The synthesis and thermal rearrangement of bis-allenyl thiosulfonates are described. Bis-γ,γ-disubstituted allenyl thiosulfonates have been prepared by disproportionation of the corresponding allenesulfinic acids. On heating, these compounds unexpectedly rearrange to a mixture of 1H,3H-thieno[3,4-d][1,2]oxathiine-3-oxide 8, 1H,3H-thieno[3,4-c]thiophene-2,2-dioxide 9, and 3-alkyl-4-alkenylthiophene 10. A tentative reaction mechanism involving sequential sigmatropic rearrangements and cyclizations is suggested.  相似文献

Phosphonomethylphenylalanine (Pmp, 2) has shown wide utility as a hydrolytically stable phosphtyrosyl (pTyr, 1) mimetic, particularly in Src homology 2 (SH2) domain-binding peptides. (2S,3R)-3-(4-(phosphonomethyl)phenyl)pipecolic acid (3) represents a variant of Pmp having φ and χ1 torsion angles constrained through incorporation into the piperidinyl ring structure contained within pipecolic acid. Reported herein is the enantioselective preparation of 3, in an orthogonally protected form (4) suitable for use in peptide synthesis. Stereochemistries at both the 2- and 3-positions are derived inductively from a single chiral center provided by the commercially available Evans chiral auxiliary, (4S)-4-benzyl-1,3-oxazolidin-2-one. Incorporation of 4 into a Grb2 SH2 domain-directed tripeptide (18) showed that Grb2 SH2 domain-binding affinity was reduced relative to the parent Pmp-containing tripeptide (19). Although conformational constraint did not enhance affinity in this case, novel amino acid analogue 4 may serve as a useful tool for the induction of defined phosphotyrosyl geometry in peptides directed at other signal transduction targets.  相似文献

The asymmetric total synthesis of (−)-dehydroclausenamide 1 was accomplished through a concise and efficient synthetic route employing as the key step the novel formation of cis-epoxy amide 7, which was resulted from the reaction of starting material 4 bearing an optically pure erythro vicinal diol unit with 5H-3-oxa-octafluoro pentanesulfonyl fluoride (HCF2CF2OCF2CF2SO2F) in the presence of 1,8-diazabicyclo[5.4.0]-7-undecene (DBU).  相似文献

In the search for novel antiplatelet agents, convenient and efficient methods for the preparation of 2,5-disubstituted pyridazin-3(2H)-ones are reported that utilise palladium-catalysed cross-coupling reactions. A post-coupling base-promoted isomerisation has been observed during Sonogashira alkynylation of 5-iodopyridazin-3(2H)-ones (3) with 1-phenyl-2-propyn-1-ol. Variable amounts of phthalazinones were isolated as by-products during the Heck alkenylation of 3. The usefulness of the hydroxymethyl fragment as a protecting group during the synthesis of 5-substituted pyridazin-3(2H)-ones has been validated.  相似文献

A new annelation procedure has been developed in a model system to introduce the cyclohexene ring of guanacastepene. Cyclohexenone 16 is prepared by sequential methylation and alkylation with allylic iodide 15. One-pot epoxidation, protodesilylation and hydrolysis of the THP forms dione 19, which cyclizes with NaOMe to 24 in 57% overall yield. Reduction, Mitsunobu inversion, and selective oxidation of the primary alcohol forms model 29. Phenyldimethylsilyl allylic iodide 36 is easier to make and more stable than trimethylsilyl allylic iodide 6 used by Stork.  相似文献

The synthesis and structural characterization of tetra(1H, 1H, 2H, 2H-polyfluoroalkyl)silaneswith the same or different chain lengths CnF2n+1 linked to Si (1n6) is reported.

When the synthesis was effected from chlorosilanes and fluorinated organomagnesiumor organolithium reagents, the trialkylsilanes were obtained. The last fluorinated chainwas introduced either via a fluoroalkyllithium reagent or by hydrosilylation of thetrialkylsilanes.

