Four new cycloartane glycosides have been isolated from a methanolic extract ofThalictrum squarrosum Stephan ex Willd.: squarroside A1 (I) — (21R, 22S, 23R)-3β-(β-D-glucopyranosyloxy)-21α-methoxy-21,23-epoxycycloart-24-ene-22β,30-diol, C30H60O10; squarroside A2 (II) — the (21S)-epimer of compound (I); squarroside B1 (III) (21R, 22S, 23R)-3gb-[O-α-L-rhamnopyranosyl-(1 → 6)-β-D-glucopyranosyloxy]-21α-methoxy-21,23-epoxycycloart-24-ene-22β,30-diol, C43H70O14; and squarroside B2 (IV) — the (21S)-epimer of compound (III). The proposed structures were determined on the basis of1H and13C NMR spectroscopy, FAB mass spectrometry, and chemical transformations. (Z)-4-Bromo-1,3-di(2-thienyl)-2-buten-1-one was obtained by the bromination of 1,3-di(2-thienyl)-2-buten-1-one by NBS in anhydrous CCl4. The starting butanone was obtained by the condensation of 1-(2-thienyl)-1-ethanone by the action of SOCl2. The reaction of (Z)-4-bromo-1,3-di(2-thienyl)-2-buten-1-one with tertiary amines such as Et3N, pyridine, 1-alkyl-1,3-diazole, 1-alkylbenzimidazole, and 1-alkyl-1,2,4-triazole leads to quaternary salts. The azolium salts cyclize by the action of base to give di(2-thienyl)azolo[a]pyridinium derivatives. 3-Methyl-6,8-di(2-thienyl)[1,3]thiazolo[3,2-a]pyridin-4-ium and 2,4-di(2-thienyl)pyrido[2,1-b]benzothiazol-10-ium bromides were obtained by the same procedure but without separating the intermediate quaternary salts.  相似文献

A method has been developed for the synthesis of 4-(2-quinolinyl)-1,2,3,3a-tetrahydropyrrolo-[1,2-a]quinolines based on the 1,3-dipolar cycloaddition reaction of α,β-unsaturated carbonyl compounds to 2,3′-biquinolinium salts. Oxidation in benzene using MnO2 gave 4-(2-quinolinyl)-pyrrolo[1,2-a]quinolines. It was found that a side product of the 1,3-dipolar cycloaddition is 7,14-di-benzoyl-6,13-di(2-quinolyl)-6a,7,13a,14-tetrahydro-7a,14a-diazadibenzo[a,h]anthracene. Reaction of 1′-phenacyl-2,3′-biquinolinium salts with hydroxylamine in acetic acid gave 4-(2-quinolyl)imidazo-[1,2-a]quinolines. *For Communication 24 see [1]. Cyclization were occurred via the coupling reactions of some mercuric chloride derivatives of sydnone with LiPdCl3-CuCl2. A unique six-membered ring, 3,3′-ethylene-4,4′-bissydnone, was obtained by the cyclization reation of 1,2-di[3-(4-chloromercuric)sydnonyl]ethane. However, the seven-membered 3,3′-trimethylene-4,4′-bissydnone and 1,3-di[3-(4-chloro)sydnonyl]-propane were obtained from the corresponding mercuric chlroide of sydnone. Onyl substitution reaction took place when 4,4′-di[3-(4-chloromercuric)sydnonyl]biphenyl, 4,4′-di[3-(4-chloromercuric)sydnonyl]benzene, di(p-[3-(4-chloromercuric)sydnonyl]-phenyl}methane and, di(p-[3-(4-chloromercuric)sydnonyl]phenyl]ether were treated using the same process.  相似文献

