A sample preparation method based on ultrasound assisted pseudo-digestion of Pb, Cu, Zn and Ni from street dust samples under ultrasonic effect has been described. Parameters influencing pseudo-digestion, such as sonication time, sample mass, particle size and solvent system were fully optimized. Final solutions obtained upon sonication were analyzed by atomic absorption spectrometry. The best conditions for metal pseudo-digestion were as follows: a 25 min sonication time, a 0.3 g sample mass (in 25 ml solvent), a particle size <63 μm and a mixture of concentrated HNO3-HClO4-HF (2:1:1, v/v/v). Analytical results for the four metals by ultrasound assisted pseudo-digestion, acid bomb and conventional wet digestion methods showed a good agreement, thus indicating the possibility of using mild conditions for sample preparation instead of intensive treatments inherent with the digestion methods. In addition, this method reduces the time required for all treatments (pseudo-digestion or digestion, heating to dryness, cooling and separation) with acid bomb and conventional wet digestion methods approximately from 9 to 1 h. The accuracy of the method was tested either by comparing obtained results with those of acid bomb and conventional wet digestion methods or by application on two standard reference materials. The average relative standard deviation of ultrasound assisted pseudo-digestion method varied between 0.9 and 1.8% for N = 12, depending on the analyte.  相似文献

Speciation of iron in milk was carried out by high performance liquid chromatography (HPLC) and electrothermal atomic absorption spectrometry (ETAAS). Milk whey was obtained and low molecular weight protein separation was performed by size exclusion chromatography (SEC) with a TSK Gel SW glass guard (Waters) pre-column and a TSK-Gel G2000 glass (Toso Haas) column. After studying water as a possible mobile phase, this mobile phase was carefully selected in order to avoid alterations of the sample and to make subsequent iron determination in the protein fractions easier by ETAAS. The proposed method is sensitive (limit of detection [LOD] and LOQ 1.4 and 4.7 μg l−1, respectively) and precise (relative standard deviation [RSD]<10%). Iron is principally found in the proteins of 3 and 76 kDa in breast milk, and it is irregularly distributed in infant formulas.  相似文献

Cadmium, lead, copper and manganese were determined in human deciduous teeth and bone ash 1400 standard reference material by electrothermal atomic absorption spectrometry (ETAAS), using a lanthanum + palladium + citric acid (CA) modifier mixture. Optimum masses and mass ratios of La, La + Pd and La + Pd + CA modifiers for analytes in bone ash 1400 sample solution were investigated. Pyrolysis and atomization temperatures of analytes in a tooth sample solution were obtained with and without modifiers. The mixture of La + Pd + CA was found to be preferable for the determination of analytes in tooth samples and bone ash 1400, dissolved in a mixture of HNO3 + H2O2. The detection limits and characteristic masses of analytes were obtained with or without modifiers based on integrated absorbance for tooth sample solution (2% m/v). The detection limits obtained with La + Pd + CA are 6,24,16 and 46 ng g?1 for Cd, Cu, Mn and Pb, respectively. Recovery tests for analytes in bone ash 1400 and a tooth solution with La and La + Pd + CA modifier mixture were studied and compared with certified and non certified values. The La + Pd + CA mixture was also applied to the determination of Cd, Pb, Cu and Mn in tooth samples.  相似文献

A slurry sampling method has been developed for the determination of Pb in marine plankton by ETAAS using a freshwater plankton certified reference material (CRM 414). Slurries were prepared in 1–3% m/v range with 1% v/v HNO3 by ultrasonic agitation for 5 min. The effects of several chemical modifiers, including Ir(NO3)2, Mg(NO3)2, Pd(NO3)2, Pd(NO3)2 + Mg(NO3)2, and Mg(NO3)2 + NH4H2PO4, were investigated for the stabilization of Pb during thermal pretreatment. Lead in slurries was effectively stabilized up to 1000 °C with Ir, Pd and Pd + Mg modifiers among which Pd + Mg provided the best results with complete atomization at 1850 °C. Firings in the presence of Ir were, problematic due to ash formation inside the atomizer. Water, dilute HNO3 and HF were examined as suspension medium. Dilute HNO3 (1–2% v/v) proved to be advantageous over water as it afforded extraction of Pb from plankton almost quantitatively in 5 min agitation. Hydrofluoric acid was the least suitable medium. Increasing HF concentration up to 5% v/v resulted in inaccuracy and substantial background absorption. Fast-heating furnace method provided comparable accuracy and precision to that of conventional-heating in slurries of CRM 414. Detection limits and characteristic masses were, respectively, 0.49 μg L− 1 and 32 pg for the conventional method and 0.62 μg L− 1 and 37 pg for the fast-heating method. However, fast-heating approach suffered from distorted peaks at high temperatures and incomplete pyrolysis of matrix at lower temperatures. Analysis of marine plankton samples for Pb was performed by using the conventional furnace program. The results showed a high correlation with those obtained by solution ICP-MS. Differences were statistically insignificant within 95% confidence interval.  相似文献

