The commonly used Zn(2+) sensors 6-methoxy-8-p-toluenesulfonamidoquinoline (TSQ) and Zinquin have been shown to image zinc proteins as a result of the formation of sensor-zinc-protein ternary adducts not Zn(TSQ)(2) or Zn(Zinquin)(2) complexes. The powerful, cell-permeant chelating agent N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine (TPEN) is also used in conjunction with these and other Zn(2+) sensors to validate that the observed fluorescence enhancement seen with the sensors depends on intracellular interaction with Zn(2+). We demonstrated that the kinetics of the reaction of TPEN with cells pretreated with TSQ or Zinquin was not consistent with its reaction with Zn(TSQ)(2) or Zn(Zinquin)(2). Instead, TPEN and other chelating agents extract between 25 and 35% of the Zn(2+) bound to the proteome, including zinc(2+) from zinc metallothionein, and thereby quench some, but not all, of the sensor-zinc-protein fluorescence. Another mechanism in which TPEN exchanges with TSQ or Zinquin to form TPEN-zinc-protein adducts found support in the reactions of TPEN with Zinquin-zinc-alcohol dehydrogenase. TPEN also removed one of the two Zn(2+) ions per monomer from zinc-alcohol dehydrogenase and zinc-alkaline phosphatase, consistent with its ligand substitution reactivity with the zinc proteome.  相似文献

Zinquin (ZQ) is a commonly used sensor for cellular Zn(2+) status. It has been assumed that it measures accessible Zn(2+) concentrations in the nanomolar range. Instead, this report shows a consistent pattern across seven mammalian cell and tissue types that ZQ reacts with micromolar concentrations of Zn(2+) bound as Zn-proteins. The predominant class of products were ZQ-Zn-protein adducts that were characterized in vivo and in vitro by a fluorescence emission spectrum centered at about 470 nm, by their migration over Sephadex G-75 as protein not low molecular weight species, by the exclusion of reaction with lipid vesicles, and by their large aggregate concentration. In addition, variable, minor formation of Zn(ZQ)(2) with a fluorescence band at about 490 nm was observed in vivo in each case. Because incubation of isolated Zn-proteome with ZQ also generated similar amounts of Zn(ZQ)(2), it was concluded that this species had formed through direct ligand substitution in which ZQ had successfully competed for protein-bound Zn(2+). Parallel studies with the model Zn-proteins, alcohol dehydrogenase (ADH), and alkaline phosphatase (AP) revealed a similar picture of reactivity: ZQ(ACID) (Zinquin acid, (2-methyl-8-p-toluenesulfonamido-6-quinolyloxy)acetate)) able to bind to one Zn(2+) and extract the other in Zn(2)-ADH, whereas it removed one Zn(2+) from Zn(2)-AP and did not bind to the other. Zinquin ethyl ester (ethyl(2-methyl-8-p-toluenesulfonamido-6-quinolyloxy)acetate); ZQ(EE)) bound to both proteins without sequestering Zn(2+) from either one. In contrast to a closely related sensor, 6-methoxy-8-p-toluenesulfonamido-quinoline (TSQ), neither ZQ(ACID) nor ZQ(EE) associated with Zn-carbonic anhydrase. A survey of reactivity of these sensors with partially fractionated Zn-proteome confirmed that ZQ and TSQ bind to distinct, overlapping subsets of the Zn-proteome.  相似文献

A highly selective and sensitive fluorescent Zn(2+) sensor, 2,6-bis(2-hydroxy-benzoic acid hydrazide)-4-methylphenol (1), was designed and synthesized. In aqueous THF (4 : 6 v/v) ligand 1 induces a 2 : 1 complex formation with respect to Zn(2+) at physiological pH. This probe features visible light excitation(390 nm) and emission (490 nm) profiles, excellent selectivity responses for Zn(2+)over other competing biological metal ions with K(d) < 1 pM(2), LOD < 1 ng L(-1) and about 680 fold enhancement in fluorescent intensity upon Zn(2+) binding. It also exhibits cell permeability and intracellular Zn(2+) sensing in A375 human melanoma cancer cell.  相似文献

