以胃癌细胞系BGC-823为研究对象,采用倒置相差显微镜观察纳米TiO2光催化氧化前后BGC-823细胞形态的变化,通过流式细胞仪考察其对BGC-823细胞凋亡率及细胞周期的影响。结果表明,纳米TiO2光催化氧化使BGC-823细胞呈受激坏死或凋亡晚期继发性坏死状态;导致BGC-823细胞周期阻滞于G2/M期并诱导细胞凋亡,从而抑制其DNA合成及增殖分化进程。当TiO2比负载量为9.8×10-7g/cm2、光照时间为40 min时,G2/M期阻滞较为显著,细胞凋亡率达24.34%。  相似文献

以3,4-二甲氧基苯甲醛与3,4-二甲氧基苯乙酸为起始原料,经Perkin缩合、自由基氧化偶联反应、Swern氧化、还原胺化及付克酰基化等7步反应全合成了娃儿藤碱类似物DCB-3501,其结构经1H NMR和ESI-MS确证。体外细胞毒性测试结果表明:DCB-3501对人结肠癌细胞HCT116、人胃癌细胞BGC-823、人肝癌细胞HepG-2、人宫颈癌细胞HeLa和人大细胞肺癌细胞H460的IC50分别为20.0 μmol·L-1, 50.9 μmol·L-1, 2.1 μmol·L-1, 65.8 μmol·L-1和30.8 μmol·L-1。  相似文献

设计并制备了一种组成和结构单一的新型两亲性酞菁锌光敏剂. 采用固相合成法合成出2-单羧基取代酞菁锌, 并使之与五聚赖氨酸偶联成五聚赖氨酸2-羰基酞菁锌. 综合运用各种分析及光谱方法对所得化合物进行表征和确认. 研究了在光照下五聚赖氨酸2-羰基酞菁锌对3种肿瘤细胞(人源肝癌细胞Bel7402、人源胃癌细胞BGC823和人源白血病细胞K562) 与一种正常细胞(人源胚肺成纤维细胞HELF)的杀灭活性. 该光敏剂不仅克服了酞菁锌在水中溶解度低的问题, 且因所偶联的五聚赖氨酸对肿瘤细胞有靶向作用, 具有较高的杀灭肿瘤细胞活性.  相似文献

设计并合成了3个含有席夫碱基团的配体(BLn,n=1~3)及相应的双核联吡啶钌配合物[Ru (BLn)(bpy)2]2(ClO44,其中bpy=2,2''-联吡啶,BLn=((PyCHN)-Ph-O-C6H42R (PyCHN=N-2-吡啶亚甲基,R=none (Ru1),—C (CH32Ru2),—SO2Ru3))。通过元素分析、核磁氢谱、红外光谱、质谱等方法对配体和配合物进行了表征。通过体外细胞毒性实验研究了Ru1~Ru3对宫颈癌细胞(Hela)、胃癌细胞(BGC823)、胃癌细胞(SGC-7901)三种肿瘤细胞和人正常胚肺成纤维细胞MRC-5的细胞毒性,实验结果表明Ru3的抗肿瘤活性最好,而Ru1~Ru3均对BGC823具有选择性。  相似文献

A step-scan Fourier-transform spectrometer coupled with a multipass absorption cell was employed to detect temporally resolved infrared absorption spectra of CH(3)OSO produced upon irradiation of a flowing gaseous mixture of CH(3)OS(O)Cl in N(2) or CO(2) at 248 nm. Two intense transient features with origins near 1152 and 994 cm(-1) are assigned to syn-CH(3)OSO; the former is attributed to overlapping bands at 1154 ± 3 and 1151 ± 3 cm(-1), assigned to the S=O stretching mixed with CH(3) rocking (ν(8)) and the S=O stretching mixed with CH(3) wagging (ν(9)) modes, respectively, and the latter to the C-O stretching (ν(10)) mode at 994 ± 6 cm(-1). Two weak bands at 2991 ± 6 and 2956 ± 3 cm(-1) are assigned as the CH(3) antisymmetric stretching (ν(2)) and symmetric stretching (ν(3)) modes, respectively. Observed vibrational transition wavenumbers agree satisfactorily with those predicted with quantum-chemical calculations at level B3P86∕aug-cc-pVTZ. Based on rotational parameters predicted at that level, the simulated rotational contours of these bands agree satisfactorily with experimental results. The simulation indicates that the S=O stretching mode of anti-CH(3)OSO near 1164 cm(-1) likely makes a small contribution to the observed band near 1152 cm(-1). A simple kinetic model of self-reaction is employed to account for the decay of CH(3)OSO and yields a second-order rate coefficient k=(4 ± 2)×10(-10) cm(3)molecule(-1)s(-1).  相似文献