The parameters of analytical procedures developed for direct ETAAS determination of Pb in wine are discussed. Atomic absorption spectrometers based on transversal and longitudinal Zeeman effect, wall and integrated platform atomization with two main approaches: (i) measurements in the presence of modifier and (ii) measurements without using any modifier are compared. The optimal temperature programs are defined according to the pre-treatment and atomization curves constructed in the presence of different types of wines. For all investigated instrumental systems, 1:1 dilution of wine sample with 0.2 mol L−1 HNO3 is recommended. Matrix interferences observed, call for standard addition calibration method for Pb quantification in wines. The detection limit (3σ) achieved for wine diluted in the ratio of 1:1 varied from 0.8 to 1.9 μg L−1 depending on the instrument used. The relative standard deviation for the concentration range of 10 to 80 μg L−1 Pb in wine is typically between 4–8%. The accuracy of the analytical procedures recommended was confirmed by comparing the results obtained with those found for wine samples previously digested with HNO3-H2O2 mixture, by added/found method and by parallel analysis using different instruments. An efficient and sensitive method for the determination of methylmercury in biological samples was developed based on acid leaching extraction of methylmercury into toluene. Methylmercury in the organic phase was determined by electrothermal atomic absorption spectrometry (ETAAS). The methylmercury signal was enhanced and the reproducibility increased by formation of certain complexes and addition of Pd-DDC modifier. The complex of methylmercury with DDC produced the optimum analytical signal in terms of sensitivity and reproducibility compared to complexes with dithizone, cysteine, 1,10-phenanthroline, and diethyldithiocarbamate. Method performance was optimized by modifying parameters such as temperature of mineralization, atomization, and gas flow rate. The limit of detection for methylmercury determination was 0.015 μg g−1 and the RSD of the whole procedure was 12% for human teeth samples (n=5) and 15.8% for hair samples (n=5). The method’s accuracy was investigated by using NIES-13 and by spiking the samples with different amounts of methylmercury. The results were in good agreement with the certified values and the recoveries were 88–95%.  相似文献

Manganese in vitamin-minerals tablets was determined by solid sampling electrothermal atomic absorption spectrometry (SS-ETAAS) using three different calibration methods, namely calibration against aqueous standards, standard addition with aqueous standards on solid samples and calibration against solid certified standards. Samples were only finely ground and introduced directly into the furnace by means of solid autosampler system without any dissolving process. Effects of different calibration techniques, temperatures and heating rates of atomization and pyrolysis steps on the accuracy and precision of the analyte elements were investigated. After optimization of the experimental parameters, there is good agreement (at 95% confidence level) between the results obtained by solid sampling and those obtained by acid digestion of samples.  相似文献

The use of boric acid as a modifier for the determination of trace amount of indium in high purity antimony by electrothermal atomic absorption is described. It was found that the negative influence of the hydrofluoric acid, used for the digestion could not be eliminated by using stabilized temperature platform furnace (STPF) alone. Due to the high dissociation energy (D0 = 506 kJ mol−1) of indium fluoride, it is difficult to dissociate in the gas phase and hence is lost. In presence of HF (used for the dissolution of antimony), the universal Pd-Mg modifier does not work satisfactorily. Additionally, rising corrosion and reduced tube lifetime were observed when the acid digested (HF-HNO3) antimony solution was injected in to the platform. Improvement in platform life and elimination of interferences were achieved by the addition of boric acid as a chemical modifier together with ruthenium coating of the platform. Corrosive changes of the transversely heated graphite atomizer (THGA) platform surface were examined by scanning electron microscopy. The standard addition method was applied. A characteristic mass of 36 pg was obtained. The detection limit of the proposed method is around 0.04 μg g−1. The developed method was applied to the determination of indium in real samples. The data obtained by this method were in good agreement with those obtained by ICP-MS.  相似文献