A novel cell-impermeable zinc sensor was synthesized by incorporating poly(ethylene glycol)(PEG) to N-(8-quinolyl)-p-aminobenzenesulfonamide (HQAS) group.The polymeric zinc sensor combines both valuable features of HQAS and PEG.The HQAS of the sensor has the similar functions to TSQ,and exhibits a good fluorescence response to Zn2+ but poor fluorescence responses to other metal ions.The PEG chain can prevent the sensor to permeate healthy cell membrane.The stained experiments with the yeast cells as model showed that the sensor cannot stain the healthy yeast cells,but only the damaged or died yeast cells. These results indicated the novel zinc probe was a typical cell-impermeable zinc sensor.  相似文献

A new 2,6-bis(5,6-dihydrobenzo[4,5]imidazo[1,2-c]quinazolin-6-yl)-4-methylphenol (1) serves as a highly selective and sensitive fluorescent probe for Zn(2+) in a HEPES buffer (50 mM, DMSO:water = 1:9 (v/v), pH = 7.2) at 25 °C. The increase in fluorescence in the presence of Zn(2+) is accounted for by the formation of dinuclear Zn(2+) complex [Zn(2)(C(35)H(25)N(6)O)(OH)(NO(3))(2)(H(2)O)] (2), characterized by X-ray crystallography. The fluorescence quantum yield of the chemosensor 1 is only 0.019, and it increases more than 12-fold (0.237) in the presence of 2 equiv of the zinc ion. Interestingly, the introduction of other metal ions causes the fluorescence intensity to be either unchanged or weakened. By incubation of cultured living cells (A375 and HT-29) with the chemosensor 1, intracellular Zn(2+) concentrations could be monitored through selective fluorescence chemosensing.  相似文献

A triazole-containing 8-hydroxyquinoline (8-HQ) ether 2 was efficiently synthesized in two steps from the "click" strategy. Compound 2 gave a strong fluorescence (Φ = 0.21) in nonprotic solvent like CH(3)CN, and a weak fluorescence (Φ = 0.06) in protic solvent like water. In water, a more than 100 nm red shift of the fluorescence maximum was observed for compound 2 in comparison with that in CH(3)CN. This fluorescence difference may be attributed to the intermolecular photoinduced proton transfer (PPT) process involving the protic solvent water molecules. Similarly, this intermolecular PPT process was also observed in the high-water-content CH(3)CN aqueous solution (e.g., CH(3)CN/H(2)O = 5/95, v/v). The water content in the CH(3)CN/H(2)O binary solvent mixture greatly affected the fluorescence intensity (e.g., Φ = 0.06 and 0.25 when CH(3)CN/H(2)O = 5/95 and 95/5, v/v, respectively) and emission wavelength. Using this interesting property, by simple variation of the water content in the CH(3)CN aqueous solution, compound 2 was tuned from a selective "turn-on" fluorescent sensor for Zn(2+) (CH(3)CN/H(2)O = 5/95, v/v) to a ratiometric one for Zn(2+) and a selective "turn-off" one for Fe(3+) (CH(3)CN/H(2)O = 95/5, v/v) over a wide range of pH value. In high-water-content (CH(3)CN/H(2)O = 5/95, v/v) aqueous solution compound 2 shows a selective "turn-on" response toward Zn(2+), with a 10-fold enhancement in the fluorescence intensity at 428 nm and a 62 nm blue shift of the emission maximum (490 to 428 nm) due to the inhibition of intermolecular PPT process upon chelating with Zn(2+). However, in a less polar solvent (CH(3)CN/H(2)O = 95/5, v/v) in which compound 2 has high fluorescence (quantum yield =0.25), it shows a ratiometric response toward Zn(2+), with a continuous decrease of the fluorescence intensity at 399 nm and an increase at 423 nm. More interestingly, in this case, it also exhibits a very sensitive, selective, and ratiometric fluorescence quenching in the presence of Fe(3+), with an 81 nm red shift of the emission maximum (399 to 480 nm) in a wide range of pH through a metal ligand charge transfer (MLCT) effect.  相似文献

