内窥式散斑类共聚焦系统层析能力分析   总被引:1,自引:0,他引:1  
本文研究了内窥式散斑类共聚焦系统(Endoscope-based speckle quasi-confocal system,EBSQCS)的层析能力。从散斑类共聚焦显微镜(Speckle quasi-confocal microscope,SQCM)成像原理出发,详细分析了内窥镜光学结构对散斑类共聚焦显微镜散斑场波动的影响规律,推导了内窥镜光学结构与内窥式散斑类共聚焦系统轴向分辨力的关系。实验测得了基于光纤束的内窥式散斑类共聚焦系统的轴向分辨力曲线。选用放大倍率4倍,光纤直径5μm的内窥镜系统的内窥式散斑类共聚焦系统轴向分辨力曲线的全峰半高是散斑类共聚焦显微镜的2.3倍,与理论计算值相符。实验结果表明内窥式散斑类共聚焦系统具有很好的轴向层析能力。  相似文献

谱域光学相干层析(spectral domain optical coherence tomography, 简记为SD-OCT)系统的轴向点扩散函数(point spread function, 简记为PSF)并不具备空不变特性,无法直接应用于解卷积运算.为实现SD-OCT系统成像质量基于解卷积算法的优化,本文采用数值校正后的轴向扫描信号和轴向有效PSF来实施基于Lucy-Richardson算法的解卷积运算,进而实现了SD-OCT系统中图像质量尤其是轴向分辨率的改善.本文理论分析了SD-OCT系统中导致轴向有效PSF随成像深度增大而下降和图像模糊的因素,阐述了利用解卷积算法实现图像质量优化的过程,基于建立的SD-OCT系统实施了不同成像深度位置处PSF的标定,并利用离散轴向位置处PSF的峰值拟合了轴向有效PSF的调制函数.利用调制函数对所有轴向扫描信号进行数值校正,然后根据轴向有效PSF进行解卷积算法.典型样品的解卷积图像重建结果表明,提出的解卷积方法能有效提高系统的轴向分辨率,同时有效抑制系统灵敏度随成像深度增大而下降的趋势. 关键词: 谱域光学相干层析 Lucy-Richardson解卷积 有效点扩散函数  相似文献

In stimulated emission depletion (STED) microscopy, the lateral resolution is in the range of tens of nanometers depending on the sample and the instrument. The axial resolution, however, is in standard systems limited by diffraction to about 500 nm. We present an approach to three-dimensional diffraction-unlimited resolution by observing the sample at two optical angles. The system is realized by using an atomic force microscope (AFM) chip as a microreflector to deflect the STED beams near the region-of-interest (ROI), thus allowing observations at an angle ∠. Consequently, the superior lateral resolution can be utilized to resolve details in the axial direction of the main optical axis of the microscope. Here, fluorescent nanoparticles 90 nm apart and biological structures 80 nm apart along axial direction were distinguished by utilizing an off-the-shelf, commercial STED microscope, coupled with an AFM and an AFM chip micro-reflector.  相似文献

本文报道了一种超高分辨率谱域光学相干层析成像(SD-OCT)系统. 该系统基于超连续谱激光光源并截取部分光谱作为宽带光源, 其中心波长为665 nm, 光谱半高全宽(FWHM) 230 nm. 系统轴向分辨率0.9 μm, 轴向扫描速率28600行/秒, 横向分辨率3.9 μm, 横向视场1 mm, 最大成像深度0.6 mm(空气中). 利用研制的超高分辨率SD-OCT系统, 对不同型号的工业砂纸精细结构进行了成像, 并与普通SD-OCT的成像结果进行对比, 充分展示了研制系统在材料无损检测中优势.  相似文献

