The time-frequency structure and dynamics of oscillatory patterns in electroencephalograms of rats is studied by means of continuous wavelet transform and the decomposition of the signal by empirical modes. A method for the automatic selection of patterns using the empirical modes is developed. The method is applied to the study of sleep spindles, and it is shown that their dynamics depends on the regularities of on-off intermittency.

Insomnia is a common sleep disorder that is closely associated with the occurrence and deterioration of cardiovascular disease, depression and other diseases. The evaluation of pharmacological treatments for insomnia brings significant clinical implications. In this study, a total of 20 patients with mild insomnia and 75 healthy subjects as controls (HC) were included to explore alterations of electroencephalogram (EEG) complexity associated with insomnia and its pharmacological treatment by using multi-scale permutation entropy (MPE). All participants were recorded for two nights of polysomnography (PSG). The patients with mild insomnia received a placebo on the first night (Placebo) and temazepam on the second night (Temazepam), while the HCs had no sleep-related medication intake for either night. EEG recordings from each night were extracted and analyzed using MPE. The results showed that MPE decreased significantly from pre-lights-off to the period during sleep transition and then to the period after sleep onset, and also during the deepening of sleep stage in the HC group. Furthermore, results from the insomnia subjects showed that MPE values were significantly lower for the Temazepam night compared to MPE values for the Placebo night. Moreover, MPE values for the Temazepam night showed no correlation with age or gender. Our results indicated that EEG complexity, measured by MPE, may be utilized as an alternative approach to measure the impact of sleep medication on brain dynamics.

Transitions in the dynamics of complex systems can be characterized by changes in the synchronization behavior of their components. Taking the human cardiorespiratory system as an example and using an automated procedure for screening the synchrograms of 112 healthy subjects we study the frequency and the distribution of synchronization episodes under different physiological conditions that occur during sleep. We find that phase synchronization between heartbeat and breathing is significantly enhanced during non-rapid-eye-movement (non-REM) sleep (deep sleep and light sleep) and reduced during REM sleep. Our results suggest that the synchronization is mainly due to a weak influence of the breathing oscillator upon the heartbeat oscillator, which is disturbed in the presence of long-term correlated noise, superimposed by the activity of higher brain regions during REM sleep.

We performed electroencephalography (EEG) for six male Wistar rats to clarify temporal behaviors at different levels of consciousness. Levels were identified both by conventional sleep analysis methods and by our novel entropy method. In our method, time-dependent pattern entropy is introduced, by which EEG is reduced to binary symbolic dynamics and the pattern of symbols in a sliding temporal window is considered. A high correlation was obtained between level of consciousness as measured by the conventional method and mean entropy in our entropy method. Mean entropy was maximal while awake (stage W) and decreased as sleep deepened. These results suggest that time-dependent pattern entropy may offer a promising method for future sleep research.

We investigate the problems that might arise when two recently developed methods for detecting interdependencies between time series using state space embedding are applied to signals of different complexity. With this aim, these methods were used to assess the interdependencies between two electroencephalographic channels from 10 adult human subjects during different vigilance states. The significance and nature of the measured interdependencies were checked by comparing the results of the original data with those of different types of surrogates. We found that even with proper reconstructions of the dynamics of the time series, both methods may give wrong statistical evidence of decreasing interdependencies during deep sleep due to changes in the complexity of each individual channel. The main factor responsible for this result was the use of an insufficient number of neighbors in the calculations. Once this problem was surmounted, both methods showed the existence of a significant relationship between the channels which was mostly of linear type and increased from awake to slow wave sleep. We conclude that the significance of the qualitative results provided for both methods must be carefully tested before drawing any conclusion about the implications of such results.

