We report on the preparation of reduction‐responsive amphiphilic block copolymers containing pendent p‐nitrobenzyl carbamate (pNBC)‐caged primary amine moieties by reversible addition–fragmentation chain transfer (RAFT) radical polymerization using a poly(ethylene glycol)‐based macro‐RAFT agent. The block copolymers self‐assembled to form micelles or vesicles in water, depending on the length of hydrophobic block. Triggered by a chemical reductant, sodium dithionite, the pNBC moieties decomposed through a cascade 1,6‐elimination and decarboxylation reactions to liberate primary amine groups of the linkages, resulting in the disruption of the assemblies. The reduction sensitivity of assemblies was affected by the length of hydrophobic block and the structure of amino acid‐derived linkers. Using hydrophobic dye Nile red (NR) as a model drug, the polymeric assemblies were used as nanocarriers to evaluate the potential for drug delivery. The NR‐loaded nanoparticles demonstrated a reduction‐triggered release profile. Moreover, the liberation of amine groups converted the reduction‐responsive polymer into a pH‐sensitive polymer with which an accelerated release of NR was observed by simultaneous application of reduction and pH triggers. In this work, the synthesis and characterization of novel amphiphilic diblock copolymers of poly(2‐dimethylamino ethyl methacrylate)‐b‐poly(lauryl methacrylate), PDMAEMA‐b‐PLMA, using the reversible addition‐fragmentation chain transfer (RAFT) polymerization technique, are reported. The diblocks were successfully derivatized to cationic and zwitterionic block polyelectrolytes by quaternization and sulfobetainization of the PDMAEMA block, respectively. Furthermore, their molecular and physicochemical characterization was performed by using characterization techniques such as NMR and FTIR, size exclusion chromatography, light scattering techniques, and transmission electron microscopy. The structure of the diblock micelles, their behavior, and properties in aqueous solution were investigated under the effect of pH, temperature, and ionic strength, as PDMAEMA and its derivatives are stimuli‐responsive polymers and exhibit responses to variations of at least one of these physicochemical parameters. These new families of stimuli‐responsive block copolymers respond to changes of their environment giving interesting nanostructures, behavioral motifs, and properties, rendering them useful as nanocarriers for drug delivery and gene therapy. © 2017 Wiley Periodicals, Inc. Well‐defined tertiary amine‐based pH‐responsive homopolymers and block copolymers were synthesized via reversible addition‐fragmentation chain transfer (RAFT) polymerization using 4‐cyanopentanoic acid dithiobenzoate (CPAD) as the RAFT agent for homopolymers and a poly(ethylene glycol) (PEG) macro‐RAFT agent for the block copolymers. 1H NMR and gel permeation chromatography results confirmed the successful synthesis of these homopolymers and block copolymers. Kinetics studies indicated that the formation of both the homopolymers and the block copolymers were well defined. The pKa titration experiments suggested that the homopolymers and the related block copolymers have a similar pKa. The dynamic light scattering investigation showed that all of the block copolymers underwent a sharp transition from unimers to micelles around their pKa and the hydrodynamic diameter (Dh) was not only dependent on the molecular weight but also on the composition of the block copolymers. The polymer solution of PEG‐b‐PPPDEMA formed the largest micelle compare to the PEG‐b‐PDPAEMA and PEG‐b‐PDBAEMA with a similar molecular weight. © 2015 Wiley Periodicals, Inc. J. Polym. Dipeptide diphenylalanine has attracted significant research interests because of its ability to self‐assemble into various nanostructures such as nanotubes, nanowires, and nanoribbons. In this article, we present the synthesis and self‐assembly of a novel diphenylalanine‐based homopolymer and block/random copolymers by the reversible addition–fragmentation chain transfer (RAFT) polymerization of an acrylamide having a dipeptide moiety. The RAFT polymerization of N‐acryloyl‐l ,l ‐diphenylalanine (A‐Phe‐Phe‐OH) afforded novel amino acid‐based polymers with predetermined molecular weights and relatively narrow‐molecular weight distributions. The hierarchical self‐assembled structures of poly(A‐Phe‐Phe‐OH), which involve nanorods, larger nanofiber‐like microcrystals, and fiber bundles, were characterized by atomic force microscopy (AFM), transmission