Two new amides, claulansamides A and B ( 1 and 2 , resp.), together with five known amides, 3 – 7 and six known quinolones, 8 – 13 , were isolated from the stems and roots of Clausena lansium. Their structures were elucidated on the basis of extensive spectroscopic methods. Their absolute configurations were determined by single‐crystal X‐ray diffraction technique, CD, and optical rotation. HPLC Chiral separation of 1 afforded two enantiomers, (+)‐ and (?)‐claulansamide A, respectively. Compounds 9, 12 , and 13 were isolated from the genus Clausena for the first time. All compounds were evaluated for their cytotoxic activities against A549, BGC‐823, and HeLa cancer cell lines. However, only 9 showed cytotoxic activity against A549 cell line with an IC50 value of 46.3 μM , and 11 against BGC‐823 and HeLa cell lines with the IC50 values of 55.0 and 14.7 μM , respectively.  相似文献

Accurate equilibrium structure, dissociation energy, global potential energy surface (PES), dipole moment surface (DMS), and the infrared vibrational spectrum in the 0-3000 cm(-1) range of the F(-)-CH4 anion complex have been obtained. The equilibrium electronic structure calculations employed second-order M?ller-Plesset perturbation theory (MP2) and coupled-cluster (CC) method up to single, double, triple, and perturbative quadruple excitations using the aug-cc-p(C)VXZ [X = 2(D), 3(T), 4(Q), and 5] correlation-consistent basis sets. The best equilibrium geometry has been obtained at the all-electron CCSD(T)/aug-cc-pCVQZ level of theory. The dissociation energy has been determined based on basis set extrapolation techniques within the focal-point analysis (FPA) approach considering (a) electron correlation beyond the all-electron CCSD(T) level, (b) relativistic effects, (c) diagonal Born-Oppenheimer corrections (DBOC), and (d) variationally computed zero-point vibrational energies. The final D(e) and D0 values are 2398 +/- 12 and 2280 +/- 20 cm(-1), respectively. The global PES and DMS have been computed at the frozen-core CCSD(T)/aug-cc-pVTZ and MP2/aug-cc-pVTZ levels of theory, respectively. Variational vibrational calculations have been performed for CH4 and F(-)-CH4 employing the vibrational configuration interaction (VCI) method as implemented in Multimode.  相似文献

合成了5个以直链和带支链烷氧基乙酸为载体配基的顺铂类配合物,通过红外光谱、核磁共振氢谱和质谱对配合物进行了表征,并测试了化合物对肺腺癌SPC-A1和胃腺癌BGC823的体外抗肿瘤活性。生物活性测试结果表明,配合物的活性与离去基团有很大关系。配合物4(顺-二(异丙氧基乙酸根)·[(1R,2R)-1,2-反式环己二胺]合铂(Ⅱ))在2个细胞系中均显示最高的体外抗肿瘤活性,甚至超过顺铂。  相似文献

Gambogic acid (GA, 1 ), the most prominent representative of Garcinia natural products, has been reported to be a promising anti‐tumor agent. In order to further explore the structure‐activity relationship of GA and discover novel GA derivatives as anti‐tumor agents, 17 novel C‐37 modified derivatives of GA were synthesized and evaluated for their in vitro anti‐tumor activities against A549, HCT‐116, BGC‐823, HepG2 and MCF‐7 cancer cell lines. Among them, 11 compounds were found to be more potent than GA against some cancer cell lines. Notably, compound 8 was almost 5–10 folds more active than GA against A549 and BGC‐823 cell lines with the IC50 values of 0.12 µmol·L?1 and 0.57 µmol·L?1, respectively. Chemical modification at C‐37 position of GA by introducing of hydrophilic amines could lead to increased activity and improved drug‐like properties. These findings will enhance our understanding of the structure‐activity relationship (SAR) of GA and lead to the discovery of novel GA derivatives as potential anti‐tumor agents.  相似文献

