Ambient ionization mass spectrometry: a tutorial   总被引:4,自引:0,他引:4  
Ambient ionization is a set of mass spectrometric ionization techniques performed under ambient conditions that allows the direct analysis of sample surfaces with little or no sample pretreatment. Using combinations of different types of sample introduction systems and ionization methods, several novel techniques have been developed over the last few years with many applications (e.g., food safety screening; detection of pharmaceuticals and drug abuse; monitoring of environmental pollutants; detection of explosives for antiterrorism and forensics; characterization of biological compounds for proteomics and metabolomics; molecular imaging analysis; and monitoring chemical and biochemical reactions). Electrospray ionization and atmospheric pressure chemical ionization are the two main ionization principles most commonly used in ambient ionization mass spectrometry. This tutorial paper provides a review of the publications related to ambient ionization techniques. We describe and compare the underlying principles of operation, ionization processes, detecting mass ranges, sensitivity, and representative applications of these techniques.  相似文献

Within this paper a sub-group of ambient ionization mass spectrometry namely direct ionization mass spectrometry techniques are reviewed. They are characterized by the generation of an electrospray directly from the sample investigated. Prominent representatives include paper spray mass spectrometry, tissue spray mass spectrometry, probe electrospray ionization or thin-layer chromatography mass spectrometry. Applications of all major direct ionization techniques within different fields such as biomedical analysis, analysis of natural products, analysis of technical products and food analysis, just to name a few, are discussed and relevant parameters are listed in five Tables.  相似文献

We report the linear retention indices of the 14 methylnitro-naphthalenes on three high resolution capillary columns and their electron impact mass spectra. The analyses of the methylnitro-naphthalenes in an ambient air sample by gas chromatography/mass spectrometry with multiple ion detection utilizing 5% phenylmethylsilicone, 50% phenylmethylsilicone, and smectic columns are shown.  相似文献

The separation of structurally related angiotensin-converting enzyme (ACE) inhibitors lisinopril, cilazapril, ramipril and quinapril and their corresponding active diacid forms (prilates) by conventional TLC silica gel 60 plates was contrasted with that afforded by monolithic ultra-thin-layer chromatographic (UTLC) plates. For the use of UTLC plates technical modifications of the commercially available equipments for the sample application, development and detection were made. Plates were developed in modified horizontal developing chamber using ethyl acetate-acetone-acetic acid-water (4:1:0.25:0.5, v/v). Detection of the separated compounds was performed densitometrically in absorption/reflectance mode at 220 nm and after exposure to iodine also by image analysis. The obtained results showed that monolithic layer is more efficient for the separation of structurally similar polar compounds, such as prilates than conventional silica layers. Identification of the compounds was confirmed by ESI-MS after their on-line extraction from the UTLC and TLC plates by means of Camag TLC-MS interface.  相似文献

为探究生物质快速热解反应历程,利用裂解气质联用仪对生物质进行逐级热解实验,考察在不同温度区间热解液体产物组分的分布规律。实验结果表明,生物质的化学组成和热解温度区间对热解液体产物都有重要影响。桉木在25℃~400℃热解液体产物较少,主要是吡喃和芳香类化合物,其中5,6-二氢-4-羟基-吡喃-2-酮相对峰面积随温度升高而降低;在450℃~500℃热解液体产物种类和产量均较多,主要以酮类和芳香类化合物为主。玉米芯热解规律和桉木的相似,但在25℃~350℃主要以呋喃类化合物为主,主要热解液体产物是2,3-二氢-苯并呋喃和2-甲氧基-4-乙烯基苯酚,在400℃~450℃热解液体产物以酮类化合物为主。生物质主要化学组分在不同温度区间热解得到不同液体产物,对其进行选择性热解,能够有效实现生物质资源的综合利用。  相似文献