A new 2-(9-anthrylmethylamino)ethyl-appended cyclen, L(3) (1-(2-(9-anthrylmethylamino)ethyl)-1,4,7,10-tetraazacyclododecane) (cyclen = 1,4,7,10-tetraazacyclododecane), was synthesized and characterized for a new Zn(2+) chelation-enhanced fluorophore, in comparison with previously reported 9-anthrylmethylcyclen L(1) (1-(9-anthrylmethyl)-1,4,7,10-tetraazacyclododecane) and dansylamide cyclen L(2). L(3) showed protonation constants log K(a)(i)() of 10.57 +/- 0.02, 9.10 +/- 0.02, 7.15 +/- 0.02, <2, and <2. The log K(a3) value of 7.15 was assigned to the pendant 2-(9-anthrylmethylamino)ethyl on the basis of the pH-dependent (1)H NMR and fluorescence spectroscopic measurements. The potentiometric pH titration study indicated extremely stable 1:1 Zn(2+)-L(3) complexation with a stability constant log K(s)(ZnL(3)) (where K(s)(ZnL(3)) = [ZnL(3)]/[Zn(2+)][L(3)] (M(-)(1))) of 17.6 at 25 degrees C with I = 0.1 (NaNO(3)), which is translated into the much smaller apparent dissociation constant K(d) (=[Zn(2+)](free)[L(3)](free)/[ZnL(3)]) of 2 x 10(-)(11) M with respect to 5 x 10(-)(8) M for L(1) at pH 7.4. The quantum yield (Phi = 0.14) in the fluorescent emission of L(3) increased to Phi = 0.44 upon complexation with zinc(II) ion at pH 7.4 (excitation at 368 nm). The fluorescence of 5 microM L(3) at pH 7.4 linearly increased with a 0.1-5 microM concentration of zinc(II). By comparison, the fluorescent emission of the free ligand L(1) decreased upon binding to Zn(2+) (from Phi = 0.27 to Phi = 0.19) at pH 7.4 (excitation at 368 nm). The Zn(2+) complexation with L(3) occurred more rapidly (the second-order rate constant k(2) is 4.6 x 10(2) M(-)(1) s(-)(1)) at pH 7.4 than that with L(1) (k(2) = 5.6 x 10 M(-)(1) s(-)(1)) and L(2) (k(2) = 1.4 x 10(2) M(-)(1) s(-)(1)). With an additionally inserted ethylamine in the pendant group, the macrocyclic ligand L(3) is a more effective and practical zinc(II) fluorophore than L(1).  相似文献