光学相干层析成像(OCT)系统的纵向分辨力不仅与光源的带宽有关,而且与系统中两干涉臂间的色散匹配有关。如果色散没有得到精确匹配,将使光学相干层析成像系统的纵向分辨力达不到所预期的理论值。色散问题在超高分辨光学相干层析成像系统中尤为突出。提出了一种基于双光栅快速扫描光学延迟线(RSOD),用于光学相干层析成像系统的色散补偿。该方法中新增的光栅引入了光栅间距这一独立变量,其与常规单光栅快速扫描光学延迟线机构中的光栅离焦量一起,可使光学相干层析成像系统中的群速度色散(GVD)和三阶色散(TOD)同时得到补偿。分析了双光栅快速扫描光学延迟线的色散特性和色散调节原则,并提供了一个典型光学相干层析成像系统中的色散补偿实例。  相似文献

为解决微小内轮廓尺寸为代表的微小尺寸的非接触式超精密测量问题,提出了将自聚焦透镜体积小的特点与共焦显微技术的高分辨率和绝对位置跟踪特性相结合的差动式自聚焦共焦微型显微技术的光探测技术,建立了相应的传感系统,介绍了系统的工作原理和构成,自聚焦透镜测头直径为1mm,两个探测器差动设置,不但消除了光源的光强漂移和探测器的电子漂移产生的共模噪声,提高了测量信噪比,而且有效地提高了系统的轴向分辨率,初步实验表明,系统轴向分辨率在倾斜率小于20度的范围内可达5nm。  相似文献

光学相十层析成像(光学相干层析成像术)的轴向分辨力由光源带宽和探测光束的聚焦条件共同决定。提高光学相干层析成像术轴向分辨力的方法主要基于带宽光源技术。提出了一种将变迹术与光学相十层析成像术相干门有机结合的方法来提高其轴向分辨力。通过适当形式的光瞳滤波器.使光学相干层析成像术系统轴向响应的主瓣宽度缩小到相干门之内,而其旁瓣则处于相干门之外.不对相干成像产生有效贡献。这样.就能在光源带宽不变的条件下,有效提高光学相十层析成像术的轴向分辨力,避免了采用宽带光源所带来的费用昂贵和系统复杂等缺陷。  相似文献

We demonstrate inhibition of the sidelobes of the axial point spread function in optical coherence tomography by shaping the power spectrum of the light source with a remaining power of 4.54 mW. A broadband amplified spontaneous emission source radiating at 1565 +/- 40 nm is employed in a free-space optical coherence tomography system. The axial point spread functions before and after optical spectral shaping are presented. Results show that spectral shaping of the source can inhibit sidelobes of the point spread function up to 12.9 dB, with an associated small increase of 2.2 dB in noise floor in the far field. The effect of spectral shaping on axial resolution is demonstrated according to three metrics. Image quality improvement is also illustrated with optical coherence tomography images of an onion before and after spectral shaping.  相似文献

建立了反射式光纤共焦扫描成像系统,分析了光纤-集光透镜参数A及物透镜有效数值孔径等对系统成像分辨率的影响。并在此基础上选择了合适参数的透镜,获得了优化的反射式光纤共焦扫描成像系统,测试结果表明,该系统具有亚微米级横向成像能力,微米级向层析能力,成像稳定性那,它将应用于材料及生物组织三维成像检测中。  相似文献

A fiber-optic-bundle-based optical coherence tomography (OCT) probe method is presented. The experimental results demonstrate this multimode optical fiber-bundle-based OCT system can achieve a lateral resolution of 12 microm and an axial resolution of 10 microm with a superluminescent diode source. This novel OCT imaging approach eliminates any moving parts in the probe and has a primary advantage for use in extremely compact and safe OCT endoscopes for imaging internal organs and great potential to be combined with confocal endoscopic microscopy.  相似文献

We demonstrate experimentally and theoretically a method for enhancing the axial resolution of optical low-coherence reflectometry (OLCR) beyond the Fourier transform limit of light source spectrum. The resolution enhancement originates from the superposition of multiple OLCR scans with different offsets of the center-wavelength light-ray with respect to the rotation axis of the mirror mounted on the galvanometer optical scanner in the used optical phase delay line. After certain software process of the multiple scan fringe patterns of different offsets, the superposition leads to an OLCR axial resolution beyond the transform limit. Experimentally, by using four different offsets for superposition, about 1.5 times enhancement of the transform-limit resolution is obtained.  相似文献

