We have synthesized poly(ε‐caprolactone‐co‐tert‐butyl glycidyl ether) (CL‐co‐BGE) statistical copolymers using 1‐tert‐butyl‐4,4,4‐tris(dimethylamino)‐2,2‐bis [tris(dimethylamino)phophoranylidenamino]‐2Λ5,4Λ5‐catenadi(phosphazene) (t‐BuP4) as the catalyst. The hydrolysis of the resulting polymers yields amphiphilic poly(ε‐caprolactone‐co‐glycidol) (CL‐co‐GD) copolymers. By use of the quartz crystal microbalance with dissipation (QCM‐D), we have investigated the enzymatic degradation of the copolymers. It is shown that the degradation rate increases with the content of hydrophilic (GD) units. (3‐(4,5‐Dimethylthiazol‐2‐yl)‐2,5‐diphenyl tetrazolium bromide) (MTT) assay experiments demonstrate that the CL‐co‐GD copolymers have low cytotoxicity. © 2015 Wiley Periodicals, Inc. J. Polym. Comb‐like amphiphilic poly(poly((lactic acid‐co‐glycolic acid)‐block‐poly(ethylene glycol)) methacrylate (poly((PLGA‐b‐PEG)MA)) copolymers were synthesized by radical polymerization. (PLGA‐b‐PEG)MA macromonomer was prepared by ring‐opening bulk polymerization of DL ‐lactide and glycolide using purified poly(ethylene glycol) monomethacrylate (PEGMA) as an initiator. (PLGA‐b‐PEG)MA macromonomer was copolymerized with PEGMA and/or acrylic acid (AA) by radical polymerization to produce comb‐like amphiphilic block copolymers. The molecular weight and chemical structure were investigated by GPC and 1H NMR. Poly((PLGA‐b‐PEG)MA) copolymer aqueous solutions showed gel–sol transition behavior with increasing temperature, and gel‐to‐sol transition temperature decreased as the compositions of the hydrophilic PEGMA and AA increased. The gel‐to‐sol transition temperature of the terpolymers of the poly((PLGA‐b‐PEG)MA‐co‐PEGMA‐co‐AA) also decreased when the pH was increased. The effective micelle diameter obtained from dynamic light scattering increased with increasing temperature and with increasing pH. The critical micelle concentration increased as the composition of the hydrophilic monomer component, PEGMA and AA, were increased. The spherical shape of the hyperbranched polymers in aqueous environment was observed by atomic force microscopy. © 2008 Wiley Periodicals, Inc. A series of amphiphilic cationic random copolymers, namely poly[2‐(methacryloyloxy)ethyl trimethylammonium chloride‐co‐stearyl methacrylate] or poly(MADQUAT‐co‐SMA), have been synthesized via conventional free‐radical copolymerization using 2,2′‐azobisisobutyronitrile (AIBN) as initiator and n‐dodecanethiol as chain transfer agent. The resultant products were then characterized by FT‐IR, 1H NMR, MALDI‐TOF MS measurements. From the number‐average molecular weights of the copolymers, we can conclude that these copolymers have oligomeric structure with a limited number of hydrophilic and hydrophobic moieties in a short polymer chain. The reactivity ratios (rMADQUAT = 0.83, rSMA = 0.25) between the hydrophilic MADQUAT monomer and the hydrophobic SMA monomer were calculated by the Finemann and Ross method, which was based on the results of 1H NMR analysis. The surface activity of the random copolymers was studied by the combination of surface tension and contact angle measurement, and the results indicated that these copolymers possess relatively high surface activity. The critical aggregation concentrations (cac) of the copolymers in aqueous solution were determined by fluorescence probe method as well as surface tension measurement. The different nanoparticles of poly(MADQUAT‐co‐SMA) copolymers formed in pure water or ethanol‐water mixture were proved by the particle size and size distribution in the measurement of dynamic light scattering (DLS). Furthermore, using transmission electron microscopy (TEM), we could observe various self‐assembly morphologies of these random copolymer. All these results show that the amphiphilic cationic random copolymers have