It was found that estradiol valerate could be adsorbed at a mercury electrode under open circuit. The adsorptive and electrochemical behaviors of estradiol valerate on a static mercury electrode were investigated by cyclic voltammetry, linear scan voltammetry and chronocoulometry. Based on this, a sensitive and selective adsorptive stripping square-wave voltammetric method was developed for the determination of estradiol valerate based on the optimization of solution conditions and electrochemical parameters. It was found that in a Britton-Robinson buffer solution containing 18% alcohol (pH 9.5), estradiol valerate gave a sensitive reductive peak at potential -1.29 V (vs. SCE) and the peak current was linear with the concentration of estradiol valerate in the range 2.0 x 10(-8)-2.5 x 10(-6) mol L-1. The detection limit was 1.1 x 10(-8) mol L-1. The interference of some common steroid estrogens was examined and it was found that they did not interfere in the determination of estradiol valerate in the present system.  相似文献

A biosensor based on vaseline/graphite modified with avocado tissue (Persea americana) as the source of polyphenol oxidase was developed and used for the chronoamperometric determination of paracetamol in pharmaceutical formulations. This enzyme catalyses the oxidation of paracetamol to N-acetyl-p-benzoquinoneimine whose electrochemical reduction back to paracetamol was obtained at a potential of -0.12 V. After addition of paracetamol reference solutions in glass cell and stirring for 60 s for the accumulation of N-acetyl-p-benzoquinoneimine at the electrode surface under open-circuit conditions, the current response was monitored by 120 s without stirring. The currents obtained at 70 s were proportional to the paracetamol concentration from 1.2x10(-4) to 5.8x10(-3) mol l(-1) (r=0.9927) with a detection limit of 8.8x10(-5) mol l(-1). The recovery of paracetamol from two samples ranged from 97.9 to 100.7% and a relative standard deviation lower than 0.5% for a solution containing 5.0x10(-3) mol l(-1) paracetamol in 0.10 mol l(-1) phosphate buffer solution (pH 7.0; n=10) was obtained. The results obtained for paracetamol in pharmaceutical formulations using the proposed biosensor and those obtained using a pharmacopoeial procedure are in agreement at the 95% confidence level.  相似文献

A homogeneous liquid-liquid extraction method for 36 metal ions with diethyldithiocarbamate was studied. As a result, 11 metal ions were extracted as metal-chelates. Under the experimental conditions, the maximum concentration factor was 500 (i.e., 0.1 mL of sedimented liquid phase was produced from 50 mL of aqueous phase). Moreover, the proposed method was utilized as a preconcentration method for X-ray fluorescence analysis of these metals. The recovery of each metal was ca. 97-100%. All calibration curves were linear over the range of 5.0 x 10(-7) mol L-1 to 1.0 x 10(-5) mol L-1. The detection limits were at the 10(-8) mol L-1 levels and the relative standard deviations were below 5% (5 determinations). When the proposed method was used for the determination of contaminants in a synthetic sample (Al-based alloy model) and of components in an Au-Pd alloy, the results were satisfactory.  相似文献

应用方波溶出伏安法研究了芦丁在0.1mol.L-1HAc-NaAc缓冲溶液中于玻碳电极上的电化学行为.考察了溶液酸度、沉积电位、沉积时间等因素对芦丁方波溶出伏安行为的影响,优化了实验参数.结果表明,在pH3.0的HAc-NaAc缓冲溶液中,芦丁在-0.46 V产生溶出峰,峰电流与芦丁的浓度在5.0×10-8mol.L-1~1.0×10-6mol.L-1范围内呈现良好的线性关系,相关系数0.997 99,检测限1.1×10-8mol.L-1.可用于药剂中芦丁含量的测定.  相似文献

A new method for the determination of the critical micelle concentration (CMC) of Triton X-100 in aqueous solution and beta-cyclodextrin solution by resonance Rayleigh scattering (RRS) has been developed. The method is based on the measurement of the RRS intensity of different concentration of Triton X-100 in aqueous solution and beta-cyclodextrin solution (6.0 x 10(-4) mol l(-1)). When the RRS intensities were plotted against the concentration of Triton X-100, an inflection point appeared at the Triton X-100 concentration of 5.0 x 10(-4) mol l(-1) in aqueous solution and 1.1 x 10(-3) mol l(-1) in beta-cyclodextrin solution, respectively. These values of concentration corresponded to the CMC of Triton X-100 in aqueous solution and beta-cyclodextrin solution, which also agreed closely with the results reported by surface tension and UV-Vis absorption spectrophotometry. Therefore, the present RRS method is very convenient, rapid and accurate and can be used as a new technology for the determination of CMC values of surfactants without any probe. The relationship between the RRS intensity and the concentration, aggregate state and the aggregate molecular size of Triton X-100 has been primarily discussed.  相似文献

