A novel fluorescent method for the detection of single nucleotide polymorphism (SNP) was developed using a hairpin DNA containing nucleotide base analog pyrrolo-deoxycytidine (P-dC) as a fluorescent probe. This fluorescent probe was designed by incorporating a fluorescent P-dC into a stem of the hairpin DNA, whose sequence of the loop moiety complemented the target single strand DNA (ss-DNA). In the absence of the target ss-DNA, the fluorescent probe stays a closed configuration in which the P-dC is located in the double strand stem of the fluorescent probe, such that there is weak fluorescence, attributed to a more efficient stacking and collisional quenching of neighboring bases. In the presence of target ss-DNA, upon hybridizing the ss-DNA to the loop moiety, a stem-loop of the fluorescent probe is opened and the P-dC is located in the ss-DNA, thus resulting in strong fluorescence. The effective discrimination of the SNP, including single base mismatch ss-DNA (A, T, G) and double mismatch DNA (C, C), against perfect complementary ss-DNA was achieved by increased fluorescence intensity, and verified by thermal denaturation and circular dichroism spectroscopy. Relative fluorescence intensity had a linear relationship with the concentration of perfect complementary ss-DNA and ranged from 50 nM to 3.0 μM. The linear regression equation was F/F0 = 2.73 C (μM) + 1.14 (R = 0.9961) and the detection limit of perfect complementary ss-DNA was 16 nM (S/N = 3). This study demonstrates that a hairpin DNA containing nucleotide base analog P-dC is a promising fluorescent probe for the effective discrimination of SNP and for highly sensitive detection of perfect complementary DNA.  相似文献

A versatile method for positive-type patterning of polyimide (PI) based on a two-layer photosensitive poly(benzoxazole) (PSPBO) and poly(amic acid) (PAA) film has been developed to provide a promising material in the field of microelectronics. This patterning system consisted of a pristine PAA thick bottom-layer and a poly(o-hydroxy amide) (PHA) thin top layer with 9,9-bis[4-(tert-butoxycarbonyl-methyloxy)phenyl]fluorene (TBMPF) as a dissolution inhibitor, and (5-propylsulfonyloxyimino-5H-thiophene-2-ylidene)-(2-methylphenyl)-acetonitrile (PTMA) as a photoacid generator (PAG). The PHA and PAA were prepared from 4,4′-(hexafluoroisopropylidene)-bis(o-aminophenol) and 4,4′-oxybis(benzoic acid) derivatives, and 3,3′,4,4′-biphenyltetracarboxylic dianhydride and 4,4′-oxydianiline, respectively, in N,N-dimethylacetamide. This two-layer system based on PHA (150-nm thickness) and PAA (1.5-μm thickness) showed high sensitivity of 35 mJ/cm2 and high contrast of 10.3 when exposed to a 365 nm line (i-line), post-baked at 100 °C for 2 min, and developed in a 2.38 wt.% tetramethylammonium hydroxide aqueous solution/5 wt.% iso-propanol at 25 °C. A clear positive image of a 4-μm line-and-space pattern was printed on a film which was exposed to 100 mJ/cm2 of i-line by a contact-printing mode and fully converted to the corresponding PBO/PI pattern upon heating at 350 °C, confirmed by FT-IR spectroscopy. This two-layer system could be applied to the patterning of various PAAs.  相似文献