a good self‐assembly behavior, even if they are ill‐defined copolymers. © 2009 Wiley Periodicals, Inc. A novel amphiphilic thermosensitive poly(ethylene glycol)45b‐poly(methyl methacrylate46co‐3‐(trimethoxysilyl)propyl methacrylate)2b‐poly(N‐isopropylacrylamide)429 (PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429) triblock copolymer was synthesized via consecutive atom transfer radical polymerization techniques. The thermoinduced association behavior of the resulting triblock copolymers in aqueous medium was further investigated in detail by 1H NMR, transmission electron microscopy, and dynamic light scattering. The results showed that at the temperature (25 °C) below the LCST, PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429 triblock copolymers self‐assembled into the core crosslinked micelles with the hydrophobic P(MMA‐co‐MPMA) block constructing a dense core, protected by the mixed soluble PEG and PNIPAAm chains acting as a hydrophilic shell simultaneously. With an increase in temperature, the resulting core‐shell micelles converted into a new type of micelles with the hydrophilic PEG chains stretching out from the hydrophobic core through the collapsed PNIPAAm shell. On the other hand, at the temperature (40 °C) above the LCST, such triblock copolymers formed the crosslinked vesicles with the hydrophobic PNIPAAm and P(MMA‐co‐MPMA) blocks constructing a membrane core and the soluble PEG chains building the hydrophilic lumen and the shell. On further decreasing the temperature, the resulting vesicles underwent transformation from the shrunken to the expanded status, leading to the formation of swollen vesicles with enlarged size. This study is believed to present the first formation of two types of hybrid crosslinked self‐assemblies by thermoinduced regulation. © 2011 Wiley Periodicals, Inc. Functional, degradable polymers were synthesized via the copolymerization of vinyl acetate (VAc) and 2‐methylene‐1,3‐dioxepane (MDO) using a macro‐xanthate CTA, poly(N‐vinylpyrrolidone), resulting in the formation of amphiphilic block copolymers of poly(NVP)‐b‐poly(MDO‐co‐VAc). The behavior of the block copolymers in water was investigated and resulted in the formation of self‐assembled nanoparticles containing a hydrophobic core and a hydrophilic corona. The size of the resultant nanoparticles was able to be tuned with variation of the hydrophilic and hydrophobic segments of the core and corona by changing the incorporation of the macro‐CTA as well as the monomer composition in the copolymers, as observed by Dynamic Light Scattering, Static Light Scattering, and Transmission Electron Microscopy analyses. The concept was further applied to a VAc derivative monomer, vinyl bromobutanoate, to incorporate further functionalities such as fluorescent dithiomaleimide groups throughout the polymer backbone using azidation and “click” chemistry as postpolymerization tools to create fluorescently labeled nanoparticles. © 2015 Wiley Periodicals, Inc. A new stratagem for the synthesis of amphiphilic graft copolymers of hydrophilic poly(ethylene oxide) as the main chain and hydrophobic polystyrene as the side chains is suggested. A poly(ethylene oxide) with pending 2,2,6,6‐tetramethylpiperidine‐1‐oxyls [poly(4‐glycidyloxy‐2,2,6,6‐tetramethylpiperidine‐1‐oxyl‐co‐ethylene oxide)] was first prepared by the anionic ring‐opening copolymerization of ethylene oxide and 4‐glycidyloxy‐2,2,6,6‐tetramethylpiperidine‐1‐oxyl, and then the graft copolymerization of styrene was completed with benzoyl peroxide as the initiator in the presence of poly(4‐glycidyloxy‐2,2,6,6‐tetramethylpiperidine‐1‐oxyl‐co‐ethylene oxide). The polymerization of styrene was under control, and comblike, amphiphilic poly(ethylene oxide)‐g‐polystyrene was obtained. The copolymer and its intermediates were characterized with size