Apnea and other breathing-related disorders have been linked to the development of hypertension or impairments of the cardiovascular, cognitive or metabolic systems. The combined assessment of multiple physiological signals acquired during sleep is of fundamental importance for providing additional insights about breathing disorder events and the associated impairments. In this work, we apply information-theoretic measures to describe the joint dynamics of cardiorespiratory physiological processes in a large group of patients reporting repeated episodes of hypopneas, apneas (central, obstructive, mixed) and respiratory effort related arousals (RERAs). We analyze the heart period as the target process and the airflow amplitude as the driver, computing the predictive information, the information storage, the information transfer, the internal information and the cross information, using a fuzzy kernel entropy estimator. The analyses were performed comparing the information measures among segments during, immediately before and after the respiratory event and with control segments. Results highlight a general tendency to decrease of predictive information and information storage of heart period, as well as of cross information and information transfer from respiration to heart period, during the breathing disordered events. The information-theoretic measures also vary according to the breathing disorder, and significant changes of information transfer can be detected during RERAs, suggesting that the latter could represent a risk factor for developing cardiovascular diseases. These findings reflect the impact of different sleep breathing disorders on respiratory sinus arrhythmia, suggesting overall higher complexity of the cardiac dynamics and weaker cardiorespiratory interactions which may have physiological and clinical relevance.

BackgroundBoth the WHO and the EC recommend the use of Lnight as the primary indicator for sleep disturbance. Still, a key question for noise policy is whether the prediction of sleep quality could be improved by taking the number of events into account in addition to Lnight.ObjectivesThe current paper investigates the association between sleep quality and the number of aircraft noise events. The first aim of this study was to investigate whether, for the purpose of predicting sleep quality measured by motility, the nummer of events is adequately represented in Lnight for the purpose of predicting sleep quality measured by motility. The second aim was to investigate whether the number of events at a given Lnight has an additional predictive value. In addition, it was explored whether the total number of events should be taken into account for the production of sleep quality, or only the number of events exceeding a certain sound pressure level.MethodsThis study is based on data of a field study among 418 people living within a range of 20 km from Amsterdam Airport Schiphol. The data from this study are well suited for this purpose, since for every subject both the number and the exposure level of events are available. Sleep quality was measured by motility, derived from actimeters worn on the wrist, and by self-reported sleep quality scored on a 11-point scale. Mixed linear regression models were built in a stepwise manner to predict sleep quality during a sleep period time.ResultsThe results show that, given a certain equivalent noise level, additional information on the overall number of events does not improve the prediction of sleep quality. However, the number of events above LAmax of 60 dB was related to an increase in mean motility, indicating lower sleep quality. No effect of number of events was found on self-reported sleep quality.ConclusionsThis study suggests that the number of events is more or less adequately represented in Lnight and only the number of high noise level events may have additional effects on sleep quality as measured by motility. This may be viewed as an indication that, in addition to Lnight, the number of events with a relatively high LAmax could be used as a basis for protection against noise-induced sleep disturbance.

Scaling in nature: from DNA through heartbeats to weather.
The purpose of this report is to describe some recent progress in applying scaling concepts to various systems in nature. We review several systems characterized by scaling laws such as DNA sequences, heartbeat rates and weather variations. We discuss the finding that the exponent alpha quantifying the scaling in DNA in smaller for coding than for noncoding sequences. We also discuss the application of fractal scaling analysis to the dynamics of heartbeat regulation, and report the recent finding that the scaling exponent alpha is smaller during sleep periods compared to wake periods. We also discuss the recent findings that suggest a universal scaling exponent characterizing the weather fluctuations.

The exposure-response relationships between subjective annoyance with sleep disturbance from railway trains and road traffic noise were established from an extensive social survey by CENVR (Center for Environmental Noise and Vibration Research) in Korea. The objectives of this research are to determine the long-term effects of noise on sleep and to compare the exposure-response relationships from different noise sources with those from other studies and to elucidate the effects of some modifying factors on subjective responses to noise. From an investigation of the percentage of a highly sleep-disturbed population (%HSD) in response to railway and road traffic noise, it was found that sleep is affected more by railway noise than by road traffic noise. The effects of non-acoustical factors on the responses were examined and sensitivity was shown to be a significant modifying factor, as it pertains to subjective sleep disturbance. A comparison of the response curves from an analysis of pooled data from predominantly European surveys by Miedema and Vos [Behav. Sleep Med. 5, 1-20 (2007)] with the response curves from this survey showed more of a subjective sleep disturbance response in this survey to railway noise, whereas there was no significant difference in terms of a response to road traffic noise.

