The entomopathogenic hyphomycete Metarhizium anisopliae has been used in programs of agricultural pest and disease vector control in several countries. Exposure to simulated solar radiation for a few hours can completely inactivate the conidia of the fungus. In the present study we determined the effect of exposures to full-spectrum sunlight and to solar ultraviolet A radiation at 320-400 nm (UVA) on the conidial culturability and germination of three M. anisopliae strains. The exposures were performed in July and August 2000 in Logan, UT. The strains showed wide variation in tolerance when exposed to full-spectrum sunlight as well as to UVA sunlight. Four-hour exposures to full-spectrum sunlight reduced the relative culturability by approximately 30% for strain ARSEF 324 and by 100% for strains ARSEF 23 and 2575. The relative UV sensitivity of the two more sensitive strains was different under solar UV from that under ultraviolet B radiation at 280-320 nm (UVB) in the laboratory. Four-hour exposures to solar UVA reduced the relative culturability by 10% for strain ARSEF 324, 40% for strain ARSEF 23 and 60% for strain ARSEF 2575. Exposures to both full-spectrum sunlight and UVA sunlight delayed the germination of the surviving conidia of all three strains. These results, in addition to confirming the deleterious effects of UVB, clearly demonstrate the negative effects of UVA sunlight on the survival and germination of M. anisopliae conidia under natural conditions. The negative effects of UVA in sunlight also emphasize that the biological spectral weighting functions for this fungus must not neglect the UVA wavelengths.  相似文献

The importance of conidial pigmentation to solar UV radiation tolerance in the entomopathogenic fungus Metarhizium anisopliae var. anisopliae, was estimated by comparing the effects of exposure to simulated solar UV radiation on the wild-type parent strain U.S. Department of Agriculture (USDA)-Agricultural Research Service (ARS) Collection of Entomopathogenic Fungal Cultures (ARSEF) 23, which has dark green conidia, and three groups of color mutants with yellow, purple and white conidia. The comparisons included inactivation levels and the kinetics of germination of conidia exposed or not exposed to simulated solar UV radiation. In addition to significantly inactivating the conidia of different mutants, exposure to radiation delayed for several hours the germination of surviving conidia of the wild type and all mutants. In general, mutants with white conidia were more sensitive to simulated solar UV radiation than mutants with purple conidia, which were more sensitive than mutants with yellow conidia, which in turn were more sensitive than the green wild strain. A significant variation in tolerance to simulated solar radiation was observed among mutants within each color group, particularly among mutants with yellow conidia. Revertants with green conidia, DWR 179 and DWR 176, were obtained from the very sensitive UV mutants DWR 148 (yellow conidia) and DWR 149 (purple conidia), respectively. These revertants had levels of tolerance to simulated solar UV radiation similar to those of the wild-type ARSEF 23. This observation is strong evidence of the importance of green conidial pigmentation for tolerance to simulated solar UV radiation, a factor that could be manipulated to produce M. anisopliae strains with more tolerance to solar UV radiation.  相似文献

The sensitivity of six mammalian cell strains to either germicidal (254 nm) or artificial "solar" simulated radiation was tested. The solar simulator used had an output similar, in some respects, to natural sunlight. Cellular capacity for Herpes simplex virus production was used as the assay procedure. The tested cells were a strain of African green monkey kidney cells and five human skin fibroblast cell strains. The latter included a "normal" cell strain, and four photosensitive cell strains; three of which were strains of xeroderma pigmentosum cells, and one strain of Bloom's syndrome cells. When comparing the D10 values, the different cell strains varied by a factor of six in response to germicidal radiation, but only by a factor of two to artificial "solar" simulated radiation. The relative sensitivity of the cells to either type of radiation also varied from 1.7 to 10.9. Large variations in response occurred even among the xeroderma pigmentosum cell strains. These responses suggest that mammalian cell sensitivity to 254 nm radiation may not be a true indicator of a cell's responses to natural sunlight.  相似文献