exclusion chromatography, 1H NMR, and electron spin resonance in detail. © 2006 Wiley Periodicals, Inc. Degradable, amphiphilic graft copolymers of poly(ε‐caprolactone)‐graft‐poly(ethylene oxide), PCL‐g‐PEO, were synthesized via a grafting onto strategy taking advantage of the ketones presented along the backbone of the statistical copolymer poly(ε‐caprolactone)‐co‐(2‐oxepane‐1,5‐dione), (PCL‐co‐OPD). Through the formation of stable ketoxime ether linkages, 3 kDa PEO grafts and p‐methoxybenzyl side chains were incorporated onto the polyester backbone with a high degree of fidelity and efficiency, as verified by NMR spectroscopies and GPC analysis (90% grafting efficiency in some cases). The resulting block graft copolymers displayed significant thermal differences, specifically a depression in the observed melting transition temperature, Tm, in comparison with the parent PCL and PEO polymers. These amphiphilic block graft copolymers undergo self‐assembly in aqueous solution with the P(CL‐co‐OPD‐co‐(OPD‐g‐PEO)) polymer forming spherical micelles and a P(CL‐co‐OPD‐co‐(OPD‐g‐PEO)‐co‐(OPD‐gpMeOBn)) forming cylindrical or rod‐like micelles, as observed by transmission electron microscopy and atomic force microscopy. © 2010 Wiley Periodicals, Inc. The differences in the polymerization abilities of N‐vinylformamide (NVF) and N‐vinylisobutyramide (NVIBA) and the synthesis of their copolymers were studied. The polymerization abilities were fairly good and quite similar to those of N‐vinyl‐ acetamide (NVA), a monomer in the same class as N‐vinylalkylamides. Since the monomer reactivity ratios were r1 = 1.08 and r2 = 0.92 (M1 = NVF, M2 = NVIBA), respectively, it is clear that the comonomers definitely were converted to random copolymers. The resulting copolymers poly(NVF‐co‐NVIBA) exhibited the cloud points sharply. The light transmittance profiles were the same as those for poly(NVIBA) although they increased from 39 °C for poly(NVIBA), with an increase in the corresponding hydrophilic NVF component. Our final objective was to produce a cloud point controlled polymer material with primary amino groups. To achieve this, we examined the hydrolysis of poly(NVF), poly(NVA), poly(NVIBA), and poly(NVF‐co‐NVIBA) to obtain poly(vinylamine) [poly(VAm)]. The hydrolytic cleavage of poly(NVF) and poly(NVA) was promoted by an increase in temperature. However, poly(NVIBA) was not cleaved appreciably. The hydrolysis of poly(NVF‐co‐NVIBA) was done under controlled conditions, and amino groups selectively were introduced to only one of two components of the copolymer. The cloud point of the hydrolyzed copolymer shifted to a higher temperature than that of the copolymer. © 2000 John Wiley & Sons, Inc. J Polym Sci A: Polym Chem 38: 3674–3681, 2000  相似文献

Novel biodegradable amphiphilic graft copolymers containing hydrophobic poly(ester‐carbonate) backbone and hydrophilic poly(ethylene glycol) (PEG) side chains were synthesized by a combination of ring‐opening polymerization and “click” chemistry. First, the ring‐opening copolymerization of 5,5‐dibromomethyl trimethylene carbonate (DBTC) and ε‐caprolactone (CL) was performed in the presence of stannous octanoate [Sn(Oct)2] as catalyst, resulting in poly(DBTC‐co‐CL) with pendant bromo groups. Then the pendant bromo groups were completely converted into azide form, which permitted “click” reaction with alkyne‐terminated PEG by Huisgen 1,3‐dipolar cycloadditions to give amphiphilic biodegradable graft copolymers. The graft copolymers were characterized by proton nuclear magnetic resonance (1H NMR), Fourier transform infrared spectra and gel permeation chromatography measurements, which confirmed the well‐defined graft architecture. These copolymers could self‐assemble into micelles in aqueous solution. The size and morphologies of the copolymer micelles were measured