In the sperm nuclei selenium is present only in form of a single selenoenzyme, the sperm nuclei glutathione peroxidase (snGPx), involved in processes to secure the structural stability of the nuclear chromatin. As changes in its expression may affect sperm function, its analysis is of interest in the diagnosis of male infertility. A method has been developed which by removal of the other selenium compounds present in other sperm components and measurement of the concentration of selenium in the purified human sperm nuclei by electrothermal atomic absorption spectrometry (ETAAS) allows the quantitative analysis of this enzyme. As the purification resulted in yields in the range of only 150 μg nuclei/ml semen and the amount of purified nuclei in the sample could only be determined by weighing, the main analytical difficulty arose from the accurate determination of the sample masses. The procedure includes preparation of purified sperm nuclei, measurement of the sample mass and direct selenium analysis in the suspensions of the compact sperm nuclei without prior digestion of the matter, using a palladium matrix modifier, a spectrometer with Zeeman background correction and a graphite atomizer with L'vov platform. The detection limit for the determination of selenium was 8.4 pg. The quality control of the results by means of instrumental neutron activation analysis (INAA) showed the reliability of the selenium determination by ETAAS. The procedure proved to be suitable to analyze selenium and thus snGPx in very small amounts of purified human sperm nuclei.  相似文献

Studies were conducted to evaluate citric acid production by solid-state fermentation (SSF) using cassava bagasse as substrate employing a fungal culture of Aspergillus niger LPB 21 at laboratory and semipilot scale. Optimization of the process parameters temperature, pH, initial humidity, aeration, and nutritive composition was conducted in flasks and column fermentors. The results showed that thermal treatment of cassava bagasse enhanced fungal fermentation efficacy, resulting in 220 g of citric acid/kg of dry cassava bagasse with only treated cassava bagasse as substrate. The results obtained from the factorial experimental design in a column bioreactor showed that an aeration rate of 60 mL/min (3 mL/[g·min]) and 60% initial humidity were optimum, resulting in 265.7 g/kg of dry cassava bagasse citric acid production. This was almost 1.6 times higher than the quantities produced under unoptimized conditions (167.4 g of citric acid/kg of dry cassava bagasse). The defined parameters were transferred to semipilot scale, which showed high promise for large-scale citric acid production by SSF with cassava bagasse. Respirometry assays were carried out in order to follow indirectly the biomass evolution of the process. Citric acid production reached 220, 309, 263, and 269 g/kg of dry cassava bagasse in Erlenmeyer flasks, column fermentors, a tray bioreactor, and a horizontal drum bioreactor, respectively.  相似文献

Abstract

The application of macro- and micro-electrodes constructed using the new ionophore were tested in a range of solution compositions reflecting concentrations found in fresh waters, and containing Cl?, NO3 ?, SO4 2-, HCO3 2-, H4SiO4 and a natural humic acid. The inhibition of the electrode responses to these ions was quantified using a mixed-solution method by optimising the agreement between the measured potentials and predictions from the Nicolsky-Eisenman equation. In addition, measurements were made in separate solutions of KC1 to enable results to be compared with the literature. Apart from the results obtained for humic acid, mean selectivity coefficients for 16 macro- and 21 micro-electrode experiments are given. The results indicate inhibition of the electrode response to phosphate for all the anions in the concentration ranges of 0.05–1 mM Cl?, 0.1–1.0 mM NO3 ?, 0.1–10.0 mM HCO3 ? and 0.1–1.5 mM SO4 2- with high selectivity for HPO4 2- in the presence of both dissolved silicon and a standard humic acid. This means that the application of the electrodes to hard waters is impracticable although studies of soft waters and laboratory studies in controlled conditions, e.g. calcium phosphate precipitation experiments, are feasible.  相似文献

