm). The concentration of UCA isomers is measured immediately after the irradiation. With 3.6 SED, the relative production of cis-UCA is close to the maximum obtainable, irrespective of skin type. A significant negative correlation is found between pigmentation and relative production of cis-UCA at 0.45 and 1.8 SED, and between pigmentation and absolute production of cis-UCA at 0.45 SED. At doses of 0.45 and 0.9 SED the relative and absolute production of cis-UCA are higher in the group with skin types I and II when compared with the group with skin types III and IV. The higher isomerization in the lightly pigmented subjects than in the more pigmented ones may indicate that people with fair skin are at a relatively higher risk of immunosuppression when exposed to low doses of UV radiation.  相似文献

We studied the photolysis of a fluoroethylene–fluoropropylene copolymer (FEP) film by vacuum ultraviolet (VUV) radiation from a resonance Xe lamp at a wavelength of 147 nm and air pressures of 0.05 and 2.5 Torr. The chemical changes in the FEP surface layer were investigated by Fourier-transform infrared spectroscopy with attenuated total reflection attachment and X-ray photoelectron spectroscopy. Double bonds were found to be the main product in the case of VUV treatment at 0.05 Torr, while photo-oxidation of FEP occurred predominantly by VUV treatment at 2.5 Torr under formation of the —CF2C(O)F group. This oxygen-containing group was more effectively formed in the FEP surface layer by VUV photo-oxidation than by conventional surface oxidation techniques such as treatments by plasma and corona discharge and ozone. Storage of the VUV-treated polymers in air at 50% relative humidity resulted in hydrolysis of —CF2C(O)F to the —CF2COOH group. Substantial improvement of the film wettability was noticed after VUV photo-oxidation. These findings suggest that VUV irradiation provides a high potential for surface modification of fluorinated polymers which are known to be particularly resistant against functionalization by conventional surface modification techniques such as plasma treatment. © 1998 John Wiley & Sons, Inc. In this work, we have studied the modulation of fibroblast-extracellular matrix interactions by physiological doses of ultraviolet A (UV-A) radiation using an adhesion assay on collagen. We have shown that low doses of UV-A (20 kJ/m2) stimulate fibroblast adhesion while higher doses (100 and 200 kJ/m2) inhibit it. By measurement of the thiobarbituric acid reactive substances (TBARS) and use of the chain-breaking antioxidant vitamin E, no role of lipid peroxidation can be detected in these effects. By incubating fibroblasts with a specific protein kinase C (PKC) inhibitor, GF109203X, we have demonstrated that the stimulation of the adhesion by low doses of UV-A involves, at least in part, a PKC-dependent mechanism. In addition, using function-blocking antibodies of alpha 1, alpha 2 or alpha 5 integrin chains involved in extracellular matrix anchorage, we have shown that they decrease the stimulation of adhesion following low doses of UV-A radiation, demonstrating the involvement of these three integrin chains in this UV-A effect. In parallel, 20 kJ/M2 of UV-A are found to rapidly stimulate membrane expression of alpha 1, alpha 2 and alpha 5 integrin chains. This work, which underlines the involvement of integrins in UV-A effects, contributes to the evaluation of the mechanisms by which cell-matrix interactions modulate cell behaviour.  相似文献

Abstract— Trypsin inactivated by u.v. radiation, gamma radiation, visible radiation in the presence of sensitizing dyes, and autolysis, was examined by the method of disc electrophoresis. Untreated Worthington twice crystallized salt-free trypsin separated into four bands which moved toward the cathode; the main band, which had the greatest mobility, contained all of the detectable tryptic activity. The next most mobile band has been assumed to be a chymotrypsin contaminant. The other two bands are of unknown nature. A progressive loss of all the bands was observed when the enzyme was inactivated by those procedures which produce a ‘damaged’ class of trypsin molecules, i.e. flavin-sensitized photooxidation, autolysis, and treatment with u.v. and gamma radiation. No loss of the main band was observed during photoinactivation with methylene blue and eosin Y as sensitizers. In this latter case, it is postulated that the trypsin inactivation products must be of such a nature that the net charge and conformation of the protein is not greatly changed, thus permitting all of the protein to remain in the same band during electrophoresis.  相似文献

Measurements of solar ultraviolet radiation (<320 nm) have been made with polysulphone film at a temperate and a tropical site and compared mean monthly values of the fraction of ultraviolet in solar radiation are calculated which allow an estimate of ultraviolet dose to be made from a knowledge of total solar radiation.While the data demonstrate the marked effect of sun elevation on the ultraviolet fraction of solar radiation they also indicate the effect of seasonal variation in the thickness of the ozone layer above the temperate site.It is also shown that whereas sunlight makes by far the major contribution to total solar radiation, skylight is of prime importance as regards ultraviolet radiation.  相似文献