光学相干层析成像的轴向分辨率和横向分辨率是互为独立的,其轴向分辨率由系统光源带宽和探测光束的聚焦条件共同决定,而横向分辨率由系统样品臂的聚焦条件决定. 提高光学相干层析成像的轴向分辨率的方法主要基于宽带光源技术以及变迹术与相干门相结合的方法,而这些方法对于横向分辨率并没有提高. 提出了一种通过光程编码与相干合成的方法,可以同时提高其轴向分辨率和横向分辨率. 通过在光学相干层析成像系统的样品臂中加入光程编码分束器形成多种对应不同光程延迟的有效响应函数,基于光学相干层析成像术固有的光程分辨能力可以得到同一样品对应于不同有效响应函数的多幅图像. 通过数字控制不同有效响应函数的相对贡献对其进行相干合成,可以同时实现轴向和横向的超分辨效果. 与以前的方法相比,光程编码与相干合成方法简单易行、成本低廉,不仅可以避免系统复杂和价格昂贵等不足,而且可以同时较大幅度地提高系统的轴向分辨率和横向分辨率. 关键词: 光学相干层析成像 轴向超分辨 横向超分辨 光程编码  相似文献

为了提高光学相干层析成像(OCT)的轴向分辨率,采用OCT与光学超分辨术相结合的方法,设计了三区相位型和振幅型两种光瞳滤波器,并将制作的光瞳滤波器应用于系统的样品臂中,分别测量不同条件下对应的轴向响应曲线,结果与理论分析基本吻合,实现了轴向分辨率10%以上的提高,从而验证了利用合适光瞳滤波器实现OCT轴向超分辨的可行性.  相似文献

In optical coherence tomography, axial and lateral resolutions are determined by the source coherence length and the numerical aperture of the sampling lens, respectively. Whereas axial resolution can be improved by use of a broadband light source, there is a trade-off between lateral resolution and focusing depth when conventional optical elements are used. We report on the incorporation of an axicon lens into the sample arm of an interferometer to overcome this limitation. Using an axicon lens with a top angle of 160 degrees , we maintained 10-microm or better lateral resolution over a focusing depth of at least 6 mm. In addition to having high lateral resolution, the focusing spot has an intensity that is approximately constant over a greater depth range than when a conventional lens is used.  相似文献

Kant reported [J. Mod. Optics 47, 905 (2000)] a formulation for solving the inverse problem of vector diffraction, which accurately models high-NA focusing. Here, Kant's formulation is adapted to the method of generalized projections to obtain an algorithm for designing diffractive optical elements (DOEs) that reshape the axial point-spread function (PSF). The algorithm is applied to design a binary phase-only DOE that superresolves the axial PSF with controlled increase in axial sidelobes. An 11-zone DOE is identified that axially narrows the PSF central lobe by 29% while maintaining the sidelobe intensity at or below 52% of the peak intensity. This DOE could improve the resolution achievable in several applications without significantly complicating the optical system.  相似文献

Spectral calibration is important for a Fourier domain optical coherence tomography (FD-OCT) system. A new spectral calibration method for FD-OCT is presented. Through the iterated mapping of spectral interferograms detected by an optical spectrum analyzer and the CCD grating spectrometer, multiple groups of calibration coefficients are obtained. By comparing the effects of these coefficients on improving the amplitude of the axial point spread function (PSF), optimum coefficients are obtained. In this method, no additional calibration laser source is needed, reducing the complexity of the system. The cross-sectional images of a glass plate in milk are acquired, and the experimental results indicate that the axial resolution and signal-to-noise ratio (SNR) are effectively improved with the proposed spectral calibration method.  相似文献
