麻疯油转酯化产物的高效液相色谱分析   总被引:2,自引:0,他引:2  
建立了高效液相色谱法同时测定麻疯油转酯化产物中4种主要脂肪酸甲酯的分析方法。样品经膜处理后用丙酮溶解,采用Hypersil ODS(C18)色谱柱进行分离。以乙腈为流动相进行等度洗脱,内标法定量,同时对色谱条件进行优化。结果表明,在优化的色谱条件下4种脂肪酸甲酯在10 min内得到良好的分离,标准曲线的线性相关系数均达到0.999以上,平均回收率为96%~98%,重现性相对标准偏差为7.2%~10.2%,重复性相对标准偏差为0.31%~2.02%。与气相色谱法相比,该方法具有较高的灵敏度,可用于麻疯油转酯化产物中脂肪酸甲酯含量的测定,为麻疯油转酯化反应制备生物柴油的定性定量分析提供了参考依据。  相似文献

以邻苯二甲醛与谷氨酸钠中的氨基进行柱前在线衍生化反应,采用C18色谱柱分离、荧光检测器(激发340nm,发射450nm)进行测定,建立了柱前衍生反相高效液相色谱测定鸡精中谷氨酸钠含量的方法。该方法相对标准偏差为0.69%,加标回收率为99.1%~101%,在0.10~50.0mg/L范围内,谷氨酸钠的峰面积和浓度之间的相关系数为0.9999,保留时间和峰面积的相对标准偏差分别为1.22%和0.71%,鸡精中谷氨酸钠定量下限为0.2μg/g。  相似文献

建立了用高效液相色谱法测定地胆草中绿原酸含量。选择Agilent TC-C18色谱柱(250 mm×4.6 mm,5μm),甲醇-0.1%磷酸溶液(20∶80)为流动相,流速1.0 mL/min,检测波长328 nm,进样量10μL。结果表明,绿原酸0.044 9~4.488 0μg范围内线性关系良好(r=0.999 9),平均加样回收率为99.63%,RSD为0.61%(n=9)。所用方法专属性强、快速准确,可作为地胆草中绿原酸含量的测定方法。  相似文献

为建立HPLC法测定怀菊花中没食子酸含量测定方法,采用Agilent C18柱(150 mm×4.6mm,5μm),以V(甲醇)+V(超纯水+0.1%磷酸)=25+75为流动相,流量:0.5 mL·min1;柱温为25℃,检测波长:258 nm;进样量:10μL,进行测定。结果表明,回归方程为y=3.054 2 x-2.207 1,r2=0.999 1(n=6),线性范围0~20μg,平均回收率97.28%,RSD为3.39%。样品中没食子酸质量分数为3.17 mg/g。方法快速、简便、准确、重现性较好,结果可靠,可为怀菊花中没食子酸的质量评价提供依据。  相似文献

建立了测定西沙必利片含量的HPLC法。以Kromasil Cl8柱为色谱柱、0.02mol/L磷酸二氢钾缓冲液(pH=4.5)-乙腈(体积比为70:30)为流动相,流动相的流速为0.8mL/min,检测波长为308nm。西沙必利的浓度在5-125μg/mL范围内与色谱峰面积呈线性关系,线性相关系数r=0.9998,测定结果的平均加标回收率为99.91%,相对标准偏差为0.33%。  相似文献

以麻疯树果壳为原料,采用超声波辅助法提取麻疯树果壳中的多糖。在单因素实验的基础上,利用正交实验优化提取条件,并采用还原能力、DPPH·有机自由基的清除能力、羟自由基的清除能力作为麻疯树果壳多糖的体外抗氧化作用评价的3个指标。结果表明,超声波辅助提取麻疯树果壳多糖的最佳工艺条件为超声功率160 W,提取温度80℃,提取时间1.0 h,料液比1∶25(g·m L-1),在此条件下多糖得率为5.96%。抗氧化实验表明,麻疯树果壳多糖具有良好的还原能力,且清除·OH及DPPH·的能力与浓度呈正相关。当麻疯树果壳多糖浓度为0.5 mg·m L-1时,对·OH及DPPH·的清除率分别达到70%和60%。因此,该本方法操作简便、高效,测定快速。  相似文献