In this study, five different HPLC columns were heated to 200 degrees C using a homemade heating system which can be operated in temperature programmed mode. The column bleed as an indicator of induced degradation of the stationary phase material was evaluated using a charged aerosol detector (CAD) and an ultraviolet diode array detector (UV-DAD) at different wavelengths. The silica based C-18 stationary phase gave the highest bleed, and the carbon clad titanium dioxide column the lowest bleed. This was independent of both the detection technique and the wavelength.  相似文献

The VUV MCD and absorption spectra of P4 isolated in an argon matrix were obtained using synchrotron radiation. Four well resolved 1A11T2J transitions are observed (J = A, B, C, D from low to high energy). A Jahn-Teller effect is clearly evident in the 1T2C excited state. The Beer–Lambert law is inadequate to describe the absorption of radiation by a medium if the absorbing component is being simultaneously destroyed by the radiation. A model of UV-induced DNA damage in oceanic bacterioplankton was developed and tested against previously published and novel measurements of cyclobutane pyrimidine dimers (CPD) in surface layers of the ocean. The model describes the effects of solar irradiance, wind-forced mixing of bacterioplankton and optical properties of the water on net DNA damage in the water column. The biological part includes the induction of CPD by UV radiation and repair of this damage through photoreactivation and excision. The modeled damage is compared with measured variability of CPD in the ocean: diel variation in natural bacterioplankton communities at the surface and in vertical profiles under different wind conditions (net damage as influenced by repair and mixing); in situ incubation of natural assemblages of bacterioplankton (damage and repair, no mixing); and in situ incubation of DNA solutions (no repair, no mixing). The model predictions are generally consistent with the measurements, showing similar patterns with depth, time and wind speed. A sensitivity analysis assesses the effect on net DNA damage of varying ozone thickness, colored dissolved organic matter concentration, chlorophyll concentration, wind speed and mixed layer depth. Ozone thickness and mixed layer depth are the most important factors affecting net DNA damage in the mixed layer. From the model, the total amplification factor (TAF; a relative measure of the increase of damage associated with a decrease in ozone thickness) for net DNA damage in the euphotic zone is 1.7, as compared with 2.1-2.2 for irradiance weighted for damage to DNA at the surface.  相似文献

An interaction mechanism for a bisphenol‐A‐based polybenzoxazine resin exposed to ultraviolet radiation is proposed. Fourier transform infrared spectroscopy indicated that a carbonyl‐containing species was formed when cured 6,6′‐bis(2,3‐dihydro‐3‐methyl‐4H‐1,3‐benzoxazinyl)isopropylidene (BA‐m) resin was exposed to ultraviolet radiation (λ > 290 nm) in the presence of ambient air at room temperature. The isopropylidene linkage was the reactive site of cleavage and oxidation. It was apparent from model compound studies that this structure was severed, resulting in the formation of a 2,6‐disubstituted benzoquinone. Model compound studies also demonstrated that the BA‐m polymer backbone structure, the Mannich bridge, which was present in and characterized all the polybenzoxazines, neither cleaved nor oxidized in the presence of ultraviolet radiation in a detectable manner under the conditions of this study. © 2000 John Wiley & Sons, Inc. Abstract Our goal was to ultimately predict the sensitivity of untested bacteria (including those of biodefense interest) to ultraviolet (UV) radiation. In this study, we present an overview and analysis of the relevant 254 nm data previously reported and available in the literature. The amount of variability in this data prevented us from determining an "average" response for any bacterium. Therefore, we developed particular selection criteria to include the data in our analysis and suggested future guidelines for reporting UV sensitivity results. We then compiled a table of the sensitivity to 254 nm UV for 38 bacteria and three bacterial spores. The UV sensitivity was quite similar (within 10%) among the spores of Bacillus anthracis (strains Vollum 1B and Sterne), Bacillus subtilis, and Bacillus megaterium. These data indicate that spores of B. subtilis and B. megaterium could be adequate simulants of B. anthracis spores in UVC experiments. Spores of B. anthracis, B. subtilis and B. megaterium were 5-10 times more resistant to UV than were their corresponding vegetative cells. The vegetative cells of B. anthracis showed similar UV sensitivity to those of Burkholderia pseudomallei, Shigella sonnei, and a wild-type strain of Escherichia coli. Yersinia enterocolitica and Vibrio cholerae appeared more sensitive to UV and Salmonella typhi slightly more resistant to UV than E. coli. The sensitivity (at 254 nm) of all vegetative bacteria ranged from 11 to 80 Jm(2) for a 1 Log(10) kill and from 25-200 Jm(2) for 4 Log(10) kill.  相似文献

