NEW PHTHALOCYANINES FOR PHOTODYNAMIC VIRUS INACTIVATION IN RED BLOOD CELL CONCENTRATES 总被引:4,自引:0,他引:4

Abstract Cationic phthalocyanines with either aluminum or silicon as the central metal were evaluated for their ability to inactivate viruses in red blood cell concentrates (RBCC) photodynamically. In addition, the virucidal potential of a substituted anionic phthalocyanine, aluminum dibenzodisulfophthalocyanine hydroxide (AlN2SB2POH) was evaluated and compared with that of the much studied anionic aluminum tetrasulfophthalocyanine hydroxide (AIPcS4OH). Based on the rate of inactivation of the lipid-enveloped vesicular stomatitisvirus (VSV), the viruci dal potential of these phthalocyanines was: HOSiPCOSi(CH3)2(CH2)3N+(CH3)3I- (Pc 5) = SiPC[OSi(CH3)2-(CH2)3N+(CH3)3I-]2 (Pc 6) > AIPCOSi(CH,)2(CH2)?N+(CH3)2(CH?)11CH3I- (Pc 21) = A1N2SB2POH = AlPcS4 > HOSiPc[OSi(CH3)2(CH2)3N+(CH3)2(CH2)11CH31–]2(Pc 14) > AIPcOSi(CH3)2(CH2)3N+(CHS)3I- (Pc 2). Phthalocy anine ligand 14 and Pc 21 are new phthalocyanines, made by quaternizing known amino analogues. Compared to VSV, the rate of inactivation of Sindbis virus (another model lipid-enveloped virus) was identical when treated in red blood cells (RBC) with Pc 5 and slightly higher when treated with Pc 6 and AlPcS4OH. Treatment of RBCC containing cell-free human immunodeficiency virus (HIV-1) with Pc 5 or AlPcS4OH required 15 min of irradiation to inactivate (>5 log10 reduction) the virus. The extent of HIV-1 inactivation with AlN2SB2POH was 3.7 log10 after 60 min of red light exposure. The RBC integrity after photosensitization was measured by the ability of the cells to bind to plates coated with poly-L-lysine, (which reflects the retention of the RBC surface negative charges) and hemolysis of the cells over a 7 day storage period. The RBC damage using these criteria was most pronounced with Pc 5 and Pc 6 but could be reduced when treatment was in plasma instead of buffer. These data indicate that lipid-enveloped viruses differ in their sensitivity to phthalocyanine photosensitization. Therefore, for virus sterilization of RBCC for transfusion the ability to inactivate human pathogenic viruses completely will have to be evaluated for each virus. The cationic Pc 5 appears to be a potentially useful virucidal agent. 相似文献

Abstract C8KC is a new ketochlorin photosensitizer that must be formulated with an emulsifier because of its poor water solubility. In this report, we compare properties of Cremophor EL (CRM) and Tween 80 as delivery vehicles for C8KC. Unlike Tween 80, CRM altered the physical properties of both human and mouse plasma lipoproteins, resulting in decreased electrophoretic mobility of the individual lipoproteins along with the formation of a l ipoprotein degradation product: a phospholipid fraction of low buoyant density. In human plasma, where there was sufficient low-density lipoprotein (LDL) for a distinction to be made, CRM caused a shift in binding of a ketochlorin from albumin to LDL and the degraded lipoprotein fraction. In mice bearing the RIF tumor, the use of CRM for drug formulation was associated with longer plasma and tissue persistence of C8KC, and enhanced photodynamic therapy (PDT) efficacy. These results indicate the importance of both sensitizer and vehicle as determinants of PDT efficacy. 相似文献

