Dihydropyrimidinethione podands, as well as podands containing 2-thienyl substituents have been synthesized via the multicomponent Biginelli reaction. Tuberculostatic activity of the novel podands was shown to depend on the polyether unit length. In particular, activity of dihydropyrimidinethione podands against laboratory strains of Mycobacterium tuberculosis, H37Rv, M. avium, M. terrae, and the clinical strain of multi-drug resistant M. tuberculosis (MDR), increases with a shortening of the polyether unit length.  相似文献

The basicity constants \((pK_{BH^ + } ,pK_{BH^{2 + } } )\) of 1,2-, 1,3-, and 1,4-bis(tetrazol-5-yl)benzenes and their N-tert-butyl derivatives in aqueous sulfuric acid and the dissociation constants (pK HB) of the corresponding H-complexes with p-fluorophenol in carbon tetrachloride were determined by UV and IR spectroscopy. Mono-and diprotonation of isomeric ditetrazolylbenzenes is observed in the acidity range (H 0) from ?1 to ?5 (\(pK_{BH^ + } \) ?2.5 to ?3.0; \(pK_{BH^{2 + } } \) ?3.8 to ?4.9). Introduction of a tert-butyl group into the 2-position of the heteroring almost does not affect the basicity of ditetrazolyl benzenes. Among the examined compounds, 1,2-bis(2-tert-butyltetrazol-5-yl)benzene is the strongest proton acceptor with respect to p-fluorophenol as standard proton donor, presumably due to formation of a complex with bifurcated (three-center) hydrogen bond.  相似文献

para-Substituted iron meso-triphenylcorrole derivatives [Fe(ms-p-R-Ph)3Cor] containing electron- donating (R = OMе) and electron-drawing (R = NO2) groups in phenyl rings are synthesized and characterized by 1H NMR, electronic absorption spectroscopy, and mass spectrometry. The effect of the nature of functional groups within iron complexes on the redox processes involving these complexes in water–alkaline solutions is analyzed. Electronic transitions in the ligand (Ered/ox = 0.820–0.850 V) and the metal (Ered/ox =–0.005 to–0.190 and–0.790 to–0.870 V for the Fe4+ ? Fe3+ and Fe3+ ? Fe2+ transitions, respectively) were found in the cyclic voltammograms. Iron in the synthesized complexes IIV under the conditions under study exists in the +4 oxidation state. The activity of iron complexes in electroreduction of molecular oxygen significantly depends on the nature of a substituent, increases in the series: Fe(ms-p-NO2Ph)3Cor (II) < Fe(ms-p-MeOPh)3Cor (I) < Fe(β-Br)8(ms-Ph)3Cor (IV) < Fe(ms-Ph)3Cor (II) and is caused by the fact that low-energy redox electron transitions occur in the molecules. The electrocatalytic activity of iron corroles is much higher than that of metal porphyrins with a similar structure.  相似文献

Background

The carbapenem subclass of β-lactams is among the most potent antibiotics available today. Emerging evidence shows that, unlike other subclasses of β-lactams, carbapenems bind to and inhibit non-classical transpeptidases (L,D-transpeptidases) that generate 3 → 3 linkages in bacterial peptidoglycan. The carbapenems biapenem and tebipenem exhibit therapeutically valuable potencies against Mycobacterium tuberculosis (Mtb).

Results

Here, we report the X-ray crystal structures of Mtb L,D-transpeptidase-2 (LdtMt2) complexed with biapenem or tebipenem. Despite significant variations in carbapenem sulfur side chains, biapenem and tebipenem ultimately form an identical adduct that docks to the outer cavity of LdtMt2. We propose that this common adduct is an enzyme catalyzed decomposition of the carbapenem adduct by a mechanism similar to S-conjugate elimination by β-lyases.

Conclusion

The results presented here demonstrate biapenem and tebipenem bind to the outer cavity of LdtMt2, covalently inactivate the enzyme, and subsequently degrade via an S-conjugate elimination mechanism. We discuss structure based drug design based on the findings and propose that the S-conjugate elimination can be leveraged to design novel agents to deliver and locally release antimicrobial factors to act synergistically with the carbapenem carrier.

