Ozonations of alkoxy‐ and (acyloxy)‐substituted alkylidene‐lactams 1 and 5 or of the alkylidene‐sultams 9 and 10 proceeded by regioselective cleavage of the exocyclic C?C bonds (Schemes 1 and 2). These bonds are part of an enamide system and, therefore, possess considerable polarity as shown by 13C‐NMR spectra. As a result, the partly known maleimides 3 and 6 or the ‘sulfonimides’ 11 were obtained. Compounds 3 and 11 reacted with diazomethane to give the highly reactive bicyclic derivatives 8 and 12 , respectively. The cinnamylidene‐lactames 16a,b were converted by selective ozonolysis mainly into the formylmethylene lactames 17a,b (Scheme 3). The amino‐substituted aldehyde 20 bears a structural relationship to the lactone antibiotic basidalin 21a . The tendency of some donor‐substituted maleimides to undergo [2 + 2] cycloadditions was assessed (Scheme 4). The configuration of the photodimers 22a,b and 24a,b was established by X‐ray crystallography.  相似文献

A convenient method for the synthesis of polyester‐containing amino substitutes on the aromatic rings of the backbone has been developed. This polyester was prepared by chemoselective polyesterification of isophthalic acid with bisphenol having an amino group in the presence of the condensing agent diphenyl(2,3‐dihydro‐2‐thioxo‐3‐benzoxazolyl)phosphonate ( 1 ) and 1,5‐diazabicyclo[4,3,0]‐5‐nonene as a base. The model reactions were carried out in detail to elucidate appropriate conditions of chemoselective polyesterification. Direct polycondensation of isopthalic acid with 4,4′‐[1‐(4‐aminophenyl)ethylidene]bisphenol proceeded smoothly under mild conditions and produced the desired polyester with a number average molecular weight of 11,000 and Mw/Mn of 2.22. A reliable micellar electrokinetic chromatographic method for the determination of oxcarbazepine and its two main metabolites, 10-hydroxycarbamazepine and 10,11-trans-dihydroxy-10,11-dihydroxycarbamazepine, in human plasma was developed. The separation and determination of the analytes was achieved using a system consisting of 60 mM SDS in phosphate buffer (30 mM, pH 8.0), to which 20% (v/v) methanol was added. Separation was carried out in an uncoated fused-silica capillary with a separation voltage of 25 kV and currents typically less than 40 microA. Spectrophotometric detection was at 205 nm. Isolation of oxcarbazepine and its metabolites from plasma was accomplished by a solid-phase extraction procedure. The mean extraction yield of the analytes from plasma was higher than 94%. The linear correlation coefficients were better than 0.994 for all analytes. The limit of detection was 0.05 microg/mL, the limit of quantitation 0.15 microg/mL. The repeatability for the spiked blank plasma samples was lower than 1.9% and the intermediate precision lower than 2.1%, both expressed as RSD%. The results obtained analysing real plasma samples from epileptic patients under therapy with Tolep were satisfactory in terms of precision, accuracy and detectability.  相似文献