This study determined the sensitivity of vaccinia virus, an orthopox virus commonly used as a surrogate for variola virus (etiological agent of smallpox), exposed to UVB radiation emitted by a solar simulator, or to direct natural sunlight. The data obtained indicate that: (1) the virucidal effect of natural sunlight can be mimicked adequately by an artificial light source with similar spectral characteristics in the UVB, (2) viral sensitivity to UVB or to solar radiation can be correlated with experimental data previously obtained with UVC, (3) the correlation factor between virus inactivation by solar radiation (measured at 300 ± 5 nm) and by UVC (254 nm) is between 33 and 60, and (4) the sensitivity of viruses either dry on glass surfaces or in liquid suspension is similar when in the presence of similar amounts of cellular debris and growth media. The findings reported in this study should assist in estimating the threat posed by the persistence of virus during epidemics or after an accidental or intentional release.  相似文献

We examined the response of pico-phytoplankton communities sampled at the equatorial, tropical and temperate Central Atlantic Ocean to subsurface underwater solar radiation in order to test the generality of the reported cell mortality for these populations when exposed to high ultra violet radiation (UVR) and photosynthetically active radiation. The natural communities of pico-phytoplankton populations tested experienced high cell mortality when exposed to high solar radiation, despite inhabiting tropical waters. Synechococcus and eukaryotes were more resistant to solar radiation than Prochlorococcus. The decay rates of all pico-phytoplankton groups examined tended to be much higher when exposed to total solar radiation than when UVB-R was filtered out. We also show that even short exposures of 30 min to high solar radiation were able to induce cell mortality in Prochlorococcus. The variability in the decay rates of living Prochlorococcus cells were strongly related to the condition of the original population. However, Synechococcus decay rates were higher in populations from the tropical area, with eukaryotes sensitivity increasing with increasing the trophic degree. The data reported in this study and in the literature revealed contrasting capacities of Prochlorococcus, Synechococcus and eukaryotes to survive under high solar radiation. Although the mechanisms involved are as yet unclear, their elucidation may help explain niche partitioning among these organisms in the ocean.  相似文献

Studies were carried out on the effects induced by the main components of airborne particulate matter (APM) as soot, inorganic and cyclohexane soluble substances and by glass fiber filters on photodegradation of pyrene, benz(a)anthracene and benzo(a)pyrene exposed to UV and solar radiation. In both experimental models tested hydrocarbons showed a higher photochemical stability when absorbed on APM. Inorganic components of APM slightly enhance UV photodegradation. In real condition (outdoor exposure to solar radiation) PAH half lives generally showed a good linear correlation with mean solar radiation intensity; only degradation rate of benzo(a)pyrene on APM, exposed to sunlight was practically constant. Pyrene, in particular, showed a higher degradation rate when high ozone concentrations (0.2 ppm) occurred.  相似文献

Because of its accessibility the human skin is constantly exposed to solar ultraviolet (UV) radiation. It is increasingly appreciated that exposure of the mammalian skin to UV plays a causal and decisive role in acute and chronic skin damage including the development of skin cancer. UV exposure causes sunburn, pigmentation, hyperplasia, immunosuppression, DNA damage, photoaging and photocarcinogenesis. To cope with constant environmental damage the skin possesses elaborate enzymatic detoxification systems. This paper briefly focuses on the effect of solar radiation, particularly UV spectrum, on detoxification pathways in the skin. Specifically the effect of solar radiation on cytochrome P450, glutathione, superoxide dismutase, glutathione peroxidase, catalase, glutathione-S-transferase and ceruloplasmin has been discussed.  相似文献