The infrared photodissociation spectra of [(CO 2) n (CH 3OH) m ] (-) ( n = 1-4, m = 1, 2) are measured in the 2700-3700 cm (-1) range. The observed spectra consist of an intense broad band characteristic of hydrogen-bonded OH stretching vibrations at approximately 3300 cm (-1) and congested vibrational bands around 2900 cm (-1). No photofragment signal is observed for [(CO 2) 1,2(CH 3OH) 1] (-) in the spectral range studied. Ab initio calculations are performed at the MP2/6-311++G** level to obtain structural information such as optimized structures, stabilization energies, and vibrational frequencies of [(CO 2) n (CH 3OH) m ] (-). Comparison between the experimental and the theoretical results reveals the structural properties of [(CO 2) n (CH 3OH) m ] (-): (1) the incorporated CH 3OH interacts directly with either CO 2 (-) or C 2O 4 (-) core by forming an O-HO linkage; (2) the introduction of CH 3OH promotes charge localization in the clusters via the hydrogen-bond formation, resulting in the predominance of CO 2 (-).(CH 3OH) m (CO 2) n-1 isomeric forms over C 2O 4 (-).(CH 3OH) m (CO 2) n-2 ; (3) the hydroxyl group of CH 3OH provides an additional solvation cite for neutral CO 2 molecules.  相似文献

This study uses infrared (IR) spectroscopic, point detection, mapping procedures to examine tissue samples from normal brain specimens and from astrocytic gliomas, the most frequent human brain tumors. Model systems were derived from cultured glioma cell lines. IR spectra of normal tissue sections distinguished white matter from gray matter by increased spectral contributions from lipids and cholesterol. Qualitatively the same differences were found in IR spectra of low and high grade glioma tissue sections pointing to a significant reduction of brain lipids with increasing malignancy. Whereas spectral contributions of proteins and lipids were similar in IR spectra of glioma cells and tissues, nucleic acid bands were more intense for cells suggesting higher proliferative activities. For statistical analyses of IR spectroscopic maps from 71 samples, a parameter for the lipid to protein ratio was introduced involving the CH(2) symmetric stretch band with lipids as main contributors and the amide I band of proteins. As this parameter correlated with the grade of gliomas obtained from standard histopathological examination, it was applied to classify brain tissue sections based on IR spectroscopic mapping.  相似文献

Eight novel Pt(II), Pd(II), Cu(II) and Zn(II) complexes with 4’‐substituted terpyridine were synthesized and characterized by elemental analysis, UV, IR, NMR, electron paramagnetic resonance, high‐resolution mass spectrometry and molar conductivity measurements. The cytotoxicity of these complexes against HL‐60, BGC‐823, KB and Bel‐7402 cell lines was evaluated by MTT assay. All the complexes displayed cytotoxicity with low IC50 values (<20 μm ) and showed selectivity. Complexes 3 , 5 , 7 and 8 exerted 9‐, 5‐, 12‐ and 7‐fold higher cytotoxicity than cisplatin against Bel‐7402 cell line. The cytotoxicity of complexes 3 , 5 , 6 , 7 and 8 was higher than that of cisplatin against BGC‐823 cell line. Complexes 3 , 7 and 8 showed similar cytotoxicity to cisplatin against KB cell line. Complex 7 exhibited higher cytotoxicity than cisplatin against HL‐60 cell line. Among these complexes, complex 7 demonstrated the highest in vitro cytotoxicity, with IC50 values of 1.62, 3.59, 2.28 and 0.63 μm against HL‐60, BGC‐823, Bel‐7402 and KB cells lines, respectively. The results suggest that the cytotoxicity of these complexes is related to the nature of the terminal group of the ligand, the metal center and the leaving groups. Endoplasmic reticulum (ER) has emerged as one of the interesting sub-cellular organelles due to its role in myriads of biological phenomena. Subsequently, visualization of the structure-function and dynamics of ER remained a major challenge to understand its involvement in different diseased states including cancer. To illuminate the ER, herein we have designed and synthesized γ-resorcyclic acid-based small molecules, which showed remarkable aggregation-induced emission (AIE) property in water. This AIE property was originated from the dual intramolecular H-bonding leading to the self-assembled 2D aggregation confirmed by pH- and temperature-dependent fluorescence quenching studies as well as scanning electron microscopy. These small molecules illuminated the sub-cellular ER in HeLa cervical cancer cells as well as non-cancerous RPE-1 human retinal epithelial cells within 1 h. These novel small molecules have the potential to light up ER chemical biology in diseased states.  相似文献