Some properties and characterization by 1H, 19F and 29Si NMR spectroscopy of the1H, 1H, 2H, 2H-polyfluoroalkylsilanes are described.  相似文献

Two novel sesquiterpenes, styxone A (1) and styxone B (2), and a novel lactone, styxlactone (3), were isolated from the Jamaican sponge Myrmekioderma styx. Their structures were elucidated by detailed 1H, 13C and 2D NMR data and the absolute stereochemistry of styxone A and B was determined by CD spectra. Styxone A represents a novel sesquiterpene skeleton. (S)-(+)-Curcuphenol (4), (S)-(+)-curcudiol (5), and abolene (6) were also isolated. An activity enhancement by cyanthiwigin B (7) to curcuphenol was observed in the antimicrobial assays when the two compounds were administered together.  相似文献

Two benzo[b]oxepine derivatives, edulisones A (1) and B (2), were isolated from the bark of Aglaia edulis, collected in Indonesia. The relative stereochemistry of 1 was determined by single-crystal X-ray diffraction analysis. Treatment of compounds 1 and 2 with lithium hydroxide produced the same hydrolysis products, 1a and 1b, as a result of cleavage of the pyrrolidine ring to an alkylated amide mixture in each case, which demonstrated that these substances are epimeric at their 2-aminopyrrolidine moiety.  相似文献

4-tert-Butyl-2-cyclohexen-1-one dimerizes in THF solution via its kinetic enolate, leading to di-tert-butylditwistane 8 in up to 36% yield (−78 °C → room temp., protonolysis, flash chromatography). X-ray crystallography shows that 8 incorporates one R and one S enantiomer of the starting ketone; none of the diastereomeric ditwistanes epi-8, epi’-8 or iso-8 was isolated. This means that the formation of 8 proceeds with mutual kinetic resolution and 100% induced diastereoselectivity.  相似文献

The reaction of N-(3,4-dichlorophenethyl)-N-methylamine (1) with 3-chloromethyl-5-phenyl-1,2,4-oxadiazole (2) was investigated. Employment of an equimolar amount of 1 and 2 in the presence of potassium carbonate led to the expected tertiary amine 3 (N-[(3,4-dichlorophenyl)ethyl]-N-methyl-N-[(5-phenyl-1,2,4-oxadiazol-3-yl)methyl]amine), whereas an excess of 1 and prolonged reaction time resulted in ring fission of the oxadiazole system in 3 and finally in the formation of N′-benzoyl-N-[(3,4-dichlorophenyl)ethyl]-N-methylguanidine (4) and N,N′-bis[(3,4-dichlorophenyl)ethyl]-N,N′-dimethylmethanediamine (5). The structures of products 3–5 were determined by means of 1H and 13C NMR-spectroscopy, mass spectrometry and IR-spectroscopy, for 3 (as picrate) and 4 also X-ray structure analysis was employed. A possible mechanism of the reaction pathway leading to compounds 4 and 5 is proposed.  相似文献

3-Arylsydnone-4-carbohydroximic acid chlorides (1) could react with N-arylmaleimides (3a–b) or 2-methyl-N-phenylmale-imide (3c) to give 3-(3-arylsydnon-4-yl)-5-aryl-3a,6a-dihydro-pyrrolo[3,4-d]isoxazole-4,6-diones (4a–h) or 6a-methyl-3-(3-arylsydnon-4-yl)-5-phenyl-3a,6a-dihydro-pyrrolo[3,4-d]isoxazole-4,6-diones (4i–l), respectively. However, 3-(arylsydnon-4-yl)-naphtho[2,3-d]isoxazole-4,9-diones (6a–d) were obtained in good yield by the reaction of carbohydroximic acid chlorides 1 with [1,4]naphthoquinone. Furthermore, 2-(3-arylsydnon-4-yl)benzoxazoles (9a–d) and 2-(3-arylsydnon-4-yl)benzothiazoles (9e–h) were obtained via the reaction of carbohydroximic acid chlorides 1 with ortho-substituted aromatic amines 7a and b.  相似文献

The transformation of the benzanilides 1 into 4-arylisochroman-3-acetic acids 8 applying the following sequence of reactions is described. At first, the 3-arylphthalides 3 were obtained via metallation [n-BuLi] of benzanilides 1 and subsequent treatment of the generated bis-lithiated anilides 2 with aromatic aldehydes. Next, the 3-arylphthalides 3 were reduced [LiBH4] to phthalanes 5 and then, via reductive metallation [Li/C10H8] and reaction of the generated bis-lithiated species 6 with dimethylformamide, 3-hydroxy-4-arylisochromans 7 were produced. In the final step the isochromans 7 were treated with 1-methoxy-1-trimethylsilyloxyethene in the presence of titanium tetrachloride and furnished 4-arylisochromans-3-acetic acid methyl esters 8 as trans stereoisomers (Ψ-e/e).  相似文献