Various modifiers (ascorbic acid, NH4NO3, EDTA, NH4SCN and a mixture of Pd/Mg(NO3)2) are compared for the accurate determination of vanadium in natural waters by electrothermal atomic absorption spectrometry. The interferences of compounds commonly present in natural waters, such as NaCl, CaCl2, MgCl2 and FeCl3 are studied. Matrix interferences were effectively eliminated by ascorbic acid or ammonium nitrate. For comparison, the standard addition method was applied without a modifier which provided satisfactory results. The accuracy of the method was confirmed by analysis of certified reference materials of waters (‘Trace Metals in Drinking Water’ and SRM 1643e ‘Trace Elements in Water’) as well as by recoveries of vanadium spiked to tap water, mineral water, synthetic riverine and synthetic sea waters. The limits of detection and characteristic masses for ascorbic acid and ammonium nitrate as the modifiers were 1.71 and 1.56?µg?L?1 and 70 and 67?pg, respectively. Recovery was in the range of 98–105% and RSD was less than 5%.  相似文献

Rapid and sensitive determination of citric acid in fermentation media by pyrolysis mass spectrometry (Py–MS) is proposed. Owing to high specificity of this method, distinguishing the citric acid from the matrix and by-products formed in the Krebs cycle is possible. Selected ion monitoring (SIM) mode is used for quantitative measurements, in which mass to charge (m/z) values of 175 of citric acid and 138 of 3-nitroaniline as internal standard are chosen. Limit of detection (LOD) for this method has been found to be 1 ng ml−1 and the linear working range was 10 ng ml−1–100 mg ml−1. Relative standard deviation (R.S.D.) of the method for five replicates was 0.84%. Results of Py–MS are compared with those obtained by UV–vis spectrophotometric method. Also, factor analysis is used for evaluating the influence of pH, molasses concentration, time and shaker intensity on the production of citric acid by Aspergillus niger.  相似文献

A method for determination of manganese and selenium in serum by simultaneous atomic absorption spectrometry (SIMAAS) is proposed. The samples (30 mul) were diluted (1+3) to 1.0% v/v HNO(3)+0.10% w/v Triton X-100 directly in the autosampler cups. A total of 20 mug Pd+10 mug Mg(NO(3))(2) was used as chemical modifier. The pyrolysis and atomization temperatures for the simultaneous heating program were 1200 and 2300 degrees C, respectively. The addition of an oxidant mixture (15% w/w H(2)O(2)+1.0% v/v HNO(3)) and the inclusion of a low temperature pyrolysis step (400 degrees C) attenuated the build-up of carbonaceous residues onto the integrated platform. An aliquot of 15 mul of the reference or sample solution was introduced into the graphite tube and heated at 80 degrees C; subsequently, 10 mul of oxidant mixture+10 mul of chemical modifier was introduced over that aliquot and the remaining heating program steps were executed. This strategy allowed at least 250 heating cycles for each THGA tube without analytical signal deterioration. The characteristic masses for manganese (6 pg) and selenium (46 pg) were estimated from the analytical curves. The detection limits were 6.5 pg (n=20, 3delta) for manganese and 50 pg (n=20, 3delta) for selenium. The reliability of the entire procedure was checked with the analysis of serum from Seronormtrade mark Trace Elements in Serum (Sero AS) and by addition and recovery tests (97+/-9% for manganese and 96+/-7% for selenium) using five serum samples.  相似文献

Most comparative studies on the efficiency of chemical modifiers have been conducted in aqueous media. In the present work, we proposed a detailed study of the use of different chemical modifiers for direct determination of arsenic in complex organic matrices by electrothermal atomic absorption spectrometry (ETAAS). Palladium, rhodium, tungsten, silver, lanthanum and a mixture of palladium and magnesium were tested. The figures of merit used for evaluation and comparison were acquired in the optimal conditions for each modifier, established by multivariate optimization of the main variables based on Doehlert designs. Singular features were observed for the chemical behaviour of some modifiers in organic matrices compared to aqueous media, such as the worse performance of Pd + Mg modifier and no notice of severe tube corrosion from La application. Lanthanum was chosen as the best chemical modifier for the present application, according to predefined criteria. Lanthanum showed the minimum limit of detection, characteristic concentration and blank signal among all tested species and no effect of the concomitants usually present in petrochemical feedstocks. Using a 200 mg L−1 lanthanum solution as a chemical modifier, the average relative standard deviations of 7 and 16% (at 3-15 μg L−1 level) and characteristic concentrations of 0.47 and 0.77 μg L−1 for naphtha and petroleum condensates, respectively, were observed.  相似文献