Presented here is the optimization and development of a desorption electrospray ionization mass spectrometry (DESI-MS) method for detecting natural products on tissue surfaces. Bromophycolides are algal diterpene-benzoate macrolide natural products that have been shown to inhibit growth of the marine fungal pathogen Lindra thalassiae. As such, they have been implicated in antimicrobial chemical defense. However, the defense mechanisms are not yet completely understood. Precise detection of these compounds on algal tissue surfaces under ambient conditions without any disruptive sample processing could shed more light onto the processes involved in chemical defense of marine organisms. Conventional DESI-MS directly on algal tissue showed relatively low sensitivity for bromophycolide detection. Sensitivity was greatly improved by the addition of various anions including Cl, Br, and CF3COO into the DESI spray solvent. Chloride adduction gave the highest sensitivity for all assayed anions. Density functional optimization of the bromophycolide anionic complexes produced during DESI supported this observation by showing that the chloride complex has the most favorable binding energy. Optimized DESI protocols allowed the direct and unambiguous detection of bromophycolides, including A, B, and E, from the surface of untreated algal tissue. Figure Desorption Electrospray Ionization, a novel technique for mass spectrometric analysis under open air conditions reveals the presence of naturally-occurring antibiotics on the surface of marine algae. Ab-initio calculations and experimental results indicate that sensitiviity could be greatly enhanced by means of dynamic complexation of these antibiotics with various small anions during the dynamic desorption process. Gas chromatography/mass spectrometry (GC/MS) after alkaline hydrolysis, solvent extraction and trimethylsilylation, and analytical pyrolysis using hexamethyldisilazane (HMDS) for in situ derivatisation followed by gas chromatographic/mass spectrometric analysis (Pyrolysis-silylation-GC/MS) were used to investigate the hydrolysable and soluble constituents, and the polymerised macromolecules of an archaeological fig (Ficus carica) recovered in Zaragoza (Spain), as well as of modern figs. The main aim was to study the compositional alterations undergone by the fig tissues in a particular archaeological environment: the fig was in a vessel and covered by a layer of a mixture of orpiment and gypsum. A comparison between the GC/MS results from modern and archaeological figs revealed that degradative reactions took place, leading to the disappearance/depletion of reactive (unsaturated fatty acids) and sensitive compounds (phytosterols and triterpenes). Py-silylation-GC/MS data provided evidence of a significant degradation of the saccharide and lipid components of the fig tissue, which left a residue enriched in polyphenols and polyesters.  相似文献

Summary When SFC was rediscovered in the early 1980s, it was frequently estimated that a strong driving force to its development would be the ease of devising a simple SFC/MS interface. This was believed to be easily achieved if analytical conditions were limited to capillary SFC columns as a general separation tool, and to the choice of neat CO2 as the unique supercritical fluid. The low flow rate of mobile phase delivered by capillary columns was easy to accommodate by the vacuum equipment of standard mass spectrometers, and the specific physical properties of CO2 made possible solute ionization by different ion-molecule reactions, especially charge exchange ionization. This approach has lived up to all of its promises. The major causes of the observed mismatch are the large variations of the MS source pressure as a result of the CO2 pressure gradient at the SFC column inlet, the low sensitivity of charge exchange ionization at these high MS source pressures, and the inability to handle polar and nonvolatile molecules. Adaptation of LC/MS interfaces, such as the thermospray interface or the particle beam interface, to SFC/MS conditions was a step forward, but these devices have their own limitations. Alternative methods to direct SFC/MS coupling have been investigated recently. They are based on the use of packed columns rather than capillaries, and on solute ionization at atmospheric pressure rather than under a vacuum, by means of either gas-phase corona discharge ionization or liquidphase electrospray ionization. These new developments may revive research into the design of reproducible and sensitive SFC/MS systems where the number of recent studies is still low compared with other chromatography/mass spectrometry coupling studies.See [1] for part III  相似文献

Ambient desorption ionization mass spectrometry (MS) allows for the direct analysis of ordinary objects in the open atmosphere of the laboratory or in their natural environment. Analyte desorption usually accompanies the ionization step and these processes are often concerted, multi-step processes. Ambient desorption ionization methods typically require little or no sample preparation, offer a much simplified work flow and deliver unprecedented ease of use to MS analyses.

