A method has been developed for the synthesis of 3-acetonyl- and 3-phenacylben-zo[b]furans by cyclization of 1-arloxy-2,4-pentanediones and 1-phenyl-4-aryloxy-1,3-butanediones in polyphosphoric acid. Acylation of 3-acetonyl(phenyl-benzo-[b]furans with aliphatic acid anhydrides in the presence of perchloric acid gives the benzo[b]furo[2,3-c]pyrylium perchlorates. The recyclization of the pyrylium salts with ammonia, aqueous alkali, and morpholine has been examined.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, pp. 889–893, July, 1987.  相似文献

[reaction: see text] 2,3-Disubstituted benzo[b]furans are readily prepared under very mild reaction conditions by the palladium/copper-catalyzed cross-coupling of various o-iodoanisoles and terminal alkynes, followed by electrophilic cyclization with I2, PhSeCl, or p-O2NC6H4SCl. Aryl- and vinylic-substituted alkynes undergo electrophilic cyclization in excellent yields. Biologically important furopyridines can be prepared by this approach in high yields.  相似文献

Several 2-alkylcarbamoyl-1-alkylvinylbenzo[b]furans were designed to find a selective leukotriene B4 (LTB4) receptor antagonist. 2-(2-Alkylcarbamoyl-1-alkylvinyl)benzo[b]furans having a substituent group at the 3-position, 4-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans having a substituent group at the 3-position, and 7-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans and 3-(2-alkylcarbamoyl-1-alkylvinyl)benzo[b]furans were prepared and evaluated for LTB4 receptor (BLT1 and BLT2) inhibitory activities. (E)-3-Amino-4-[2-[2-(3,4-dimethoxyphenyl)ethylcarbamoyl]-1-methylvinyl]benzo[b]furan ((E)-17c) showed potent and selective inhibitory activity for BLT2. On the other hand, (E)-7-(2-diethylcarbamoyl-1-methylvinyl)benzo[b]furan ((E)-27a) showed potent inhibitory activity for both BLT1 and BLT2.  相似文献

The UV irradiation of substituted 2,3-diphenylbenzo[b]furans and of 2,3-diphenyl-5-t-butylfuran in n-propylamine gives 1,4-dihydro derivatives of the corresponding aromatic cyclized compounds. This reaction does not involve a photoreduction. It can be concluded from deuterium labelling experiments that H atoms of the alkyl chain of n-propylamine are incorporated in the product. When the solvent is not an unhindered primary amine, only totally aromatic photocyclized products are obtained. It is proposed that hydrogens eliminated during formation of the isolated compounds are released in a reductive form and not as radicals. This is supported by the fact that acenaphthylene is reduced when irradiated in n-propylamine together with 2,3-diphenylbenzo[b]furan under conditions which do not promote, photoreduction. This seems to be a general finding in that the irradiation of stilbene in n-propylamine to acenaphthylene is the same as that of 2,3-diphenylbenzo[b]furan.  相似文献

Novel 3-acetoacetylaminobenzo[b]furan derivatives having a modified triene system at the 3-position were synthesized starting with 3-aminobenzo[b]furans. The enol isomers, 3-[(3-hydroxybut-2-enonyl)amino]benzo[b]furans (), of the 3-acetoacetylaminobenzo[b]furans were obtained as stable isomers owing to formation of a hydrogen bonding between the enol hydroxyl group and the amidocarbonyl group. The planarity of the C-2 substituent through the C-3 side chain suggested the existence of a modified conjugational triene system in the enol compound. Cysteinyl leukotriene 1 and 2 receptor antagonistic activities for these compounds were evaluated. 2-(4-Cyanobenzoyl or ethoxycarbonyl)-3-[(2-cyano-3-hydroxybut-2-enonyl)amino]benzo[b]furans (, ) were moderately active.  相似文献

Treatment of benzyl 2-halophenyl ethers with 3 equiv of t-BuLi results in Li-halogen exchange and lithiation at benzylic methylene simultaneously. These dianions do not undergo Wittig rearrangement and can be trapped with electrophiles. Their reactions with carboxylic esters afford the corresponding 2-aryl-3-hydroxy-2,3-dihydrobenzo[b]furans as a mixture of diastereoisomers. Subsequent acid-catalyzed or mediated dehydration gives moderate to good overall yield of a variety of 2-aryl-3-substituted benzo[b]furans.  相似文献

In this paper, a fully regiocontrolled synthesis of either 2- and 3-substituted benzo[b]furans is described. Direct reaction between phenols and α-bromoacetophenones in the presence of neutral alumina yields 2-substituted benzo[b]furans with complete regiocontrol. When a basic salt such as potassium carbonate is used, the corresponding 2-oxoether is obtained. Cyclization of these latter compounds promoted by neutral alumina yields the corresponding 3-substituted benzo[b]furans. Using the former method, Moracin M and other analogues can be obtained from commercial sources in two preparative steps. DFT calculations provide reasonable reaction paths to understand the formation of 2-substituted benzo[b]furans.  相似文献