electron microscopy, scanning electron microscopy, and dynamic light scattering measurements. The circular dichroic measurements of poly(A‐Phe‐Phe‐OH) revealed its characteristic chiroptical property, which is affected by the nature of the solvents and the addition of urea and salts via hydrophobic, hydrogen bonding, and electrostatic interactions. Thermo‐ and pH‐responsive block and random copolymers composed of A‐Phe‐Phe‐OH and N‐isopropylacrylamide were synthesized by RAFT polymerization, and the thermoresponsive properties and assembled structures of the resulting copolymers were investigated by AFM and turbidity measurements. © 2019 Wiley Periodicals, Inc. The azobenzene‐based amphiphilic copolymers have drawn significant attention as a kind of multi‐responsive smart materials. The demand on deeper investigation of how the azobenzene group influences the micelles formation and light‐responsive behavior on molecular level is very urgent. In this article, multi‐responsive block copolymers, poly (acrylic acid)‐block‐poly[4'‐[[(2‐Methacryloyloxy)ethyl]ethylainino]azobenzene‐co‐poly (ethylene glycol) methyl ether methacrylate] (PAA‐b‐P (AzoMA‐co‐PEGMA)), with pH‐, light‐ and reduction‐responsiveness were synthesized by the monomers of AzoMA, PEGMA and acrylic acid via reversible addition‐fragmentation chain transfer polymerization (RAFT). The amphiphilic block copolymer presented aggregation‐induced emission effect, and it was pH, light, and reduction responsive. The results showed that the micelle size decreased with the decreasing of pH within a certain range. However, the particle size of micelles increased significantly when the pH was 4. Once adding reduction agent, the micelles were disassembly. Fluorescent molecule of Nile red was selected as a hydrophobic guest molecule to study the properties of encapsulating and releasing abilities of block copolymer micelles for guest molecules. The results showed that the loading capacity of three kinds of copolymer micelles was closely related to the aggregates formed by the hydrophobic block, mainly azobenzene block. Besides, the block copolymer micelles could release a certain amount of Nile red under the irradiation of UV light, the reduction with Na2S2O4 as reductant, and the exposure to alkaline environment. The mechanism of how the different status of azobenzene group influenced the self‐assembly and multi‐responsive behavior was explored on molecular level.  相似文献

We describe herein the synthesis and self‐assembly characteristics of a doubly responsive AB diblock copolymer comprised of N‐isopropylacrylamide (NIPAM) and 4‐vinylbenzoic acid (VBZ). The AB diblock copolymer was prepared via reversible addition‐fragmentation chain transfer (RAFT) radical polymerization in DMF employing a trithiocarbonate‐based RAFT agent. PolyNIPAM was employed as the macroRAFT agent. The NIPAM homopolymerization was shown to possess all the characteristics of a controlled process, and the blocking with VBZ was judged, by size exclusion chromatography, to be essentially quantitative. The NIPAM‐VBZ block copolymer was subsequently demonstrated to be able to form normal and inverse micelles in the same aqueous solution by taking advantage of the stimuli responsive characteristics of both building blocks. Specifically, and as judged by NMR spectroscopy and dynamic light scattering, raising the temperature to 40 °C (above the lower critical solution temperature of the NIPAM block), while at pH 12 results in supramolecular self‐assembly to yield nanosized species that are composed of a hydrophobic NIPAM core stabilized by a hydrophilic VBZ corona. Conversely, lowering the solution pH to 2.0 at ambient temperature results in the formation of aggregates in which the VBZ block is now hydrophobic and in the core, stabilized by the hydrophilic NIPAM block. © 2007 Wiley Periodicals, Inc. New thermoresponsive polydisulfides of POEOMA multiblocks linked with disulfide bonds having redox‐responsive properties are reported. These POEOMA‐multisegmented polydisulfides were synthesized by a new method employing a combined RAFT/aminolysis and reversible thiol‐disulfide redox reaction that centers on the synthesis of new disulfide‐labeled difunctional RAFT agent. RAFT polymerization proceeded in living fashion, yielding well‐defined POEOMA copolymers with middle disulfides and terminal RAFT species. They were then used as precursors for thiol‐disulfide polyexchange induced by aminolysis and reductive reaction followed by oxidation: these polydisulfides with different molecular weights and end groups ex hibited tunable thermoresponsive properties and thiol‐responsive degradation.  相似文献

