The thermal migration of cyclic and acyclic organoboranes were studied. In most cases, a stereoselective 1,2-dyotropic migration was observed, allowing the stereocontrol of three contiguous chiral centers. Scope and limitations of this thermal migration are presented.  相似文献

Self-disproportionation of enantiomers of heterocycles having a tertiary trifluoromethyl alcohol center on an achiral silica-gel stationary phase is discussed. During the chromatographic separation of an enantiomerically enriched mixture of 1-(3,4-dimethoxyphenethyl)-3-hydroxy-3-(trifluoromethyl)-6,7-dihydro-1H-indole-2,4(3H,5H)-dione (1) by eluting with ether on a non-chiral regular silica-gel significant enantiomeric enrichment was observed. Separation of non-racemic samples of 1 with enantiomeric excess values of 10-54% was carefully investigated: enantiomerically pure 1 with 99.9% ee was obtained by the use of 1 with at least 40% ee. A remarkable enantiomeric enrichment in the faster eluting fractions was also observed for compound 1 with only 30% ee to transform into 80% ee. Other enantiomeric mixtures of heterocyclic molecules containing a trifluoromethyl alcohol moiety at their quaternary carbon center were also examined from an SDE view point.  相似文献

The use of cyclodextrin derivatives for the efficient separation of enantiomeric drugs is described. Hydroxypropylation, methylation or carboxymethylation of the cyclodextrin not only result in a better solubility of the cyclodextrin in aqueous solutions, but also favor, via additional hydrogen bonding, the stabilization of one of the cyclodextrin-analyte complexes. The influence of the background electrolyte on peak shape is also described here. Carboxymethylated cyclodextrin can be used in similar manner to uncharged cyclodextrins at low pH values (below 4). At pH values above 5, however, its charge also allows the separation of uncharged enantiomers as in a micellar-like system.  相似文献

Summary The calculation of capacity factors, k, from net retention times, tR, and the corresponding dead times, tM, at different temperatures suffers from the limited accuracy of the tM values. If the temperature coefficient racy of the tM values. If the temperature coefficient d ln k/d (1/T) only is required, it is sufficient to determine net retention times (tR)p at constant inlet pressure pi for different temperatures, since the temperature dependence of (tM)p can be assumed as (tM)p=A·eB/T, with B being approximately independent of the column inlet pressure and of the nature of the carrier gas. The extrapolation and interpolation of (tR)p may be either performed by linear regression or graphically with a nomogram for ln (tR)p versus 1/T. The resolution factor, , of two components, e.g. enantiomers which are resolved on a chiral stationary phase, can be treated in a similar way. Examples are given for the resolution of enantiomers of two non-proteinogenic amino acids on the new polysiloxane phase L-Chirasil-CPG.  相似文献

Pinellic acid from the tuber of Pinellia ternate, an active herbal component of the traditional Japanese herbal (Kampo) medicine Sho-seiryu-to, is a C18 trihydroxy fatty acid whose absolute stereochemistry has now been determined. All stereoisomers of pinellic acid were synthesized via regioselective asymmetric dihydroxylation, regioselective inversion, and stereoselective reduction in order to determine their absolute stereochemistries and adjuvant activities. Among this series of isomers, the (9S,12S,13S)-compound, which is a natural product, exhibited the most potent adjuvant activity. Spectral data for all of the stereoisomers of the 1,2-allylic diols were compared and related to their stereochemistries.  相似文献

Summary The chromatographic characteristics of the thiazolidine carboxylic acids formed by the reaction of D- and L-penicillamine with various substituted benzaldehydes and heterocyclic aldehydes have been studied on Chiralplate layers. It has been found that the nature and position of substituents strongly affect the RF values and resolution factors (RS) of the isomers. An unambiguous relationship has been established between the RS values of the L- and D-enantiomers and the dipole moment of the aldehydes. The transformation to thiazolidine carboxylic acids with benzaldehyde and substituted benzaldehydes, however, is not complete, thus the reaction with formaldehyde is still more suitable for quantitative determination of penicillamine isomers.  相似文献

