利用基质辅助激光解析电离飞行时间质谱技术、SDS-聚丙烯酰胺凝胶电泳技术以及Shot-gun蛋白质组学策略对贝伐珠单克隆抗体药物及其糖基化修饰进行了全面表征,共发现17种糖型并确定了特征肽段序列.利用液相色谱-串联质谱联用技术,采用平行反应检测模式,通过测定单抗水解后产生的特征肽段,实现了对大鼠血浆样品中单抗药物含量的绝对定量分析,获得不同给药计量下的贝伐珠单抗的药物浓度-时间曲线,同时实现了对单抗糖基化修饰各糖型的相对定量分析.本实验首先建立标准工作曲线,对大鼠血浆样品中的贝伐珠单抗进行定量分析,在线性范围内,相关系数R2=0.998,定量限为66 fmol,线性关系良好.对大鼠血浆样品的测定结果表明,高、低计量给药的贝伐珠单抗药物浓度-时间曲线趋势基本一致,浓度均为直接下降,与理论趋势相符.但是大多数糖型呈现浓度先上升的现象,之后的代谢情况因糖型差异而有所不同.  相似文献

The thermal inactivation kinetics of chymotrypsin, trypsin, and—-amylase bound to silica, polyacrylamide, and polystyrene were studied at different temperatures. The inactivation curves were analyzed by a kinetic model, assuming a first-order reaction of differently stable enzyme fractions on the matrix. In all cases the assumption of two enzyme fractions with distinctly different inactivation constants was sufficient for describing the inactivation progress (standard deviations between experimental and calculated inactivation curves, 1-4%). Both the inactivation constants as well as the relative concentrations of the enzyme fractions were found to change in dependence on temperature.  相似文献

Adsorption of proteins on surfaces of hydrophobic interaction chromatography media is at least a two-stage process. Application of pure protein pulses (bovine serum albumin and beta-lactoglobulin) to hydrophobic interaction chromatography media yielded two chromatographic peaks at low salt concentrations. At these salt concentrations, the adsorption process is affected by a second reaction, which can be interpreted as protein spreading or partial unfolding of the protein. The kinetic constants of the spreading reaction were derived from pulse response experiments at different residence times and varying concentrations by applying a modified adsorption model considering conformational changes. The obtained parameters were used to calculate uptake and breakthrough curves for spreading proteins. Although these parameters were determined at low saturation of the column, predictions of overloaded situations could match the experimental runs satisfactorily. Our findings suggest that proteins which are sensitive to conformational changes should be loaded at high salt concentrations in order to accelerate the adsorption reaction and to obtain steeper breakthrough curves.  相似文献

Laser-induced breakdown spectroscopy (LIBS) is demonstrated as a quantitative technique for geochemical analysis. This study demonstrates the applicability of LIBS to multielemental analysis of minerals using argon as an internal standard. Laser-induced breakdown spectroscopy has been applied to measure elements in oxide form. In the present study, the contents of several oxides, such as Fe2O3, CaO and MgO, in geological samples from the Tierga Mine (Zaragoza, Spain) were analyzed by LIBS. An argon environment was used to eliminate interference from air at atmospheric pressure. Furthermore, argon was used as an internal standard. The result was enhanced signal and enhanced linearity of the calibration curves. The Fe2O3, CaO and MgO concentrations determined by LIBS were compared with the results obtained using another analytical technique, inductively coupled plasma optical emission spectrometry (ICP-OES). The concentrations found using LIBS were in good agreement with the values obtained by ICP-OES.  相似文献

Relative peak-height ratios of products to substrates determined by MALDI-TOFMS allow the quantitative analysis of enzyme catalyzed reactions for screening purposes. Two examples were investigated: the first one was a lipase catalyzed reaction which produces 2-methoxy-N-[(1R)-1-phenylethyl]acetamide (MET) using rac-alpha-phenylethylamine (PEA) as substrate. The second one was the pyruvate decarboxylase catalyzed formation of (1R)-1-hydroxy-1-phenyl-2-propanone (PAC) with benzaldehyde (BzA) as substrate. Here the corresponding oximes were analyzed after derivatization using hydroxylamine. The standard curves (r2 = 0.985 for MET, r2 = 0.991 for PAC) were linear over two orders of magnitude for MET and PAC concentrations. After optimization of the sample preparation an average relative standard deviation of 12.5% was obtained in both cases.  相似文献