A wool‐palladium complex has been found to be able to catalyze the asymmetric hydration of 1‐octene to (S)‐(+)‐2‐octanol and 1‐decene to (R)‐(+)‐2‐decanol under 1 atm N2 and at 70°C. The optical yields were greatly affected by Pd content in wool‐palladium complex, reaction time and so on, when the proper conditions were selected, (S)‐(+)‐2‐octanol and (R)‐(+)‐2‐decanol could be obtained in 83.2 and 75.6%e.e. optical yield respectively. This chiral natural biopolymer‐palladium complex catalyst was very easy to prepare and could be reused several times without appreciable change in catalytic activity. The biotransformation of nodakenetin (NANI) by rat liver microsomes in vitro was investigated. Two major polar metabolites were produced by liver microsomes from phenobarbital‐pretreated rats and detected by reversed‐phase high‐performance liquid chromatography (RP‐HPLC) analysis. The chemical structures of two metabolites were firmly identified as 3′(R)‐hydroxy‐nodakenetin‐3′‐ol and 3′(S)‐hydroxy‐nodakenetin‐3′‐ol, respectively, on the basis of their 1H‐NMR, MS and optical rotation analysis. The latter was a new compound. A sensitive, selective and simple RP‐HPLC method has been developed for the simultaneous determination of NANI and its two major metabolites in rat liver microsomes. Chromatographic conditions comprise a C18 column, a mobile phase with MeOH‐H2O (40 : 60, v/v), a total run time of 40 min, and ultraviolet absorbance detection at 330 nm. In the rat heat‐inactivated liver microsomal supernatant, the lower limits of detection and quantification of metabolite I, metabolite II and NANI were 5.0, 2.0, 10.0 ng/mL and 20.0, 5.0, 50.0 ng/mL, respectively, and their calibration curves were linear over the concentration range 50–400, 20–120 and 150–24000 ng/mL, respectively. Protein hydrolysates have been used as active principles in cosmetic products conferring different properties to the final formulations, which are mostly controlled by the peptide size and its amino acid sequence. In this work, capillary electrophoresis coupled to mass spectrometry analyses were carried out in order to investigate such characteristics of protein hydrolysates. Samples of different origins (milk, soy and rice) were obtained from a local company, and were analyzed without a previous preparation step. The background electrolyte (BGE) and sheath liquid compositions were optimized for each sample. The best BGE composition (860 mmol/L formic acid--pH 1.8--in 70:30 v/v water/methanol hydro-organic solvent) was chosen based on the overall peak resolution whereas the best sheath liquid was selected based on increased sensitivity and presented different compositions to each sample (10.9-217 mmol/L formic acid in 75:25-25:75 v/v water/methanol hydro-organic solvent). Most of the putative peptides in the hydrolysate samples under investigation presented molecular masses of 1000 Da or less. De novo sequencing was carried out for some of the analytes, revealing the hydrophobicity/polarity of the peptides. Hence, the technique has proved to be an advantageous tool for the quality control of industrial protein hydrolysates.  相似文献

A new and very efficient route for the synthesis of aliphatic primary amine terminated polystyrene (PS) is reported. In contrast to most known methods, only traditional commercially available reagents are used. PS is synthesized by anionic polymerization with a lithium counter ion and the living chains are end‐capped by a hydroxyl group upon addition of ethylene oxide followed by protonation. The ω‐hydroxyl end group is tosylated and the tosylate is then reacted with sodium azide. The azide terminal group is finally reduced into primary amine. The different steps of functionalization have been fully characterized by SEC, ToF‐SIMS, FTIR, and 1H NMR. The amine content (= 98%) has been determined by acid‐base titration with perchloric acid. It clearly shows the efficiency of the synthetic method reported in this article although it is a multistep method. © 2000 John Wiley & Sons, Inc. This paper proposes a solid-phase microextraction (SPME) method for the passive sampling and determination of N,N-dimethylacetamide (DMA) and N,N-dimethylformamide (DMF) in air by capillary GC with flame ionization detection. The optimized conditions for SPME method were grab sampling, polydimethylsiloxane/divinylbenzene fiber, extraction for 90 min at 25 +/- 2 degrees C, desorption for 3 min at 220 degrees C, and relative humidity 45 +/- 2%. Under these conditions, the method presented good linearity (R = 0.996), repeatability (%RSD 2.79 and 9.85 for DMF and DMA, respectively), and detection limit (0.021 and 0.024 mg/m3 for DMF and DMA, respectively).  相似文献

A preparative high‐speed counter‐current chromatography method for isolation and purification of flavonoid compounds from Oroxylum indicum was successfully established by using ionic liquids as the modifier of the two‐phase solvent system. Two flavonoid compounds including baicalein‐7‐O‐diglucoside and baicalein‐7‐O‐glucoside were purified from the crude extract of O. indicum by using ethyl acetate–water–[C4mim][PF6] (5:5:0.2, v/v) as two‐phase solvent system. 36.4 mg of baicalein‐7‐O‐diglucoside and 60.5 mg of baicalein‐7‐O‐glucoside were obtained from 120 mg of the crude extract. Their purities were 98.7 and 99.1%, respectively, as determined by HPLC area normalization method. The chemical structures of the isolated compounds were identified by 1H‐NMR and 13C‐NMR.  相似文献