A capillary zone electrophoresis (CZE) method for analyzing 11 chelating agents [β-alaninediacetic acid (β-ADA), trans-1,2-diaminocyclohexane-N,N,N′,N′-tetraacetic acid (CDTA), diethylenetriaminepentaacetic acid (DTPA), ethylenediaminetetraacetic acid (EDTA), N-(2-hydroxyethyl)ethylenediamine-N,N′,N′-triacetic acid (HEDTA), N-(2-hydroxyethyl)iminodiacetic acid (HEIDA), iminodiacetic acid (IDA), methylglycinediacetic acid (MGDA), nitrilotriacetic acid (NTA), 1,3-diaminopropane-N,N,N′,N′-tetraacetic acid (PDTA) and triethylenetetraaminehexaacetic acid (TTHA)] as negatively charged copper(II) complexes has been established. Both conventional and alternative chelating agents were included in this study, because they are used side by side in industrial applications. In this study, didecyldimethylammonium bromide (DMDDAB) was successfully used as a flow reversal reagent for the first time in an aqueous CZE method based on phosphate BGE with UV spectrophotometric detection. In addition this new flow modifier was compared to common TTAB. Method development was done using a fused silica capillary (61 cm × 50 μm i.d.). The optimized BGE was a 105 mmol L−1 phosphate buffer with TTAB or DMDDAB in the concentration 0.5 mmol L−1 at pH 7.1. The measurements were done with −20 kV voltage using direct UV detection at 254 nm. In both CZE methods all 11 analyte zones were properly separated (resolutions ≥2.4), and the calibrations gave excellent correlation coefficients (≥0.998; linear range tested 0.5-2.0 mmol L−1). The limits of detection were ≤34 and ≤49 μmol L−1 with the method of DMDDAB and TTAB, respectively. A clear benefit of both methods was the short analysis time; all 11 complexes were detected in less than 6 and 5.5 min with the methods of TTAB and DMDDAB, respectively. The two methods were tested with dishwashing detergents and paper mill wastewater samples and proved to be suitable for practical use.  相似文献

Chlorophyll derivative possessing a trifluoroacetyl group at the 3-position was synthesized as a new chemosensor for alcohols and amines. Intense Qy peak of the trifluoroacetyl-chlorin (701 nm in CHCl3) showed blue shifts to 667 nm in MeOH and 665 nm in n-BuNH2 due to the formation of the corresponding hemiacetal and hemiaminal with visible color changes. Thermodynamic parameters for the complexation between trifluoroacetyl-chlorin and n-BuNH2 in CDCl3 were determined to be ΔH = −48 kJ mol−1 and ΔS = −147 J K−1 mol−1. Ratiometric fluorescence sensing of n-BuNH2 in THF was also demonstrated.  相似文献

A novel progesterone immunosensor using a colloidal gold-graphite-Teflon-tyrosinase composite biosensor as amperometric transducer is reported. A sequential competitive configuration between the analyte and progesterone labelled with alkaline phosphatase (AP) was used. Phenyl phosphate was employed as the AP-substrate and the enzyme reaction product, phenol, was oxidized by tyrosinase to o-quinone, which is subsequently reduced at −0.1 V at the biocomposite electrode. Variables such as the concentration of phenyl phosphate, the amount of antibody attached to the electrode surface, immersion time in a 2% BSA solution, working pH and incubation times in progesterone and AP conjugate were optimized. A linear calibration graph for progesterone was obtained between 0 and 40 ng mL−1 with a slope value of −82.3 nA ng−1 mL, and a detection limit of 0.43 ng mL−1. The time needed to reach the steady-state current from the addition of phenyl phosphate was 30-40 s. These analytical characteristics improve substantially those reported for other progesterone immunosensors. A lifetime of 14 days with no need to apply any regeneration procedure was also achieved. The usefulness of the immunosensor was evaluated by determining progesterone in milk samples spiked with the analyte at 5.0 and 1.5 ng mL−1 concentration levels. Following a very simple procedure, involving only sample dilution, mean recoveries (n = 7) of 98 ± 3% and 99 ± 3%, respectively, were obtained.  相似文献