A simple flow injection system is proposed for the determination of thiamine in pharmaceutical formulations. The determination is based on the precipitation reaction of thiamine with silicotungstic acid in acidic medium to form a thiamine silicotungstate suspension that is measured at 420 nm. Adding 0.05% (w/v) poly(ethyleneglycol) in the carrier solution (0.5 mol l(-1) hydrochloric acid), an improvement in the sensitivity, repeatability and baseline stability of the flow injection system was obtained. The calibration graph was linear in the thiamine concentration range from 5.0x10(-5) to 3.0x10(-4) mol l(-1) with a detection limit of 1.0x10(-5) mol l(-1). The relative standard deviations for ten successive measurements of 1.0x10(-4) mol l(-1) and 2.5x10(-4) mol l(-1) thiamine were less than 1% and an analytical frequency of 90 h(-1) was obtained.  相似文献

A flow injection (FI) enzymatic system, based on the use of immobilized salicylate hydroxylase in glass beads, was developed for the determination of salicylate. Salicylate hydroxylase and nicotinamide adenine dinucleotide (NADH) are used to convert salicylate to catechol. The reaction of catechol with 4-aminophenol at high pH yields a colored product which is detected spectrophotometrically at 565 nm. Ten samples of human serum containing from 5.0 x 10(-4) to 5.0 x 10(-3) mol/L added salicylate were analyzed and the recovery was determined. Eight additional serum samples containing salicylate were analyzed by the Trinder test and the proposed method. The results obtained with the 2 methods showed good agreement by the statistical Student's t-test. The relative precision of the method is about 3.4% (RSD of the mean recovery). Considering the lowest concentration analyzed, the quantitative limit of detection is about 0.2 x 10(-5) mol/L (3 x SD). The volume of the sample used was 150 microL. The proposed method was also used to analyze medicines containing acetylsalicylic acid. The results were statistically compared with those obtained through the U.S. Pharmacopoeia procedure and showed excellent agreement.  相似文献

A chemiluminescence (CL) detection for the determination of sulfite using the reaction of Ru(bipy)(3)(2+) (bipy=2,2'-bipyridyl) -SO(3)(2-)-KMnO(4) is described. The concentration of sulfite is proportional to the CL intensity from 5.0x10(-8) to 1.25x10(-4) mol l(-1). The limit of detection is 2.5x10(-8) mol l(-1) and the relative standard deviation is 4.9% for the 2x10(-5) mol l(-1) sulfite solution in six repeated measurements. This method has been successfully applied to the determination of sulfite in sugar and sulfur dioxide in air by using triethanolamine (TEA) as the absorbent material.  相似文献

A fibre-optic based on immobilized acetylcholinesterase is described and its application in the detection of carbamate and organophosphate pesticides through enzyme inhibition measurements is discussed. The bioactive component of the sensor consists of acetylcholinesterase covalently immobilized on preactivated isothiocyanate glass mixed with thymol blue indicator bound on aminopropyl glass and the sensor was constructed by packing a thin layer of the glass bead mixture at the tip of a bifurcated fibre-optic sensor head, which was then integrated with a flow-through cell. The response of the sensor to acetylcholine was highly reproducible (RSD<2%) and readily reversible. The sensor exhibited a linear response to acetylcholine in the concentration range 2.5-25 mM (r(2)=0.992). Inhibition plots obtained for test organophosphate (paraoxon) and carbamate (carbofuran) pesticides exhibited concentration-dependent behaviour and showed linear profiles in the concentration ranges 5x10(-8)-5x10(-7) M for carbofuran and 5x10(-7)-5x10(-6) M for paraoxon. The detection limits, calculated at I(10%), are 1.5x10(-8) M (3.1 ppb) and 1.1x10(-7) M (24.7 ppb) for carbofuran and paraoxon, respectively. The regeneration of paraoxon-inhibited sensor was possible using 2-pyrimidine aldoxime, while repetitive substrate injection was necessary to reactivate the carbofuran-inhibited optrode. The factors affecting the inhibition and reactivation processes were investigated.  相似文献