Many laboratory and field studies of the effect of noise on sleep have been performed where subjects sleep whole nights. It was suggested from our former studies that the most serious effect of noise on sleep is disturbance in falling asleep and that people have to make efforts to try to sleep in noisy situations. In this study, the effort to fall asleep was used as an index of sleep and the effect of various physical properties of sounds was examined. Subjects were asked to try to sleep listening to sounds presented with a mini-disk and they were allowed to switch off the sound after 1 h if they could not sleep. The results suggest that (1) whether subjects can sleep within 1 h after they start to try to sleep is a good index of the effect of noise on sleep and that LAeqis a good index of the effect of noise on sleep except for the sounds which have meanings such as songs and people's talk.

Different sleep stages are associated with distinct dynamical patterns in EEG signals. In this article, we explored the relationship between the sleep architecture and fractal dimension (FD) of sleep EEG. In particular, we applied the FD analysis to the sleep EEG of patients with obstructive sleep apnea-hypopnea syndrome (OSAHS), which is characterized by recurrent oxyhemoglobin desaturation and arousals from sleep, a disease which received increasing public attention due to its significant potential impact on health. We showed that the variation of FD reflects the macrostructure of sleep. Furthermore, the fast fluctuation of FD, as measured by the zero-crossing rate of detrended FD (zDFD), is a useful indicator of sleep disturbance, and therefore, correlates with apnea-hypopnea index (AHI), and hourly number of blood oxygen saturation (SpO2) decreases greater than 4%, as obstructive apnea/hypopnea disturbs sleep architecture. For practical purpose, a modified index combining zDFD of EEG and body mass index (BMI) may be useful for evaluating the severity of OSAHS symptoms.

The aim of this study was to explore and compare the effect of noise from railway and road traffic on sleep in subjects habitually exposed to nocturnal noise. Forty young and middle aged healthy subjects were studied with polysomnography (PSG) during two consecutive nights in their own bedroom. Noise measurements and recordings were conducted concurrently outside of the bedroom fac?ade as well as inside the bedroom of each participant. Different noise exposure parameters were calculated (L(p,A,eq,night), L(p,A,Fmax, night), and L(AF5,night)) and analyzed in relation to whole-night sleep parameters. The group exposed to railway noise had significantly less Rapid eye movement, (REM) sleep than the group exposed to road traffic noise. A significant association was found between the maximum level (L(p,A,Fmax, night)) of railway noise and time spent in REM sleep. REM sleep was significantly shorter in the group exposed to at least a single railway noise event above 50 dB inside the bedroom. These results, obtained in an ecological valid setting, support previous laboratory findings that railway noise has a stronger impact than road traffic noise on physiological parameters during sleep, and that the maximum noise level is an important predictor of noise effects on sleep assessed by PSG, at least for railway noise.

When subjected to alternating quiet nights (32 dB) and noise nights (equivalent levels of 47 dB) a group of 14 subjects showed an average increase in the fraction of deep sleep of about 2.5% resulting from the traffic noise. Another group of 12 subjects whose noise nights were at 60 dB had an average of 4.6% increase in deep sleep during these nights. The number of wakings also increased for both groups but, as was found before, this adapted rapidly with the number of nights. The average latency of sleep onset does not appear to be affected by the traffic noise but individual differences are great and may be of opposite sign. Latency of sleep onset and waking both show appreciable "laboratory effect" which takes longer to disappear than the one or two nights usually assumed.

Due to the ongoing discussion about the relevance of sleep studies performed in the laboratory, the aim of this study was to assess the effects of road traffic noise exposure on sleep in laboratory and in field settings. Eighteen healthy young subjects participated in the study. They were exposed to noise from road traffic in the laboratory and exposed to the same recorded traffic noise exposure in their own homes. Their sleep was evaluated with wrist actigraphs and questionnaires on sleep. No significant increase in effects of noise on sleep in the laboratory was found. The results indicate that laboratory experiments do not exaggerate effects of noise on sleep.