A series of environmentally sensitive ABA triblock copolymers with different block lengths were prepared by reversible addition‐fragmentation chain transfer (RAFT) polymerization from acrylic acid (AA) and N‐isopropylacrylamide (NIPAAm). The GPC and 1H NMR analyses demonstrated the narrow molecular weight distribution and precise chemical structure of the prepared P(AA‐b‐NIPAAm‐b‐AA) triblock copolymers owing to the controlled/living characteristics of RAFT polymerization. The lower critical solution temperature (LCST) of the triblock copolymers could be tailored by adjusting the length of PAA block and controlled by the pH value. Under heating, the triblock copolymers underwent self‐assemble in dilute aqueous solution and formed nanoparticles revealed via TEM images. Physically crosslinked nanogels induced by inter‐/intra‐hydrogen bonding or core‐shell micelle particles thus could be obtained by changing environmental conditions. With a well‐defined structure and stimuli‐responsive properties, the P(AA‐b‐NIPAAm‐b‐AA) copolymer is expected to be employed as a nanocarrier for biomedical applications in controlled‐drug delivery and targeting therapy. © 2015 Wiley Periodicals, Inc. The synthesis and self‐assembly properties in aqueous solutions of novel amphiphilic block copolymers composed of one hydrophobic poly (lauryl methacrylate), (PLMA) block and one hydrophilic poly (oligo ethylene glycol methacrylate) (POEGMA) block are reported. The block copolymers were prepared by RAFT polymerization and were molecularly characterized by size exclusion chromatography, NMR and FT‐IR spectroscopy, and DSC. The PLMA‐b‐POEGMA amphiphilic block copolymers self‐assemble in nanosized complex nanostructures resembling compound micelles when inserted in aqueous media, as supported by light scattering and TEM measurements. We report here a novel direct method for the syntheses of primary aminoalkyl methacrylamides that requires mild reagents and no protecting group chemistry. The reversible addition‐fragmentation chain transfer polymerization (RAFT) of the aminoalkyl methacrylamide revealed to be highly efficient with 4‐cyanopentanoic acid dithiobenzoate (CTP) as chain transfer agent and 4,4′‐azobis(4‐cyanovaleric acid) (ACVA) as initiator. Cationic amino‐based homopolymers of reasonably narrow polydispersities (Mw/Mn < 1.30) and predetermined molecular weights were obtained without recourse to any protecting group chemistry. A range of block and random copolymers were also synthesized via the RAFT process. The homopolymers and copolymers were characterized by aqueous conventional and triple detection gel permeation chromatography systems. Furthermore, the primary amine‐based methacrylamide monomers and polymers revealed to be highly stable both with the primary amino group in the protonated and deprotonated form. We have also demonstrated that stabilized gold nanoparticles can be generated with the RAFT‐synthesized amine‐based polymers via a photochemical process. © 2008 Wiley Periodicals, Inc. A novel POSS‐containing methacrylate monomer (HEMAPOSS) was fabricated by extending the side chain between polyhedral oligomeric silsesquioxane (POSS) unit and methacrylate group, which can efficiently decrease the steric hindrance in free‐radical polymerization of POSS‐methacrylate monomer. POSS‐containing homopolymers (PHEMAPOSS) with a higher degree of polymerization (DP) can be prepared using HEMAPOSS monomer via reversible addition–fragmentation chain transfer (RAFT) polymerization. PHEMAPOSS was further used as the macro‐RAFT agent to construct a series of amphiphilic POSS‐containing poly(N, N‐dimethylaminoethyl methacrylate) diblock copolymers, PHEMAPOSS‐b‐PDMAEMA. PHEMAPOSS‐b‐PDMAEMA block copolymers can self‐assemble into a plethora of morphologies ranging from irregular assembled aggregates to core‐shell spheres and further from complex spheres (pearl‐necklace‐liked structure) to large compound vesicles. The thermo‐ and pH‐responsive behaviors of the micelles were also investigated by dynamic laser scattering, UV spectroscopy, SEM, and TEM. The synthesis and self‐assembly properties in aqueous solutions of novel amphiphilic block copolymers composed of one hydrophilic, pH and temperature responsive poly(dimethyl amino ethyl methacrylate) (PDMAEMA) block and one weakly hydrophobic, water insoluble, potentially thermoresponsive poly(hydroxy propyl methacrylate) (PHPMA) block, are