Abstract— The carcinogenic and melanogenic effects of a filtered metal halide source (UVASUN) that emits UV radiation in a range from 340 to 400 nm and a bank of Philips TL 09R tubes (TL 09) emitting in a range from 310 to 400 nm were studied in lightly pigmented hairless hr/hr C3H/Tif mice. Both the carcinogenic effect of the two UVA radiation sources alone and in combination with a UV source, consisting of one Philips TL 12 and five Bellarium-S SA-1–12 tubes emitting radiation somewhat similar to the UV part of the solar spectrum (SOLAR UV), were investigated. Finally, the melanogenic effect of exposure to the two UVA sources were studied. The mice were exposed to the UVA sources 30 min/ day, 5 days/week, in equal erythemogenic doses, calculated by using the Commission Internationale de 1'Eclairage human erythema action spectrum. Equal erythemogenic doses of TL 09 and UVASUN induced the same degree of skin pigmentation, but skin tumor development was enhanced in mice exposed to TL 09 compared with UVASUN ( P < 0.0005). For all but one tumor, endpoint pretreatment with TL 09 or UVASUN for 91 days did not influence tumor development during subsequent exposure to SOLAR UV radiation 10 min/day, 4 days/week. Exposure to the two UVA radiation sources after 91 days of SOLAR UV exposure significantly enhanced skin tumor development. Overall, the data on the interaction between exposure to the UVA sources and SOLAR UV indicated that the risk of SOLAR UV-induced carcinogenesis was independent of the type of prior-UVA exposure and post-UVA exposure.  相似文献

Solar radiation is one of the major factors responsible for the control of fungus populations in the environment. Inactivation by UVA and UVB radiation is especially important for the control of fungi that disperse infective units through the air, including fungi such as Cryptococcus spp. that infect their vertebrate hosts by inhalation. Cryptococcus neoformans produces melanin in the presence of certain exogenous substrates such as l-3,4 dihydroxyphenylalanine and melanization may protect the fungus against biotic and abiotic environmental factors. In the present study, we investigated the effect of exposure to an UVB irradiance of 1000 mW m(-2) (biologically effective weighted irradiance) on the survival of melanized and nonmelanized cells of four strains of C. neoformans and four strains of C. laurentii. The relative survival (survival of cells exposed to radiation in relation to cells not exposed) of cells grown 2, 4, 6 or 8 days on medium with or without L-dopa was determined after exposure to UVB doses of 1.8 and 3.6 kJ m(-2). Both the irradiance spectrum and the intensities of those doses are environmentally realistic, and, in fact, occur routinely during summer months in temperate regions. Differences in tolerance to UVB radiation were observed between the C. neoformans and C. laurentii strains. The C. neoformans strains were more susceptible to UVB radiation than the C. laurentii strains. In C. neoformans, differences in tolerance to radiation were observed during development of both melanized and nonmelanized cells. For most treatments (strain, time of growth and UVB dose), there were virtually no differences in tolerances between melanized and nonmelanized cells, but when differences occurred they were smaller than those previously observed with UVC. In tests with two strains of C. laurentii, there was no difference in tolerance to UVB radiation between melanized and nonmelanized cells during 8 days of culture; and in tests with four strains for less culture time (4 days) there were no significant differences in tolerance between melanized and nonmelanized cells of any strain of this species.  相似文献

There is persuasive evidence that each of the three main types of skin cancer, basal cell carcinoma (BCC), squamous cell carcinoma (SCC) and melanoma, is caused by sun exposure. The incidence rate of each is higher in fairer skinned, sun-sensitive rather than darker skinned, less sun-sensitive people; risk increases with increasing ambient solar radiation; the highest densities are on the most sun exposed parts of the body and the lowest on the least exposed; and they are associated in individuals with total (mainly SCC), occupational (mainly SCC) and non-occupational or recreational sun exposure (mainly melanoma and BCC) and a history of sunburn and presence of benign sun damage in the skin. That UV radiation specifically causes these skin cancers depends on indirect inferences from the action spectrum of solar radiation for skin cancer from studies in animals and the action spectrum for dipyrimidine dimers and evidence that presumed causative mutations for skin cancer arise most commonly at dipyrimidine sites. Sun protection is essential if skin cancer incidence is to be reduced. The epidemiological data suggest that in implementing sun protection an increase in intermittency of exposure should be avoided, that sun protection will have the greatest impact if achieved as early as possible in life and that it will probably have an impact later in life, especially in those who had high childhood exposure to solar radiation.  相似文献