In attempts to investigate whether the photoluminescence properties of the Zn-based heterometal-organic frameworks (MOFs) could be tuned by doping different Ln(3+) (Ln = Sm, Eu, Tb) and Mn(2+) ions, seven novel 3D homo- and hetero-MOFs with a rich variety of network topologies, namely, [Zn(mip)](n) (Zn-Zn), [Zn(2)Mn(OH)(2)(mip)(2)](n) (Zn-Mn), [Mn(2)Mn(OH)(2)(mip)(2)](n) (Mn-Mn), [ZnSm(OH)(mip)(2)](n) (Zn-Sm), [ZnEu(OH)(mip)(2)](n) (Zn-Eu1), [Zn(5)Eu(OH)(H(2)O)(3)(mip)(6)·(H(2)O)](n) (Zn-Eu2), and [Zn(5)Tb(OH)(H(2)O)(3)(mip)(6)](n) (Zn-Tb), (mip = 5-methylisophthalate dianion), have been synthesized hydrothermally based on a single 5-methylisophthalic acid ligand. All compounds are fully structurally characterized by elemental analysis, FT-IR spectroscopy, TG-DTA analysis, single-crystal X-ray diffraction, and X-ray powder diffraction (XRPD) techniques. The various connectivity modes of the mip linkers generate four types of different structures. Type I (Zn-Zn) is a 3D homo-MOF with helical channels composed of Zn(2)(COO)(4) SBUs (second building units). Type II (Zn-Mn and Mn-Mn) displays a nest-like 3D homo- or hetero-MOF featuring window-shaped helical channels composed of Zn(4)Mn(2)(OH)(4)(COO)(8) or Mn(4)Mn(2)(OH)(4)(COO)(8) SBUs. Type III (Zn-Sm and Zn-Eu1) presents a complicated corbeil-like 3D hetero-MOF with irregular helical channels composed of (SmZnO)(2)(COO)(8) or (EuZnO)(2)(COO)(8) heterometallic SBUs. Type IV (Zn-Eu2 and Zn-Tb) contains a heterometallic SBU Zn(5)Eu(OH)(COO)(12) or Zn(5)Tb(OH)(COO)(12), which results in a 3D hetero-MOF featuring irregular channels impregnated by parts of the free and coordinated water molecules. Photoluminescence properties indicate that all of the compounds exhibit photoluminescence in the solid state at room temperature. Compared with a broad emission band at ca. 475 nm (λ(ex) = 380 nm) for Zn-Zn, compound Zn-Mn exhibits a remarkably intense emission band centered at 737 nm (λ(ex) = 320 nm) due to the characteristic emission of Mn(2+). In addition, the fluorescence intensity of compound Zn-Mn is stronger than that of Mn-Mn as a result of Zn(2+) behaving as an activator for the Mn(2+) emission. Compound Zn-Sm displays a typical Sm(3+) emission spectrum, and the peak at 596 nm is the strongest one (λ(ex) = 310 nm). Both Zn-Eu1 and Zn-Eu2 give the characteristic emission transitions of the Eu(3+) ions (λ(ex) = 310 nm). Thanks to the ambient different crystal-field strengths, crystal field symmetries, and coordinated bonds of the Eu(3+) ions in compounds Zn-Eu1 and Zn-Eu2, the spectrum of the former compound is dominated by the (5)D(0) → (7)F(2) transition (612 nm), while the emission of the (5)D(0) → (7)F(4) transition (699 nm) for the latter one is the most intense. Compound Zn-Tb emits the characteristic Tb(3+) ion spectrum dominated by the (5)D(4) → (7)F(5) (544 nm) transition. Upon addition of the different activated ions, the luminescence lifetimes of the compounds are also changed from the nanosecond (Zn-Zn) to the microsecond (Zn-Mn, Mn-Mn, and Zn-Sm) and millisecond (Zn-Eu1, Zn-Eu2, and Zn-Tb) magnitude orders. The structure and photoluminescent property correlations suggest that the presence of Mn(2+) and Ln(3+) ions can activate the Zn-based hetero-MOFs to emit the tunable photoluminescence.  相似文献