Reaction of 2-hydroxybenzaldehydes with -methylfuran (sylvan) under acid-catalysis conditions was investigated. It is shown that the reaction does not stop at the step involving formation of 2-hydroxyarylfuryl-methanes but proceeds further with opening of one of furan rings and recyclization to 3-furylbenzo-furan derivatives. The latter in turn undergo transformations that lead to new heterocyclic systems — 5,6-dihydro-2,4-dimethyl-4-(5-methyl-2-furyl)-4H-benzo[b]furo[2,3-h]cyclopenta[b]furans. A novel series of some novel 5-substituted-1,2,4-triazolo[4,3-c]8,9,10-trihydrocyclopenta/8,9,10,11,12-pentahydrocyclohepta[b]thieno[3,2-e]pyrimidin-3-thiones has been synthesized. The intermediates 4-chloro-2-substituted-5,6,7-trihydrocyclopenta/5,6,7,8,9-pentahydrocyclohepta[b]thieno[2,3-d]pyrimidines were prepared by warming 2-substituted-5,6,7-trihydrocyclopenta/5,6,7,8,9-pentahydrocyclohepta[b]thieno[2,3-d]pyrimidin-4[3H]-ones with oxalyl chloride. Thieno[2,3-d]pyrimidin-4[3H]-ones were prepared by a novel, microwave assisted, solvent free, synthetic route under basic conditions hitherto unreported in the literature from ortho amino ester of thiophene. The chloro derivatives, without further purification, were hydrazinated to yield 2-substituted-4-hydrazino-5,6,7-trihydrocyclopenta/5,6,7,8,9-pentahydrocyclohepta[b]thieno[2,3-d]pyrimidines. These compounds were cyclized with carbon disulphide to give the title compounds in quantitative yields. The final compounds were screened for antibacterial activity by Kirby Bauer's method using ampicillin as the standard against various gram positive and gram negative bacteria. All the compounds showed antibacterial activity comparable with the standard.  相似文献

The solution-phase parallel synthesis of a 121-member library of multi-substituted benzo[ b]furans is described. 2,3,5-Trisubstituted benzo[ b]furans have been prepared by the palladium-catalyzed substitution of 3-iodobenzofurans by Suzuki-Miyaura, carbonylative Suzuki, Sonogashira, Heck, and carboalkoxylation chemistry. The 3-iodobenzofurans are readily prepared in good to excellent yields by the palladium/copper-catalyzed cross-coupling of various o-iodoanisoles and terminal alkynes, followed by electrophilic cyclization with ICl.  相似文献

Benzo[b]furans were prepared in one pot based on the addition/palladium-catalyzed C-H bond functionalization of phenols with bromoalkynes. The addition reactions of phenols to bromoalkynes generated (Z)-2-bromovinyl phenyl ethers in high yields with excellent regio- and stereoselectivity. The obtained (Z)-2-bromovinyl phenyl ethers subsequently proceeded by intramolecular cyclization to afford 2-substituted benzo[b]furans in good yields through palladium-catalyzed direct C-H bond functionalizations. It is important to note that the transformation of phenols with bromoalkynes into benzo[b]furans could be carried out in one pot with a simple and efficient tandem procedure.  相似文献

Tris[4-(substituted)furan-3-yl]boroxines 2 , prepared from the corresponding 4-(substituted)-3-(trimethylsilyl) furan 1, were converted successfully to 4-(substituted)-3-(tributylstannyl)furans 3 through palladium-catalyzed cross-coupling reactions with tributylstannyl chloride. Palladium-catalyzed cross-coupling reactions of 3 with organohalides afforded 3,4-disubstituted furans 4 . Regiospecific iodination of 4-(trimethylsilyl)-3-((tributylstannyl) furan ( 3a ) gave 4-iodo-3-(trimethylsilyl)furan ( 5 ), which reacted with excess ethyl acrylate under a common Heck-condition to produce 2,3-bis(trans-ethoxycarbonylvinyl)-4-(trimethylsilyl)furan ( 6 ). A thermal 6-electrocyclic reaction followed by dehydration converted 6 into benzo[2,3-6]furan 8 . Oxidation of 2 generated the corresponding 4-substituted-3(2H)-furanones 9 .  相似文献

A novel seven-membered lactam formation method has been established by intramolecular ring closure reaction of 4-bromo-(E)-3-[(2-alkylvinyl)carbonylamino]benzo[b]furans under Heck coupling conditions. A number of furo[2,3,4-jk][2]benzazepin-4(3H)-ones, tricyclicbenzo[b]furans, have been prepared by this method and evaluated for their leukotriene B(4) (LTB(4)) receptor and poly(ADP-ribose)polymerase-1 (PARP-1) inhibitory activities.  相似文献