Abstract

Ionic surfactants and near-infrared laser dyes formed complexes which were extracted into organic solvents as ion pairs. Surfactants were determined spectrophotometrically by measuring the near-infrared absorbance and fluorescence of the ion pair in the organic solvent. Several of the commercially available near infrared dyes have been found suitable for surfactant determination in water using this technique. The excess near-infrared dye coextracted into the organic solvent was determined by blank extractions. The calibration curves were linear within two orders of magnitude of surfactant concentrations. Non-linear calibration curves are obtained for wider concentration range of surfactants. This method using the recently developed near-infrared laser diode intracavity technique was applied to the determination of SDS in water. Lower detection limits and ease of operation are the major advantages of using this new laser diode technique. The extraction efficiency of different solvent systems was evaluated.  相似文献

A series of thioether‐functionalised imidazolium salts have been prepared and characterized. Subsequent reaction of the thioether‐functionalised imidazolium salts with iodomethane affords imidazolium–sulfonium salts composed of doubly charged cations and two different anions. Imidazolium–sulfonium salts containing a single anion type are obtained either by a solvent extraction method or by anion exchange. The imidazolium–sulfonium salts undergo a methyl‐transfer reaction on exposure to water, giving rise to a new, singly charged imidazolium salt with iodide introduced at the 2‐position of the imidazolium ring. Crystal structures of some of the imidazolium–sulfonium salts were determined by X‐ray crystallography providing the topology of the interactions between the dications and the anions. Electrospray ionization mass spectrometry and quantum‐chemical calculations were used to rationalise the relative strength of these interactions.  相似文献

Fast and convenient CE assays were developed for the screening of adenosine kinase (AK) inhibitors and substrates. In the first method, the enzymatic reaction was performed in a test tube and the samples were subsequently injected into the capillary by pressure and detected by their UV absorbance at 260 nm. An MEKC method using borate buffer (pH 9.5) containing 100 mM SDS (method A) was suitable for separating alternative substrates (nucleosides). For the CE determination of AMP formed as a product of the AK reaction, a phosphate buffer (pH 7.5 or 8.5) was used and a constant current (95 microA) was applied (method B). The methods employing a fused-silica capillary and normal polarity mode provided good resolution of substrates and products of the enzymatic reaction and a short analysis time of less than 10 min. To further optimize and miniaturize the AK assays, the enzymatic reaction was performed directly in the capillary, prior to separation and quantitation of the product employing electrophoretically mediated microanalysis (EMMA, method C). After hydrodynamic injection of a plug of reaction buffer (20 mM Tris-HCl, 0.2 mM MgCl2, pH 7.4), followed by a plug containing the enzyme, and subsequent injection of a plug of reaction buffer containing 1 mM ATP, 100 microM adenosine, and 20 microM UMP as an internal standard (I.S.), as well as various concentrations of an inhibitor, the reaction was initiated by the application of 5 kV separation voltage (negative polarity) for 0.20 min to let the plugs interpenetrate. The voltage was turned off for 5 min (zero-potential amplification) and again turned on at a constant current of -60 microA to elute the products within 7 min. The method employing a polyacrylamide-coated capillary of 20 cm effective length and reverse polarity mode provided good resolution of substrates and products. Dose-response curves and calculated K(i) values for standard antagonists obtained by CE were in excellent agreement with data obtained by the standard radioactive assay.  相似文献

The aim of the present study is to compare available surface titration curves of kaolinite, to explain the differences between them, and to constrain their interpretation based on predictions of surface protonation that emerged from dissolution experiments. Comparison of six surface titration curves obtained at 25 degrees C reveals significant discrepancies, both in the shape of the curves and in the pH of the point of zero net proton charge (pH(PZNPC)). Based on an analysis of the different sites available for adsorption on kaolinite surfaces we conclude that different kaolinite samples are expected to have similar pH(PZNPC). Therefore, the major reason for the differences in the observed surface protonation is related to the different ways in which the pH(PZNPC) was determined. To compare the titration curves, some of the curves were recalculated so that the proton surface concentrations of all the titration curves would be zero around pH 5. As a result, we obtained a good agreement between the titration curves. A prediction of the molar fraction of protonated sites was retrieved from modeling of kaolinite dissolution reaction and was compared to the protonation data obtained from surface titration. The model successfully predicts the surface protonation data of most of the surface titrations.  相似文献