The filamentous rhodophytes Callithamnion gaudichaudi Agardh and Ceramium sp. were utilized to study the effects of solar radiation (PAR, 400-700 nm, UV-B, 280-315 nm and UV-A, 315-400 nm) on the photosynthetic performance in situ in Patagonia waters (Argentina). A pulse amplitude modulated (PAM) fluorometer was used to determine the fluorescence parameters. The two species grew in different habitats in the eulittoral: Ceramium sp. was found only in rock pools while C. gaudichaudii grew on exposed rocks and fell dry during low tide. Both species differed in their fluorescence parameters and their sensitivity to solar radiation exposure. The photosynthetic quantum yield had its lowest values at noon, but it recovered in the afternoon/evening hours, when irradiances were lower. PAR (irradiance of about 400 W m(-2) at noon) was responsible for most of the decrease in the yield on clear days, especially in Ceramium sp., but UVR (280-400 nm) also accounted for a significant decrease. Fluence rate response curves indicated that both species were adapted to low fluence rates and showed a pronounced non-photochemical quenching at intermediate and higher irradiances. Both species showed a rapid adaptation during measurement of fast induction kinetics but differed significantly in their fluorescence components. All photosynthetic pigments were bleached after 8 h exposure to solar radiation over a full day. Strong absorption in the UV-A range, most likely due to mycosporine-like amino acids, was detected in both strains. The pronounced sensitivity to solar radiation in situ and the recovery capacity of these two filamentous Rhodophyte species, as well as the presence of protective compounds, suggests that these algae have the ability to adapt to the relatively high radiation levels and changes in irradiance found in the Patagonia waters.  相似文献

Measurements of solar ultraviolet radiation (<320 nm) have been made with polysulphone film at a temperate and a tropical site and compared mean monthly values of the fraction of ultraviolet in solar radiation are calculated which allow an estimate of ultraviolet dose to be made from a knowledge of total solar radiation.While the data demonstrate the marked effect of sun elevation on the ultraviolet fraction of solar radiation they also indicate the effect of seasonal variation in the thickness of the ozone layer above the temperate site.It is also shown that whereas sunlight makes by far the major contribution to total solar radiation, skylight is of prime importance as regards ultraviolet radiation.  相似文献

The combination of salt water baths and solar radiation is known as an effective treatment for patients with psoriasis and atopic dermatitis. To determine whether increased susceptibility to UVB radiation may contribute to this therapeutic effect we have studied the effect of bathing the skin in salt water prior to UVB irradiation. Twelve subjects were phototested on the volar aspects of their forearms with increasing doses of UVB radiation. One forearm was exposed to 5% salt water prior to irradiation. The minimal erythema dose (MED) was determined and the erythema index and skin pigmentation were assessed by photometric measurement. The combination of salt water bath and irradiation yielded a significant decrease of the MED when compared to UVB alone (median 90 mJ/cm2 vs 130 mJ/cm2, P < 0.01). Analysis of variance showed a significant influence of salt water bath on erythema (P < 0.05) but not on skin pigmentation. Within the MED test area the erythema index of the salt water exposed forearms was elevated significantly (P < 0.05) while skin pigmentation was not affected. Thus, bathing the skin in salt water leads to a decreased threshold level for the elicitation of UVB-induced erythema and a selective increase of the erythemal response. This sensitization to the effects of shortwave UVB radiation may increase immunosuppressive effects of UVB radiation and may lead to an increased efficacy of UVB phototherapy. However, there is also an increased sunburn risk when salt water baths are followed by exposure to UV radiation.  相似文献

Our goal was to derive a quantitative factor that would allow us to predict the solar sensitivity of vegetative bacterial cells to natural solar radiation from the wealth of data collected for cells exposed to UVC (254 nm) radiation. We constructed a solar effectiveness spectrum for inactivation of vegetative bacterial cells by combining the available action spectra for vegetative cell killing in the solar range with the natural sunlight spectrum that reaches the ground. We then analyzed previous studies reporting the effects of solar radiation on vegetative bacterial cells and on bacterial spores. Although UVC-sensitive cells were also more sensitive to solar radiation, we found no absolute numerical correlation between the relative solar sensitivity of vegetative cells and their sensitivity to 254 nm radiation. The sensitivity of bacterial spores to solar exposure during both summer and winter correlated closely to their UVC sensitivity. The estimates presented here should make it possible to reasonably predict the time it would take for natural solar UV to kill bacterial spores or with a lesser degree of accuracy, vegetative bacterial cells after dispersion from an infected host or after an accidental or intentional release.  相似文献

