An efficient and eco-friendly protocol for synthesizing difluoromethylated oxindoles through a visiblelight induced one-pot tandem reaction of N-arylacrylamides, difluoroacetic acid and Ph I(OAc)_2 was developed. This reaction proceeded in the absence of any additive, base, metal-catalyst and external photosensitizer, using cheap and easily available CHF_2CO_2H as the difluoromethylation reagent and bulk biomass-derived 2-Me THF as the sole solvent. 26 Examples of N-arylacrylamide substrates were investigated, and all of them successfully underwent difluoromethylation to deliver the target products in good to excellent yields.  相似文献

An unprecedented radical (phenylsulfonyl)difluoromethylation of terminal alkenes with PhSO2CF2I has been achieved by using Et3B/air as an initiator. This synthetic methodology was also used in the one-pot regioselective preparation of PhSO2CF2-substituted alkanes, and in the regio- and stereoselective preparation of PhSO2CF2-substituted alkenes with high E/Z ratio (up to > or =100:1).  相似文献

Due to its unique physical and electrophilic properties, the difluoromethyl group (−CF2H) has been playing an irreplaceable role in the field of pharmaceutical and agrochemical industry. Methods that could efficiently incorporate the difluoromethyl group into the target molecules are increasing in the recent years. Developing a stable and efficient difluoromethylating reagent is thus highly attractive. In this review, we describe the development of a nucleophilic difluoromethylation reagent [(SIPr)Ag(CF2H)], including its elemental reaction, difluoromethylation reaction with different types of electrophiles, and its application in the synthesis of a nucleophilic and an electrophilic difluoromethylthiolating reagent.  相似文献

An efficient method for the selective C‐difluoromethylation of carbon acids with the reagent TMSCF2Br has been developed. A variety of structurally diverse sp3‐ and sp‐hybridized carbon nucleophiles, including esters, amides, fluorenes, terminal alkynes, β‐ketoesters, malonates, and other activated C?H nucleophiles, could be efficiently and selectively transformed into the corresponding C‐difluoromethylated products under mild conditions. This protocol is also effective for the late‐stage difluoromethylation of pharmaceutically relevant molecules and can be readily scaled up. Moreover, ambident substrates with more than one reactive site towards difluorocarbene can be difluoromethylated orthogonally using TMSCF2Br.  相似文献

A new and efficient method for the preparation of tri- and difluoromethylsilanes using magnesium metal-mediated reductive tri- and difluoromethylation of chlorosilanes is reported using tri- and difluoromethyl sulfides, sulfoxides, and sulfones. The byproduct of the process is diphenyl disulfide. Since phenyl trifluoromethyl sulfone, sulfoxide, and sulfide are readily prepared from trifluoromethane (CF(3)H) and diphenyl disulfide, the method can be considered to be catalytic in diphenyl disulfide for the preparation of (trifluoromethyl)trimethylsilane (TMS-CF(3)) from non-ozone-depleting trifluoromethane.  相似文献

The diastereoselective nucleophilic (phenylsulfonyl)difluoromethylation and (phenylsulfonyl)monofluoromethylation of alpha-amino N-tert-butanesulfinimines (3) by using PhSO2CF2H and PhSO2CH2F reagents gave products 4 or 5 in high yields (73-99%) and with excellent diastereoselectivity (dr up to >99:1). After subsequent reductive desulfonylation and acid-catalyzed alcoholysis, compounds 4 and 5 could be readily transformed to chiral alpha-difluoromethylated or alpha-monofluoromethylated ethylenediamines in good yields.  相似文献

A palladium‐catalyzed C(sp2)?H difluoromethylation of aldehyde‐derived hydrazones using bromodifluoromethylated compounds to afford the corresponding functionalized difluoromethylketone hydrazones has been established. It is proposed that a radical/SET mechanism proceeding via a difluoroalkyl radical may be involved in the catalytic cycle. Applications of the methodology to the synthesis of α,α‐difluoro‐β‐ketoesters and α,α‐difluoroketones (RCOCF2H) have been illustrated.  相似文献

Despite a growing interest in CHF2 in medicinal chemistry, there is a lack of efficient methods for the insertion of CHF18F into druglike compounds. Herein described is a photoredox flow reaction for 18F‐difluoromethylation of N‐heteroaromatics that are widely used in medicinal chemistry. Following the two‐step synthesis for a new 18F‐difluoromethylation reagent, the photoredox reaction is completed within two minutes and proceeds by C?H activation, circumventing the need for pre‐functionalization of the substrate. The method is operationally simple and affords straightforward access to radiolabeled N‐heteroaromatics with high molar activity suitable for biological in vivo studies and clinical application.  相似文献

A new method is proposed to determine the calibration factor (CF) of methyl and ethyl peroxy radicals in a chemical amplifier. The radical source comes from the reactions of excess methane and ethane, respectively, with known concentrations of OH radicals generated by the photolysis of water vapor at 184.9 nm in air in a flow tube. This yields a mixed radical source with equal amounts of HO2 and RO2 (R = CH3, C2H5). The CF for RO2 can be derived from the CF for HO2 and an average CF for the mixed radicals. The reliability of the method was evaluated by comparing the CF ratios of RO2 to HO2 obtained from both the experiments and theoretical calculations.  相似文献

