The reactivity of 4-aryl-1-(2-chloroethyl)azetidin-2-ones and 4-aryl-1-(3-bromopropyl)azetidin-2-ones with regard to lithium aluminium hydride has been evaluated for the first time. 4-Aryl-1-(2-chloroethyl)azetidin-2-ones were transformed into novel 1-(1-aryl-3-hydroxypropyl)aziridines through an unprecedented conversion of beta-lactams into 2,3-unsubstituted aziridine derivatives. Unexpectedly, 4-aryl-1-(3-bromopropyl)azetidin-2-ones underwent dehalogenation towards 3-aryl-3-(N-propylamino)propan-1-ols upon treatment with LiAlH(4). 1-(1-Aryl-3-hydroxypropyl)aziridines were further elaborated by means of ring opening reactions using benzyl bromide in acetonitrile towards 3-aryl-3-[N-benzyl-N-(2-bromoethyl)amino]propan-1-ols and using aluminium(iii) chloride in diethyl ether, affording 3-aryl-3-[N-(2-chloroethyl)amino]propan-1-ols.  相似文献

Reactions of 2-benzylidenemalononitrile and 2-nitro-3-phenylacrylonitrile with aryl azides in diethyl ether at room temperature gave mixtures of regioisomeric 1(3)-aryl-5-phenyl-4,5-dihydro-1(3)H-1,2,3-triazole-4,4-dicarbonitriles and 1-aryl-5(4)-phenyl-1H-1,2,3-triazole-5(4)-carbonitriles, respectively. 2-Benzylidenemalononitrile reacted with the same arylazides on heating in boiling chloroform to produce 1-aryl-2-phenylaziridine-2,2-dicarbonitriles.  相似文献

Various substituted iodoarenes underwent arylation with diethyl 2-pentenedicarboxylate under the Heck reaction conditions to give the expected diethyl 3-aryl-2-pentenedicarboxylates. Extension of this reaction to 2-iodoaniline gave the hitherto unreported ethyl quinolin-2-one-4-acetate.  相似文献

Alkyl 4-oxo-2-phenylamino-4,5-dihydrofuran-3-carboxylates were used for the preparation of alkyl 5-amino-7-aryl-2-{[aryl(hydroxy)methyl](phenyl)amino}-4,6-dicyano-1-benzofuran-3-carboxylates, 4-oxo-2-phenylamino-N-(p-tolyl)-4,5-dihydrofuran-3-carboxamide, and ethyl 4-chloro-5-formyl-2-(phenylamino)furan-3-carboxylate. [reaction: see text] The arylation of ethyl acetoacetate, ethyl benzoyl acetate, and diethyl malonate under the catalysis of CuI/L-proline in DMSO proceeds smoothly at 40-50 degrees C in the presence of Cs2CO3 to provide the 2-aryl-1,3-dicarbonyl compounds in good yields. Michael addition of ethyl N-arylmalonamates to ethyl 2-(ethoxymethylidene)-3-oxobutanoate in ethanol in the presence of triethylamine at room temperature afforded the corresponding adducts which underwent cyclization to diethyl 1-aryl-6-methyl-2-oxo-1,2-dihydropyridine-3,5-dicarboxylates in 17–65% yield. In silico evaluation of various regioisomeric 5- and 3-hydroxy-substituted alkyl 1-aryl-1H-pyrazole-4-carboxylates and their acyclic precursors yielded promising results with respect to their binding in the active site of dihydroorotate dehydrogenase of Plasmodium falciparum (PfDHODH). Consequently, four ethyl 1-aryl-5-hydroxy-1H-pyrazole-4-carboxylates and their 3-hydroxy regioisomers were prepared by two-step syntheses via enaminone-type reagents or key intermediates. The synthesis of 5-hydroxy-1H-pyrazoles was carried out using the literature protocol comprising acid-catalyzed transamination of diethyl [(dimethylamino)methylene]malonate with arylhydrazines followed by base-catalyzed cyclization of the intermediate hydrazones. For the synthesis of isomeric methyl 1-aryl-3-hydroxy-1H-pyrazole-4-carboxylates, a novel two-step synthesis was developed. It comprises acylation of hydrazines with methyl malonyl chloride followed by cyclization of the hydrazines with tert-butoxy-bis(dimethylamino)methane. Testing the pyrazole derivatives for the inhibition of PfDHODH showed that 1-(naphthalene-2-yl)-5-hydroxy-1H-pyrazole-4-carboxylate and 1-(naphthalene-2-yl)-, 1-(2,4,6-trichlorophenyl)-, and 1-[4-(trifluoromethyl)phenyl]-3-hydroxy-1H-pyrazole-4-carboxylates (~30% inhibition) were slightly more potent than a known inhibitor, diethyl α-{[(1H-indazol-5-yl)amino]methylidene}malonate (19% inhibition).  相似文献