by transmission electron microscopy and dynamic light scattering, which are influenced by the length of PEG and grafting density. © 2011 Wiley Periodicals, Inc. Well‐defined amphiphilic graft copolymers containing hydrophilic poly((meth)acrylic acid) (PMAA) or poly(acrylic acid) (PAA) side chains with gradient and statistical distributions were synthesized. For this purpose, the hydroxy‐functionalized copolymers with various gradient degrees, in which 2‐(6‐hydroxyhexanoyloxy)ethyl (meth)acrylate units (caprolactone 2‐[methacryloyloxy]ethyl ester, CLMA) formed strong gradient with tert‐butyl acrylate (tBA), slight gradient copolymers with tert‐butyl (meth)acrylate (tBMA), and statistical copolymers with methyl (meth)acrylate (MMA) were modified to bromoester multifunctional macroinitiators, P(tBMA‐grad‐BrCLMA), P(BrCLMA‐grad‐tBA), and P(BrCLMA‐co‐MMA). In the next step, they were applied in controlled radical polymerization of tBMA and tBA yielding graft copolymers with various lengths of side chains as well as graft densities. Further, the tert‐butyl groups in copolymers were successfully removed via acidolysis in the presence of trifluoracetic acid, which caused transformation of the hydrophobic graft copolymers into amphiphilic ones with ability of self‐assembly for the future biomedical applications. The spontaneous hydrogel formation of a sort of biocompatible and biodegradable amphiphilic block copolymer in water was observed, and the underlying gelling mechanism was assumed. A series of ABA‐type triblock copolymers [poly(D,L ‐lactic acid‐co‐glycolic acid)‐b‐poly(ethylene glycol)‐b‐poly(D,L ‐lactic acid‐co‐glycolic acid)] and different derivatives end‐capped by small alkyl groups were synthesized, and the aqueous phase behaviors of these samples were studied. The virgin triblock copolymers and most of the derivatives exhibited a temperature‐dependent reversible sol–gel transition in water. Both the poly(D,L ‐lactic acid‐co‐glycolic acid) length and end group were found to significantly tune the gel windows in the phase diagrams, but with different behaviors. The critical micelle concentrations were much lower than the associated critical gel concentrations, and an intact micellar structure remained after gelation. A combination of various measurement techniques confirmed that the sol–gel transition with an increase in the temperature was induced not simply via the self‐assembly of amphiphilic polymer chains but also via the further hydrophobic aggregation of micelles resulting in a micelle network due to a large‐scale self‐assembly. The coarsening of the micelle network was further suggested to account for the transition from a transparent gel to an opaque gel. © 2006 Wiley Periodicals, Inc. A series of amphiphilic graft copolymers of poly (vinylidene fluoride‐co‐chlorotrifluoroethylene)‐g‐poly(2‐vinyl pyridine), P (VDF‐co‐CTFE)‐g‐P2VP, with different degrees of P2VP grafting (from 26.3 to 45.6 wt%) was synthesized via one‐pot atom transfer radical polymerization (ATRP). The amphiphilic properties of P (VDF‐co‐CTFE)‐g‐P2VP graft copolymers allowed itself to self‐assemble into nanoscale structures. P (VDF‐co‐CTFE)‐g‐P2VP graft copolymers were introduced into neat P (VDF‐co‐CTFE) as additives to form blending membranes. When two different solvents, N‐methyl‐2‐pyrrolidone (NMP) and dimethylformamide (DMF), were used, specific organized crystalline structures were observed only in the NMP systems. P (VDF‐co‐CTFE)‐g‐P2VP played a pivotal role in controlling the morphology and pore structure of membranes. The water flux of the membranes increased from 57.2 to 310.1 L m?2 h?1 bar?1 with an increase in the PVDF‐co‐CTFE‐g‐P2VP loading (from 0 to 30 wt%) due to increased porosity and hydrophilicity. The flux recovery ratio (FRR) increased from 67.03% to 87.18%, and the irreversible fouling (Rir) decreased from 32.97% to 12.82%. Moreover, the pure gas permeance of the membranes with respect to N2 was as high as 6.2 × 104 GPU (1 GPU = 10–6 cm3[STP]/[s cm2 cmHg]), indicating their possible use as a porous polymer support for gas separation applications.  相似文献

