Based on the lipase-catalysed kinetic resolution of the silyloxyalcohol (1RS,2SR)-5 by transesterification with vinyl acetate in the presence of lipase from Pseudomonas cepacia a synthesis of both enantiomers of the β-amino acid cispentacin (1R,2S)-1 and (1S,2R)-1 using simple functional group interconversions is described.  相似文献

A new, facile preparation of enamines was achieved a two-step sequence: (i) the ready formation of an N-(-aminoalkyl)benzotriazole derivative from equimolar amounts of benzotriazole, an aldehyde, and a secondary amine, (ii) the elimination of benzotriazole from the derivative by sodium hydride in tetrahydrofuran (THF). The method provides enamines in good overall yields based on the quantity of amine used.  相似文献

(+)-(9S)-Dihydroerythronolide A, 1, is prepared in 8 steps from macrolide 3 by exploiting the conformational preferences of the (5E,11E)-diene intermediate 2. The stereocontrolled introduction of the hydroxyl groups at C6, C11, and C12 is achieved by osmylation, 2 → 13 and 15 → 1, while that at C5 is obtained by a Zn(BH4)2 reduction, 13 → 14.  相似文献

A Pd-catalyzed enantioselective synthesis of 2-methyl-3-methyleneindoline in up to 89% yield and 84% ee from racemic vinyl benzoxazinanones has been developed with the help of(R,R)-BenzP*ligand.Mechanism studies support the formation of palladacyclobutane as the key intermediate via C2 attack to π-allyl Pd complex.The β-hydride elimination provides a new reaction pathway for the palladacyclobutane.  相似文献

(±)-1-Cyclohexyl-c-2-hydroxymethyl-r-1-cyclohexanol 3, a precursor of the antimuscarinic drug Rociverine 1, was obtained diastereospecifically in very high yield, from the Grignard reaction between C6H11MgCl and an appropriately protected 2-(hydroxymethyl)cyclohexanone. The preparation of enantiomerically enriched cis diol (+)-(1R,2S)-3 and the corresponding 2-acetoxymethyl derivative (+)-(1S,2R)-12 was achieved by lipase PPL-catalyzed transesterification of racemic diol (±)-3.  相似文献

The enzymatic resolution products [(1R,4aR,8aR)-1,2,3,4,4a,5,6,7,8,8a-decahydro-5,5,8a-trimethyl-2-oxo-trans-naphthalene-1-methanol-2-ethylene acetal (8aR)-7 (98% ee) and {acetate of (1S,4aS,8aS)-1,2,3,4,4a,5,6,7,8,8a-decahydro-5,5,8a-trimethyl-2-oxo-trans-naphthalene-1-methanol-2-ethylene acetal} (8aS)-9 (>99% ee)] obtained by the lipase-catalyzed enantioselective acetylation of (±)-7 in the presence of vinyl acetate as an acyl donor were converted to the ,β-unsaturated ketones (8aR)-6 and (8aS)-6, respectively. Concise syntheses of (+)-totarol 1, (+)-podototarin 2 and (+)-sempervirol 3 were achieved based on Michael reactions between (8aS)-6 and the appropriate β-keto ester followed by aldol condensation. The first chiral syntheses of (+)-jolkinolides E 4 and D 5 were achieved from (5R,10R,12R)-12-hydroxypodocarpa-8(14)-en-13-one 15 derived from (8aR)-6.  相似文献

The first total synthesis of (3S,4S,5S,6R)-5-benzyloxy-6-hydroxy-3,4-(isopropylidendioxy)-cyclohex-1-enecarbaldehyde from d-glucose is described. The key steps of this synthesis are the stereoselective Michael addition of 2-lithio-1,3-dithiane to 3-O-benzyl-5,6-dideoxy-1,2-O-isopropylidene-6-nitro--d-xilo-hex-5-enofuranose followed by the enantioselective two-step transformation of 3-O-benzyl-5,6-dideoxy-5-C-(1,3-dithian-2-yl)-6-nitro-β-l-idofuranose into (1S,2S,3S,4S,5S,6R)-5-benzyloxy-6-hydroxy-3,4-(isopropylidendioxy)-2-nitro-cyclohexanecarbaldehyde propylene dithioacetal, which was finally converted into the target compound.  相似文献