Frequency dependent NMR relaxation studies have been carried out on Miglyol molecules confined inside core shell polymeric capsules to obtain a correlation between capsule dimension and the measurable parameters. The polymeric capsules were prepared using an interfacial polymerization technique for three different concentrations of Miglyol. It was shown that the variation of Miglyol concentration influences the capsule dimension. Their average size was estimated using the pulsed field gradient diffusometry technique. The relaxation dispersion curves were obtained at room temperature by a combined use of a fast field cycling instrument and a high-field instrument. The frequency dependence of relaxation rate shows a transition from a diffusion-limited to a surface-limited relaxation regime.  相似文献

An efficient organocatalytic route has been developed to synthesize novel substituted thioxanthenes (2a2v) starting from diaryl thioether alcohols (1a1v) using the intramolecular Friedel–Crafts reaction. The starting materials were obtained in two stages via a coupling reaction followed by the Grignard reaction. In this study, we tried for the first time to use some organic Brønsted acids as organocatalysts (3a3h) in the intramolecular Friedel–Crafts cyclization reaction of thioether alcohols. The synthesis of original substituted thioxanthenes was achieved within 15?minutes by using N-triflylphosphoramide (3h) with quantitative yields in THF at room temperature.  相似文献

Aromatic poly(thioether ketone)s were prepared by the direct polycondensation of aromatic dicarboxylic acids with aryl compounds containing ether or sulfide structures using phosphorus pentoxide/methanesulfonic acid (PPMA) as a condensing agent and solvent. Polycondensation proceeded smoothly and produced aromatic poly(thioether ketone)s with inherent viscosities up to 0.73 dL/g. The synthesis of substituted aryl ketones by the reaction of substituted benzoic acids with aryl compounds in PMMA was studied in detail to demonstrate the feasibility of the reaction for polymer formation. The thermogravimetry of the aromatic poly(thioether ketone)s showed a 10% weight loss in air and nitrogen at around 450 and 460°C, respectively. © 1992 John Wiley & Sons, Inc.  相似文献

A simple and sensitive fluorometric method for the determination of nalidixic acid was established by using 0.75M sodium borohydride and 7.5% hydrogen peroxide solution as fluorogenic reagents. Analyte concentrations of 0.0232-11.6 pg/mL could be determined with high precision and accuracy by the method. A relative standard deviation of 1.75% was obtained for a nalidixic acid concentration of 0.232 pg/mL. The method was satisfactorily applied to the determination of nalidixic acid in human serum, fish muscle, and chicken muscle, and the calibration curves were linear from 0.23 to 58.00 pg/mL, from 9.28 to 32.48 mg/kg, and from 4.64 to 23.20 mg/kg, respectively. Laser ablation inductively coupled plasma mass spectrometry using a quadrupole-based mass spectrometer (LA-ICP-QMS) was applied for the analysis of powdered zeolites (microporous aluminosilicates) used for clean-up procedures. For the quantitative determination of trace element concentrations three geological reference materials, granite NIM-G, lujavrite NIM-L and syenite NIM-S, from the National Institute for Metallurgy (South Africa) with a matrix composition corresponding to the zeolites were employed. Both the zeolites and reference materials were fused with a lithium borate mixture to increase the homogeneity and to eliminate mineralogical effects. In order to compare two different approaches for the quantification of analytical results in LA-ICP-MS relative sensitivity coefficients (RSCs) of chemical elements and calibration curves were measured using the geostandards. The experimentally obtained RSCs are in the range of 0.2-6 for all elements of interest. Calibration curves for trace elements were measured without and with Li or Ti as internal standard element. With a few exceptions the regression coefficients of the calibration curves are better than 0.993 with internal standardization. NIM-G granite reference material was employed to evaluate the accuracy of the technique. Therefore, the measured concentrations were corrected with RSCs which were determined using lujavrite reference material NIM-L. This quantification method provided analytical results with deviations of 1-11% from the recommended and proposed values in granite reference material NIM-G, except for Co, Cs, La and Tb. The relative standard deviation (RSD) of the determination of the trace element concentration (n = 5) is about 1% to 6% using Ti as internal standard element. Detection limits of LA-ICP-QMS in the lower microg/g range (from 0.03 microg/g for Lu, Ta and Th to 7.3 microg/g for Cu, with the exception of La) have been achieved for all elements of interest. Under the laser ablation conditions employed (lambda: 266 nm, repetition frequency: 10 Hz, pulse energy: 10 mJ, laser power density: 6 x 10(9) W/cm2) fractionation effects of the determined elements relative to the internal standard element Ti were not observed.  相似文献