Thermoresponsive double hydrophilic diblock copolymers poly(2‐(2′‐methoxyethoxy)ethyl methacrylate‐co‐oligo(ethylene glycol) methacrylate)‐b‐poly(6‐O‐methacryloyl‐D ‐galactopyranose) (P(MEO2MA‐co‐OEGMA)‐b‐PMAGP) with various compositions and molecular weights were obtained by deprotection of amphiphilic diblock copolymers P(MEO2MA‐co‐OEGMA)‐b‐poly(6‐O‐methacryloyl‐1,2:3,4‐di‐O‐isopropylidene‐D ‐galactopyranose) (P(MEO2MA‐co‐OEGMA)‐b‐PMAlpGP), which were prepared via reversible addition‐fragmentation chain transfer (RAFT) polymerization using P(MEO2MA‐co‐OEGMA) as macro‐RAFT agent. Dynamic light scattering and UV–vis studies showed that the micelles self‐assembled from P(MEO2MA‐co‐OEGMA)‐b‐PMAlpGP were thermoresponsive. A hydrophobic dye Nile Red could be encapsulated by block copolymers P(MEO2MA‐co‐OEGMA)‐b‐PMAGP upon micellization and released upon dissociation of the formed micelles under different temperatures. The galactose functional groups in the PMAGP block have specific interaction with HepG2 cells, and P(MEO2MA‐co‐OEGMA)‐b‐PMAGP has potential applications in hepatoma‐targeting drug delivery and biodetection. © 2011 Wiley Periodicals, Inc. A three‐step process, combining nitroxide‐mediated polymerization (NMP) and reversible addition‐fragmentation chain transfer (RAFT) polymerization techniques, for synthesizing well‐defined amphiphilic and thermosensitive graft copolymers with fluorescence poly(styrene‐co‐(p‐chloromethylstyrene))‐g‐poly(N‐isopropylacrylamide) (P(St‐co‐(p‐CMS))‐g‐PNIPAAM), was conducted. Firstly, the NMP of styrene (St) and p‐chloromethylstyrene (p‐CMS) were carried out using benzoyl peroxide (BPO) as the initiator to obtain the random copolymers of P(St‐co‐(p‐CMS)). Secondly, the random copolymers were converted into macro‐RAFT agents with fluorescent carbazole as Z‐group through a simple method. Then the macro‐RAFT agents were used in the RAFT polymerization of N‐isopropylacrylamide (NIPAAM) to prepare fluorescent amphiphilic graft copolymers P(St‐co‐(p‐CMS))‐g‐PNIPAAM with controlled molecular weights and well‐defined structures. The copolymers obtained were characterized by gel permeation chromatography (GPC), 1H nuclear magnetic resonance (NMR) spectroscopy, and FT‐IR spectroscopy. The size of self‐assembly micelles of the resulting graft copolymers in deionized water was studied by high performance particle sizer (HPPS), the results showed that the Z‐average size of the micelles increased with the increase of molecular weights of PNIPAAM in side chains. The aqueous solution of the micelles prepared from P(St‐co‐(p‐CMS))‐g‐PNIPAAM using a dialysis method showed a lower critical solution temperature (LCST) at ~ 27.5 °C, which was below the value of NIPAAM homopolymer (32 °C). © 2007 Wiley Periodicals, Inc. The synthesis of a new cyclic carbonate monomer containing an allyl group was reported and its biodegradable amphiphilic block copolymer, poly(ethylene glycol)‐block‐poly(L ‐lactide‐co‐5‐methyl‐5‐allyloxycarbonyl‐propylene carbonate) [PEG‐b‐P(LA‐co‐MAC)] was synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and 5‐methyl‐5‐allyloxycarbonyl‐1,3‐dioxan‐2‐one (MAC) in the presence of poly (ethylene glycol) as a macroinitiator, with diethyl zinc as a catalyst. 