Metal complexes are recently being hybridized with different moieties to discover new drugs due to their advantageous attributes. Among the metals, copper is a good one to synthesize a metal complex due to its being endogenous, redox and DNA cleavage potential, reported anti-cancer efficacies and selective permeability for cancer cells. In this study, first we synthesized a new copper (II) complex and determined its toxic doses on NIH/3T3 normal fibroblast cells, SPC212 mesothelioma and DU145 prostate cancer cells. Then, we ascertained anti-proliferative, apoptotic, morphological, oxidative and endoplasmic reticulum (ER) stress inducing effects of these newly synthesized compounds on DU145 prostate cancer cells. A novel Copper(II)/1-(4-(trifluoromethyl)benzyl)-1H-benzimidazole/2,2′-bipyridyl complex was synthesized and mainly characterized by single crystal X-ray diffraction analysis. Anti-proliferative effect of copper(II) complex was gauged by MTT. Oxidative and ER stress were evaluated by ELISA and Western blot. The morphological effect was examined by microscope analysis. Besides, immunocytochemistry of Bax, a pro-apoptotic protein and PCNA, a proliferation marker protein was performed. As a result, the inhibitory effect of newly synthesized substance was superior to the chemicals from which it was synthesized. Its IC50s against DU145 were 37.0, 21.1 and 10.0 µM for 24, 48 and 72 h-treatments, respectively. Oxidative and ER stress increased after treatment. In microscopy, we observed apoptotic hallmarks like nuclear condensation, cellular shrinkage and membrane blebbings. In immunochemistry, increased Bax and decreased PCNA were apparent. Copper(II) complex with its relatively low IC50 can also be tested on other cancer and normal cell lines.  相似文献

The rate constant of the reaction Cl + CF(3)CF═CH(2) (k(1)) has been measured relative to several reference species using the relative rate technique with either gas chromatographic analysis with flame-ionization detection (GC/FID) or Fourier transform infrared (FTIR) analysis. Cl atoms were generated by UV irradiation of Cl(2)/CF(3)CF═CH(2)/reference/N(2)/O(2) mixtures. At 300-400 K in the presence of >20 Torr O(2), k(1) = 1.2 × 10(-11) e((+1100/RT)) cm(3) molecule(-1) s(-1). In N(2) diluent, k(1) has a sharp negative temperature coefficient resulting from the relatively small exothermicity of the following reactions: (1a) Cl + CF(3)CF═CH(2) ? CF(3)CFClCH(2)(?); (1b) Cl + CF(3)CF═CH(2) ? CF(3)CF(?)CH(2)Cl (reaction 1), which were determined in these experiments to be ~16.5 (±2.0) kcal mol(-1). This low exothermicity causes reaction 1 to become significantly reversible even at ambient temperature. The rate constant ratio for the reaction of the chloroalkyl radicals formed in reaction 1 with Cl(2) (k(2)) or O(2) (k(3)) was measured to be k(2)/k(3) = 0.4 e(-(3000/RT)) for 300-400 K. At 300 K, k(2)/k(3) = 0.0026. The reversibility of reaction 1 combined with the small value of k(2)/k(3) leads to a sensitive dependence of k(1) on the O(2) concentration. Products measured by GC/FID as a function of temperature are CF(3)CFClCH(2)Cl, CF(3)COF, and CH(2)Cl(2). The mechanism leading to these products is discussed. The rate constant for the reaction Cl + CF(3)CFClCH(2)Cl (k(11)) was measured as a function of temperature (300-462 K) at 760 Torr to be k(11) = 8.2 × 10(-12) e(-(4065/RT)) cm(3) molecule(-1) s(-1). Rate constants relative to CH(4) for the reactions of Cl with the reference compounds CH(3)Cl, CH(2)Cl(2), and CHCl(3) were measured at 470 K to resolve a literature discrepancy. Two kinds of highly ordered mesoporous silica materials (FDU-11, FDU-13) with novel three-dimensional (3-D) tetragonal and orthorhombic structures were synthesized by using tetra-headgroup rigid bolaform quaternary ammonium surfactant [(CH(3))(3)NCH(2)CH(2)CH(2)N(CH(3))(2)CH(2)(CH(2))(11)OC(6)H(4)C(6)H(4)O(CH(2))(11)CH(2)N(CH(3))(2)CH(2)CH(2)CH(2)N(CH(3))(3).4Br] (C(3-12-12)(-)(3)) as a template under alkaline conditions. High-resolution transmission electron microscopy (HRTEM), small-angle X-ray scattering (SAXS), and X-ray diffraction (XRD) show that mesoporous silica FDU-11 has primitive tetragonal P4/mmm structure with cell parameters a = b = 8.46 nm, c = 5.22 nm, and c/a ratio = 0.617. N(2) sorption isotherms show that calcined FDU-11 has a high BET surface area of approximately 1490 m(2)/g, a uniform pore size of approximately 2.72 nm, and a pore volume of approximately 1.88 cm(3)/g. Mesoporous silica FDU-13 has primitive orthorhombic Pmmm structure. The cell parameters are a = 9.81, b = 5.67, and c = 3.66 nm. N(2) sorption isotherms show that calcined FDU-13 has a high BET surface area of 1210 m(2)/g, a uniform mesopore size of approximately 1.76 nm, and a large pore volume of approximately 1.83 cm(3)/g. Such low symmetries for 3-D mesostructures (tetragonal and orthorhombic system) have not been observed before even in amphiphilic liquid crystals, which maybe resulted from an oblate aggregation of the bolaform surfactant and its strong electrostatic interaction with inorganic precursor. A probable mechanism has been proposed for the formation of such a 3-D low symmetrical mesostructure. These results will further extend the synthesis of mesoporous materials and may open up new opportunities for their new applications in catalysis, separation, and nanoscience.  相似文献