In order to assess the short- and long-term impacts of UV radiation (UVR, 280-400nm) on the red tide alga, Heterosigma akashiwo, we exposed the cells to three different solar radiation treatments (PAB: 280-700nm, PA: 320-700nm, P: 400-700nm) under both solar and artificial radiation. A significant decrease in the effective quantum yield (Y) during high irradiance periods (i.e., local noon) was observed, but the cells partially recovered during the evening hours. Exposure to high irradiances for 15, 30, and 60min under a solar simulator followed by the recovery (8h) under dark, 9 and 100micromolphotonsm(-2)s(-1) of PAR, highlighted the importance of the irradiance level during the recovery period. Regardless the radiation treatments, the highest recovery (both in rate and total Y) was found at a PAR irradiance of 9micromolphotonsm(-2)s(-1), while the lowest was observed at 100micromolphotonsm(-2)s(-1). In all experiments, PAR was responsible for most of the observed inhibition; nevertheless, the cells exposed only to PAR had the highest recovery in any condition, as compared to the other radiation treatments. In long-term experiments (10 days) using semi-continuous cultures, there was a significant increase of UV-absorbing compounds (UV(abc)) per cell from 1.2 to >4x10(-6)microgUV(abc)cell(-1) during the first 3-5 days of exposure to solar radiation. The highest concentration of UV(abc) was found in samples exposed in the PAB as compared to PA and P treatments. Growth rates (mu) mimic the behavior of UV-absorbing compounds, and during the first 5 days mu increased from <0.2 to ca. 0.8, and stayed relatively constant at this value during the rest of the experiment. The inhibition of the Y decreased with increasing acclimation of cells. All our data indicates that H. akashiwo is a sensitive species, but was able acclimate relatively fast (3-5 days) synthesizing UV-absorbing compounds and thus reducing any impact either on photosystem II or on growth.  相似文献

The degradation of an aliphatic-aromatic biodegradable polyester film was studied under conditions of solar exposure and soil burial in a tropical area. Film samples were evaluated for changes over 40 weeks by visual examination, scanning electronic microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy, mechanical properties, molecular weight, gel content, and thermal properties. Photodegradation played a major role in the atmospheric degradation of the film, causing it to lose integrity and mechanical properties after week 8 due to main chain scission and crosslinking. SEM micrographs and FTIR spectra indicated that photodegradation started at the exposed side of the film and propagated through the polymer matrix after week 8. FTIR spectra also indicated that subsequent photooxidation processes took place. The reduction of molecular weight of the soil burial samples was much slower than that of the non-crosslinked portion of solar exposed film samples. The reduction of number average molecular weight of the non-crosslinked solar exposed samples followed a first order reaction, whereas the soil burial samples show a surface erosion biodegradation behavior. The relationship among total solar radiation, gel content and number average molecular weight indicated that an accumulated total solar radiation of 800 MJ/m2, reached in approximately 7 weeks at the exposure site, is required for PBAT mulch film integrity loss.  相似文献

Human skin is the outer covering of the body, and its composition changes with overexposure to environmental pollution and solar radiation. Infrared (IR) radiation is capable of penetrating more deeply into the skin producing free radicals causing irreversible damage. Confocal Raman spectroscopy was considered as a potential tool for the in vivo analysis of the different metabolic conditions with respect to different depths of the skin. In this regard, this work verifies the influence of infrared radiation on the skin dermis after having been exposed to 432 J cm?2 which corresponds to the dose received in a day in the summer time in a tropical region. This study was performed with 17 female volunteers who were divided into two groups. The marked skin area was exposed twice to IR radiation for a duration of 30 min each with an interval of 30 min. The spectral signatures were collected in the fingerprint region before (T0) and after 60 min (T60) of IR irradiation. The analysis shows that, on average, no significant variations occurred in group I and decreased collagen was observed in group II. However, when considering the effect seen in each individual, collagen degradation was detected in 60% of volunteers.  相似文献