N-Alkenylidenetrifluoromethanesulfonamides TfN=CH–CR=C(Me)R′ (R, R′ = H, Me) have been synthesized by reaction of N-sulfinyltrifluoromethanesulfonamide TfNSO with (E)-but-2-enal, (E)-2-methylbut- 2-enal, and 3-methylbut-2-enal. Despite greater stability of N-alkenylidenetrifluoromethanesulfonamides relative to their propargyl isomers TfNHCH2C≡CR, no rearrangement of the latter into the former occurs due to the presence of an acidic NH proton, which hampers formation of carbon-centered anion.  相似文献

The temperature dependence of the heat capacity C p o of the [(Me3Si)7C60]2 fullerene complex was measured for the first time using precision adiabatic vacuum calorimetry over the temperature range 6.7–340 K and high-accuracy differential scanning calorimetry at 320–635 K. For the most part, the error in the C p o values was about ±0.5%. An irreversible endothermic effect caused by the splitting of the dimeric bond between fullerene fragments and the thermal decomposition of the complex was observed at 448–570 K. The thermodynamic characteristics of this transformation were calculated and analyzed. Multifractal analysis of the low-temperature (T < 50 K) heat capacity was performed, and conclusions were drawn concerning the character of the heterodynamicity of the structure. The experimental data obtained were used to calculate the standard thermodynamic functions C p o (T), H o (T) ? H o (0), S o (T) ? S o (0), and G o (T) ? H o (0) over the temperature range from T → 0 to 445 K and estimate the standard entropy of formation of the compound from simple substances at 298.15 K. The standard thermodynamic properties of [(Me3Si)7C60]2 are compared with those of the (C60)2 dimer, the [(η6-Ph2)2Cr]+[C60]?? fulleride, and the initial C60 fullerene.  相似文献

The syntheses and crystal structures of the layered coordination polymers M(C8H8NO2)2 [M = Mn (1), Co (2), Ni (3) and Zn (4)] are described. These isostructural compounds contain centrosymmetric trans-MN2O4 octahedra as parts of infinite sheets; the ligand bonds to three adjacent metal ions in μ3-N,O,O′ mode from both its carboxylate O atoms and its amine N atom. In each case, weak intra-sheet N–H?O and C–H?O hydrogen bonds may help to consolidate the structure. The trans(O)-cis(C)-bis(pyridine-2-carboxylato)dicyanocobaltate(III) ions, [Co(Pic)2(CN)2]?, crystallize from acid medium with three water molecules per two crystallographically nonequivalent complexes whose charge is compensated by protons. One of the water molecules forms an oxonium ion (H3O+) with a proton. The other two water molecules bound to each other through a short hydrogen bond O-H…O (2.403(2) Å) and thus forming (H5O2)+ cations is another proton site. The (H3O)(H5O2)[Co(Pic)2(CN)2]2 crystals are monoclinic: a = 10.7027(7) Å, b = 25.786(1) Å, c = 11.4865(8) Å, β = 91.411(9)°, Z = 4, space group P21/n.  相似文献

The mechanism of aniline oxidation by singlet oxygen was studied by the DFT-PBE/L2 method. According to the calculations, aniline endoperoxide cannot participate in the reaction because of its energy instability. The addition of 1O2 to aniline proceeds with the simultaneous proton transfer to the oxygen molecule from the NH2 group (for the syn-approach of oxygen) or from the aromatic ring (for the anti-approach). For the syn-approach of the 1O2 molecule, the HNC6H4(H)OOH intermediate is formed, whose decomposition leads to aniline p-hydroperoxide (predominantly) or p-iminoquinone. In the case of the anti-approach, the 1O2 molecule is inserted at the C–H bond to form aniline p-hydroperoxide (H2NC6H4OOH). The decomposition of aniline p-hydroperoxide with the formation of p-aminophenol and H2O2 molecule proceeds via concerted mechanism.  相似文献

X-ray structural analysis has been performed for two complex compounds: Cu(en)2(o-HB)2H2O (I) (a = 16.873(4) Å, b = 8.713(2) Å, c = 14.803(3) Å, β = 91.15(2)°, V = 2175.8(8) Å3, C2/c, Z = 4, R(F) = 0.0263, 1516 reflections with I > 3σ (I)) and [Cu(en)2(OH2)2]2+(o-AB?)2 (II) (a = 7.488(5) Å, b = 22.122(8) Å, c = 7.856(5) Å, β = 118.77(2)°, V = 1140.7(11) Å3, P21/n, Z = 2, R(F) = 0.0432, 1684 reflections with I > 3σ(I)) synthesized under identical conditions (en is ethylenediamine, o-HB is o-hydroxybenzoate, and o-AB is o-aminobenzoate). Although the compounds were assumed to have similar structures and the Cu-Lig bond lengths and the cis and trans angles are acceptable for an octahedral structure, the geometric parameters of o-HB suggest that the copper atom has a plane square environment.  相似文献