The Raman spectroscopy of n-tetradecane was investigated in a Moissanite anvil cell at pressure from 0.1 MPa to 1.4 GPa and ambient temperature. The result shows that the liquid-solid phase transition of n-tetradecane takes place at around 302.8 MPa and the corresponding DeltaV(m) obtained is about -9.6 cm(-3)/mol. Above 302.8 MPa, the frequencies of CH(2) and CH(3) symmetric stretching and asymmetric stretching vibration shift to higher wave numbers in a linear manner with increasing pressure, which can be expressed as: nu(s)(CH(3))=0.013P+2882.0; nu(as)(CH(3))=0.014P+2961.6; nu(s)(CH(2))=0.013P+2850.8; nu(as)(CH(2))=0.009P+2923.2. This relationship indicates that n-tetradecane can be a reliable pressure gauge for the experimental study within the pressure range of 0.3-1.4 GPa.  相似文献

A series of novel bis-naphalenyl compounds with different diamine linkers were synthesized and characterized by 1H NMR, 13C NMR, and HR-MS. The DNA binding abilities of the compounds were studied by using flourescence titration, DNA thermal denaturation experiments, viscosity titration, and NMR studies. The DNA binding abilities of all the bis-naphalenyl compounds were on the same order of magnitude. Compared with the groove binding mode of the monomer, the bis-naphalenyl compounds exhibited partial intercalating binding mode. The cytotoxicity activities of the compounds were evaluated by MTT assay in vitro. According to the results of MTT assay, bis-naphalenyl compound 3c with hexamethylenediamine linker, and 3d with p-xylylenediamine linker were found to be more toxic against BGC823 cells. The IC50 values of the two compounds were similar to that of the control drug (5-Fluorouracil) on BGC823 cells. Compared with the results on BGC823 cells, better results were found on SW480 cells. Compounds 3c and 3d exhibited smaller IC50 values than that of control drug (5-Fluorouracil). The IC50 values of 3c, 3d, and 5-Fluorouracil were 52.01, 66.09, and 230.11 μM, respectively.  相似文献

Two new Co(II) and Ni(II) complexes exhibiting DNA cytotoxic activities with 3-(2-pyridyl)pyrazole-based ligand, [Co(L)(3)](ClO(4))(2) (1) and [Ni(L)(3)](ClO(4))(2) (2) (L=1-[3-(2-pyridyl)-pyrazol-1-ylmethyl]-naphthalene) were synthesized and structurally characterized. Both 1 and 2 crystallized in the monoclinic system, space group P2(1)/c, for 1, a=12.8324(8), b=12.1205(8), c=33.27(2) A, beta=93.92(3) degrees and Z=4; for 2, a=12.8764(3), b=12.1015(3), c=33.2415(9) A, beta=93.998(1) degrees and Z=4. Among them, the Co(II) and Ni(II) ions were all coordinated by six N donors from three distinct L ligands. In addition, the cytotoxic activities of 1, 2 and L in vitro were evaluated against three different cancer cell lines HL-60 (human leukemia), BGC-823 (stomach cancer) and MDA-MB-435 (mammary cancer), respectively. The results showed that 1 exhibited significantly high cytotoxic activities against HL-60 and moderate activities against BGC-823 and MDA-MB-435. In order to further investigate the relationships between structures and DNA-binding behaviors of these complexes, the interactions of 1, 2 and L with calf thymus DNA (CT-DNA) were then subjected to thermal denaturation, viscosity measurements and spectrophotometric methods. The results indicated that 1 and 2 intercalated with DNA via L ligand. The intrinsic binding constants of 1, 2 and L with DNA were 1.6x10(4), 5.6x10(3) and 2.76x10(3) M(-1), respectively.  相似文献