Alcohol dehydrogenase (ADH) has been immobilized on a nickel hexacyanoferrate modified microband gold electrode surface by a glutaraldehyde/bovine serum albumin (BSA) cross-linking procedure to provide a new amperometric sensor for the assay of ethanol. The resulting enzyme electrode exhibits excellent electrocatalysis for the oxidation of reduced nicotinamide-adenine dinucleotide (NADH). The amperometric determination is based on the electrochemical detection of NADH which is generated in the enzymatic reaction of ethanol with NAD(+) under catalysis of ADH. The influence of various experimental conditions was examined for the determination of the optimum analytical performance. The sensor responds rapidly to ethanol with a detection limit of (5.0 +/- 0.3) x 10(-7) mol 1(-1). The response current increases linearly with ethanol concentration up to 5 mmol 1(-1). The sensor remains relatively stable for about 1 week.  相似文献

A pressurized CEC (pCEC) coupled with on-column chemiluminescence (CL) detection was developed for direct determination of amino acids, which was based on the principle of an enhanced effect of Cu(II)-amino acid complexes on the CL reaction between luminol and hydrogen peroxide in alkaline solution. The effects of some important factors on pCEC separation and CL intensity were systemically investigated. Baseline separation of amino acids including L-histidine (L-His), L-threonine (L-Thr), and L-tyrosine (L-Tyr) was achieved by using a monolithic column with a mobile phase of 5.0x10(-3) mol/L phosphate buffer at pH 8.0 that contained 25% v/v methanol and 5.0x10(-4) mol/L luminol and 1.0x10(-5) mol/L Cu(II) at an applied voltage of -5 kV. The calibration curves of the analytes by plotting the peak height against corresponding concentration were linear over the range of 3.2x10(-6)-3.2x10(-4) mol/L for L-His, 4.1x10(-6)-4.1x10(-4) mol/L for L-Thr, and 6.0x10(-7)-3.0x10(-4) mol/L for L-Tyr. The LODs for L-His, L-Thr, and L-Tyr were 6.4x10(-7), 8.4x10(-7), and 3.0x10(-7) mol/L (S/N = 2), respectively. The proposed method was applied to the analysis of amino acid injection sample with satisfactory results. Mean recoveries for three amino acids were from 84.3 to 89.6%.  相似文献

A novel covalently modified glassy carbon electrode with glutamic acid has been fabricated via an electrochemical oxidation procedure and was applied to the catalytic oxidation of uric acid (UA) and ascorbic acid (AA), reducing the overpotentials by about 0.2 V and 0.3 V, respectively. Based on its strong catalytic function toward the oxidation of UA and AA, the modified electrode resolved the overlapping voltammetric response of UA and AA into two well-defined voltammetric peaks with both cyclic voltammetry (CV) and differential pulse voltammetry (DPV), which can be used for the simultaneous determination of these species in a mixture. The catalytic peak current obtained from DPV was linearly dependent on the UA and AA concentration in the range 2 x 10(-6)-4 x 10(-4) mol L-1 and 1.0 x 10(-6)-4 x 10(-4) mol L-1 with correlation coefficients of 0.996 and 0.997, respectively. The detection limits (3 delta) for UA and AA were 1.1 x 10(-6) mol L-1 and 9.2 x 10(-7) mol L-1, respectively. The modified electrode shows good sensitivity, selectivity and stability, and has been applied to the determination of UA and AA simultaneously in human urine samples with satisfactory results.  相似文献

Terbium sensitized fluorescence was used to develop a sensitive and simple spectrofluorimetric method for the determination of the anthranilic acid derivatives furosemide and mefenamic and tolfenamic acids. The method makes use of radiative energy transfer from anthranilates to terbium ions in alkaline methanolic solutions. Optimum conditions for the formation of the anthranilate-Tb3+ complexes were investigated. Under optimized conditions, the detection limits are 6 x 10(-9), 1.4 x 10(-8) and 9.0 x 10(-9) mol l-1 for furosemide, mefenamic acids and tolfenamic acid, respectively. The range of application is 2.5 x 10(-8)-5.0 x 10(-5) mol l-1 for all three drugs. The method was successfully applied to the determination of furosemide and mefenamic and tolfenamic acids in serum after extraction of the samples with ethyl acetate, evaporation of the organic layer under a stream of nitrogen at 40 degrees C and reconstitution of the residue with alkaline methanolic terbium solution prior to instrumental measurement. The mean recoveries from serum samples spiked with furosemide (5.0 x 10(-7), 2.0 x 10(-6) and 8.0 x 10(-6) mol l-1), mefenamic acid (3.0 x 10(-6), 9.0 x 10(-6) and 3.0 x 10(-5) mol l-1) and tolfenamic acid (3.1 x 10(-6), 12.5 x 10(-6) and 2.5 x 10(-5) mol l-1) were 96 +/- 8, 101 +/- 5 and 98 +/- 7%, respectively. The within-run precision (RSD) for the method for two serum samples of each drug varied from 2 to 8% and the day-to-day precision for two concentration levels varied from 2 to 13%.  相似文献