A simple, efficient and eco-friendly procedure for the selective monohydrolysis of diethyl 2,6-dimethyl-4-aryl-4H-pyran-3,5-dicarboxylates under quaternary ammonium salt catalysis conditions is presented. The catalytic activities of various quaternary ammonium salts were investigated using different molar ratios of NaOH and water-organic solvent mixtures. The results indicate that the combination of 1.0 equivalent of tetraethyl-ammonium bromide (TEAB) with 1.2 equivalents of NaOH in a 10% water-ethanol media at 40 °C displays remarkable selectivity for the monohydrolysis of diethyl 2,6-dimethyl-4-aryl-4H-pyran-3,5-dicarboxylates. The utility of this process is demonstrated by the monohydrolysis of a series of 4-aryl-4H-pyran-3,5-dicarboxylate esters to afford the corresponding monoesters in 20-80% yields under the optimized conditions.  相似文献

Reactions of ethoxymethylidenemalonate with malonic acid N-arylamidoesters in the presence of triethylamine afforded diethyl 1-aryl-6-hydroxy-2-oxo-1,2-dihydropyridine-3,5-dicarboxylates via the cyclization of Michael adducts. In the presence of sodium ethoxide the formed adducts may be also involved into simultaneous retro-Michael reaction leading to the formation of ethyl 1-aryl-5-arylcarbamoyl-6-hydroxy-2-oxo-1,2-dihydropyridine-3-carboxylates. Direction of heterocyclization or retro-Michael reaction is determined by the acidity of the hydrogen atoms of adducts and basicity of catalyst.  相似文献

5-Pyridyl- and 5-aryl-2-furaldehydes are prepared from furaldehyde diethyl acetal in a four-step, one-pot procedure:(i) deprotonation; (2) Li to Zn transmetalation; (3) Pd-mediated cross-coupling; (4) aldehyde deprotection. Treatment of ethyl 4-aryl-6-(trifluoromethyl)-2-oxo-2H-pyran-3-carboxylates, prepared from 4,4,4-trifluorobutane-1,3-diones, PCl5 and sodium diethyl malonate, with sulfuric acid afforded the intramolecular Friedel–Crafts acylation products, 3-(trifluoromethyl)indeno[2,1-c]pyran-1,9-diones, from which 2-(trifluoromethyl)-6H-pyrano[3,4-c]quinoline-4,5-diones were obtained via the Schmidt reaction in moderate yields. In reaction with acylating agents, 2-aryl-2-methyl-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones are acylated at the methyl group and also enter into reaction with diethyl oxalate. 1-Aryl-2-phenacyl-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones undergo dehydration under the influence of concentrated sulfuric acid. On the basis of the PMR and UV spectra, it was concluded that 1-aryl-2-acetonyl(phenacyl)-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones exist in two tautomeric forms with strong intramolecular hydrogen bonds of the chelate type — enaminocarbonyl and enol.  相似文献

Diethyl 2-aryl-4-hydroxy-4-methyl-6-(thio)semicarbazonocyclohexane-1,3-dicarboxylates were synthesized, and their structure was determined by 1H and 13C NMR spectroscopy, including the HSQC technique.