The KPb2Cl5 and KPb2Br5 crystals are monoclinic (P21/c) with a microtwinned structure. X-ray analysis of chloride resulted in the parameters a = 8.854(2) Å, b = 7.927(2) Å, c = 12.485(3) Å; β = 90.05(3)°, dcalc = 4.78(1) g/cm3 (STOE STADI4, MoKα, 2θmax = 80°), R1 = 0.0702 for 4094 F ≥ 4 σ(F) reflections. Positional selectivity (α:β ratio) of electrophilic substitution in pyrrole, N-methylpyrrole, and N-tert-butylpyrrole was analyzed by ab initio [RHF/6-31G(d), MP2/6-31G(d)//RHF/6-31G(d)] and DFT [B3LYP/6-31G(d)] calculations of some σ-complexes derived from the substrates. The results of calculations with the use as model electrophilic species of trimethylsilyl cation [MP2/6-31G(d)//RHF/6-31G(d) and B3LYP/6-31G(d)] and SO3 molecule [B3LYP/6-31G(d)] instead of proton are fairly consistent with the experimental data, according to which trimethylsilylation of pyrrole and its N-substituted derivatives with trimethylsilyl trifluoromethanesulfonate, as well as sulfonation with pyridine-sulfur trioxide complex, gives the corresponding β-substituted products.  相似文献

Hydrated alkaline-earth metal tungstates Ba4Ca2 + x W2 ? x O12 ? 2x with perovskite structure were studied by the thermogravimetry, 1H NMR, IR, and Raman spectroscopy methods. Electrical conductivity and transfer numbers were measured with varying T, \(p_{O_2 } \) and \(p_{H_2 O} \). The solid solutions are capable of reversibly intercalating water and can exhibit high-temperature proton transport. The localization of protons on oxygen results in the appearance of energetically nonequivalent OH groups; a small fraction of protons are present in the form of H2O and H3O+.  相似文献

The subsolidus phase composition of the M2O-CdO-V2O5 systems with M = Li or Na is studied. Double orthovanadates MCdVO4 and MCd4(VO4)3 form solid solutions of composition Li1 ? 2x/3Cd x/3CdVO4 (0 ≤ x ≤ 1, orthorhombic space group Cmcm, modulation at x = 0.6) and Na3 ? 2x Cd3 + x (VO4)3 (0 ≤ x ≤ 0.10 and 0.30 ≤ x ≤ 1, orthorhombic space group Cmcm and Pn21 a or Pnma, respectively). Novel synthetic isoprenoids have been synthesized in engineered microbial hosts by evolving terpene synthase or expressing heterologous terpene synthases. Recently, the native operon, crtNaNcM derived from Planococcus sp. PAMC 21323, has isolated for potential industrial applications of C35 carotenoids. For the first time, novel C35 carotenoids (sesquarterpene) were synthesized in Corynebacterium glutamicum expressing the crtNaNcM genes. The recombinant strains accumulate various sesquarterpene including 4-apolycopene (red color), 4-aponeurosporene (yellow color), and no pigmentation, depending on the expression of the genetic elements of the crtNaNcM genes. Subsequently, the carotenoid extract from the cells harboring pCES-H36-CrtNaNcM was analyzed, resulting in significantly higher antioxidant activity than those of other strains harboring pCES-H36-CrtNcM and pCES-H36-CrtNaNc, respectively. This study will promote further engineering of C. glutamicum to increase sesquarterpene productions.  相似文献

Steric structure of fluorinated 2-arylhydrazono-3-oxo esters was studied by 1H, 19F, and 13C NMR spectroscopy and X-ray analysis. It was found that these compounds in the crystalline state and in solutions in acetone-d 6, DMSO-d 6, and CDCl3 exist as Z isomers with the ester fragment involved in intramolecular hydrogen bond with the hydrazone NH proton. Exceptions are alkyl 2-arylhydrazono-4,4-difluoro-3-oxobutanoates which exist in acetone-d 6 as mixtures of Z and E isomers, the former prevailing. Unlike fluorinated analogs, ethyl 2-(4-methylphenyl)hydrazono-3-oxobutanoate in crystal has the structure of E isomer in which intramolecular hydrogen bond is formed between the NH proton and acetyl carbonyl group. The same compound in acetone-d 6, DMSO-d 6, and CDCl3 gives rise to a mixture of Z and E isomers, the latter prevailing.  相似文献

The treatment of o-diaminobenzene with Zn ( OAc )2 · 2H2O in alcohol results in the formation of mononuclear bis(o-diaminobenzene)diacetate Zinc, Zn[C8H11N2O2]2. Its structure was determined by X-ray diffraction analysis. The complex is also characterized by elemental analysis, 1H NMR and IR. Solid solutions Bi3Nb1–yWyO7 ± δ, Bi3Nb1–yVyO7 ± δ, Bi3Nb1–yFeyO7 ± δ (y = 0.1–0.5; Δy = 0.1), and Bi3–xYxNb1–yWyO7 ± δ (x = 0.05, 0.1; y = 0–0.3; Δy = 0.1) have been studied. The homogeneity ranges of the solid solutions and crystal-chemical parameters have been determined by means of X-ray powder diffraction. The electrical conductivity of sintered samples has been studied by impedance spectroscopy. The joint introduction of yttrium and tungsten into the niobium sublattice does not lead to an increase in the conductivity of solid solutions, and the change of the dopant type has no noticeable effect on this conductivity.  相似文献