13C NMR and 1H NMR were used for microstructure identification of the copolymers. The copolymer could form micelles in aqueous solution. The core of the micelles is built of the hydrophobic P(LA‐co‐MAC) chains, whereas the shell is set up by the hydrophilic PEG blocks. The micelles exhibited a homogeneous spherical morphology and unimodal size distribution. By using the cyclic carbonate monomer containing allyl side‐groups, crosslinking of the PEG‐b‐P(LA‐co‐MAC) inner core was possible. The adhesion and spreading of ECV‐304 cells on the copolymer were better than that on PLA films. Therefore, this biodegradable amphiphilic block copolymer is expected to be used as a biomaterial for drug delivery and tissue engineering. © 2007 Wiley Periodicals, Inc. A successive method for preparing novel amphiphilic graft copolymers with a hydrophilic backbone and hydrophobic side chains was developed. An anionic copolymerization of two bifunctional monomers, namely, allyl methacrylate (AMA) and a small amount of glycidyl methacrylate (GMA), was carried out in tetrahydrofuran (THF) with 1,1‐diphenylhexyllithium (DPHL) as the initiator in the presence of LiCl ([LiCl]/[DPHL]0 = 2), at −50 °C. The copolymer poly(AMA‐co‐GMA) thus obtained possessed a controlled molecular weight and a narrow molecular weight distribution (Mw /Mn = 1.08–1.17). Without termination and polymer separation, a coupling reaction between the epoxy groups of this copolymer and anionic living polystyrene [poly(St)] at −40 °C generated a graft copolymer with a poly(AMA‐co‐GMA) backbone and poly(St) side chains. This graft copolymer was free of its precursors, and its molecular weight as well as its composition could be well controlled. To the completed coupling reaction solution, a THF solution of 9‐borabicyclo[3.3.1]nonane was added, and this was followed by the addition of sodium hydroxide and hydrogen peroxide. This hydroboration changed the AMA units of the backbone to 3‐hydroxypropyl methacrylate, and an amphiphilic graft copolymer with a hydrophilic poly(3‐hydroxypropyl methacrylate) backbone and hydrophobic poly(St) side chains was obtained. © 2000 John Wiley & Sons, Inc. Novel and well‐defined amphiphilic dendrimer‐star copolymer poly(ε‐caprolactone)‐block‐(poly(2‐(2‐methoxyethoxy)ethylmethacrylate‐co‐oligo(ethylene glycol) methacrylate))2 with Y‐shaped arms were synthesized by the combination of ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP). The investigation of thermal properties and the analysis of crystalline morphology indicate that the high‐branched structure of dendrimer‐star copolymers with Y‐shaped arms and the presence of amorphous P(MEO2MA‐co‐OEGMA) segments together led to the complete destruction of crystallinity of the PCL segments in the dendrimer‐star copolymer. In addition, the hydrophilicity–hydrophobicity transition of the dendrimer‐star copolymer film can be achieved by altering the external temperatures. The amphiphilic copolymers can self‐assemble into spherical nanomicelles in water. Because the lower critical solution temperature of the copolymers can be adjusted by varying the ratio of MEO2MA and OEGMA, the tunable thermosensitive properties can be observed by transmittance, dynamic laser light scattering, and transmission electron microscopy (TEM). The release rate of model drug chlorambucil from the micelles can be effectively controlled by changing the external temperatures, which indicates that these unique high‐branched amphiphilic copolymers have the potential applications in biomedical field. © 2011 Wiley Periodicals, Inc. Novel block–graft copolymers [poly(styrene‐b‐ethylene‐co‐butylene‐b‐styrene)‐g‐poly(tert‐butyl acrylate)] were synthesized by the atom transfer radical polymerization (ATRP) of tert‐butyl acrylate (tBA) with chloromethylated poly(styrene‐b‐ethylene‐co‐butylene‐b‐styrene) (SEBS) as a macromolecular initiator. The copolymers were composed of triblock SEBS as the backbone and tBA as grafts attached to the polystyrene end blocks. The macromolecular initiator (chloromethylated SEBS) was prepared by successive hydrogenation and chloromethylation of SEBS. The degree of chloromethylation, ranging