Four spectroradiometers located at latitudes from 55 degrees to 90 degrees S conducted near-continuous measurements of ground-level solar ultraviolet irradiance from 1990 through 2001. The behavior during months from October through December is of special interest because this period includes the springtime loss in column ozone and the naturally large irradiances of early summer. Monthly integrated irradiances using biological weightings for erythema and damage to DNA show a distortion of the normal annual cycle in irradiance, with enhanced values occurring in October and November. In some cases, these irradiances exceed those near summer solstice in December. Changes in local cloudiness and column ozone both contribute significantly to interannual variability in erythemal irradiance. This is particularly the case at Palmer Station, near 65 degrees S, where the monthly integrated erythemal irradiance in November 1997 was more than double that observed 5 years earlier. In general, at sites on the Antarctic continent, interannual variability in monthly integrated erythemal irradiance is greatest in November, when the observation for any given year can fall 40% above or below the multiyear mean. Near the tip of South America, interannual variability is approximately half that seen in Antarctica.  相似文献

The results of studies on the use of an excimer laser with a wavelength of 193 nm are presented for the treatment of surface of polymers to improve adhesion. It is shown that the radiation of coherent waves in the ultraviolet range leads to a change in surface topography and increase in its specific area, which contributes to increasing the mechanical adhesion; in addition, laser exposure activates the surface of the polymers, intensifying adsorption adhesive interaction.  相似文献

Using our newly built extreme ultraviolet (XUV) photoelectron and photoion spectrometer, we have obtained the pulsed field ionization zero kinetic energy (ZEKE) photoelectron spectra of SO2+(X2A1)<--SO2(X1A1) by coherent XUV radiation in the energy range of 12.29-12.82 eV. The adiabatic ionization potential (IP) of SO2 is 12.3458+/-0.0002 (eV), which was determined by comparing the partially resolved rotational branch contour with the simulated one. Besides the bending vibrational mode (upsilon2) which was found to be exclusive in the photoelectron spectra (PE) reported previously, we also observed the other two modes: the symmetric stretching (upsilon1) and the antisymmetric stretching (upsilon3) vibrations. The fundamental of the symmetric stretching (upsilon(1)) is 1057 cm(-1) and the overtone of the antisymmetric stretching (2upsilon(3)) is 2494 cm(-1). The new vibrational progressions (upsilon(1)00)+, (1upsilon(2)0)+, (2upsilon(2)0)+, and (0upsilon(2)2)+ have also been observed, and these new observations suggested that the irregular structure of (0upsilon(2)0)+ assigned to the previous PE spectra should be reconsidered. The comparison of the intensities of these vibrational bands with the calculated Franck-Condon factors with harmonic approximation was also made.  相似文献

Measurements were conducted at San Ya, China (18.4°N, 109.7°E, altitude 18 m) to investigate the diurnal variation of ocular exposure to ultraviolet (UV) radiation. The experimental apparatus was composed of a manikin and a dual-detector spectrometer to simultaneously measure ocular and ambient UV data. The experimental apparatus was rotated clockwise to simulate three different types of exposure. When the manikin was facing into the sun, the ocular exposure to UV radiation on a summer day was bimodally distributed. The maximum ocular UV irradiance occurred at solar elevations of around 40° and 50° for UVA and UVB respectively. The spectral irradiances were measured at specific wavelength to obtain the ocular biologically effective UV (UV(BE) ) irradiances for photokeratitis, photoconjunctivitis and cataract, and the UV index (UVI) was calculated at the same time point for comparison. When the manikin faced the sun, the maximal ocular UV(BE) irradiance values were obtained at the solar elevation where the UVI value was 8. The results of this study showed that protection against ocular overexposure during outdoor activities should be taken not only at noon but also at other times.  相似文献

The control of unspecific adsorption of proteins to natural and technical surfaces plays an important role in biology and also for many applications. Organic model surfaces, e.g., self-assembled monolayers, are often used to identify fundamental surface and/or protein properties that rule protein adsorption. Some techniques involved in biointerface research require the use of heavy water, e.g. neutron scattering techniques. Also in NMR studies D(2)O is the solvent of choice when focusing on biomolecular and hydration dynamics. So far several studies have been concerned with the characterization of the unspecific adsorption of proteins from normal water buffers. In the present work, we report a comparison of the unspecific protein adsorption from normal and heavy water buffers. So far it has been assumed that the surface kinetic of the unspecific adsorption is unaffected by the substitution of water by D(2)O. However, for the four proteins investigated here, this assumption does not hold. The ratio k(H)/k(D) of the adsorption rate constants of the different buffer conditions describe the strength of the isotope effect. We have measured ratios between 1.0 and 2.6, indicating that the adsorption kinetics are strongly affected by a H(2)O-D(2)O substitution.  相似文献