Since the introduction of desorption electrospray ionization (DESI [Z. Takats, J.M. Wiseman, B. Gologan, R.G. Cooks, Science (Washington, D. C.) 306 (2004) 471]) in 2004 and the direct analysis in real time (DART [R.B. Cody, J.A. Laramee, H.D. Durst, Anal. Chem. 77 (2005) 2297]) in 2005, this new field of MS has developed rapidly. Numerous permutations of the various options for analyte desorption and ionization have been demonstrated. Desorption steps, such as momentum transfer, dissolution into ricocheting droplets and thermal desorption, have been combined with ionization steps, including ESI, atmospheric pressure chemical ionization and photo-ionization. The large number of possible combinations of desorption and ionization components that have already been applied is creating a proliferation of techniques and acronyms that is becoming ever more complex.

Here, we provide a logical framework for the classification of these related experiments, based on the desorption and ionization processes involved in each.  相似文献

A new method was developed for the analysis of nitrate and nitrite in a variety of water matrices by using reversed-phase liquid chromatography/electrospray ionization/mass spectrometry in the negative ion mode. For this direct analysis method, nitrate and nitrite anions were well separated under the optimized LC conditions, detected by monitoring m/z 62 and m/z 46 ions, and quantitated by using an isotope dilution technique that utilized the isotopically labeled analogs. The method sensitivity, accuracy, and precision were investigated, along with matrix effects resulting from common inorganic matrix anions. The isotope dilution technique, along with sample pretreatment using barium, silver, and hydrogen cartridges, effectively compensated for the ionization suppression caused by the major water matrix anions, including chloride, sulfate, phosphate, and carbonate. The method detection limits, based on seven reagent water replicates fortified at 0.01 mg N/L nitrate and 0.1 mg N/L nitrite, were 0.001 mg N/L for nitrate and 0.012-0.014 mg N/L for nitrite. The mean recoveries from the replicate fortified reagent water and lab water samples containing the major water matrix anions, were 92-103% for nitrate with an imprecision (relative standard deviation, RSD) of 0.4-2.1% and 92-110% for nitrite with an RSD of 1.1-4.4%. For the analysis of nitrate and nitrite in drinking water, surface water, and groundwater samples, the obtained results were generally consistent with those obtained from the reference methods. The mean recoveries from the replicate matrix spikes were 92-123% for nitrate with an RSD of 0.6-7.7% and 105-113% for nitrite with an RSD of 0.3-1.8%.  相似文献

An assay based on protein precipitation and liquid chromatography/tandem mass spectrometry (LC-MS/MS) has been developed and validated for the quantitative analysis of lisinopril in human plasma. After the addition of enalaprilat as internal standard (IS), plasma samples were prepared by one-step protein precipitation using perchloric acid followed by an isocratic elution with 10 mm ammonium acetate buffer (pH adjusted to 5.0 with acetic acid)-methanol (70:30, v/v) on a Phenomenex Luna 5 mu C(18) (2) column. Detection was performed on a triple-quadrupole mass spectrometer utilizing an electrospray ionization (ESI) interface operating in positive ion and selected reaction monitoring (SRM) mode with the precursor to product ion transitions m/z 406 --> 246 for lisinopril and m/z 349 --> 206 for enalaprilat. Calibration curves of lisinopril in human plasma were linear (r = 0.9973-0.9998) over the concentration range 2-200 ng/mL with acceptable accuracy and precision. The limit of detection and lower limit of quantification in human plasma were 1 and 2 ng/mL, respectively. The validated LC-MS/MS method has been successfully applied to a preliminary pharmacokinetic study of lisinopril in Chinese healthy male volunteers.  相似文献

Reversed-phase TLC systems using ion-pairing reagents are described for the separation without prior derivatization of the porphyrins of rat Harderian gland. Porphyrins with R(f) values greater than protoporphyrin but smaller than coproporphyrin, representing harderoporphyrin, were labile to 1 m NaOH, which converted them to protoporphyrin, inconsistent with the established literature that harderoporphyrin is a tricarboxylic acid porphyrin. It was confirmed by HPLC/electrospray ionization MS that this 'harderoporphyrin' fraction consists of the recently characterized glycoconjugate, protoporphyrin-1-O-acyl beta-xyloside, with trace amounts of protoporphyrin-1-O-acyl beta-glucoside.  相似文献