An enantiodivergent preparation of (+)-(R)- and (−)-(S)-3-amino-4,4-dimethyl-1-phenylpyrrolidin-2-one, (R)- and (S)-9, and several derivatives, from 4,4-dimethyl-1-phenylpyrrolidin-2,3-dione, 4, and (R)- or (S)-1-phenylethylamine, (R)- or (S)-5, as the chirality transfer agents, is described. The total synthesis of optically active chanoclavine-I, an ergot alkaloid, was accomplished using palladium-catalyzed intramolecular cyclization (Heck reaction) as a key step. The conjugate ester (6) was obtained in 2 steps from optically active 4-bromotryptophan (10), and the cyclization of 6 proceeded smoothly without racemization to give the key intermediate, tricyclic tetrahydrobenz[c,d]indole derivative (7), in high yield.  相似文献

A practical synthetic method for a xanthine dehydrogenase inhibitor, (S)-(−)-BOF-4272, was established utilizing an asymmetric oxidation of diaryl sulfide BOF-4269. The oxidation of the sulfide with 1-chlorobenzotriazole carried out in the presence of 4-cyanopyridine and chiral 2-phenylcyclohexanol gave a high enantiomeric excess (73%ee). The sulfoxides in each enantiomerically pure form could be obtained by treating with alkaline hydrolysis or thermolysis of one the diastereomeric intermediate sulfonium salts (>99%de). Thus the transformation into the sulfoxides occur with virtually perfect inversion (alkaline hydrolysis) or retention (thermolysis). It is therefore possible to obtain the target sulfoxide, (S)-(−)-BOF-4272, from both the two diastereomeric sulfonium intermediates.  相似文献

A series of new 2D-layered structural rare-earth coordination polymers with the general formal [Ln(C8H4O5)(H2O)5]·(H2O)·(C8H4O5)1/2 (Ln=Eu for (1); Gd for (2); Tb for (3); Dy for (4); and Er for (5)) have been yielded by hydrothermal synthesis. The coordination polymers crystallize in monoclinic space group C/2c with a=19.838(16), b=10.529(8), c=17.752(14) Å, β=107.503(14)° for (1), with a=19.823(7), b=10.552(4), c=17.762(6) Å, β=107.443(6)° for (2), with a=19.770(4), b=10.519(2), c=17.698(4) Å, β=107.52(3)° for (3), with a=19.632(2), b=10.492(2), c=17.617(3) Å, β=107.470(12)° for (4), with a=19.648(7), b=10.480(3), c=17.598(6) Å, β=107.502(6)° for (5), respectively. And the metal ions (Ln3+) are located in nine-member coordination environment. The carboxyl groups from 5-hydroxyisophthalate chelate the metal ions to form 1D helical cation chains. It is interesting that these helical cation chains are arranged to form 2D anion–cation layers by the uncoordinated ligands' anions as template. And the luminescence properties of the rare-earth ions are studied in the paper.  相似文献

A new practical method for the enzymatic synthesis of 1,2-diols has been developed by employing a lipase catalyzed one-pot transesterification protocol. A series of substituted -acetoxyphenylethanones 3a–g have been reduced to the corresponding alcohols under mild conditions employing sodium borohydride and moist neutral alumina, and further subjected for lipase catalyzed irreversible transesterification in the same pot to give mono- and diacetate diols (R)-4 and (S)-5, which on hydrolysis afforded terminal 1,2-diols, (R)- and (S)-6 in high enantiomeric excess.  相似文献

(R)-Oxynitrilase from almonds (Prunus amygdalus) catalyzes the enantioselective addition of HCN to ethyl alkyl ketones 1 in diisopropyl ether yielding (R)-ethyl alkyl ketone cyanohydrins (R)-2, which are hydrolyzed under acid catalysis to give the -hydroxy acids (R)-3. This (R)-oxynitrilase also catalyzes the enantioselective addition in aqueous citrate buffer (50 mM, pH 4.0), as demonstrated for the preparation of (R)-methyl alkyl ketone cyanohydrins (R)-5 which are obtained in high enantiomeric excesses comparable to those in diisopropyl ether as solvent.  相似文献