The relative importance of type I and type II mechanisms in the photodynamic treatment of red blood cell concentrations (RBCC) to inactivate viruses was studied using aluminum phthalocyanine tetrasulfonate (AlPcS4), visible light and quenching or enhancing agents of reactive forms of oxygen. Treatment of a human RBCC with 10-13 microM AlPcS4 and 25-26 mW/cm2 visible light resulted in the rapid and complete inactivation of added vesicular stomatitis virus (VSV). The addition of mannitol, glycerol, reduced glutathione (GSH), or superoxide dismutase (SOD), known quenching agents of type I mechanisms, had little to no effect on the rate of inactivation of VSV. Significant inhibition of VSV kill was observed on addition of tryptophan or sodium azide, known quenchers of type II mechanisms. Additionally, the rate of VSV kill was enhanced in the presence of D2O. Taken together, these results indicate a predominant role of singlet oxygen in the inactivation of VSV on photodynamic treatment of RBCC. The relative importance of type I and type II mechanisms on cellular toxicity was also evaluated. Little, if any hemoglobin release was observed on treatment of human or rabbit RBCC with 10 microM AlPcS4 and 44 J/cm2 of visible light in the presence or absence of the above mentioned quenchers. Abstract The post-illumination photohemolysis of human red blood cells was used as a model system for studying membrane damage sensitized by various phthalocyanine sulfonates. With a constant fluence and photolysis time at 670 ± 10 nm and with a fixed optical density of the sensitizer, the percentage of hemolysis was measured as a function of time after illumination; and from the usual sigmoidal shaped curves, the times for 50% hemolysis were determined. The most effective central cation was Al, followed by Zn; the metal-free compound and those containing the paramagnetic ions Cu, Fe and Co were inactive. The sensitizer bound to the red blood cells was found to be effective for photohemolysis. Studies of the Al compound in 98.0% deuterated media (as measured by NMR) showed a definite deuterium isotope effect, suggesting the participation of singlet oxygen. This result was supported by the large protective effects of 3.3 raM and 13.3 mM tryptophan present during photolysis. Tryptophan reacts rapidly with singlet oxygen and with OH radicals. The much smaller effects observed with the same concentrations of mannitol and glycerol, OH radical scavengers which do not react with singlet oxygen, are consistent with the predominant role of singlet oxygen in photohemolysis sensitized by Al phthalocyanine sulfonate. 相似文献

Abstract— The photodynamic action of protoporphyrin on red cell ghosts is reflected by extensive cross-linking of membrane proteins to very high molecular weight protein aggregates. This process was studied with sepharose gel chromatography and sodium dodecyl sulphate polyacrylamide gel electrophoresis.
Most sensitive to this photodynamic effect are spectrin and band 2. 1, 2. 2, 2.3 and 4.1. polypeptides, which are cross-linked after very brief illumination periods, with a concomitant loss of spectrin-associated ATPase activity. Band 6 protein, representing the monomeric form of glyceraldehyde-3-phosphate dehydrogenase, is also very sensitive to protoporphyrin-induced cross-linking. The enzymatic activity decreased even faster than the amount of band 6 polypeptides, suggesting that modification(s) of the enzyme other than cross-linking, possibly by rapid photooxidation of a thiol group, may be responsible for inactivation.
Extracted and purified spectrin was cross-linked with about the same velocity as membrane-bound spectrin, reinforcing our previously drawn conclusion that membrane lipids are not involved in the cross-linking reaction. Eluted band 6 polypeptides on the other hand exhibited a relatively fast photo-oxidative modification but a much slower cross-linking to dimers and tetramers. Abstract— Phthalocyanines are being studied as photosensitizers for virus sterilization of red blood cells (RBC). During optimization of the reaction conditions, we observed a marked effect of the irradiance on production of RBC damage. Using a broad-band light source (600–700 nm) between 5 and 80 mW/ cm2 , there was an inverse relationship between irradiance and rate of photohemolysis. This effect was observed with aluminum sulfonated phthalocyanine (AlPcSn ) and cationic silicon (HOSiPc-OSi[CH3 ]2 [CH2 ]3 N+ [CH3 ]3 I- phthalocyanine (Pc5) photosensitizers. The same effect occurred when the reduction of RBC negative surface charges was used as an endpoint. Under the same treatment conditions, vesicular stomatitis virus inactivation rate was unaffected by changes in the irradiance. Reduction in oxygen availability for the photochemical reaction at high irradiance could explain the effect. However, theoretical estimates suggest that oxygen depletion is minimal under our conditions. In addition, because the rate of photohemolysis at 80 mW/cm2 was not increased when irradiations were carried out under an oxygen atmosphere this seems unlikely. Likewise, formation of singlet oxygen dimoles at high irradiances does not appear to be involved because the effect was unchanged when light exposure was in D2 O. While there is no ready explanation for this irradiance effect, it could be used to increase the safety margin of RBC virucidal treatment by employing exposure at high irradiance, thus minimizing the damage to RBC. 相似文献