reported. The block copolymers were prepared by RAFT polymerization and were molecularly characterized by size exclusion chromatography, NMR, and FTIR spectroscopies. The PDMAEMA‐b‐PHPMA amphiphilic block copolymers self‐assemble in different nanostructured aggregates when inserted in aqueous media. The effects of different solubilization protocols, as well as the effects of solution temperature and pH on the structure of the aggregates, are studied by light scattering and fluorescence spectroscopy measurements. For the precision synthesis of primary amino functional polymers, cationic polymerization of a phthalimide‐containing vinyl ether monomer precursor, 2‐vinyloxyethyl phthalimide (PIVE), was examined using a base‐assisting initiating system. Living polymerization of PIVE in CH2Cl2 in the presence of 1,4‐dioxane as an added base yielded nearly monodispersed polymers (Mw/Mn < 1.1) and higher molecular weight polymers, which have never been obtained using other initiating systems. Furthermore, block copolymers with hydrophobic or hydrophilic groups could be prepared. The deprotection of the pendant phthalimide groups gave well‐defined pH‐responsive polymers with pendant primary amino groups. Dual‐stimuli–responsive block copolymers having a pH‐responsive polyamine segment and a thermosensitive segment self‐assembled in water in response to both pH and temperature. © 2010 Wiley Periodicals, Inc. Thiol‐responsive symmetric triblock copolymers having single disulfide linkages in the middle blocks (called mono‐cleavable block copolymers, ss‐ABP2) were synthesized by atom transfer radical polymerization in the presence of a disulfide‐labeled difunctional Br‐initiator. These brush‐like triblock copolymers consist of a hydrophobic polyacrylate block having pendent oligo(propylene oxide) and a hydrophilic polymethacrylate block having pendent oligo(ethylene oxide). Gel permeation chromatography and 1H NMR results confirmed the synthesis of well‐defined mono‐cleavable block copolymers and revealed that polymerizations were well controlled. Because of amphiphilic nature, these copolymers self‐assembled to form colloidally stable micelles above critical micellar concentration of 0.032 mg · mL−1. In response to reductive reactions, disulfides in thiol‐responsive micelles were cleaved. Atomic force microscopy and dynamic light scattering analysis suggested that the cleavage of disulfides caused dissociation of micelles to smaller‐sized assembled structures in water. Moreover, in a biomedical perspective, the mono‐cleavable block copolymer micelles are not cytotoxic and thus biocompatible.

Self‐immolative polymers (SIPs) undergo depolymerization in response to the cleavage of stimuli‐responsive end‐caps from their termini. Some classes of SIPs, including polycarbamates, have depolymerization rates that depend on environmental factors such as solvent and pH. In previous work, hydrophobic SIPs have been incorporated into amphiphilic block copolymers and used to prepare nanoassemblies. However, stimuli‐responsive hydrophilic blocks have not previously been incorporated. In this work, we synthesized amphiphilic copolymers composed of a hydrophobic polycarbamate SIP block and a hydrophilic poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) block connected by a UV light‐responsive linker end‐cap. It was hypothesized that after assembly of the block copolymers into nanoparticles, chain collapse of the PDMAEMA above its lower critical solution temperature (LCST) might change the environment of the SIP block, thereby altering its depolymerization rate. Self‐assembly of the block copolymers was performed, and the depolymerization of the resulting assemblies was studied by fluorescence spectroscopy, dynamic light scattering, and NMR spectroscopy. At 20 °C, the system exhibited a selective response to the UV light. At 65 °C, above the LCST of PDMAEMA, the systems underwent more rapid depolymerization, suggesting that the increase in rate arising from the higher temperature dominated over environmental effects arising from chain collapse. © 2018 Wiley Periodicals, Inc. A carboxylic acid based reversible additionfragmentation transfer (RAFT) agent is used to prepare gels composed of worm‐like diblock copolymers using two non‐ionic monomers, glycerol monomethacrylate (GMA) and 2‐hydroxypropyl methacrylate (HPMA). Ionization of the carboxylic acid end‐group on the PGMA stabilizer block induces a worm‐to‐sphere transition, which in turn causes immediate degelation. This morphological transition is fully reversible as determined by TEM and rheology studies and occurs because of a subtle change in the packing parameter for the copolymer chains. A control experiment where the methyl ester derivative of the RAFT agent is used to prepare the same diblock copolymer confirms that no pH‐responsive behavior occurs in this case. This end‐group ionization approach is important for the design of new pH‐responsive copolymer nano‐objects as, unlike polyacids or polybases, only a minimal amount of added base (or acid) is required to drive the morphological transition.  相似文献