The employment of pyridine-2-carbaldehyde oxime (paoH) in zinc(II) benzoate chemistry, in the absence or presence of azide ions, is described. The syntheses, crystal structures and spectroscopic characterization are reported for the complexes [Zn(O(2)CPh)(2)(paoH)(2)] (1), [Zn(12)(OH)(4)(O(2)CPh)(16)(pao)(4)] (2) and [Zn(4)(OH)(2)(pao)(4)(N(3))(2)] (3). The Zn(II) centre in octahedral 1 is coordinated by two monodentate PhCO(2)(-) groups and two N,N'-chelating paoH ligands. The metallic skeleton of 2 describes a tetrahedron encapsulated in a distorted cube. The {Zn(12)(μ-OH)(4)(μ(3)-ΟR)(4)}(16+) core of the cluster can be conveniently described as consisting of a central {Zn(4)(μ(3)-ΟR)(4)}(4+) cubane subunit (RO(-) = pao(-)) linked to four {Zn(2)(μ-OH)}(3+) subunits via the OH(-) group of each of the latter, which becomes μ(3). The molecule of 3 has an inverse 12-metallacrown-4 topology. Two triply bridging hydroxido groups are accommodated into the metallacrown ring. Each pao(-) ligand adopts the η(1)?:?η(1)?:?η(1)?:?μ coordination mode, chelating one Zn(II) atom and bridging a Zn(II)(2) pair. Complexes 1 and 2 display photoluminescence with maxima at ~355 nm and ~375 nm, upon maximum excitation at 314 nm; the origin of the photoluminescence is discussed.  相似文献

Three fluorescent probes CdABA', CdABA and ZnABA', which are structural isomers of ZnABA, have been designed with N,N-bis(2-pyridylmethyl) ethylenediamine (BPEA) as chelator and 2-aminobenzamide as fluorophore. These probes can be divided into two groups: CdABA, CdABA' for Cd(2+) and ZnABA, ZnABA' for Zn(2+). Although there is little difference in their chemical structures, the two groups of probes exhibit totally different fluorescence properties for preference of Zn(2+) or Cd(2+). In the group of Zn(2+) probes, ZnABA/ZnABA' distinguish Zn(2+) from Cd(2+) with F(Zn)(2+)-F(Cd)(2+) = 1.87-2.00. Upon interchanging the BPEA and carbamoyl groups on the aromatic ring of the fluorophore, the structures of ZnABA/ZnABA' are converted into CdABA/CdABA'. Interestingly, the metal ions selectivity of CdABA/CdABA' was switched to discriminate Cd(2+) from Zn(2+) with F(Cd)(2+)-F(Zn)(2+) = 2.27-2.36, indicating that a small structural modification could lead to a remarkable change of the metal ion selectivity. (1)H NMR titration and ESI mass experiments demonstrated that these fluorescent probers exhibited different coordination modes for Zn(2+) and Cd(2+). With CdABA' as an example, generally, upon addition of Cd(2+), the fluorescence response possesses PET pathway to display no obvious shift of maximum λ(em) in the absence or presence of Cd(2+). However, an ICT pathway could be employed after adding Zn(2+) into the CdABA' solution, resulting in a distinct red-shift of maximal λ(em).  相似文献

Attapulgite, an extremely stable fibrillar mineral present in nature, is a promising new carrier of luminescent lanthanide complexes for further applications. A europium complex Eu(DBM)(3)(H(2)O)(2) (HDBM = dibenzoylmethane) was covalently coupled onto modified attapulgites (or silica nanoparticles) via a ligand exchange reaction, generating attapulgite-based ternary europium complexes. The composites were characterized by CHN elemental analysis, inductively coupled plasma-atomic emission spectroscopy (ICP-AES) for Eu(3+) content, powder X-ray diffraction (XRD), thermogravimetry (TG) and UV-vis absorption spectra. The results indicate that the Eu(3+) complex was grafted covalently to the outer surfaces of attapulgites (or silica nanoparticles) and modifications by coupling agents containing different alkoxide groups (aminopropyltriethoxysilane (APTES) or aminopropylmethyldiethoxysilane (APMDES)) led to different grafting ratios. The structures of these composites were further evidenced by the determination of photophysical behaviours and coordinated water molecules of the complexes linked to matrices. Attapulgite-based lanthanide composites linked by APTES can be excited by visible light, with a wide excitation wavelength range from UV to visible light (a maximum at 398 nm), long luminescence lifetime (503 μs), high quantum yield (48%) and improved exposure durability. When incubated with HeLa cells at 37 °C, the fluorescence of matrix-APTES-cpa-Eu(DBM)(3) is observed on the cell membrane. Moreover, the low cytotoxicity of our present system results in potential applications for cell imaging in biological systems.  相似文献