Rubella virus is very sensitive to photodynamic action. When tested with 1.2 X 10(-5) M toluidine blue and 8 W fluorescent lamp at a fluence of 11 W/m2, inactivation kinetics showed a linear single hit curve with a k value of 1.48 min-1. Photodynamic inactivation of rubella virus greatly enhanced recombination with a latent virus (R-virus) of baby hamster kidney BHK21 cells. In contrast, no hybrids were detected in lysates of the cells infected with either UV-treated or untreated rubella virus. Therefore, hybrid viruses were readily detected only in lysates of BHK21 cells infected with photodynamically treated rubella virus. Photodynamic damage of rubella virus genomes generated a new hybrid type (hybrid type 3) in addition to a previously described type 2 hybrid (formerly designated as HPV-RV variant). Although both of these hybrid types carry the CF antigens of rubella virus, plaque forming ability of type 3 hybrid is neutralized neither by anti-rubella serum nor by anti-latent virus serum while type 2 hybrid is neutralized by anti-latent virus serum. 相似文献

Abstract— Absorption and fluorescence spectra of chlorphyll-a in aqueous alcohol were determined as a function of temperature between 173° and 293°K. From a comparison of these spectra with ones obtained in pure methanol, it is suggested that changes in molecular association occur within aggregates of chlorophyll-a in aqueous-alcoholic solution induced by variation of the dielectric constant. Abstract The dependence of radiation transmission on sample thickness was studied in isolated samples of human stratum corneum and full-thickness epidermis. The investigation also included samples of skin repeatedly exposed to UV-B. Transmission was measured in the ultraviolet and in the visible from 248–546 nm. Two methods, one microscopic and the other mechanical, were used to measure thickness. There was a good correlation between the results.
The dependence of transmission on thickness in these samples could be described satisfactorily by an exponential function, implying that the Lambert-Beer law is approximately valid. Thus, a single parameter, such as the half-value layer ( d½ ), is sufficient to characterize absorption in the skin samples.
Water content of the isolated stratum corneum was influenced by maintenance conditions: samples floating on water containing a small amount of NaCl were more hydrated than samples floating on a more concentrated salt solution, or stored in air. Changes in water content of the samples resulted in changes of thickness and, to a lesser extent, of transmission. Approximate in vivo values of d½ were computed after estimating the in vivo water content of stratum corneum.

Abstract Zinc phthalolocyanine photosensitized donor-acceptor systems for light energy conversion and for the design of photoelectrochemical molecular devices are presented. Covalently linked phthalocyanine complexes were incorporated in bilayer lipid membranes (BLM) and deposited on SnO2 transparent electrodes. Their photovoltages were measured and compared. It has been found that a more favorable orientation and closer proximity are attained in the diad compounds between the donor (phthalocyanine)-acceptor (anthraquinone) pair than in the reference compound for efficient light-induced charge separation and transfer. The triad compound is the best among all tested compounds. The decrease in the fluorescence yield and lifetime induced by quinones was examined and the apparent electron-transfer rate constants were calculated. 相似文献

THE FUNCTION OF P700 AND CYTOCHROME f IN THE PHOTOSYNTHETIC REACTION CENTER OF SYSTEM 1 IN RED ALGAE
Abstract— Kinetics and quantum yields of light-induced oxidation of P700 and the f -type cytochrome were measured in marine red algae from absorbance changes in the region 420, 435 and 705 nm. The quantum yield for cytochrome oxidation in Iridaea splendens and Schizymenia pacifica was 0.5-0.65 in far-red light, at 20 and at 0C.
Oxidation rates of P700 measured when varying amounts of cytochrome were in the oxidized state indicated that a reaction center of system 1 in Iridaea contains 1 P700 and 4 cytochrome molecules. Oxidized P700 only accumulates when all 4 cytochromes are oxidized. The rate of photochemistry of system 1, measured as the sum of the rates of P700 and cytochrome oxidation, was independent of the oxidation level of cytochrome, but decreased with accumulation of oxidized P700. This decrease was less than proportional to the fraction of P700 that was in the oxidized state, which suggested transfer of excitation energy between reaction centers.
The quantum yield for cytochrome oxidation after dark periods of 1 min or more was only about 0.2. This effect was tentatively ascribed to a dark reduction of the cytochrome coupled to phosphorylation. 相似文献