The oxidation of oils and biodiesels occurs due to several factors: the quantity of double bonds and the presence of allylic and bis-allylic hydrogens. Esters (biodiesel) that have large amounts of unsaturated fatty acids are more susceptible to oxidation than saturated. The aim of this work was to analyze the thermal and oxidative stability of ethyl biodiesel from Jatropha curcas L. and beef tallow by thermogravimetric, pressure differential scanning calorimetry, and PetroOxy methods. The samples of biodiesel from beef tallow present higher oxidation stability compared to biodiesel from J. curcas. In relation to calorimetric curves of biodiesel from J. curcas and beef tallow stored by 60 days without and with antioxidant, there was verified displacement of peak temperature of the transition to higher temperatures, respectively. Just a sample of biodiesel from beef tallow stored for 60 days with 3,000 ppm of antioxidant t-butyl-hydroxyquinone was within the standard established by Brazilian National Agency of Petroleum, Natural Gas, and Biofuels (ANP). The biodiesel from beef tallow was more stable in terms of thermal and oxidative stability than biodiesel from J. curcas. The thermal and oxidative stability of biodiesel depends on its chemical structure; this corroborates the fact that the oils with a predominance of saturated fatty acids are more stable than the unsaturated.  相似文献

The present investigation aimed to evaluate the reliability of in vitro propagation methods for elite genotypes of Jatropha curcas L., that maintain genetic integrity of tissue culture (TC) regenerates among two regeneration systems developed through direct shoot bud regeneration using nodal/apical shoot segments (protocol-A) and in vitro-derived leaves (protocol-B) as explants. Random amplified polymorphic DNA (RAPD), intersimple sequence repeat (ISSR), simple sequence repeat (SSR) molecular markers, and flow cytometery (FCM) were employed to evaluate genetic homogeneity in TC-regenerates at different passages of subcultures. RAPD markers showed genetic homogeneity in fifth-generation TC-regenerates of both protocols. ISSR markers showed genetic stability of leaf regenerates (protocol-B) at 10th generation. FCM analysis of TC-regenerates at 10th generation in protocol-B and at 20th generation in both protocols, showed stability of ploidy level. SSR assessment of TC-regenerates at 20th generation in both protocols confirmed genetic homogeneity. The results confirmed the genetic stability of the TC-regenerates and demonstrated the reliability of the regeneration systems developed so far using explants of two different origins, for large-scale multiplication of elite genotypes of Jatropha.  相似文献

Ferula communis is a showy herbaceous plant typical of the Mediterranean area where it is used as a traditional medicine. The plant is a source of bioactive compounds such as daucane sesquiterpenes and prenylated coumarins. In Italy, most of phytochemical studies focused on Sardinian populations where poisonous and nonpoisonous chemotypes were found, while investigations on peninsular populations are scarce. In this work, we report the chemical characterisation of the essential oils obtained from different parts of F. communis growing in central Italy. The chemical profiles of the plant parts, as detected by GC-FID and GC-MS, were different from each other and from those reported in insular populations. Notably, α-pinene (10.5%), γ-terpinene (7.6%) and hedycariol (8.4%) were the major volatile constituents in flowers; α-pinene (55.9%), β-pinene (16.8%) and myrcene (5.9%) in fruits; β-eudesmol (12.1%), α-eudesmol (12.1%) and hedycariol (10.3%) in leaves; (E)-β-farnesene (9.5%), β-cubebene (8.2%) and (E)-caryophyllene (7.2%) in roots. The volatile profiles detected did not allow to classify the investigated central Italy population into the poisonous and nonpoisonous chemotypes previously described in Sardinia.  相似文献