from 1.6 to 36.5 mol % according to the styrene units in SEBS, was attained with adjustments in the amount of SnCl4 and the reaction time with a slight effect on the monodispersity of the starting material (SEBS). The ATRP mechanism of the copolymerization was supported by the kinetic data and the linear increase in the molecular weights of the products with conversion. The graft density was controlled with changes in the functionality of the chloromethylated SEBS. The average length of the graft chain, ranging from a few repeat units to about two hundred, was adjusted with changes in the reaction time and alterations in the initiator/catalyst/ligand molar ratio. Incomplete initiation was detected at a low conversion; moreover, for initiators with low functionality, sluggish initiation was overcome with suitable reaction conditions. The block–graft copolymers were hydrolyzed into amphiphilic ones containing poly(acrylic acid) grafts. The aggregation behavior of the amphiphilic copolymers was studied with dynamic light scattering and transmission electron microscopy, and the aggregates showed a variety of morphologies. © 2002 Wiley Periodicals, Inc. Although biodegradable amphiphilic block copolymer micelles have been widely applied in the clinical applications as drug delivery nanocarriers, low‐efficiency cellular internalization frequently reduces therapeutic efficacy of the loaded drugs. Here, photothermal effect‐promoted cellular internalization of finely tuned thermo‐responsive amphiphilic biodegradable block copolymer nanocarriers via noninvasive stimuli of near‐infrared (NIR) light irradiation is demonstrated. Amphiphilic block copolymers, poly(ε‐caprolactone)‐block‐poly(N‐isopropylacrylamide‐coN,N‐dimethylacrylamide) (PCL‐b‐P(NIPAM‐co‐DMA)), are prepared with finely tuned compositions of P(NIPAM‐co‐DMA) for desirable lower critical solution temperature of the block copolymer micelles in aqueous solution. The block copolymers are then used to co‐encapsulate doxorubicin and indocyanine green, which show high encapsulation efficiency and significant photothermal effect upon exposure to NIR light irradiation. The photothermal effect‐induced collapse and hydrophilic‐to‐hydrophobic transition of P(NIPAM‐co‐DMA) shells significantly enhance the interactions between drug‐loaded micelles and cell membranes, which dramatically promote the cellular internalization of the micelles and therapeutic efficacy of loaded anticancer drugs.

A series of novel temperature‐ and pH‐responsive graft copolymers, poly(L ‐glutamic acid)‐g‐poly(N‐isopropylacrylamide), were synthesized by coupling amino‐semitelechelic poly(N‐isopropylacrylamide) with N‐hydroxysuccinimide‐activated poly(L ‐glutamic acid). The graft copolymers and their precursors were characterized, by ESI‐FTICR Mass Spectrum, intrinsic viscosity measurements and proton nuclear magnetic resonance (1H NMR). The phase‐transition and aggregation behaviors of the graft copolymers in aqueous solutions were investigated by the turbidity measurements and dynamic laser scattering. The solution behavior of the copolymers showed dependence on both temperature and pH. The cloud point (CP) of the copolymer solution at pH 5.0–7.4 was slightly higher than that of the solution of the PNIPAM homopolymer because of the hydrophilic nature of the poly(glutamic acid) (PGA) backbone. The CP markedly decreased when the pH was lowered from 5 to 4.2, caused by the decrease in hydrophilicity of the PGA backbone. At a temperature above the lower critical solution temperature of the PNIPAM chain, the copolymers formed amphiphilic core‐shell aggregates at pH 4.5–7.4 and the particle size was reduced with decreasing pH. In contrast, larger hydrophobic aggregates were formed at pH 4.2. © 2008 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 46: 4140–4150, 2008  相似文献
