A simple and efficient method for the synthesis of ethyl 5-alkanoyl- and 5-aroyl-4-pyrone-2-carboxylates was developed, which is based on the condensation of 1-R-2-(dimethyl-aminomethylidene)butane-1,3-diones, obtained from 1,3-diketones and dimethylformamide dimethyl acetal, with diethyl oxalate in the presence of NaH in THF. Ethyl 5-acyl-4-pyrone-2-carboxylates were used in the synthesis of 6-R- and 5-RCO-comanic acids.  相似文献

Several new 1,2,3-triazolo[4,5-b][1,4]diazepines were prepared starting from 1-benzyl-1 and 1-phenethyl-4,5-diamino-1,2,3-triazole 2 (Scheme 1), by condensation reactions with β-diketones (Scheme 2), β-ketoesters (Scheme 3), and diethyl malonates (Scheme 4). In the first case we obtained compounds 3 and 4 with basic properties, while the ester function condensations gave cyclic amide derivatives 7, 8, 10, 12 and 13 with acid properties. Some N-methyl derivatives 11, 14 and 15 were obtained from the cyclic amide compounds. Most of compounds were tested for their ability to displace [3H]flunitrazepam from bovine brain membranes but no compound showed benzodiazepine receptor binding affinity.  相似文献

The reaction of α,β-unsaturated β-diketones, such as 3-(o-chloro, m-nitro, and o-nitrobenzylidene)acetylacetones (I, II, and III) with hydroxylamine hydrochloride was carried out. Among them, compound I and II in acetonitrile, methanol, and acetic acid afforded 4-(α-hydroxy, methoxy, acetoxy, and chlorobenzyl)-3,5-dimethylisoxazoles (IV-XI) in fairly good yield. On the other hand, III yielded 3-(3′,5′-dimethylpyrazolo)-5-chloroanthranil (XV) under the almost same conditions by the participation of o-nitro group.  相似文献

It has been shown that nitration with a KNO3 + H2SO4 mixture of 2,5-dimethyl- and 5-methyl-2-phenylindoles proceeds strictly selectively with the formation of 6-nitro derivatives, by reduction of which the corresponding aminoindoles were synthesized. It was established that the condensation of the aminoindoles obtained with β-diketones leads initially to enamino ketones, which in the presence of various acidic reagents are not subject to cyclization, i.e. may not be used for the synthesis of the corresponding pyrroloquinolines.  相似文献

Spiro[1,3-benzodioxole-2,3′-pyrrolidine] was synthesized in several steps from ethyl 2-ethoxycarbonyl-1, 3-benzodioxole-2-acetate, prepared by condensation of pyrocatechol with either diethyl meso-dibromosuccinate, diethyl acetylenedicarboxylate or diethyl bromo maleate. The structural factors leading to the formation of the benzodioxole rather than the benzodioxan and some possible reaction mechanisms are discussed.  相似文献

[reaction: see text] The condensation of ketones or aldehydes with sulfones was shown to give a variety of products. Condensation of 2-methylcyclohexanone with dimethyl sulfone using potassium t-butoxide as base gave useful yields of 1,2-dimethylenecyclohexane. Under the same conditions, cycloheptanone, 3-methyl-2-butanone, and 2-butanone were converted to dienes. Remarkably, these reaction conditions converted acetophenone into p-terphenyl (10%) and (E)-1,4-diphenyl-3-penten-1-one (44%). Propiophenone was converted to 2'-methyl-p-terphenyl (61%). Using alpha-tetralone produced 1-methynaphthalene and naphthalene. No reaction took place with beta-tetralone. Using diethyl sulfone with alpha-tetralone lead to pure naphthalene. Condensation of isobutyraldehyde and dimethyl sulfone using potassium t-butoxide gave isoprene in low yield. Using benzaldehyde and benzyl phenyl sulfone in N,N-dimethylacetamide gave 1,2-diphenyl-1-phenylsulfonylethylene, N,N-dimethylcinnamide, and a complex condensation product. Only 1,2-diphenyl-1-phenylsulfonylethylene was obtained when the solvent was THF.  相似文献

Stobbe condensation of 2,3-dimethoxybenzaldehyde with diethyl succinate, followed by reduction and cyclization, gave 3-carbethoxy-5,6-dimethoxy-l-tetralone (7a). Subsequent reduction, aromatization and side chain conversion yielded the key intermediate 7,8-dimethoxy-2-naphthalenecarboxaldehyde (8c). 6,7-Dimethoxybenz[g]isoquinoline (1) was prepared from 8c via condensation with nitromethane, reduction, formylation, cyclization, and aromatization reactions; the isomeric 8,9-dimethoxybenz[g]isoquinoline (2) was obtained by the condensation of 8c with the diethyl acetal of aminoacetaldehyde (12) followed by reduction, tosylation, cyclization and aromatization. The methiodides of both compounds were also prepared.  相似文献