A novel thiosalen ligand based on a thioether has been prepared and readily coordinated with copper(I) salts (CuCl, CuBr, CuI, and CuCN). The new organometallic catalyst was used for the direct and enantioselective alkynylations of imines in an A3-coupling reaction. Quasi-isothermal analysis (QIA) is a very useful technique. Compared to conventional non-isothermal thermogravimetry, close-lying reactions can easily be separated by use of this method and kinetic data can be obtained for each intermediate reaction in a single run. This paper discusses the shape of the weight and temperature curves expected in QIA for different controlling mechanisms, and a method is given for calculating the activation energies from these curves. As an example, the QIA curves obtained in thermal decomposition of ammonium-uranyl carbonate (AUC) are analysed and the kinetics of some of the intermediate reactions are determined.  相似文献

A simple technique for quantitative analysis of four urinary biomarkers, tryptophan (TRP), 5-hydroxytryptophan (5-HTP), 5-hydroxytryptamine (5-HT) and 5-hydroxyindole acetic acid (5-HIAA) of carcinoid tumors is developed using gold nanoparticles as the assisted matrix in surface-assisted laser desorption/ionization time-of-flight mass spectrometry (SALDI–TOF MS). The optimal SALDI conditions for the efficient ionization of those biomarkers are systematically explored by the adjustments of the concentrations of gold nanoparticles and internal standards. The mass spectra with strong signals and minimal background noise are obtained using 1-naphthaleneacetamide (NAD) as the internal standard. The calibration curves of the biomarker concentrations are determined using SALDI–TOF MS and the high linearity is obtained in all samples. For future clinical testing, multiplexed detection of those biomarkers in the urine samples of healthy males is performed. The successful quantitative detections of TRP, 5-HTP, 5-HT and 5-HIAA indicate that our technique provided great potentials to be developed a simple and rapid platform for the tumor biomarker detections.  相似文献

Novel 4-vinyl guaiacol based thioether derivatives were synthesized in a three-step reaction procedure by thiol-ene coupling as the key step. The synthesized compounds were characterised by spectroscopic techniques and evaluated for their tribological and antioxidant properties in two different base oils namely epoxy2-ethylhexyl esters of karanja fatty acids (EKE) and dioctyl sebacate (DOS). It was found that the synthesized products were exhibited superior antioxidant performance compared to butylated hydroxytoluene (BHT). All the three synthesized additives were improved the tribological properties of the base oil EKE and DOS. Dithio derivative at 0.75 wt% reduced the wear scar diameter by 36% and at 1 wt% improved the weld point by 33% of base oil EKE. Surface and elemental analysis result suggests that in the tribochemical process the synthesized thioether derivatives decompose and form an effective tribofilms on interacting surfaces. X-ray Photoelectron Spectroscopy (XPS) of the surface lubricated with base oil containing DMFD was evidence for the formation of tribofilm with FeS, FeSO4 and Fe2O3. The antiwear behaviour of the additives was well correlated with quantum chemical calculations. Overall the dithio derivative is more effective as antiwear, extreme pressure and antioxidant bio lubricant additive than other thioether derivatives.  相似文献

A new series of thioether–phosphinite ligands, easily prepared in a few steps from inexpensive d-(+)-xylose, was tested in the Pd-catalyzed allylic substitution of substrates with different steric properties. The results show that the enantiomeric excesses are strongly dependent on the steric properties of the substituent in the thioether moiety. Enantiomeric excesses of up to 93% with high activities were obtained for the substrate rac-1,3-diphenyl-3-acetoxyprop-1-ene 7 with dimethyl malonate as the nucleophile.  相似文献