A rapid, selective, and sensitive kinetic flow-injection method for iodide content determination with amperometric detection on a platinum electrode was developed. The method is based on the catalytic effect of iodide on the Mn3+ reaction with As3+ in the presence of sulfuric acid. The calibration curve was linear in the concentration range from 5.0 x 10(-7) to 1.0 x 10(-4) mol/L iodide. The limit of detection (LOD) was found to be 5.0 x 10(-9) mol/L iodide. The relative standard deviations (RSD) were 1.68% and 3.03% for 1.0 x 10(-3) mol/L standard and 1.0 x 10(-6) mol/L iodide solution (n = 6), respectively. The method has been successfully applied for determination of iodide in waters, table salts, fodder, organic substances and human blood sera. The results were compared with those obtained by a standard AOAC (Association of Official Analytical Chemists) method, as well as with those obtained by a kinetic spectrophotometric procedure for determination of iodide.  相似文献

For the first time, a new method is described for the determination of scandium based on the cathodic adsorptive stripping of the scandium-alizarin red S complex onto a carbon paste electrode. The second-order derivative linear scan voltammograms of the complex are recorded by use of model JP-303 polarographic analyzer from 0.0 to -1.0 V (versus SCE). Optimum conditions are found to be: an acetic acid (0.36 mol l(-1))-potassium biphthalate (0.064 mol l(-1)) buffer solution (pH 4.0) containing 2.0x10(-5) mol l(-1) alizarin red S, a preconcentration potential of 0.0 V, a preconcentration time of 60 s, a rest time of 10 s and a scan rate of 100 mV s(-1). The results show that the complex can be adsorbed on the surface of a carbon paste electrode, yielding one peak at -0.58 V, corresponding to the reduction of alizarin red S in the complex at the electrode. The detection limit is found to be 6.0x10(-10) mol l(-1) for 3 min of preconcentration time. The linear range is 1.0x10(-9) to 4.0x10(-7) mol l(-1). Application of the procedure to the determination of scandium in the ore samples gave good results.  相似文献

A new ammonia optical sensor was designed using bis(acetylacetoneethylendiamine)tributylphosphin cobalt(III) tetraphenylborate complex, coated on transparent triacetylcellulose film as membrane. The change in the absorbance of the optode at the maximum wavelength of 408 nm was related to ammonia concentration in aqueous samples. A buffer solution with a pH of 9 (sodium borate-HCl) was used. The optode was fully regenerated in pH 2. The linear dynamic range for determination of ammonia was 3.3 x 10(-4) to 6.9 x 10(-3) mol l(-1) with a detection limit of 5.0 x 10(-5) mol l(-1) and a response time range of 4 - 6 min. This membrane was successfully applied for determination of ammonia in drinking water.  相似文献

A flotation spectrophotometric method for the determination of germanium with isochromatic dye ion-pairs is described. The molar ratio of germanium to rhodamine 6G to tetrabromofluorescein is 1:5:5. The apparent molar absorptivity is 5.8 x 10(5) l mol(-1) cm(-1) at 531 nm. Beer's law is obeyed over the concentration range of 5.0 x 10(-8)-1.25 x 10(-6) mol l(-1). The proposed method is sensitive and accuracy and can be applied satisfactorily to the determination of germanium in vegetables.  相似文献

The voltammetric behavior of Vitamin B(2) (VB(2)) has been studied at the gold electrode modified with a self-assembled monolayer of l-cysteine. The voltammetric responses are evaluated with respect experimental conditions, such as composition and pH of the supporting electrolyte, concentration of VB(2), accumulation potential and accumulation time. On basis of the voltammetric behavior a highly sensitive method is present for the determination of VB(2) by using linear sweep stripping volammetry. The method is suitable for the determination of VB(2) concentrations between 5.0x10(-11) and 5.0x10(-6) mol l(-1). And the detection limit can be reached to 2.5x10(-11) mol l(-1). The method is applied to determine the concentration of VB(2) in the tablets with satisfactory results.  相似文献