Dimethyl sulfoxide (DMSO) was simply immobilized to neutral alumina via quite strong hydrogen bonding between sulfoxide oxygen and surface alumina hydroxo groups. The produced alumina-modified dimethyl sulfoxide (AMDMSO) solid phase (SP)-extractor experienced high thermal and medium stability. Moreover, the small and compact size of DMSO moiety permit high surface coverage evaluated to be 2.1 ± 0.1 mmol g−1 of alumina. Hg(II) uptake was 1.90 mmol g−1(distribution coefficient log Kd = 5.658) at pH 1.0 or 2.0, 1.68 mmol g−1 (log Kd = 4.067) at pH 3.0 or 4.0 while the metal ions Ca(II), Fe(III), Co(II), Ni(II), Cu(II), Zn(II), Cd(II) and Pb(II) showed low values 0.513-0.118 mmol g−1 (log Kd < 3.0) in the pH range 4.0-7.0. A mechanism was suggested to explain the unique uptake of Hg(II) ions by binding as neutral and chloroanionic species predominate at pH values ≤ 3.0 of a medium rich in chloride ions. A direct and fast batch separation mode was achieved successfully to retain selectively Hg(II) in presence of other eight coexisting metal ions. Thus, Hg(II) was completely retained; Ca(II), Co(II), Ni(II) and Cd(II) were not retained, while Pb(II), Cu(II), Zn(II) and Fe(III) exhibited very low percentage retention evaluated to be 0.42, 0.49, 1.4 and 5.43%, respectively. The utility of the new modified alumina sorbent for concentrating of ultratrace amounts of Hg(II) was performed by percolating 2 l of doubly distilled water, drinking tap water, and Nile river water spiked with 10 ng/l over 100 mg of the sorbent packed in a minicolumn used as a thin layer enrichment bed prior to the determination by CV-AAS. The high recovery values obtained (98.5 ± 0.5, 98.5 ± 0.5 and 103.0 ± 1.0) based on excellent enrichment factor 1000, along with a good precision (R.S.D.% 0.51-0.97%, N = 3) demonstrate the accuracy and validity of the new modified alumina sorbent for preconcentrating ultratrace amounts of Hg(II) with no matrix interference.  相似文献

An amperometric artemisinin (ARN) sensor based on the supramolecular recognition of glycosylated metalloporphyrin, which is included in the Au-nanoparticles-chitosan film coated on the glass carbon electrodes, was developed. For the improvement of the selectivity of artemisinin detection, 5,10,15,20-tetrakis[2-(2,3,4,6-tetraacetyl-β-d-glucopyranosyl)-1-O-phenyl]porphyrin (T(o-glu)PPH) metal complex [FeT(o-glu)PPCl] was synthesized and employed as a ARN-sensitive and -selective material in the amperometric sensors. The proposed [FeT(o-glu)PPCl]/Au-nanoparticles modified electrodes showed excellent selectivity and sensitivity toward ARN with respect to a number of interferents and exhibited stable current response, which can be attributed to the coordination of ARN with the [FeT(o-glu)PPCl] in the electrodes. The calibration graph obtained with the proposed sensor was linear over the range of 1.8 × 10−7-1.7 × 10−9 mol l−1, with a detection limit of 1.7 × 10−9 mol l−1 for ARN. Significant advantages of the proposed procedure over the conventional reductive electrochemical methods are the selective detection and the relatively low applied potential requirement of the ARN-sensor. The prepared sensor is applied for the determination of ARN in plant samples and the results agreed with the values obtained by the pharmacopoeia method.  相似文献

The efficiency of niobium(V) oxide as a sorbent phase for solid phase microextraction (SPME) was investigated. The thin glass-ceramic rod was coated with niobium(V) oxide using chemical vapor deposition and Nb2O5 as a chemical precursor. Optimum conditions for the preparation and conditioning of the fibers are presented. The fibers were used for the extraction of a mixture of alcohols and a mixture of phenols from the headspace samples. The results obtained proved the suitability of niobium(V) oxide as a new SPME fiber. The calibration graphs for alcohols and phenols in a concentration range of 50-1000 μg l−1 were linear (r > 0.995) and the detection limits were below 0.8 μg l−1 level. The repeatability for one fiber (n = 6) under similar conditions was between 3 and 10.4%. The fiber-to-fiber reproducibility (n = 6) was between 5 and 15%.  相似文献