Abstract— We have examined the relationship between the survival curves obtained with UV light (254 nm) and those obtained with a near-UV sburce (Westinghouse Sun Lamps, FS20) simulating sunlight for cultured Chinese hamster cells, C3H mouse cells, and HeLa cells. In no case do the "sunlight" survival curves have the same shape as the UV survival curves. Also, the combined lethal effects of UV and near-UV, sunlight-like exposures are not entirely additive. Hence, we conclude that (1) the cell-killing photolesions induced by these radiations are at most only partly the same, and (2) in view of (I), caution should be exercised in predicting near UV-induced dose-dependencies of other end points based upon observations with UV. 相似文献

Abstract— Conidia of differing ages from wild type and the fluffy ( fl ) mutant of Neurospora were inactivated with both blacklight and shortwave UV. Five-day-old cultures of both wild type and fluffy conidia were equally sensitive to inactivation by shortwave UV while the fluffy mutant was more sensitive to blacklight inactivation. As the cultures age, both wild type and the fluffy mutant become more sensitive to inactivation by blacklight and shortwave UV. Abstract— Studies have been made of the inactivation of potato virus X (PVX), free PVX-RNA, and a phenotypically mixed virus particle composed of PVX-RNA and tobacco mosiac virus (U-1 strain) protein (PVXRNA : TMVPRO ) by ultraviolet radiation of 254, 280 and 302 nm wavelengths. Rate constants and quantum yields of inactivation have been determined under conditions of photoreactivation and non-photoreactivation in H2 O and D2 O. The ratios of the rate constants in H2 O to those in D2 O obtained for PVX-RNA were greater than unity at the above three wavelengths, as was the case for PVX at 254 and 302 nm. The ratios of the rate constants were, within experimental error, equal to unity for PVXRNA :TMVPRO at all three wavelengths and for PVX at 280 nm. It is concluded that, in contrast to the situation observed with TMV and PVXRNA :TMVPRO , the behavior of the intact PVX viron upon irradiation closely approximates the behavior of free RNA in solution, on the basis of quantum yields of inactivation, photoreactivated sector, and the ratio of kinetic isotope effects in H2 O and D2 O. 相似文献

Abstract— Effects of active oxygen scavengers on cell inactivation and petite induction of yeast by the photodynamic action of euflavine were examined. Histidine, sodium azide, 1,3-diphenyl-isobenzofuran and p-carotene, which are singlet oxygen scavengers, inhibited photodynamic cell killing. Histidine and sodium azide inhibited petite induction, too. These results suggested that photobiological effects of euflavine are induced via singlet oxygen-mediated Type II reaction process. In this work, however, we found that ethanol, which is reported to be a hydroxyl radical scavenger, notably inhibited photodynamic cell inactivation and petite induction by euflavine. Inhibition of petite induction was increased with increasing concentration of ethanol. Decrease of absorbance of euflavine by irradiation was also inhibited by the addition of ethanol.
These results suggested that ethanol possibly acts as a singlet oxygen scavenger. 相似文献

Abstract— Human cells of the line NHIK 3025 were exposed to hematoporphyrin derivative (Hpd) and light and analysed with respect to; (i) the mobility of membrane proteins as determined by electron spin resonance measurements of a protein-bound spin label, (ii) fluorescence excitation spectra, (iii) relative number of DTNB-reactive SH-groups on their surface and in sonicated cell homogenates, (iv) survival, and (v) morphologic appearance as seen by ordinary phase contrast microscopy. A significant fraction of the porphyrins bound to the outer cell membrane was in close contact with proteins. 5,5'-Dithiobis-2-nitrobenzoic acid reactive SH-groups on the outer cell membrane were very sensitive to the treatment with Hpd + light and were degraded according to non-exponential kinetics. When the cells were irradiated after spin labelling, the labelled proteins became less mobile during the irradiation, indicating protein cross linking. Irradiation before spinlabelling resulted in a selective degradation of low-mobility proteins. 相似文献