Ratiometric sensors for the detection of metal ions have gained increasing attention due to its self-calibration tendency for the environmental effects. In this context, we have synthesized and characterized a dual emitting ratiometric Zn(2+) probe (1) having acridinedione as a fluorophore and N,N-bis(2-pyridylmethyl)amine (BPA) as a receptor unit. Existence of two different conformation of the molecule with photoinduced electron transfer (PET) from amine moiety to the acridinedione fluorophore leads to dual emission, namely locally excited (425 nm) and anomalous charge transfer emission (560 nm) in aprotic solvents. In the presence of one equivalent of Zn(2+), a 15-fold fluorescence enhancement in the locally excited state together with the quenching of charge transfer emission is observed. The intensity changes at the two emission peaks allow a ratiometric detection of Zn(2+) under PET signaling mechanism. The utilization of PET process for the ratiometric fluorescence change will further signify the importance of PET mechanism in sensing action. Addition of Zn(2+) to 1 in acetonitrile/water mixtures shows a single emission peak with fluorescence enhancement.  相似文献

A novel host inclusion complex of beta-cyclodextrin-o-vanillin benzoylhydrazone (beta-CDP-OVBH) was prepared and characterized by IR and 1H-NMR spectra. A highly selective and sensitive fluorescent determination of trace amounts of zinc was proposed based on the reaction between Zn(2+) and beta-CDP-OVBH in sodium hydroxide-ammonium acetate buffer medium of pH 8.3. The molar ratio of beta-CDP-OVBH to Zn(2+) was 1:1.The maximum excitation and emission wavelengths were 396 and 486 nm, respectively. The linear range of this method was 2.5-500 microg l(-1) with a detection limit of 0.60 microg l(-1). The effect of interferences in the determination of zinc was investigated, the results showed that the host reagent had a quite high identifying capacity on Zn(2+). This method was successfully applied to the determination of trace amounts of Zn(2+) in tea and human hairs.  相似文献

A ratiometric fluorescent receptor with a C(3) symmetric structure based on a pyrene-linked triazole-modified homooxacalix[3]arene (L) was synthesized and characterized. This system exhibited an interesting ratiometric detection signal output for targeting cations and anions through switching the excimer emission of pyrene from the "on-off" to the "off-on" type in neutral solution. (1)H NMR titration results suggested that the Zn(2+) center of receptor L·Zn(2+) provided an excellent pathway of organizing anion binding groups for optimal host-guest interactions. It is thus believed that this receptor has potential application in sensing, detection, and recognition of both Zn(2+) and H(2)PO(4)(-) ions with different optical signals. In addition, the fluorescence emission changes by the inputs of Zn(2+) and H(2)PO(4)(-) ions can be viewed as a combinational R-S latch logic circuit at the molecular level.  相似文献

A novel two-photon fluorescence probe for Zn(2+) derived from dicyanostilbene as a two-photon fluorophore and 4-(pyridine-2-ylmethyl)piperazine as a novel Zn(2+) ligand was developed. The probe shows a 72.5-fold fluorescence enhancement in response to Zn(2+), a large two-photon action cross-section (580 GM), a noncytotoxic effect, and pH insensitivity in the biologically relevant range, and its dissociation constant (K(d)(TP)) is 0.52 ± 0.01 μM. The probe can selectively detect free Zn(2+) ions in live cells for 1500 s or so and in living tissues at a depth of 80-150 μm without interference from other metal ions and the membrane-bound probes.  相似文献

设计合成了荧光传感分子2-吡啶甲醛-4-乙氧基苯甲酰肼腙(PAEB),采用红外光谱法、氢核磁共振和元素分析法对其结构进行了表征,用紫外可见吸收光谱和荧光发射光谱研究了PAEB对不同金属离子的配位作用的机理。结果表明:PAEB与锌(Ⅱ)形成物质的量的比为1比1配合物。由于此配合物的形成,在发射波长470nm(激发波长为400nm)处出现明显的荧光增强,且对锌(Ⅱ)而言具有较高的选择性。初步探讨了受体分子与锌(Ⅱ)的荧光增强机理。  相似文献