A new strategy to 6-hydroxy-2-naphthoic acid (HNPA) and 4-hydroxybenzoic acid from 2,6-diisopropylnaphthalene and p-cymene, respectively, was developed using the NHPI-catalyzed aerobic oxidation as a principal reaction. 2,6-Diisopropylnaphthalene was oxidized by the oxidation with O2 (1 atm) by NHPI (10 mol %) combined with Co(OAc)2 (0.5 mol %) to give 6-acetyl-2-isopropylnaphthalene, which then was converted to 6-isopropyl-2-naphthoic acid under O2 (1 atm) in the presence of Co(OAc)2 (0.5 mol %) and Mn(OAc)2 (0.5 mol %). Esterification of the resulting acid followed by the aerobic oxidation produced methyl 6-hydroxy-2-naphthoate whose hydrolysis led to the desired HNPA. An alternative route involves the oxidation of 6-acetyl-2-isopropylnaphthalene to 6-acetyl-2-naphthol on which subsequent oxidation and deacetylation gave HNPA. This method was successfully extended to the synthesis of 4-hydroxybenzoic acid from p-cymene.  相似文献

A chemically modified electrode (CME) was prepared and studied as a potentiometric sensor for the end-point detection in the automatic titration of vanadium(V) with EDTA. The CME was constructed with a paste prepared by mixing spectral-grade graphite powder, Nujol oil and N-2-naphthoyl-N-p-tolylhydroxamic acid (NTHA). Buffer systems, pH effects and the concentration range were studied. Interference ions were separated by applying a liquid-liquid extraction procedure.The CME did not require any special conditioning before using. The electrode was constructed with very inexpensive materials and was easily made. It could be continuously used, at least two months without removing the paste.Automatic potentiometric titration curves were obtained for V(V) within 5 × 10−5 to 2 × 10−3 M with acceptable accuracy and precision. The developed method was applied to V(V) determination in alloys for hip prothesis.  相似文献

Plasticized membranes using 1-phenyl-3-(2-thiazolyl)-2-thiourea (PTT) and 1-phenyl-3-(2-thiazolyl)-2-urea (PTU) have been prepared and explored as ytterbium ion-selective sensors. Effect of various plasticizers, viz. chloronaphthalene (CN), o-nitrophenyloctyl ether (o-NPOE), dibutylphthalate (DBP), dioctylsebacate (DOS) and anion excluders, sodium tetraphenylborate (NaTPB) and oleic acid (OA) was studied and improved membrane performance was observed. Optimum performance was noted with membrane of PTT having composition of PTT (3.5):PVC (80):DOS (160):NaTPB (1.5) in mg. The sensor works satisfactorily in the concentration range 1.2 × 10−7 to 1.0 × 10−2 M (detection limit 5.5 × 10−8 M) with a Nernstian slope of 19.7 mV decade−1 of activity. Wide pH range (3.0-8.0), fast response time (10 s), non-aqueous tolerance (up to 20%) and adequate shelf life (12 weeks) indicate the vital utility of the proposed sensor. The proposed electrode comparatively shows good selectivity for Yb3+ ion with respect to alkali, alkaline earth, transition and rare earth metals ions and can be used for its determination in binary mixtures and sulfite determination in white and red wine samples.  相似文献