Summary A new expression which permits the prediction of the net retention volumes in gas chromatography with column packings of coated porous polymers is reported. The porous polymers Chromosorb 101 and Chromosorb 102 were used as supports and squalane, Ethofat and Carbowax 20M as stationary phases at three different column temperatures of 80°C, 100°C and 150°C. Several organic compounds of various polarity were used as test compounds and the net retention volumes, VNtheor, have been calculated according to this new expression. The VNtheor values were compared with the experimental net retention volumes, VNexp, and it was found that the difference between these two values is dependent on the type of compound and column packing. Nevertheless, the elution order could be predicted in most cases.  相似文献

Reaction of trialkylboranes with organic azides in refluxing xylene, followed by hydrolysis, leads to good yields of secondary amines. This reaction is highly dependent on the steric effects around both boron and the azide moiety. The reaction becomes much slower when the steric bulk of one of the reagents is increased and fails when both are hindered. The dialkylchloroboranes are more reactive than trialkylboranes and provide better yields of the desired secondary amines with a11 azides tested. The alkyldichloroboranes react with organic azides at temperatures between room temperature and 60°C and produce excellent yields of secondary amines. Furthermore, the stereochemistry of the original carbon-boron bond is retained. The mechanism of these reactions is discussed and the reaction applied to the synthesis of N-alkyl- and N-arylaziridines.  相似文献

Summary Chiral recognition of many enantiomeric solutes by a chiral amide stationary phase is based mainly on hydrogen bonding. A chiral-recognition-factor CHI is proposed, given by the difference of the enthalpy change in the enantiomer discrimination, standardized with respect to the specific interaction of the solutes with the diamide core of the stationary phase. The r?le of the entropy part is also discussed. By extrapolation of the retention behaviour to elevated temperature, peak inversion of enantiomers is predicted. Summary Enantiomeric separation of mosapride and a structurally related compound was performed using chiral chromatography and experimental design. Unique effects of mobile phase pH and column temperature made it possible to control the elution order of the enantiomers when using Chiral-AGP as the solid phase. At a low mobile phase pH (<6) the (R)-enantiomer of mosapride elutes before the (S)-form whereas the (S)-enantiomer elutes first at a high mobile phase pH (>6). By using a mobile phase pH around 6, the column temperature could also be used to control the elution order of the enantiomers of mosapride. Similar effects of mobile phase pH and column temperature were obtained for the enantiomers of a structurally related compound, a metabolite (M1). Isocratic chromatographic systems made it possible to determine enantiomeric impurities less than 0.1% in the respective enantiomer of mosapride. The enantiomers of mosapride as well as the enantiomers of M1 could easily be separated simultaneously using Chiral-AGP and a simple gradient elution. 根据前期得到的梯度液相色谱保留时间计算公式,在不指定溶剂强度模型形式的前提下,探讨了梯形梯度洗脱的一些特点。对于溶质在梯形梯度坡度上流出时的情形,推导得到溶质流出色谱柱所对应的流动相组成(φR)随梯度斜率(B)变化的表达式。该公式表明,在该情形中φR将会随着B值的增加而增加。对于溶质在梯形梯度最后一个等度区间流出时的情形,如果初始和终止流动相组成保持不变而仅有梯度的斜率发生变化时,从理论上证明了溶质保留时间(tR)与梯度斜率的倒数(1/B)之间呈线性关系。实验中以C18色谱柱为固定相,甲醇-水为流动相,联苯为样品,测定了不同流动相组成以及梯形梯度条件下的保留时间,所得到的实验值与理论值吻合,从而验证了理论方法的正确性。  相似文献