8-Benzenesulfonyloxy-5- N,N-dimethylaminosulfonylquinolin-2-ylmethyl-pendant cyclen (BS-caged-L(4), BS = benzenesulfonyl) was designed and synthesized as a "caged" derivative of a previously described Zn(2+) fluorophore, 8-hydroxy-5- N,N-dimethylaminosulfonylquinolin-2-ylmethyl-pendant cyclen (L(4)) (cyclen = 1,4,7,10-tetraazacyclododecane). In the absence of metal ions and in the dark, BS-caged-L(4) (10 microM) showed negligible fluorescence emission at pH 7.4 (10 mM HEPES with I = 0.1 (NaNO3)) and 25 degrees C (excitation at 328 nm). Addition of Zn(2+) induced an increase in the UV/vis absorption of BS-caged-L(4) (10 microM) at 258 nm and a significant increase in fluorescence emission at 512 nm. These responses are results from the formation of Zn(H-1L(4)) by the hydrolysis of the sulfonyl ester at the 8-position of the quinoline unit promoted by the Zn(2+)-bound HO(-). Improvement of cell membrane permeation in comparison with L(4) is also described.  相似文献

The characteristics as a chemosensor of the compound 3-methyl-6,8-di(2-pyridyl)-[1,2,3]triazolo[5',1':6,1]pyrido[2,3-]pyrimidine (1) have been analyzed. Interaction with Cu(2+) produces a quenching of the fluorescence, while interaction with Zn(2+) leads to a quenching of the fluorescence followed by a bathochromic shift. The crystal structure of the Zn(1)(H(2)O)(3)(ClO(4))(2) x H(2)O complex shows the coordination of Zn(2+) through the terpyridine moiety. The octahedral site is completed by three water molecules. Interactions of the Zn(2+) complex with the anions sulfate, nitrate, nitrite, and dihydrogenphosphate in ethanol produce hypsochromic shifts and restoration of the fluorescence whose magnitude depends on the anion involved. The maximum interaction is observed for H(2)PO(4)(-). Interactions of the Zn(2+) complex with the amino acids l-aspartate and l-glutamate have also been explored showing a higher interaction with l-aspartate.  相似文献

Hydrothermal treatment of racemic atropisomeric ligand diethyl (R,S)-7,7'-dimethyl-4,4'-biquinoline-3,3'-dicarboxylate (DDBD) in the presence of pyridine over 4 days at 140 ℃ with Zn(OAc)2 offers zinc coordination polymer [{Zn(DBD)(pyridine)2(H2O)}n{Zn(DBD)(H2O)1/2}]n (1), which shows weak fluorescence at about 490 nm at solid state at room temperature. Zn(2+) plays important roles in various biological systems; as a result, the development of tools that can visualize chelatable Zn(2+) has attracted much attention recently. We report here newly synthesized fluorescent sensors for Zn(2+), ZnAF-Rs, whose excitation maximum is shifted by Zn(2+) under physiological conditions. Thus, these sensors enable ratiometric imaging, which is a technique to reduce artifacts by minimizing the influence of extraneous factors on the fluorescence of a probe. Ratiometric measurement can provide precise data, and some probes allow quantitative detection. ZnAF-Rs are the first ratiometric fluorescent sensors for Zn(2+) that enable quantitative analysis under physiological conditions. ZnAF-Rs also possess suitable K(d) for applications, and high selectivity against other biologically relevant cations, especially Ca(2+). Using these probes, changes of intracellular Zn(2+) concentration in cultured cells were monitored successfully. We believe that these probes will be extremely useful in studies on the biological functions of Zn(2+).  相似文献