The ESR spectrum of the chain-end radical RCF2CF2* detected in Nafion perfluorinated membranes exposed to the photo-Fenton reagent was accurately simulated by an automatic fitting procedure, using as input the hyperfine coupling tensors of the two F alpha and two F beta nuclei as well as the corresponding directions of the principal values from density functional theory (DFT) calculations. An accurate fit was obtained only for different orientations of the hyperfine coupling tensors for the two F alpha nuclei, indicating a nonplanar structure about the C alpha radical center. The fitted isotropic hyperfine splittings for the two F beta nuclei in the Nafion radical, 24.9 and 27.5 G, are significantly larger than those for the chain-end radical in Teflon (15 G), implying different radical conformations in the two systems. The excellent fit indicated that the geometry and electronic structure of free radicals can be obtained not only from single-crystal ESR spectroscopy, but also, in certain cases, from powder spectra, by combination with data from DFT calculations. The optimized structures obtained by DFT calculations for the CF3CF2CF2CF2* or CF3OCF2CF2* radicals as models provided additional support for the pyramidal structure determined from the spectral fit. Comparison and analysis of calculated and fitted values for the hyperfine splittings of the two F beta nuclei suggested that the radical detected by ESR in Nafion is ROCF2CF2*, which originates from attack of oxygen radicals on the Nafion side chain. The combination of spectrum fitting and DFT is considered important in terms of understanding the hyperfine splittings from 19F nuclei and the different conformations of fluorinated chain-end-type radicals RCF2CF2* in different systems, and also for elucidating the mechanism of Nafion fragmentation when exposed to oxygen radicals in fuel cell conditions.  相似文献

Although bromodifluoromethane (BrCF2H) is a simple and readily available fluorine source, direct formation of difluoromethylated arenes with BrCF2H has not been reported. Herein, we describe an efficient method to access difluoromethylated arenes through a nickel‐catalyzed difluoromethylation of arylboronic acids with BrCF2H. The reaction exhibits high efficiency, good functional group tolerance and broad substrate scope, thus providing an efficient route for applications in drug discovery and development. Preliminary mechanistic studies reveal that a difluoromethyl radical is involved in the reaction.  相似文献

Free radical difluoromethylation of protonated heteroaromatic bases was accomplished using sodium difluoromethanesulfinate in combination with tert-butyl hydroperoxide in a two-phase system (methylene chloride–water) at room temperature. A new, stable and scalable reagent for deuteriodifluoromethylthiolation (deuterated N-difluoromethylthiophthalimide, PhthSCF2D) has been developed. This reagent can be applied for the photocatalytic radical deuteriodifluoromethylthiolation of various olefins and aldehydes (30 examples). Meanwhile, it can achieve the electrophilic deuteriodifluoromethylthiolation of a series of electrophilic substrates including electron-rich arenes, aryl/vinylboronicacids, alkynes, amines, thiols and β-ketoesters (22 examples). Some complex molecules can also be applied in both radical and electrophilic deuteriodifluoromethylthiolation using PhthSCF2D as the reagent.  相似文献

A general method for the efficient difluoromethylation of alcohols using commercially available TMSCF2Br (TMS=trimethylsilyl) as a unique and practical difluorocarbene source is developed. This method allows primary, secondary, and even tertiary alkyl difluoromethyl ethers to be synthesized under weakly basic or acidic conditions. The reaction mainly proceeds through the direct interaction between a neutral alcohol and difluorocarbene, which is different from the difluoromethylation of phenols. Moreover, alcohols containing other moieties that are also reactive toward difluorocarbene can be transformed divergently by using TMSCF2Br. This research not only solves the synthetic problem of difluorocarbene‐mediated difluoromethylation of alcohols, it also provides new insights into the different reaction mechanisms of alcohol difluoromethylation and phenol difluoromethylation with difluorocarbene species.  相似文献

Selective incorporation of a fluoroalkyl moiety to modulate the properties of an organic molecule has become a frequently used strategy in life science- and materials science-related applications. In this context, selective introduction of a (phenylsulfonyl)difluoromethyl group (PhSO2CF2) into organic molecules has attracted much attention, since the PhSO2CF2 group can be regarded as a “chemical chameleon” that can be readily transformed into difluoromethyl (CF2H), difluoromethylene (–CF2–), and difluoromethylidene (CF2) functionalities. This article overviews the recent development of (phenylsulfonyl)difluoromethylation reactions from 2003, including the nucleophilic (phenylsulfonyl)difluoromethylations with PhSO2CF2H, PhSO2CF2SiMe3 and PhSO2CF2Br reagents, free radical (phenylsulfonyl)difluoromethylations with PhSO2CF2I reagent, and electrophilic (phenylsulfonyl)difluoromethylations with a hypervalent iodine(III)-CF2SO2Ph reagent.  相似文献