Cholesterol-modified chitosan conjugate with succinyl linkages (CHCS) was synthesized and characterized by fourier transform infrared (FTIR) and proton nuclear magnetic resonance (1H NMR). The degree of substitution (DS) of cholesterol moiety determined by elemental analysis was 7.3%. The self-aggregation behavior of CHCS was evaluated by the fluorescence probe technique and the critical aggregation concentration (CAC) was 1.16 × 10−2 mg mL−1 in 0.1 M acetic acid solution. CHCS formed monodisperse self-aggregated nanoparticles with a roughly spherical shape and a mean diameter of 417.2 nm by probe sonication in aqueous media. Epirubicin (EPB), as a model anticancer drug, was physically entrapped inside CHCS self-aggregated nanoparticles by the remote loading method and the characteristics of EPB-loaded CHCS self-aggregated nanoparticles were analyzed using dynamic laser light scattering (DLLS), transmission electron microscopy (TEM) and fluorescence spectroscopy. EPB-loaded CHCS self-aggregated nanoparticles were almost spherical in shape and their size increased from 338.2 to 472.9 nm with the EPB-loading content increasing from 7.97% to 14.0%. The release behavior of EPB from CHCS self-aggregated nanoparticles was studied in vitro by dialysis method. The results showed that EPB release rate decreased with the pH increase of the release media. In phosphate buffered saline (PBS, pH 7.4), the EPB release was very slow and the total release amount was about 24.9% in 48 h.  相似文献

A novel fluorescence (FL) reaction for N-terminal Gly-containing peptides has been developed using 3,4-dihydroxyphenylacetic acid (3,4-DHPAA). The reaction of the peptides with 3,4-DHPAA was carried out in borate buffer (pH 8.0) in the presence of sodium periodate at 37 °C for 10 min, and the FL was measured with a spectrofluorimeter at the excitation and emission wavelengths of 370 nm and 465 nm, respectively. The 3,4-DHPAA reagent generated particularly strong FL for peptides containing Gly at their N-termini. When various other bio-substances, such as amino acids, sugars, nucleic bases, nucleotides, and proteins, were reacted with 3,4-DHPAA, no FL was observed. Under optimized reaction conditions, the lower detection limit of 0.25 μmol L−1 was obtained for the N-terminal Gly-containing peptides of Gly-Pro (GP) and Gly-Pro-Pro (GPP), which gave 3 times greater FL intensity than that observed for the reagent blank. The proposed reaction with 3,4-DHPAA as a fluorogenic reagent is selective and sensitive for the detection of N-terminal Gly-containing peptides, and therefore, this method could be a useful tool for the determination of these particular oligopeptides.  相似文献

In this study a new method for determination of cadmium in alcohol fuel using Moringa oleifera seeds as a biosorbent in an on-line preconcentration system coupled to flame atomic absorption spectrometry (FAAS) was developed. Flow and chemical variables of the proposed system were optimized through multivariate designs. The limit of detection for cadmium was 5.50 μg L−1 and the precision was below 2.3% (35.0 μg L−1, n = 9). The analytical curve was linear from 5 to 150 μg L−1, with a correlation coefficient of 0.9993. The developed method was successfully applied to spiked alcohol fuel, and accuracy was assessed through recovery tests, with recovery ranging from 97.50 to 100%.  相似文献

A novel biomimetic sensor for rutin determination based on a dinuclear complex [MnIIIMnII(Ldtb)(μ-OAc)2]BPh4 containing an unsymmetrical dinucleating ligand, 2-[N,N-bis(2-pyridylmethyl)-aminomethyl]-6-[N-(3,5-di-tert-butyl-2-oxidoben-zyl)-N-(2-pyridylamino)aminomethyl]-4-methylphenol (H2Ldtb), as a manganese peroxidase mimetic was developed. Several parameters were investigated to evaluate the performance of the biomimetic sensor obtained after the incorporation of the dinuclear complex in a carbon paste. The best performance was obtained in 75:15:10% (w/w/w) of the graphite powder:Nujol:MnIIIMnII complex, 0.1 mol L−1 phosphate buffer solution (pH 6.0) and 4.0 × 10−5 mol L−1 hydrogen peroxide. The response of the sensor towards rutin concentration was linear using square wave voltammetry in the range of 9.99 × 10−7 to 6.54 × 10−5 mol L−1 (r = 0.9998) with a detection limit of 1.75 × 10−7 mol L−1. The recovery study performed with pharmaceuticals ranged from 96.6% to 103.2% and the relative standard deviation was 1.85% for a solution containing 1.0 × 10−3 mol L−1 rutin (n = 6). The lifetime of this biomimetic sensor was 200 days (at least 750 determinations). The results obtained for rutin in pharmaceuticals using the biomimetic sensor and those obtained with the official method are in agreement at the 95% confidence level.  相似文献