(N,N-Dimethyldithiocarbamato)(n-butyl)diphenyltin(IV), n-BuPh2SnS2NMe2, crystallizes in the monoclinic space group P21/n with a 9.772(5), b 9.895(4), c 21.418(9) Å, β 95.81(3)0, V 2060 Å3 Z = 4, μ 14.4 cm−1 The structure was determined by the heavy-atom technique from 3103 independent reflections measured at room temperature on an Enraf-Nonius four-circle CAD-4 diffractometer using monochromatized Mo-K radiation and refined to a final R value of 5.8%. The tin atom is essentially four-coordinated with a weak fifth tin-sulphur bond (Sn---S(2) 3.079(1) Å) considerably longer than the other (Sn---S(1) 2.466(1) Å). A comparison with the complex n-BuPhSn(C1)S2CNEt2 (Sn---S(1) 2.454(1) Å; Sn---S(2) 2.764(1) Å) suggests that enhanced steric factors are responsible for the preferential monodentate behaviour of the dithiocarbamate ligand in the title complex.  相似文献

4,4′-Unsymmetrically substituted biphenyls can be synthesized by cross-coupling reactions of substituted aromatic organometallic reagents and aromatic halides catalyzed by palladium complexes. This two-step method from commercially available aromatic halides has been used for the synthesis of a series of donor/acceptor para-substituted biphenyls, D---C6H4---C6H4---A, where D is an electron donor group and A an electron acceptor group, which are of interest as liquid crystal precursors and as having potential in non-linear optics. Biaryls in which the donor-phenyl moiety is replaced by a 2-furyl or 2-thienyl can be synthesized similarly. The method can also be used for the convergent synthesis of previously unreported unsymmetrically substituted polyparaphenylenes (n = 3,4).  相似文献

Inokosterone, a phytoecdysone isolated from Achyranthes fauriei (Amaranthaceae), has been partially acetylated to give the 2,26-diacetate (4) which has been converted into methyl 5 - acetoxy - 4 - methylpentanoate (7), showing no apparent []D, and 2β - acetoxy - 3β,14 - dihydroxy - 5β - pregn - 7 - ene - 6,20 - dione (8). Chemical and physiochemical studies have shown the configurations at C-20 and C-22 to be R. Inokosterone has thus been concluded to be a mixture of C-25 epimers of (20R,22R) - 2β,3β,14,20,22,26 - hexahydroxy - 5β - cholest - 7 - en - 6 - one (1). After the synthesis of the model compound, a C-25 epimeric mixture of (20R,22R) - 3β,20,22,26 - tetrahydroxy - 5 - cholestane (23), inokosterone has been synthesized via (20R) - 2β,3β,14,20 - tetrahydroxy - 20 - formyl - 5β - pregn - 7 - en - 6 - one (25) by Grignard reaction with 4 - (tetrahydrofuran - 2 - yloxy) - 3 - methylbutynylmagnesium bromide (15) followed by hydrogenation and hydrolysis. The use of an NMR shift reagent with the inokosterone acetates (9, 29) and the optical activity measurement of - methylglutaric acid (3) derived from inokosterone have established that inokosterone is a 1:2 mixture of the C-25 R and S epimers.  相似文献

Dolastatin D (1), a cytotoxic cyclic depsipeptide possessing the novel β-amino acid (2R,3R)-3-amino-2-methylbutanoic acid [(2R,3R]-3] as a component, has been isolated from the Japanese sea hare Dolabella auricularia. New insights into the reaction of diethylzirconocene with an alkyne, an isocyanate, and an acyl chloride in this order are reported. The products of 5-hydroxypyrrol-2(5H)-ones are obtained in good yields after hydrolysis. It is an efficient method for the synthesis of 5-hydroxypyrrol-2(5H)-ones.  相似文献

Treatment of [Pd{CH2C(CH3)CH2}(Ph2PPy)Cl] (Ph2PPy = 2-(diphenylphosphino)pyridine) with cis-[Pd(tBuNC)2Cl2] in dichloromethane affords the mixed isocyanide-tertiary phosphine complex cis-[Pd(tBuNC)Ph2PPy)Cl2], in which the Ph2PPy is a monodentate P-donor, and [{Pd[CH2C(CH3)CH2]Cl}2]. The steric effects of the Ph2PPy bridging ligand in determining the reaction course is discussed. The complex cis-[Pd(tBuNC)(Ph2PPy)Cl2] was crystallographically characterized: P21/n, a = 15.143(2), b = 9.527(1), c = 17.517(4) Å, β = 113.96(1)°, V= 2309.4(7) Å3, Z = 4. The final R value was 0.044, Rw= 0.046 for the 3078 reflections with I > 3σ(I).  相似文献
