The possibility of bringing consistency to the Δr G, Δr H, and Δr S values for the same chemical reaction was found. The conditions were determined and the procedure for bank data processing suggested when the sought value could be calculated by several methods. Aspects of the current status of and research in analytical chemistry are briefly discussed and the need for cost effective analytical procedures is emphasized. The present and future of a number of aspects of flow-injection analysis are considered. These include the basic theory, the kinetic features, the control features, time-based methodology, and the coupling of sample pretreatment with instrumentation. Several aspects of this latter topic are considered with particular reference to the flow-injection atomic spectrometry combination. Problems of kinetic mismatch between chemistry, manifold residence, and instrument operation are discussed and some possible solutions proposed including the use of closed loop manifolds.  相似文献

Arborescent polymers are characterized by a dendritic, multilevel branched architecture derived from successive grafting reactions. In spite of their much larger size, these materials display properties analogous to dendrimers and hyperbranched polymers, the two other dendritic polymer families. The distinguishing features of arborescent polymers are their assembly from polymeric building blocks of uniform size and their very high molecular weights attained in few synthetic steps. This article offers an overview of the historical aspects of the development of dendrigraft polymers, starting from our initial efforts on the synthesis of arborescent polystyrenes. Major subsequent developments in the synthetic techniques from our and other research groups allowing the synthesis of dendrigraft copolymers, tailoring of the structural characteristics of the molecules, and further simplifications to their synthesis are also reviewed, with emphasis over the broad range of architectures attainable in these systems. © 2007 Wiley Periodicals, Inc. We present an overview of the current status of experimental, theoretical, molecular dynamics (MD), and density functional theory (DFT) studies of argon vapor-to-liquid nucleation. Since the experimental temperature-supersaturation domain does not overlap with the corresponding MD and DFT domains, separate comparisons have been made: theory versus experiment and theory versus MD and DFT. Three general theoretical models are discussed: Classical nucleation theory (CNT), mean-field kinetic nucleation theory (MKNT), and extended modified liquid drop model-dynamical nucleation theory (EMLD-DNT). The comparisons are carried out for the area below the MKNT pseudospinodal line. The agreement for the nucleation rate between the nonclassical models and the MD simulations is very good--within 1-2 orders of magnitude--while the CNT deviates from simulations by about 3-5 orders of magnitude. Perfect agreement is demonstrated between DFT results and predictions of MKNT (within one order of magnitude), whereas CNT and EMLD-DNT show approximately the same deviation of about 3-5 orders of magnitude. At the same time the agreement between all theoretical models and experiment remains poor--4-8 orders of magnitude for MKNT, 12-14 orders for EMLD-DNT, and up to 26 orders for CNT. We discuss possible reasons for this discrepancy and the ways to carry out experiment and simulations within the common temperature-supersaturation domain in order to produce a unified picture of argon nucleation.  相似文献

The biosynthesis of human blood group B antigens is accomplished by a highly specific galactosyltransferase (GTB). On the basis of NMR experiments, we propose a "molecular tweezers mechanism" that accounts for the exquisite stereoselectivity of donor substrate selection. Transferred NOE experiments for the first time reveal the bioactive conformation of the donor substrate UDP-galactose (UDP-Gal) and of its enzymatically inactive analogue, UDP-glucose (UDP-Glc). Both bind to GTB in a folded conformation that is sparsely populated in solution, whereas acceptor ligands bind in a conformation that predominates in solution. The bound conformations of UDP-Gal and UDP-Glc are identical within experimental error. Therefore, GTB must discriminate between the two activated sugars on the basis of a hitherto unknown transition state that can only be formed in the case of UDP-Gal. A full relaxation and exchange matrix analysis of STD NMR experiments reveals that acceptor substrates dissociate significantly faster (k(off) > 100 Hz) from the binding pocket than donor substrates (k(off) approximately 10 Hz). STD NMR experiments also directly show that proper recognition of the hexopyranose rings of the UDP sugars requires bivalent metal cations. At the same time, this analysis furnishes the complete three-dimensional structure of the enzyme with its bound donor substrate UDP-Gal on the basis of a prior crystal structure analysis. We propose that, upon acceptor binding, GTB uses the Asp 302 and Glu 303 side chains as "molecular tweezers" to promote bound UDP-Gal but not UDP-Glc into a transition state that leads to product formation.  相似文献

Fluorescence correlation spectroscopy has been increasingly used in polymer science. In the present perspective, the principles of the method are briefly reviewed, and the temporal and spatial resolutions are critically discussed. Examples of recent findings are summarized, focusing on polymer solutions, environmental parameters, combination with other techniques, near-interface measurements, simulations, and modeling. A method is described for the introduction of structural diversity into the thiocarbonyl group of 6-thioguanine within support-bound, fully protected oligonucleotides via ‘on-column′ conjugation. 2′-Deoxy-6-thioguanosine with a chemically-labile trigger at its 6-thio function was incorporated at defined sites into chemically synthesized oligonucleotides. Following selective removal of the thio-protection group the support-immobilized oligonucleotides were conjugated with various groups on-column, and then deprotected and purified to produce a number of oligomers each containing a different modified base. Since the modification is accomplished on-column without affecting other functional and protecting groups in the oligomers this method is compatible with introducing structural diversity at multiple sites in DNA.  相似文献

The solid phase synthesis of the cyclic depsipeptide antibiotic lysobactin is described. The natural product was synthesized via a linear approach using mostly an Fmoc-strategy solid phase peptide synthesis (SPPS) with a single purification. A lysobactin analog has also been synthesized displaying nanomolar membrane disruption activity not seen with the natural product.  相似文献

Microglial cells play a dual role in the central nervous system as they have both neurotoxic and neuroprotective effects. Uncontrolled and excessive activation of microglia often contributes to inflammation-mediated neurodegeneration. Recently, much attention has been paid to therapeutic strategies aimed at inhibiting neurotoxic microglial activation. Pharmacological inhibitors of microglial activation are emerging as a result of such endeavors. In this review, natural products-based inhibitors of microglial activation will be reviewed. Potential neuroprotective activity of these compounds will also be discussed. Future works should focus on the discovery of novel drug targets that specifically mediate microglial neurotoxicity rather than neuroprotection. Development of new drugs based on these targets may require a better understanding of microglial biology and neuroinflammation at the molecular, cellular, and systems levels.  相似文献

We introduce a novel siloxane-terminated solubilizing group and demonstrate its effectiveness as a side chain in an isoindigo-based conjugated polymer. An average hole mobility of 2.00 cm(2) V(-1) s(-1) (with a maximum mobility of 2.48 cm(2) V(-1) s(-1)), was obtained from solution-processed thin-film transistors, one of the highest mobilities reported to date. In contrast, the reference polymer with a branched alkyl side chain gave an average hole mobility of 0.30 cm(2) V(-1) s(-1) and a maximum mobility of 0.57 cm(2) V(-1) s(-1). This is largely explained by the polymer packing: our new polymer exhibited a π-π stacking distance of 3.58 ?, while the reference polymer showed a distance of 3.76 ?.  相似文献