For preparing a berberine-sensitive electrode, 5,10,15,20-tetrakis[2-(2,3,4,6-tetraacetyl-β-D-glucopyranosyl)-1-O-phenyl]porphyrin (T(o-glu)PPH2) was synthesized from the reaction of pyrrole with ortho-acetylglycosylated benzaldehyde by Lindsay’s method. The electrode based on T(o-glu)PPH2 with an optimized membrane composition exhibits Nernstian response to berberine in the concentration range 2.4 × 10–7–5.0 × 10–3 mol L–1, with a pH range from 3.9 to 10.2, and a fast response time of 30 s. The electrode shows fair selectivity towards berberine with respect to common co-existing species. T(o-glu)PPH2 shows better potentiometric response characteristics comparing to chloro[5,10,15,20-tetrakis[2-(2,3,4,6-tetraacetyl-β-D-glucopyranosyl)-1-O-phenyl]-porphinato]-manganese (MnT(o-glu)PPCl) and better selectivity towards berberine than tetraphenylporphyrin (TPPH2). The effect of the composition of the electrode membrane has been studied and the experimental conditions optimized. The contents of berberine in pharmaceutical tablets were determined by direct potentiometry and the results agreed with values obtained by the pharmacopoeia method. The synthesis of 1,2-, 1,3-, and 1,4-phenylene-bis[(4R,5R)-4,5-di(hydroxydiphenylmethyl)-1,3-dioxolane]s (ortho-, meta-, and para-bis-(R,R)-TADDOLs) and bis[4-{[(4R, 5R)-4,5-di(hydroxydiphenylmethyl)]-1,3-dioxolan-2-yl}phenyl]methane was carried out. The possibilities of the use of these compounds as catalysts for the C-C bond formation in the addition of Me3SiCN to benzaldehyde and the oxirane ring opening in cyclohexene oxide by Me3SiCN were investigated. Heating of 8-aminotheophylline with methyl (Z)-2-benzoylamino-3-(dimethylamino)propenoate in acetic acid afforded in a one-pot synthesis a new pyrimido[2,1-f]theophylline derivative. Methylation of this by using CH3I/NaH furnished in good yield the double methylated derivative. Furthermore, glycosidation of the former with 1-α-bromo-2,3,4,6-tetra-O-acetyl-d-glucose gave the β-glucoside derivative. Reaction of 8-aminotheophylline with [bis(methylthio)methylene]malonitrile, ethyl[bis(methylthio)methylene]cyanoacetate, 1,3-diphenylprop-2-en-1-one, 2-cyano-1,3-diphenylprop-2-en-1-one, 1-(4-nitrophenyl)-3-(dimethylamino)prop-2-ennitrile, 1-phenyl-3-(dimethylamino)prop-2-en-1-one, 2-substituted 3-aryl or heteroarylprop-2-ennitrile and ethyl(arylmethylene)cyanoacetate in N,N-dimethylformamide in the presence of anhydrous potassium carbonate afforded also the corresponding new derivatives of pyrimido-[2,1-f]theophylline. However, 8-aminotheophylline reacted in similar manner with 3-chloropentan-2,4-dione and 2-bromo-1-phenylethanone to give the corresponding imidazo[2,1-f]theophyllines. Furthermore, azo-coupling of one of these with 4-methylphenyldiazonium chloride was performed. The antimicrobial activity of the products has been evaluated. A series of tetrathiafulvalene-substituted 2,3-di(2-pyridyl)quinoxaline (dpq) ligands, 2-(4,5-bis(methylthio)-1,3-dithiol-2-ylidene)-6,7-di(pyridin-2-yl)- [1,3]dithiolo[4,5-g]quinoxaline (L1), dimethyl-2-(6,7-di(pyridin-2-yl)-[1,3]dithiolo[4,5-g]quinoxalin-2-ylidene)-1,3-dithiole-4,5-dicarboxylate (L2), and 2-(5,6-dihydro-[1,3]dithiolo[4,5-b] [1,4]dithiin-2-ylidene)-6,7-di(pyridin-2-yl)-[1,3]dithiolo[4,5-g]quinoxaline (L3), have been prepared. Reactions of these ligands with Re(CO)5Cl afford the corresponding dinuclear rhenium(I) carbonyl complexes, Re2(L)(CO)6Cl2 (L = L1, 5a; L = L2, 5b; L = L3, 5c). All new compounds are fully characterized by 1H NMR, IR and mass spectroscopies. The crystal structures of 5a and 5b have been studied. Optimized conformations and molecular orbital diagrams of 5a5c have been calculated with density functional theory (DFT). The spin-allowed singlet−singlet electronic transitions of all complexes have been calculated with time-dependent DFT (TDDFT), and the UV-Vis−NIR spectra are discussed based on the theoretical calculations.  相似文献