A chelate complex of zinc(II) and 2,2′-bipyridine-6,6′-diyl bis(tert-butyl nitroxide) (bpybNO) with a metal/ligand ratio of 1/2 was structurally characterized to be [Zn(bpybNO)2][Zn(hfac)3]2, where Hhfac stands for 1,1,1,5,5,5-hexafluoropentane-2,4-dione. The magnetic susceptibility measurement indicates the presence of considerable antiferromagnetic interaction among the four S = 1/2 spins. The exchange parameter J was estimated as 2J/kB = −103(1) K, on the basis of a tetrahedral coupling model. The antiferromagnetic coupling is stronger after complexation than before. The density-functional theory calculation on related model compounds supports the present analysis and clarified the role of the zinc ion as a superexchange coupler.  相似文献

In this work, we utilized polyethyleneimine-capped silver nanoclusters (PEI-Ag nanoclusters) to develop a new fluorometric method for the determination of hydrogen peroxide and glucose with high sensitivity. The PEI-Ag nanoclusters have an average size of 2 nm and show a blue emission at 455 nm. The photostable properties of the PEI-Ag nanoclusters were examined. The fluorescence of the PEI-Ag nanoclusters could be particularly quenched by H2O2. The oxidization of glucose by glucose oxidase coupled with the fluorescence quenching of PEI-Ag nanoclusters by H2O2 can be used to detect glucose. Under optimum conditions, the fluorescence intensity quenched linearly in the range of 500 nM–100 μM with high sensitivity. The detection limit for H2O2 was 400 nM. And a linear correlation was established between fluorescence intensity (F0 − F) and concentration of glucose in the range of 1.0 × 10−6 to 1.0 × 10−5 M and 1.0 × 10−5 to 1.0 × 10−3 M with a detection limit of 8.0 × 10−7 M. The method was used for the detection of glucose in human serum samples with satisfactory results. Furthermore, the mechanism of sensitive fluorescence quenching response of Ag nanoclusters to glucose and H2O2 has been discussed.  相似文献

Facilitated transport of Am(III) in nitric acid medium using tetra(2-ethyl hexyl) diglycolamide (TEHDGA) in n-dodecane as carrier was studied. It was aimed at finding out the physico-chemical model for the transport of Am(III) using TEHDGA/n-dodecane as carrier under various experimental parameters like feed acidity, carrier concentration, varying strippant, varying membrane pore size, etc. The feed acidity and carrier concentrations were varied from 1 M to 6 M HNO3 and 0.1 M to 0.3 M TEHDGA/n-dodecane, respectively. The transport of Am(III) increased with increase in feed acidity and carrier concentration reaching maximum at 3 M HNO3 and 0.2 M TEHDGA/n-dodecane, respectively. Several stripping agents were tested and 0.1 M HNO3 was found to be the most suitable stripping agent for this system. Almost quantitative transport of Am(III) was observed at about 180 min with feed acidity of 3 M HNO3, 0.1 M HNO3 as strippant and 0.2 M TEHDGA/n-dodecane as carrier. The pore size of the membrane support was varied from 0.20 μm to 5 μm and the permeation coefficient increased with increase in pore size up to 0.45 μm (2.43 × 10−3 cm/s), and then decreased with further increase in pore size. The plot between permeation coefficient vs. (membrane thickness)−1 was linear which showed that the Am(III) transport was membrane diffusion limited. The membrane diffusion coefficient calculated from the graph was found to be 1.27 × 10−6 cm2/s and its theoretical value was 1.22 × 10−6 cm2/s. The stability of the carrier against leaching out of the membrane support as well as the integrity of membrane support was studied over a period of 30 days and was found to be satisfactory within the studied time period.  相似文献