A series of amphiphilic temperature‐responsive star‐shaped poly(D,L‐lactic‐co‐glycolic acid)‐b‐methoxy poly(ethylene glycol) (PLGA‐mPEG) block copolymers with different arm numbers were synthesized via the arm‐first method. Gel permeation chromatography data confirmed that star‐shaped PLGA‐mPEG copolymers had narrow polydispersity index, indicating the successful formation of star‐shaped block copolymers. Indirectly, the 1H NMR spectra in two kinds of solvents and dye solubilization method had confirmed the formation of core‐shell micelles. Further, core‐shell micelles with sizes of about 30–50 nm were directly observed by transmission electron microscopy. Subsequently, the micellar sizes and distributions as a function of concentrations and temperature were measured. At various copolymer concentrations, individual micelles with size of 20–40 nm and grouped micelles with size of 600–700 nm were found. Micellar mechanism of star‐shaped block copolymers in aqueous solution was simultaneously discussed. In addition, sol–gel transition of star‐shaped block copolymers in water was also investigated via the inverting test method. The critical gel temperature (CGT) and critical gel concentration (CGC) values of two‐arm, three‐arm and four‐arm copolymer solutions were markedly higher than ones of one‐arm copolymer. Moreover, the same CGC values of copolymer solution with different molecular weight and the same arm composition were ~15 wt %, and CGT values increased from ~38 to ~47°C with increasing arm numbers. One‐step synthesis of block‐graft copolymers by reversible addition‐fragmentation chain transfer (RAFT) and ring‐opening polymerization (ROP) by using a novel initiator was reported. Block‐graft copolymers were synthesized in one‐step by simultaneous RAFT polymerization of n‐butylmethacrylate (nBMA) and ROP of ε‐caprolacton (CL) in the presence of a novel macroinitiator (RAFT‐ROP agent). For this purpose, first epichlorohydrin (EPCH) was polymerized by using H2SO4 via cationic ring‐opening mechanism. And then a novel RAFT‐ROP agent was synthesized by the reaction of potassium ethyl xanthogenate and polyepichlorohydrin (poly‐EPCH). By using the RAFT‐ROP agent, poly[CL‐b‐EPCH‐b‐CL‐(g‐nBMA)] block‐graft copolymers were synthesized. The principal parameters such as monomer concentration, initiator concentration, and polymerization time that affect the one‐step polymerization reaction were evaluated. The block lengths of the block‐graft copolymers were calculated by using 1H‐nuclear magnetic resonance (1H NMR) spectrum. The block length could be adjusted by varying the monomer and initiator concentrations. The characterization of the products was achieved using 1H NMR, Fourier‐transform infrared spectroscopy, gel‐permeation chromatography, thermogravimetric analysis, differential scanning calorimetry, elemental analysis, and fractional precipitation (γ) techniques. © 2013 Wiley Periodicals, Inc. Poly(pentafluorophenyl acrylate)‐block‐poly(N‐isopropylacrylamide) (PPFPA‐b‐PNIPAM) is synthesized by reversible addition‐fragmentation chain transfer (RAFT) polymerization. Light‐responsive moieties of ortho‐nitrobenzyl (ONB)‐protected diamine are partially introduced to the PFPA moieties via postpolymerization modification. The amphiphilic block copolymers are assembled into micelles in water. The ONB‐protected diamine group in the micelle core is released upon UV irradiation, which subsequently induces an in situ cross‐linking by a spontaneous reaction with the remaining PFPA groups in the core and yields stable cross‐linked micelles. Micellization of the copolymers is confirmed by dynamic light scattering (DLS) and transmission electron microscopy (TEM). 4‐Nitro‐7‐piperazino‐2,1,3‐benzoxadiazole (NBD) and pyrene are loaded in the core of cross‐linked micelles to demonstrate the possibility for additional post‐functionalization of residual PFPA moieties and hydrophobic molecule encapsulation, respectively.