Abstract—There is evidence to indicate that an increased exposure to solar radiation in the UV-B region (specifically, 290–320 nm) may occur as a result of anthropogenic degradation of stratospheric ozone. The fact that present levels of solar UV radiation can detrimentally affect marine organisms led to experiments to quantify the impact of increased UV radiation upon a marine community. Two 720–l seawater chambers (continuous flow-through design) were exposed to simulated solar UV radiation. Fluorescent sunlamps filtered by a 290 nm cutoff filter (a 0.13 mm thickness of cellulose triacetate film) were used as the radiation source. Utilization of three different weighting factors for the spectral irradiances at the surface of the chambers yielded differences of 18%, 35% and 40% in biologically effective fluence rate between the two chambers. Analysis of attached forms of algae at various depths demonstrated that a surface exposure of 1.4W/m2 in the 290–315nm waveband as contrasted with the chamber receiving a surface exposure of 1.0W/m2 resulted in depressed Chl a concentrations, reduced biomass, increased autotrophic indices, and decreased community diversity. These results indicate a potential for adverse effects of increased solar UV-8 radiation: decreased community diversity, community structure shifts, and decreased productivity.  相似文献

Experiments (6-8 days) were conducted during late summer, late fall and late winter, 2003 with waters collected off Bahía Nueva, Chubut, Argentina (42.7 degrees S, 65 degrees W) to determine the combined effects of solar ultraviolet radiation (UVR, 280-400 nm) and nutrient addition on phytoplankton communities. Samples were put in UVR-transparent containers and incubated under two radiation treatments: (a) Samples exposed to full solar radiation (PAB treatment, 280-400 nm) and (b) Samples exposed only to PAR (PAR treatment, 400-700 nm). At the beginning of the experiments, nutrients (i.e., NaPO(4)H(2) and NaNO(3)) were added to one set of samples from each radiation treatment (N cultures) whereas in the other set, nutrients remained at the concentration of the seawater. Chlorophyll a, biomass, UV-absorbing compounds and taxonomic composition were recorded throughout the experiments. N cultures always had significantly higher growth rates (P<0.05) than that in non-enriched cultures. At the beginning of experiments, phytoplankton communities were generally dominated by monads and flagellates but by the end, diatoms comprised the bulk of biomass, with only one to four taxa dominating, suggesting a selection towards more tolerant/less sensitive species. Over the experimental time frame, the observed taxonomic changes were mostly due to nutrient availability, and to a lesser extent to solar UVR exposure. Overall, the results indicate that environmental conditions (i.e., light history, nutrient concentration) together with the physiological status of the cells play a very important role at the time to assess the combined effect of nutrient addition and solar radiation on coastal phytoplankton assemblages from Patagonia.  相似文献

Ultraviolet‐B radiation (280–320 nm) has long been associated with the inactivation of microorganisms in the natural environment. Determination of the environmental inactivation kinetics of specific indicator organisms [used as tools in the field of microbial source tracking (MST)] is fundamental to their successful deployment, particularly in geographic regions subject to high levels of solar radiation. Phage infecting Bacteroides fragilis host strain GB124 (B124 phage) have been demonstrated to be highly specific indicators of human fecal contamination, but to date, little is known about their susceptibility to UV‐B radiation. Therefore, B124 phage (= 7) isolated from municipal wastewater effluent, were irradiated in a controlled laboratory environment using UV‐B collimated beam experiments. All B124 phage suspensions possessed highly similar first order log‐linear inactivation profiles and the mean fluence required to inactivate phage by 4 ? log10 was 320 mJ cm?2. These findings suggest that phage infecting GB124 are likely to be inactivated when exposed to the levels of UV‐B solar radiation experienced in a variety of environmental settings. As such, this may limit the utility of such methods for determining more remote inputs of fecal contamination in areas subject to high levels of solar radiation.  相似文献