A precise and accurate stable isotope dilution liquid chromatography/tandem mass spectrometry method for the analysis of intracellular homocysteine has been developed. An internal standard, [(2)H(8)]-homocystine, was added to cell pellets from EA.hy 926 cells grown in culture under low and high folate concentrations. D,L-dithiothreitol was used to reduce cellular homocystine to homocysteine. Cellular proteins were precipitated by the addition of formic acid in acetonitrile. After centrifugation, a portion of the supernatant was analyzed by liquid chromatography/tandem mass spectrometry. Using a Supelcosil cyano column with an Applied Biosystems API 4000 triple quadrupole mass spectrometer, the SRM transitions for homocysteine (m/z 136 to m/z 90) and [(2)H(4)]-homocysteine (m/z 140 to m/z 94) were monitored. The method was validated by conducting five replicate analyses on three different days at four different concentrations (concentrations at the lower limit of quantitation and expected lower quartile, mid-range and upper quartile). The limit of detection was 2 ng/10(6) EA.hy 926 cells. Using this method, the intracellular homocysteine concentration in EA.hy 926 cells ranged from 10 to 36 ng/10(6) cells.  相似文献

Combining electrochemical conversion, liquid chromatography and electrospray ionization mass spectrometry (EC/LC/ESI-MS) on-line allows the rapid identification of possible oxidation products of clozapine (CLZ) in the absence and in the presence of glutathione. CLZ is, depending on the applied potential, oxidized to various products in an electrochemical flow-through cell using a porous glassy carbon working electrode. Several hydroxylated and demethylated species are detected on-line using LC/MS. While hydroxy-CLZ is most abundant at a potential of 400 mV, demethylation occurs more readily at higher potentials (at around 700 mV versus Pd/H2 reference). In the presence of glutathione (GSH), various isomeric glutathione adducts and respective products of further oxidation can be identified. The thioadducts are characterized by tandem MS. Mono-GSH and bis-GSH derivatives can be seen in the chromatograms. The results correlate well with the cyclic voltammetric profile of CLZ. The data are relevant from a pharmacological point of view, since similar metabolites (phases I and II) have been reported in the literature. The EC/LC/MS and EC/MS methods should be valuable tools that can be used to anticipate and understand the metabolization patterns of molecules of pharmacological interest and to point out reactive intermediates.  相似文献

薄层色谱(TLC)是一类非常实用的液相色谱方法,由于其装置简单、操作便捷、灵活、通量高、成本低,以及样品前处理简单等优点,在许多行业的检测中都有广泛的应用并扮演着重要的角色。随着现代检测技术的不断发展以及各种检测技术综合应用程度的加深,薄层色谱与质谱的联用(TLC-MS)也成为这一方法的重要发展方向。随着我国医药、食品、科学仪器等事业的不断发展和升级,相信薄层色谱-质谱联用技术可以起到更好的作用,并迎来发展的契机。该综述将目前薄层色谱-质谱的联用形式分为3类,一是接口仪器的间接联用,二是质谱对薄层板的原位检测,三是质谱对薄层分离过程的实时监测,并按此分类对典型的联用形式进行了总结和简要描述。随着薄层色谱-生物自显影技术的广泛使用,薄层色谱与质谱联用的技术方法极大地提高了食品、药用生物活性物质的研发效率。目前,薄层色谱与质谱联用发展的主要瓶颈是“即插即用”型部件的设计和商品化。具有实时监测功能,同时又兼备灵活扫描功能和高通量特点的TLC-MS技术也很令人期待。此外,不同种类TLC-MS解吸-电离技术的对比研究也是有待讨论的应用问题。  相似文献

A coupled capillary column system was developed for the qualitative and quantitative determination of melamine with isotope internal standard in dairy products by gas chromatography/mass spectrometry (GC/MS) without derivatization. A 30 m of DB-5ms ((5%-phenyl)-methylpolysiloxane, 0.25 mm i.d., 0.25 μm df) coupled with a 1.5 m of Innowax (polyethylene glycol, 0.32 mm i.d., 0.25 μm df) by a quartz capillary column connector was introduced as separation column. Three advantages were discussed for the coupled system. The sample was fortified with a ring-labeled 13C315N3-melamine as an isotope internal standard and extracted by 1% of trichloroacetic acid aqueous solution. 2.2% of lead acetate solution was then added to deposit protein in the sample matrix. After purification by cation exchange cartridge, the sample solution was directly injected and detected by GC/MS. A six-point calibration curve ranging from 0.05 to 2 mg kg−1 of melamine in sample was used to establish instrument response. The recovery was 93.9-102% with relative standard deviation from 3.1 to 8.7% when isotope internal standard used. The calculated method detection limit was 0.01 mg kg−1.  相似文献