4a-Substituted dihydroindenopyridines undergo cleavage of the C(9b)=N bond in water-containing acidic medium, resulting in the formation of diastereomeric ethyl α-acetyl-β-(2,3-dihydro-1,3-dioxo-1H-inden-2-yl)-β-phenylpropionates and 9a-substituted 1,9a-dihydrofluore-none derivatives. The cyclization of α-acetyl-β-(2,3-dihydro-1,3-dioxo-1H-inden-2-yl)-β-phenyl-propionates with benzylammonium acetate affords 3-benzylamino-1,9a-dihydrofluorenones. The nucleophilic addition of 2,4-dithiobiurete, 1- and 1,4-substituted 2,4-dithiobiuretes (2 a-e) with benzoylacetylene (1) has been studied. 2-(Benzoylmethyl)-4-(R1-imino)-6-(R2-imino)dihydro-4H-1,3,5-dithiazinium perchlorates (3 a-e) are obtained in glacial acetic acid (AcOH) in the presence of equimolar quantities of HClO4. The reaction of benzoylacetylene with 1,5-diphenyl-2,4-dithiobiurete in either of MeOH, C6H6, or MeCN solvents proceeds non-selectively to give a mixture of products such as 2-(benzoylmethyl)-4,6-di(phenylimino)dihydro-4H-1,3,5-dithiazine (5), 2-(benzoylmethyl)-4-(β-benzoylvinyl)thio-3-phenyl-6-(phenylimino)-3,6-dihydro-2H-1,3,5-thiadiazine (8), 2-(benzoylmethyl)-1,3-diphenylhexahydro-1,3,5-triazine-4,6-dithione (7) and N-(β-benzoylvinyl)-N-phenylthioureas (6).  相似文献

The corresponding 1-(2-quinoxalyl)-, 1-[3,5-di(trifluoromethyl)phenyl]-, and 1-ethoxycarbonyl-3-methyl-4-oxo-4,5,6,7-tetrahydroindazoles have been obtained from reactions of 2-acetyl-1,3-cyclohexanedione, its 5,5-dimethyl and 5-(2-furyl) derivatives, with 2-hydrazinoquinoxaline, 3,5-di(trifluoromethyl)phenylhydrazine, and ethoxycarbonylhydrazine. On interaction with ethoxycarbonylhydrazine the intermediate 2-[1-(-ethoxycarbonyl)hydrazino]ethylidene-1,3-cyclohexanediones were also isolated. From the potassium salt of 2-formyldimedone and 2-carboxyphenylhydrazine hydrochloride, 2-(2-carboxyphenyl)hydrazinomethylene-5,5-dimethyl-1,3-cyclohexanedione was obtained, the cyclization of which in ethanol in the presence of HCl led to 1-(2-carboxyphenyl)- and 1-(2-ethoxycarbonylphenyl)-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole.  相似文献

Addition of ethyl-, propyl-, and n-butylmercaptans to 1-O-allyl-2,3,4,6-tetra-O-acetyl-β-D-galactopyranose in the presence of benzoyl peroxide catalyst was studied for the first time. The products were 1-O-(3-ethylthiopropyl)-2,3,4,6-tetra-O-acetyl-β-D-galactopyranose, 1-O-(3-propylthiopropyl)-2,3,4,6-tetra-O-acetyl-β-D-galactopyranose, and 1-O-(3-butylthiopropyl)-2,3,4,6-tetra-O-acetyl-β-D-galactopyranose. The Knoevenagel condensations of 5-[3-(trifluoromethyl)phenyl]furan-2-carbaldehyde with seven compounds containing an active methyl or methylene group have been studied. The compounds used were: methyl 