The reactions of 1-boraadamantane and 2-methyl-, 2-ethyl-, 2,2-dimethyl-, and 3,5-dimethyl-substituted 1-boraadamantanes with their 1-azaadamantane analogs afforded a series of 1:1 adducts, which are stable to atmospheric air and moisture.IH,13C, and31B NMR spectra as well as mass spectra of the compounds synthesized were investigated. Only the adduct of 2,2-dimethyl-1-boraadamantane with 2,2-dimethyl-1-azaadamantane readily dissociates into the initial components due to steric hindrances that prevent strong B→N coordination. The structure and geometric parameters of the 1-boraadamantane complex with 3,5-dimethyl 1-azaadamantane were established by X-ray diffraction analysis. Translated fromIzvestiya Akademii Nauk. An HPLC‐MS/MS method has been developed and validated for the determination of venlafaxine enantiomers in human plasma and applied to a pharmacokinetic study in healthy Chinese volunteers. The method was carried out on a vancomycin chiral column (5 µm, 250 × 4.6 mm) maintained at 25°C. The mobile phase was methanol–water containing 30 mmol/L ammonium acetate, pH 3.3 adjusted with aqueous ammonia (8:92, v/v) at the flow rate 1.0 mL/min. A tandem mass spectrometer with an electrospray interface was operated in the multiple reaction monitoring mode to detect the selected ions pair at m/z 278.0 → 120.8 for venlafaxine enantiomers and m/z 294.8 → 266.7 for estazolanm (internal standard). The method was linear in the concentration range of 0.28–423.0 ng/mL. The lower limit of quantification was 0.28 ng/mL. The intra‐and inter‐day relative standard deviations were less than 9.7%. The method was successfully applied for the evaluation of pharmacokinetic profiles of venlafaxine enantiomers in 18 healthy volnteers. Validation parameters such as the specificity, linearity, precision, accuracy and stability were evaluated, giving results within the acceptable range. Pharmacokinetic parameters of the venlafaxine enantiomers were measured in the 18 healthy Chinese volunteers who received a single regimen with venlafaxine hydrochloride capsules. The results show that AUC(0–∞), Cmax and t1/2 between S‐venlafaxine and R‐venlafaxine are significantly different (p < 0.05). Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Summary Program temperature retention indices for fifteen nonalkane solutes have been determined by cubic splines, by other procedures found in the literature and by interpolation of the n-alkanes retention time logarithm for eleven temperature programs. A comparison in terms of variance of the differences between PTRI calculated by CS and each of the remaining methods is made for each of the eleven program runs, for each of the three stationary phases used and for many of the programs. The smallest variances obtained result when the Zenkevich, van den Dool & Kratz and Chen et al. methods are tested. The stationary phase polarity is of no relevance since it has no effect on the specific PTRI found by the different methods employed in this work.  相似文献

Chiral substituted phenylethyl-1H-tetrazoles were built-up from the corresponding carboxylic acid derivatives by a useful three-step synthesis. The procedure, that preserves the chiral center from racemization, was successfully applied to a selection of several hit compounds by conversion of the carboxylic acid moiety to the nitrile derivatives and subsequent reaction with trimethylstannyl azide, under microwave conditions. A useful application to the corresponding tetrazole analogue has been found also in the conversion of the aminoacidic moiety like (R)-N-Cbz-phenylglycine showing a wide potential synthetic application.  相似文献

The values of the gas-chromatographic indicator reflecting the capacity of analytes for self-association in pure liquids, δT b.p., were estimated for C1–C9 and C11 n-alkanols by capillary gas chromatography on a nonpolar stationary phase under isothermal conditions. The δT b.p. values ofn-alkanols, found as the difference between the boiling points measured directly and those calculated from GC data, are correlated with thermodynamic characteristics of the formation ofn-alkanol associates in pure liquids. Usingn-alkanols as analytes with insignificant temperature increments of the retention indices, it was shown that the δT b.p. values can be determined under conditions used in gas chromatography with temperature programming. In this way a single chromatographic run can be used to compare the capacities for self-association of analytes boiling over a wide temperature range. The C2–C9 n-alkanethiols, which are not associated in neat liquids, have negative δT b.p. values. An interpretation of this finding is proposed.