An aziridinomitosene skeleton, a basic core of mitomycin antibiotics, was straightforwardly prepared from N‐(p‐toluenesulfonyl)indole‐2‐carboxaldehyde in 16% overall yield by successive reactions of guanidinium ylide‐mediated aziridination, InCl3‐catalyzed epimerization of trans‐3‐(indol‐2‐yl)aziridine‐2‐carboxylate, leading to the cis‐derivative, and dehydrative cyclization.  相似文献

The modification of CaCO(3) crystal growth by synthetic L-Ser(PO(3)H(2)) and L-Thr(PO(3)H(2)) containing polypeptides is described. The amino acids Gly, L-Glu, L-Asp, L-Ser, L-Ala, and L-Lys induced rhombohedral calcite with a rough surface. Dipeptides, Xaa-L-Ser(PO(3)H(2)) (Xaa = Gly, L-Glu, L-Asp, L-Ser, L-Ala and L-Lys) induced vaterite crystals in the lower [Ca(2+)]. On the other hand, L-Ser(PO(3)H(2))-containing polypeptides formed spherical vaterite and fibrous calcite. The characteristic helical calcite was found in the presence of copoly[L-Ser(PO(3)H(2))(75)L-Asp(25)] or poly[L-Ser(PO(3)H(2))(3)-L-Asp]. Fibrous calcite, spherical vaterite, and helical calcite crystals were subjected to XRD and EDX analysis. XRD revealed the specific faces of these crystals. EDX spectra and surface analysis visualized the localization of the polypeptides and CaCO(3) components. Together with TEM and SAED data, we propose hypothetical growth mechanisms for the fibrous and helical calcite crystals.  相似文献

2-(2-phenyl-1H-phenanthro-[9,10-d]imidazole-1-yl)-acetic acid (PPIA) and 2-(9-acridone)-acetic acid (AAA), two novel precolumn fluorescent derivatization reagents, have been developed and compared for analysis of primary aromatic amines by high performance liquid chromatographic fluorescence detection coupled with online mass spectrometric identification. PPIA and AAA react rapidly and smoothly with the aromatic amines on the basis of a condensation reaction using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC) as dehydrating catalyst to form stable derivatives with emission wavelengths at 380 and 440 nm, respectively. Taking six primary aromatic amines (aniline, 2-methylaniline, 2-methoxyaniline, 4-methylaniline, 4-chloroaniline, and 4-bromoaniline) as testing compounds, derivatization conditions such as coupling reagent, basic catalyst, reaction temperature and time, reaction solvent, and fluorescent labeling reagent concentration have also been investigated. With the better PPIA method, chromatographic separation of derivatized aromatic amines exhibited a good baseline resolution on an RP column. At the same time, by online mass spectrometric identification with atmospheric pressure chemical ionization (APCI) source in positive ion mode, the PPIA-labeled derivatives were characterized by easy-to-interpret mass spectra due to the prominent protonated molecular ion m/z [M + H](+) and specific fragment ions (MS/MS) m/z 335 and 295. The linear range is 24.41 fmol-200.0 pmol with correlation coefficients in the range of 0.9996-0.9999, and detection limits of PPIA-labeled aromatic amines are 0.12-0.21 nmol/L (S/N = 3). Method repeatability, precision, and recovery were evaluated and the results were excellent for the efficient HPLC analysis. The most important argument, however, was the high sensitivity and ease-of-handling of the PPIA method. Preliminary experiments with wastewater samples collected from the waterspout of a paper mill and its nearby soil where pollution with aromatic amines may be expected show that the method is highly validated with little interference in the chromatogram.  相似文献

A new class of pyrazolone derivatives has been isolated in good to excellent yields from the 2:1 condensation reaction between 3‐methyl‐1‐phenyl‐5‐pyrazolone and arylaldehydes in the presence of ionic liquid [HMIM]HSO4. The compounds were characterised by their IR, NMR spectra, MS and elemental analyses. The important features of the methodology are a wide application range of substrates, higher yields and shorter reaction time.  相似文献
