含环氧基的硅与二乙胺或聚乙烯亚胺反应,其产物再与碘甲烷或氯化苄反应,得到含季铵离子的强阴离子交换色谱填料,该埴料可用于蛋白质和生物工程产品(如基因重组人干扰素γ,粒细胞-巨噬细胞集落刺激因子和人绒毛膜促性腺素等)的分离纯化,其中,环氧基硅胶与二乙胺加成后再用氯化季铵化的产物是较好的强阴离子交换色谱填料。 相似文献

将硅胶与环氧基有机硅烷反应的产物与聚乙烯亚胺交联,再与环状酸酐反应制得高效弱阳离子交换色谱填料WCX-5和WCX-6;交联产物与聚琥珀酰亚胺反应,然后碱性水解制得填料WCX-7,研究了它们的性能,WCX-5的分离性能优于WCX-5,WCX-7有一些不同于前两者的色谱特性,三者皆可用于蛋白质的分离,溶菌酶的活性回收率大于94%。 相似文献

以大孔微球硅胶(30nm,8μm)为基体,经γ-氨丙基三乙氧基硅烷活化与间隔臂氯代环氧丙烷反应后,再与螯合剂亚氨基二乙酸键合,后者与铜离子(Ⅱ)螯合后,构成定位金属离子亲和色谱固定相。用磷酸盐缓冲溶液(pH=7.0)作流动相,经紫外吸收检测(260nm)对核苷酸混合物进行了高效液相色谱分析,取得较满意的结果。 相似文献

固相配位化学反应研究(ⅩⅩⅤ)——草酸高铁与路易氏间碱的固相反应 总被引:1,自引:1,他引:1
用气相色谱法研究了草酸铁(Ⅲ)与路易氏碱在氢气氛和氦气氛中的固相反应。结果表明草酸铁与氢氧化钠在室温下明显发生固相反应,其产物随温度升高而变化。同时发现α-Fe对草酸钠有催化分解作用,与四硫化钾低于220°C时发生固相反应,生成KFeS2,但只有1/4草酸铁参加了固相反应,反应不完全,对固相反应的机制及反应气氛的影响也进行了探讨。 相似文献

用气相色谱分析由Me2SiCl2(1)与Li在各阶段反应的生成物, 发现除偶联反应外, 还有环化、环转化平衡、聚合和裂解等反应, 它们随反应条件的改变而变化。当反应液中含有催化量的Me3SiSiPh3(2)时, 可以促进热力学稳定的(Me2Si)6的生成, 进一步证明了亲核试剂对合成(MeSi)6的催化作用。在一定的反应条件下, 产物的收率达80%以上。 相似文献

采用全新的两步原位合成法,先通过将带有双活性反应基团γ—三甲氧基硅丙基异丁烯酸酯的CH3-O-Si-基团与石英毛细管内壁表面的Si-0H反应,在石英毛细管内壁键合上带有活性基团的中间有机层,再原位合成多孔聚合物,制成高效一体化Q型聚合物颗粒多孔层开管柱(PLOT)Pora-Q毛细管色谱柱;该色谱柱具有良好的惰性、较强的分离能力;与一般的商品化PLOT-Q色谱柱相比,热稳定性和机械强度有明显的改善和提高,最高使用温度提高了50℃达到300℃,扩展了该种色谱柱的应用范围。 相似文献

Summary The chromatographic behaviour of anions on paper strips treated with tri-n-octylamine (TOA) salts or Aliquat 336 and developed with aqueous solutions of acetic, formic, monochloroacetic or trifluoroacetic acids and their sodium salts was investigated. Liquid-liquid extraction of organic acids by 0.1 M solution of TOA in benzene as well as the anion exchange between benzene solutions of TOA salts or Aliquat 336 (in acetate form) in benzene and aqueous solutions of sodium halides was also studied. It was found that extraction increases in the following order of the acids: CH3COOOH<HCOOH<CH2CICOOH<CF3COOH; the relative affinity of organic anions to the quaternary alkyl-ammonium cation also increase in a similar order. The extraction of acid in excess over the amount necessary to neutralize the amine was observed for all four acids. The RF values of anions investigated depend markedly on the type of organic acids or their salts and their concentration in the mobile phase. Halide ions are more strongly retained on paper treated with Aliquat 336 as compared with TOA salts. 将阴阳离子交换色谱柱在灌注色谱工作站上串联,使苦瓜籽粗提液中未被阴离子色谱柱吸附的蛋白质直接在阳离子色谱柱上吸附,用盐线性梯度洗脱从阳离子色谱柱上分离出两个具有抗真菌活性的蛋白。两个抗真菌蛋白经鉴定都达到了电泳纯,相对分子质量均为30000左右,N-端氨基酸序列分别为DVSFRLSGADPRSYGMFI与DVNFDLSTATAK,表明所得到的两种蛋白为苦瓜籽中的2个Ⅰ型核糖体失活蛋白α-苦瓜素(α-MMC) 和β-苦瓜素(β-MMC)。抗菌实验表明,α-MMC对香蕉枯萎菌和果腐霉菌均具有较强的抑制作用,β 相似文献

Continuous perfusion cell culture, using albumin containing medium, offers the potential advantages of higher recombinant Prourokinase (r-ProUK) yields, higher initial product purity and increased throughput compared to batch culture technology using medium supplemented with fetal bovine serum. We have characterized the production of r-ProUK in medium supplemented with a lipid rich bovine serum albumin (Albumax) in a perfusion system. The results of these studies showed that it was necessary to modify the r-ProUK batch recovery scheme to process r-ProUK from a perfusion system. To accommodate large volumes of perfusate harvested over a ten to fourteen day production cycle, cation exchange and hydrophobic interaction chromatography (HIC) resins were identified that had increased product binding capacity, better flow characteristics and wider pH ranges which allowed caustic cleaning. The mobile phase composition, pH and ionic strength were modified to improve r-ProUK yields from the identified resins, and procedures were developed to eliminate r-ProUK degradation products. Strategies were defined for processing continuous harvest, which contained four to seven times the amount of r-ProUK of batch harvests. 相似文献

Surface‐activated chemical ionization (SACI) has been widely used in recent years for the analysis of different compounds (e.g. peptides, street drugs, amino acids). The main benefits of this technology are its high sensitivity and its effectiveness under different chromatographic conditions [i.e. ion exchange chromatography and reversed‐phase (RP) chromatography]. Here we used SACI in conjunction with quadrupole time‐of‐flight mass spectrometry to analyze enterotoxin A, which is produced by Staphylococcus aureus, in milk matrix using both RP and ion exchange chromatographies. SACI had increased sensitivity as compared with electrospray ionization. Moreover, the higher quantitation efficiency of this technique, mainly in terms of limit of detection (0.01 ng/ml), limit of quantitation (0.05 ng/ml), linearity range (0.05–50 ng/ml), matrix effect, accuracy (intraday and interday accuracy errors were 9.2% and 10.3%, respectively) and precision (intraday and interday precision errors were 5.3% and 12.8%, respectively), is shown and discussed.