应用傅立叶变换近红外漫反射光谱仪,从500个样品中按高、中、低含量挑选出具代表性的150~172个烟草样品建立了近红外光谱与烟草中的苹果酸、柠檬酸和石油醚提取物成分含量间的数学模型,用建立的模型对36个样品进行预测,结果表明,各成分近红外预测值与实测值之间的平均偏差:苹果酸为0.090,柠檬酸为0.040,石油醚提取物为0.124;且近红外预测值与化学法不存在显著性差异,近红外光谱分析技术可初步用于烟草部分香气成分的快速定量分析.  相似文献

Silver nanoparticles (AgNPs) were proposed as a new chemical modifier for the elimination of interferences when determining arsenic and antimony in aqueous NaCl or Na2SO4 solutions and in sea-water by electrothermal atomic absorption spectrometry. For this purpose, the AgNPs were prepared simply by reducing silver nitrate with sodium citrate. The effects of pyrolysis and atomization temperatures and the amounts of interferents and modifiers on the sensitivities of these elements were investigated. In the presence of the proposed modifier, a pyrolysis temperature of at least 1100 °C for arsenic and 900 °C for antimony could be applied without the loss of analytes, and the interferences were greatly reduced to allow for interference-free determination. The detection limits (N = 10, 3σ) for arsenic and antimony were 0.022 ng and 0.046 ng, respectively. AgNPs are cheaper and more available compared to many other modifiers. No background was detected, and the blank values were negligible.  相似文献

The examination of the effect of interfering ions on the analytical signal of ruthenium measured by electrothermal atomic absorption spectrometry was initially performed in this work. The complexes of ruthenium(III) with thiosemicarbazide (TSd) and acetaldehyde thiosemicarbazone (AcTSn) were prepared and imprinted in polymeric network. The ion-imprinted polymers were synthesized by copolymerization of methacrylic acid, as functional monomer and ethylene glycol dimethacrylate, as crosslinking agent in the presence of 2,2-azobisisobutyronitrile as initiator. The effects of sample volume, pH, and flow rate on the extraction of analyte were studied in dynamic mode. The optimum pH for quantitative retention of ruthenium on each of the studied sorbents was 7.5 ± 0.5. The elution of analyte was completed with 0.2 mol L−1 thiourea in 0.2 mol L−1 HCl. The effect of matrix ions on ruthenium(III) separation process was studied. The analytical performance of the Ru-TSd polymer in the presence of competing ions was better than Ru-AcTSn polymer, considering recovery of analyte, reproducibility of results, selectivity coefficients, and sorbent capacity. The detection limit of the proposed method (0.16 ng mL−1 on Ru-TSd and 0.25 ng mL−1 on Ru-AcTSn) is lower in comparison with the previously published methods. The developed separation method was successfully applied to the determination of trace amounts of ruthenium in spiked water samples, sludge, grass, and human hair.  相似文献

A comparative study of pretreatment methods for the determination of 10 elements (As, Cd, Pb, V, Ni, Mn, Cr, Cu, Fe, Al) in atmospheric aerosols by electrothermal atomic absorption spectrometry (ETAAS) was conducted. For the digestion of the particulates collected in filters, six methods were compared using a mixture of HNO3 and HF with or without the addition of various oxidative agents (HClO4 or H2O2) or acids (HCl). The comparative study was performed using loaded cellulose filter samples, which were digested in Parr bombs and heated in a conventional oven at 170 °C for 5 h. The extraction efficiency and blanks were compared and it was proved that the digestion method using only HNO3–HF extracted most of the metals and gave the lowest blanks. The HNO3–HF mixture was selected for the development of an improved microwave digestion method specific for aerosol-loaded filters. The operating parameters were optimized, so that quantitative recovery of the reference materials NIST 1649a urban dust and NIST 1648 urban particulate matter was achieved. The blank of cellulose and teflon filters were also determined and compared. Teflon filters present the lowest blanks for all the elements. The obtained limits of detection for each type of filters were adequate for environmental monitoring purposes. ETAAS instrumental operation was also optimized for the compensation and the elimination of interferences. The temperature optimization was performed for each metal in every type of filter and optimized parameters are proposed for 10 elements.  相似文献