A benchtop SFC/MS system is described which utilizes supercritical carbon dioxide in a 50 micron diameter open tubular column interfaced directly to an unchanged commercially available benchtop capillary GC/MS system equipped with a chemical ionization (CI) source. A small amount of methane reagent gas was admitted co-axially to a capillary restrictor at the exit of the capillary chromatographic column. This make-up gas served as the CI reagent gas and appeared to optimize the sensitivity of the system while providing abundant (M+1) ions for the analytes investigated in this study. Good chromatographic intergrity was obtained for the GC/MS test compound, decafluorotriphenylphosphine (DFTPP), but the capillary restrictor appeared to cause some tailing of the ion current profiles resulting from low nanogram levels of caffeine and some fatty acid esters. Improvements in the SFC/MS capillary restrictor interface and the pumping system of the benchtop GC/MS system should increase the capability of this system for future applications.  相似文献

A method for the determination of ethyl glucuronide (EtG) in hair samples, using liquid chromatography/electrospray tandem mass spectrometry (LC/ESI-MS/MS), was developed and validated. The treatment of hair samples was as follows: to 100 mg of washed (dichloromethane followed by methanol, 1 ml each) and cut (1-2 mm) material, 700 microl of water, 20 microl of internal standard solution (pentadeuterated EtG, D(5)-EtG, 500 microg/l) and 20 microl of methanol were added. Samples were incubated at 25 degrees C overnight and then ultrasonicated for 2 h. Finally, 8 microl of the centrifuged solution (13,000 rpm) were analyzed by LC/ESI-MS/MS in negative ion mode. The surviving ions of EtG and D(5)-EtG were monitored together with the following MRM transitions: m/z 221 --> 75, m/z 221 --> 85 (EtG) and m/z 226 --> 75, m/z 226 --> 85 (D(5)-EtG). The method exhibited a mean correlation coefficient better than 0.9998 over the dynamic range (3-2000 pg/mg). The lower limit of quantification (LLOQ) and the limit of detection (LOD) were 3 and 2 pg/mg respectively. The intra- and interday precision and accuracy were studied at four different concentration levels (3, 5, 56 and 160 pg/mg) and were always better than 7% (n = 5). Matrix effects did not exceed 20%. The method was applied to several hair samples taken from autopsies of known alcoholics, from patients in withdrawal treatment, from social drinkers, from adult teetotalers and from children not exposed to ethanol, with EtG concentrations globally ranging from < or =2 to 4180 pg/mg.  相似文献

N-acetylaspartic acid (NAA) is a specific urinary marker for Canavan disease, an autosomal recessive leukodystrophy. We developed a 'dilute and shoot' stable isotope dilution liquid chromatography tandem mass spectrometry (LC-MS/MS) method for determination of NAA in urine. Deuterated internal standard d(3)-NAA was added to untreated urine and the mixture was injected into the LC-MS/MS system operated in the negative ion mode. Chromatography was carried out on a C(8) minibore column using 50% acetonitrile solution containing 0.05% formic acid at a flow rate of 0.25 mL/min. The retention time was 1.6 min and the turnaround time was 2.2 min. NAA and d(3)-NAA were analyzed in multiple reaction monitoring mode. Calibrators and quality control samples were prepared in pooled control urine. The assay was linear up to 2000 micromol/L with limit of quantification at 1 micromol/L (S/N = 12). Interassay and intraassay coefficients of variation were less than 7% and recovery at three different concentrations was 98.9-102.5%. The LC-MS/MS method for NAA as described involves no extraction and no derivatization, showed no interference and gave excellent recovery with low variability and short analytical time. The method was successfully applied for the retrospective analysis of urine from 21 Canavan disease cases.  相似文献