通过分析不同部位万寿菊挥发性化学成分,为万寿菊的开发利用提供实验依据.采用同时蒸馏.萃取法(SDE)提取不同部位万寿菊挥发油,气相色谱法分离,质谱法鉴定结构.结果表明万寿菊花、叶、茎挥发油的含量分别为3.7%、3.5%和2.9%.在花、叶和茎挥发油中分别鉴定出40、33和35种化学成分.万寿菊不同部位挥发油的含量及其化学成分存在一定的差异,其中万寿菊花挥发油的含量最高,万寿菊花、叶、茎挥发油中柠檬烯、3,7-二甲基-1,6-辛二烯.3-醇、1-环己基-2.甲基-丙烯-2-酮和3-甲基-6-(1-甲乙基)-2-环己烯-1-酮含量较高.  相似文献

The water vapor permeability (WVP) and hardness of biodegradable nanocomposite films based on glutelin from Jatropha curcas L. were determined and the results were analyzed by response surface methodology (RSM). The effects of independent variables constituted by the plasticizer (glycerol), nanofiller (cloisite), and pH on the physical parameters were evaluated. The regression models obtained for each physical property adequately described the measurements used in the experimental design, with regression coefficients of 0.9513 and 0.9421 for WVP and hardness. The regression coefficients show that pH has the most significant effect on the WVP, whereas the cloisite content has the largest influence on the hardness of the nanocomposite film. Fourier transform infrared spectroscopy provided confirmation of the results obtained by RSM. Corroboration that the gluteline structure is in a similar form as the WVP and primarily influenced by pH was obtained using conventional and second derivative infrared spectra. However, the hardness of the film primarily depends on the cloisite content. This methodology may be useful for the design and elaboration of nanocomposite films based on J. curcas L. with specific barrier and mechanical properties.  相似文献

We apply the Riccati–Padé method and the Rayleigh–Ritz method with complex rotation to the study of the resonances of a one-dimensional well with two barriers. The model exhibits two different kinds of resonances and we calculate them by means of both approaches. While the Rayleigh–Ritz method reveals each set at a particular interval of rotation angles the Riccati–Padé method yields both of them as roots of the same Hankel determinants.  相似文献

Three new abietane diterpenes, sugikurojins D (1), E (2), and F (3), and two new abietanes which incorporate cadinane, sugikurojins G (4) and H (5) were isolated from the bark of Cryptomeria japonica. These structures were elucidated primarily by extensive NMR experiments. The structure of sugikurojin D (1) was deduced to be 6alpha-acetoxy-7beta,11-dihydroxy-12-methoxy-8,11,13-abietatriene. Sugikurojin E (2) was deduced to be 6alpha-acetoxy-7beta,12-dihydroxy-8,11,13-abietatriene. Sugikurojin F (3) was 7alpha-methoxy-8,13-abietadien-11,12-dione. Sugikurojins G (4) and H (5) had a unique skeleton incorporating an alpha-cadinol or a 1alpha-hydroxy-T-cadinol in ferruginol, respectively. Also obtained in this investigation were the known diterpenes (6-14). An antibacterial activity of ten among these against Staphylococcus aureus and Escherichia coli was inactive at the (MIC: 125 microg/ml) level. Meanwhile, in the cytotoxic activity against HL-60, compounds 4, 8, and 11 showed moderate (IC50: 4, 35.4; 8, 28.0; 11, 52.4 microM) though weak (IC50: 4, 100; 8, 80.8; 11, 100 microM) activity against HCT-15.  相似文献

用沉淀浸渍法制备了固体酸催化剂SO2-4/ZrO2-CeO2,用于催化小桐籽油脂肪酸与甲醇酯化制备生物柴油.考察了焙烧温度和CeO2负载量对催化剂活性的影响,并进行了单因素实验和动力学研究.研究表明,SO2-4/ZrO2 -CeO2有较高的催化活性,当甲醇与脂肪酸体积比2∶1,反应温度150℃,催化剂用量为脂肪酸质量的8%,反应60 min时,脂肪酸转化率可达0.940 3.动力学计算表明,该酯化反应的表观活化能为45.31 kJ/mol,动力学模型为-dcA/dt=38.371e(45310/RT C 1.44 A ).  相似文献