A pyrimethanil-imprinted polymer (P1) was prepared by iniferter-mediated photografting a mixture of methacrylic acid and ethylene dimethacrylate onto homemade near-monodispersed chloromethylated polydivinylbenzene beads. The chromatographic behaviour of a column packed with these imprinted beads was compared with another column packed with irregular particles obtained by grinding a bulk pyrimethanil-imprinted polymer (P2). The comparison was made using the kinetic model of non-linear chromatography, studying the elution of the template and of two related substances, cyprodinil and mepanipyrim. Extension of the region of linearity, capacity factors for the template and the related substances, column selectivity, binding site heterogeneity, apparent affinity constant (K) and lumped kinetic association (ka) and dissociation rate constant (kd) were studied during a large interval of solute concentration, ranging between 1 and 2000 μg/ml. From the experimental results obtained, in the linearity region of solute concentration column selectivity and binding site heterogeneity remained essentially the same for the two columns, while column capacity (at 20 μg/ml, P1 = 23.1, P2 = 11.5), K (at 20 μg/ml, P1 = 8.3 × 106 M−1, P2 = 2.5 × 106 M−1) and ka (at 20 μg/ml, P1 = 3.5 μM−1 s−1, P2 = 0.47 μM−1 s−1) significantly increased and kd (at 20 μg/ml, P1 = 0.42 s−1, P2 = 0.67 s−1) decreased for the column packed with the imprinted beads. Molybdate was examined as a complex-forming additive to the CE background electrolytes (BGE) to affect the selectivity of separation of polyhydric phenols such as flavonoids (apigenin, hyperoside, luteolin, quercetin and rutin) and hydroxyphenylcarboxylic acids (ferulic, caffeic, p-coumaric and chlorogenic acid). Effects of the buffer concentrations and pH and the influence of molybdate concentration on the migration times of the analytes were investigated. In contrast to borate (which is a buffering and complex-forming agent generally used in CE at pH ≥9) molybdate forms more stable complexes with aromatic o-dihydroxy compounds and hence the complex-formation effect is observed at considerably lower pH. Model mixtures of cinnamic acid, ferulic acid, caffeic acid and 3-hydroxycinnamic acid were separated with 25 mM morpholinoethanesulfonic acid of pH 5.4 (adjusted with Tris) containing 0.15 mM sodium molybdate as the BGE (25 kV, silica capillary effective length 45 cm × 0.1 mm I.D., UV-vis detection at 280 nm). With 25 mM 2-hydroxy-3-[4-(2-hydroxyethyl)-1-piperazinyl]propanesulphonic acid/Tris of pH* 7.4 containing 2 mM sodium molybdate in aqueous 25% (v/v) methanol as the BGE mixtures of all the above mentioned flavonoids, p-coumaric acid and chlorogenic acid could be separated (the same capillary as above, UV-vis detection at 263 nm). The calibration curves (analyte peak area versus concentration) were rectilinear (r > 0.998) for ≈8-35 μg/ml of an analyte (with 1-nitroso-2-naphthol as internal standard). The limit of quantification values ranged between 1.1 mg l−1 for p-coumaric acid and 2.8 mg l−1 for quercetin. The CE method was employed for the assay of flavonoids in medicinal plant extracts. The R.S.D. values ranged between 0.9 and 4.7% (n = 3) when determining luteolin (0.08%) and apigenin (0.92%) in dry Matricaria recutita flowers and rutin (1.03%) and hyperoside (0.82%) in dry Hypericum perforatum haulm.