2-cyanoacetate, malononitrile, 2-furylacetonitrile, acetophenone, 2-thioxo-1,3-thiazolidin-4-one (rhodanine), 5,5-dimethylcyclohexane-1,3-dione (dimedone), and methyl 2-azidoacetate. The effect of microwave irradiation on the condensation reactions was studied and compared with “’classical”’ conditions. Thermolysis of methyl 2-azido-3-{5-[3-(trifluoromethyl)phenyl]-2-furyl}propenoate afforded methyl 2-[3-(trifluoromethyl)phenyl)]-4H-furo[3,2-b]pyrrole-5-carboxylate. (2E)-3-{ 5-[3-(Trifluoromethyl)phenyl]-2-furyl}propenoic acid was converted to the corresponding azide, which was cyclized on heating into 2-[3-(trifluoromethyl)phenyl)]-4,5-dihydrofuro[3,2-c]pyridin-4-one. The latter after successive action of POCl3 and NH2NH2-Pd/C gave 2-[3-(trifluoromethyl)-phenyl]furo[3,2-c]pyridine. In this study, we observed unprecedented cleavages of the Cβ–Cγ bonds of tryptophan residue side chains in a series of hydrogen-deficient tryptophan-containing peptide radical cations (M•+) during low-energy collision-induced dissociation (CID). We used CID experiments and theoretical density functional theory (DFT) calculations to study the mechanism of this bond cleavage, which forms [M – 116]+ ions. The formation of an α-carbon radical intermediate at the tryptophan residue for the subsequent Cβ–Cγ bond cleavage is analogous to that occurring at leucine residues, producing the same product ions; this hypothesis was supported by the identical product ion spectra of [LGGGH – 43]+ and [WGGGH – 116]+, obtained from the CID of [LGGGH]•+ and [WGGGH]•+, respectively. Elimination of the neutral 116-Da radical requires inevitable dehydrogenation of the indole nitrogen atom, leaving the radical centered formally on the indole nitrogen atom ([Ind]-2), in agreement with the CID data for [WGGGH]•+ and [W1-CH3GGGH]•+; replacing the tryptophan residue with a 1-methyltryptophan residue results in a change of the base peak from that arising from a neutral radical loss (116 Da) to that arising from a molecule loss (131 Da), both originating from Cβ–Cγ bond cleavage. Hydrogen atom transfer or proton transfer to the γ-carbon atom of the tryptophan residue weakens the Cβ–Cγ bond and, therefore, decreases the dissociation energy barrier dramatically.  相似文献

The reaction of propynals X-C≡C-CHO, where X = Alk or Ar, with 2,3-di(N-hydroxyamino)-2,3-dimethylbutane gives exclusively 1-X-2-(1-hydroxy-4,4,5,5-tetramethylimidazolidin-2-ylidene)ethanones. In the case of X = Me3Si, the reaction affords 2-(2-trimethylsilylethynyl)-4,4,5,5-tetramethylimidazolidine-1,3-diol. The reaction of propynal containing X = Et3Ge yields both types of the products. The resulting imidazolidine-1,3-diol can be quantitatively isomerized to imidazolidin-2-ylideneethanone, oxidized to 2-[2-(triethylgermyl)ethynyl]-4,4,5,5-tetramethyl-4,5-dihydro-1H-imidazole-3-oxide-1-oxyl, or transformed into ethynyl-substituted nitronyl nitroxide. The direct and indirect stereochemical resolution of the enantiomers of free and N-protected (R,S)-2′,1′:1,2;1″,2″:3,4-dinaphthcyclohepta-1,3-diene-6-amino-6-carboxylic acid (Bin) was achieved by high-performance liquid chromatographic methods.