The photochemical reactivity of cis- and trans-2-(p-carboxybenzyl)-2,6-diphenyl-6-vinylcyclohexanone, cis-1 and trans-1, was investigated in solution and in the crystalline solid state. Photochemical decarbonylation in solution proceeded in excellent yields to give cis- and trans-1-(p-carboxybenzyl)-1,2-diphenyl-2-vinylcyclopentanes cis-2 and trans-2 along with 3-(p-carboxybenzyl)-1,3-diphenylcycloheptene 3. Reactions in crystals were suppressed by a stereospecific quenching interaction between the benzyl substituent and the carbonyl oxygen in the crystalline ketone.  相似文献

A total synthesis of the marine sesquiterpene helianane 1 is described involving the thermal rearrangement of the benzoxabicyclo[4.2.0]octenone 4 to generate the dienone 5 incorporating the benzoxocane ring system of 1. This dienone was converted to the key ketone 11, which on interaction with methylmagnesium iodide followed by hydrogenation of the resulting alkene 18 furnished helianane 1.  相似文献

A crystalline dipeptide, (R)-phenylglycyl-(R)-phenylglycine (RR-1), recognized p-halobenzyl methyl sulfoxides with high R-enantioselectivity (86–99% ee) to form inclusion compounds. The single-crystal X-ray analyses showed that RR-1 molecules are arranged in parallel and zigzags via hydrogen bonding to construct a pleated sheet. The guest molecules that form hydrogen bond with +NH3 of RR-1 are accommodated in the channel cavity between the layers. In contrast to the inclusion crystals of parent benzyl methyl sulfoxide, in which a rectangular cavity is formed, the cavity including p-halobenzyl methyl sulfoxides becomes rhomboidal. We also examine the guest exchange in these inclusion compounds and it was found that the guest exchanges occur when the host structure changes.  相似文献

The simple synthesis in nearly quantitative yield of dimesitylneopentylsilene 3, the first stable silene in the neopentyl series, was performed using t-butyllithium and dimesitylvinyifiuorosilane. 3 was isolated by crystallization from pentane and characterized by 13C (doubly bonded carbon at +110.4 ppm) and 29Si (+77.6 ppm) NMR spectroscopy and analysis.  相似文献

Enantioenriched, diastereomerically pure (2-carbamoyloxy-1-alkenyl)cyclopropanes 22 are easily prepared via deprotonation of different allyl carbamates with n-butyllithium and (−)-sparteine (4). The mechanism of the cyclization reaction was determined and several substituted (S)-configured vinylcyclopropanes 32 were synthesized by two different methods. The configurational stability of the intermediate lithiated allyl carbamates and the half-time of epimerization were investigated in a series of silylation experiments, achieving up to 90% ee in the kinetically controlled enantiotopos-differentiating deprotonation.  相似文献

Tripodal aza crown ether calix[4]arenes, 5a, 5b, 6a and 6b, have been synthesized. The structure of protonated 5a was elucidated by X-ray crystallography to be a self-threaded rotaxane. Complexation studies of 5a and 5b towards anions using Na+ as countercation were carried out by 1H NMR titration in dimethylsulfoxide-d6 and the mixture of chloroform-d and methanol-d4, respectively. Ligands 5a and 5b were able to form 1:1 complexes with Br, I and NO3 and the complexation stability varied as follows: NO3>I>Br. The effect of countercation on anion complexation was also investigated. The results showed that the association constants of 5a towards Br in the presence of various cations varied as K+>Bu4N+>Na+. The enhancement in anion complexation ability of 5a may result from the rearrangement of the tripodal ammonium unit in the presence of K+. The neutral forms, 6a and 6b, were able to form complexes with transition metal ions such as Co2+, Ni2+, Cu2+ and Zn2+. The stability of the complexes followed the sequence: Ni2+2+Cu2+Zn2+. Compounds 6a and 6b may, therefore, potentially be used as either transition metal ion or anion receptors that can be controlled by pH of the solution.  相似文献

The rare earth(III) salt catalysed asymmetric Diels–Alder reaction of cyclopentadiene with a chiral dienophile in supercritical carbon dioxide (scCO2) proceeded rapidly to give the adduct with a higher diastereoselectivity than that in dichloromethane; Yb(ClO4)3 gave the endo adduct with value up to 77% de at 40°C, 8 MPa. The chiral rare earth diketonate catalyzed hetero Diels–Alder reaction of the Danishefsky's diene with benzaldehyde gave a higher yield and an enantioselectivity in scCO2 than that in dichloromethane. Scandium/pybox 8a complex catalysed asymmetric Diels–Alder reaction of 3-crotonoyl-2-oxazolidinone with cyclopentadiene in the presence of MS4A proceeded smoothly in scCO2 to give the endo adduct 10 in a good yield with up to 88% ee.  相似文献