The synthesis and transition temperatures of a series of 5-(4-alkyl- and 4-alkoxy-phenyl)2-cyanobenzo[b]furans and a 5-(4'-alkylbiphenyl-4-yl)-2-cyanobenzo[b]furan are presented. The 2-cyanobenzo[b]furans show similar mesophase types to the analogous biphenyl and terphenyl compounds, which are obtained by replacing the benzo[b]furan unit with a phenyl ring. The transition temperatures for the 2-cyanobenzo[b]furan compounds are always higher than for their biphenyl and terphenyl counterparts, but they are much lower than for the corresponding phenylnaphthalenes. Five mesogenic benzo[b]furans without a cyano group were prepared as intermediates and these compounds have lower clearing points than their biphenyl analogues.  相似文献

6,7-Dichloro-2,3-dihydro-2-benzo[b]furancarboxylic acid derivatives having a 3,3-N,S-disubstituted-2-propenoyl group at the 5-position were prepared by alkylation of 5-(thiocarbamoyl)acetyl derivatives of the 2,3-dihydro-2-benzo[b]furancarboxylic acid ester or by acetal exchange reaction of 5-[3,3-bis(alkylthio)-2-propenoyl] derivatives. Synthesis of 5-[4 and/or 5-(di)substituted-4-thiazolin-2-ylidene]acetyl-2,3- dihydro-2-benzo[b]furancarboxylic acids was also achieved by the reaction of 2-halo-1-methoxyethyl isothiocyanate with the 5-acetyl derivative in the presence of base or through sulfide contraction of 2-[[6,7-dichloro-2-methoxycarbonyl-2,3-dihydrobenzo[b]furan-5-yl) carbonyl)-methylthio]thiazolium bromide. Some of the compounds which were synthesized showed potent natriuretic activities in rats and mice. The structure-activity relationship is also discussed.  相似文献

Various 3-alkyl-2-nitrobenzo[b]furans were synthesized from common intermediate 2-(2-nitroethyl)phenols via a hypervalent iodine-induced oxidative cyclization, with good to excellent yields. This facile route is able to efficiently functionalize 2-nitrobenzo[b]furans, which are difficult to obtain by classical methods.  相似文献

A W(CO)(5)·THF-catalyzed cycloisomerization of bicyclo[4.1.0] substrates to afford mono C4-substituted 4,5-dihydro-benzo[b]furans and -indoles is reported. The title compounds are versatile intermediates that lead to a range of fused bicycles including the cores of various furan-, benzofuran-, and indole-containing natural products. In many cases, the functionalization of the dihydro-benzo[b]furans and -indoles is orthogonal to that of the corresponding benzofurans and indoles and, thus, offers complementary approaches for synthesis.  相似文献

Abstract

The synthesis of acetic 2-{[1-methyl-2-(4-oxo-5,6,7,8-tetrahydro-4H-benzo [4,5]-thieno [2,3-d] [1,3-oxazin-2-yl)ethylidene]amino}-4,5,6,7-tetrahydrohenzo[b]thiophene ?3-carboxylic acid anhydride 5 and 2-(oxopropyl)-5,6,7,8-tetrahydro-4H-benzo-[4,5] thieno[2,3-d][1,3]oxazin-4-one 7, has been achieved in three steps from ethyl 2-amino-4,5,6,7-tetrahydrobenzo(b]thiophene-3-carboxlate 1 via the reaction with ethyl acetoacetate followed by hydrolysis and acetic anhydride-induced cyclization. The 2-substituent in compound 5 has two functional groups i.e. active methylene and acid anhydride which are suitably located for intramolecular transformation. Thermal and/or base catalyzed intramolecular cyclization of 5 afforded 2-(4-acetoxy-(hydroxyl)-2-methyl-5,6,7,8-tetrahydrobenzo[4,5] thieno[2,3-b]pyridin-3-yl)-5,6,7,8- tetrahydro-4H-benzo[4,5]thieno[2,3-d] [1,3]oxazin-4-one 10 and 9 respectively. Treatment of 5 with hydrazine hydrate, aromatic and/or heterocyclic amines induced the same intramolecular cyclization with a concomitant oxazine-pyrimidine interconversion to give 3-amino(aryl or heteryl)-2-(4-hydroxy-5,6,7,8-tetrdhydrobenzo-[4,5]thieno[2,3-b]pyridin-3-yl)-3,4,5,6,7,8-hexahydrobenzo[4,5] thieno[2,3-d] pyrimid in-4-one 11–14 respectively.  相似文献