The development of circadian sleep-wakefulness rhythm was investigated by a longitudinal study of six normal infants. We propose an entropy based measure for the sleep/wake cycle fragmentation. Our results confirm that the sleep/wake cycle fragmentation and the sleep/wake ratio decrease, while the circadian power increases during the maturation process of infants. In addition to these expected linear trends in the variables devised to quantify sleep consolidation, circadian power and sleep/wake ratio, we found that they present infradian rhythms in the monthly range.

Recently, measuring the complexity of body movements during sleep has been proven as an objective biomarker of various psychiatric disorders. Although sleep problems are common in children with autism spectrum disorder (ASD) and might exacerbate ASD symptoms, their objectivity as a biomarker remains to be established. Therefore, details of body movement complexity during sleep as estimated by actigraphy were investigated in typically developing (TD) children and in children with ASD. Several complexity analyses were applied to raw and thresholded data of actigraphy from 17 TD children and 17 children with ASD. Determinism, irregularity and unpredictability, and long-range temporal correlation were examined respectively using the false nearest neighbor (FNN) algorithm, information-theoretic analyses, and detrended fluctuation analysis (DFA). Although the FNN algorithm did not reveal determinism in body movements, surrogate analyses identified the influence of nonlinear processes on the irregularity and long-range temporal correlation of body movements. Additionally, the irregularity and unpredictability of body movements measured by expanded sample entropy were significantly lower in ASD than in TD children up to two hours after sleep onset and at approximately six hours after sleep onset. This difference was found especially for the high-irregularity period. Through this study, we characterized details of the complexity of body movements during sleep and demonstrated the group difference of body movement complexity across TD children and children with ASD. Complexity analyses of body movements during sleep have provided valuable insights into sleep profiles. Body movement complexity might be useful as a biomarker for ASD.

Background

In humans, rapid eye movements (REM) density during REM sleep plays a prominent role in psychiatric diseases. Especially in depression, an increased REM density is a vulnerability marker for depression. In clinical practice and research measurement of REM density is highly standardized. In basic animal research, almost no tools are available to obtain and systematically evaluate eye movement data, although, this would create increased comparability between human and animal sleep studies.

Methods

We obtained standardized electroencephalographic (EEG), electromyographic (EMG) and electrooculographic (EOG) signals from freely behaving mice. EOG electrodes were bilaterally and chronically implanted with placement of the electrodes directly between the musculus rectus superior and musculus rectus lateralis. After recovery, EEG, EMG and EOG signals were obtained for four days. Subsequent to the implantation process, we developed and validated an Eye Movement scoring in Mice Algorithm (EMMA) to detect REM as singularities of the EOG signal, based on wavelet methodology.

Results

The distribution of wakefulness, non-REM (NREM) sleep and rapid eye movement (REM) sleep was typical of nocturnal rodents with small amounts of wakefulness and large amounts of NREM sleep during the light period and reversed proportions during the dark period. REM sleep was distributed correspondingly. REM density was significantly higher during REM sleep than NREM sleep. REM bursts were detected more often at the end of the dark period than the beginning of the light period. During REM sleep REM density showed an ultradian course, and during NREM sleep REM density peaked at the beginning of the dark period. Concerning individual eye movements, REM duration was longer and amplitude was lower during REM sleep than NREM sleep. The majority of single REM and REM bursts were associated with micro-arousals during NREM sleep, but not during REM sleep.

Conclusions

Sleep-stage specific distributions of REM in mice correspond to human REM density during sleep. REM density, now also assessable in animal models through our approach, is increased in humans after acute stress, during PTSD and in depression. This relationship can now be exploited to match animal models more closely to clinical situations, especially in animal models of depression.

Disturbance of sleep by train and road noises was studied through in situ physiological recordings. For the same value of Leq three times as many disturbances due to the noise from road traffic were found as there were due to the train noise. The data on sleep reactions for all the noise events does not show a better train noise adaptation than that for the road noise.
