Summary.  Various sulfa drugs were condensed with 4-isothiocyanato-4-methyl-2-pentanone at pH∼3–5 by refluxing in methanol to give various substituted mercaptopyrimidines. On condensation with 9-chloro-2-substituted or -unsubstituted acridines, sulfathiazole gave the corresponding condensed products. N-Ethylaminoadenosine reacted with 9-chloroacridine to the coupled product. Condensation of sulfathiazole with 9-isothiocyanato-2,4-substituted or -unsubstituted acridines afforded the corresponding condensed compounds. The structures of all synthesized compounds were confirmed by spectroscopic methods. Anticancer, antiinflammatory, and analgesic activities of all compounds were investigated. Abstract  

C-2-unsubstituted imidazoline-3-oxides were reduced with NaBH4 in THF to give the corresponding trans-3,5-diarylimidazolidin-1-ols, while under the same conditions C-2-substituted derivatives gave the corresponding ring–chain–ring tautomers. Treatment of the crude reaction mixture from the reduction of C-2-unsubstituted imidazoline-3-oxides with a MeOH–H2O mixture provided reductive C–N bond cleavage to give hydroxylamines, while under the same conditions ring–chain–ring tautomers remained unchanged.  相似文献

Lithium aluminum hydride reduction of 5-substituted or unsubstituted ethyl or methyl 1-aryl-1H-pyrazole-4-carboxylates gave, generally in excellent yields, 5-substituted or unsubstituted 1-aryl-1H-pyrazole-4-methanols which afforded the corresponding 1-aryl-4-(bromomethyl)-1H-pyrazoles with hydrobromic acid in acetic acid solution. These crude intermediates gave by reaction with potassium cyanide in dimethylsulfoxide solution 1-aryl-1H-pyrazole-4-acetonitriles only in the case of 5-unsubstituted compounds, otherwise mixtures of 5-substituted 1-aryl-1H-pyrazole-4-acetonitriles and 4-methyl-1-phenyl-1H-pyrazole-3-carbonitriles were generally obtained. Acetonitriles IIIa,b,i,l gave in excellent yields the corresponding 1-aryl-1H-pyrazole-4-acetic acids Va,b,i,l by alkaline hydrolysis. Compounds Vb,i,l showed in the writhing test appreciable analgesic properties, associated with low acute toxicity; moreover, compound VI exhibited a statistically significant antiinflammatory activity in the carrageenan-induced edema assay.  相似文献

Solid-state techniques allow for waste-free quantitative syntheses. The solid–solid reactions of α-haloketones with several pyrazolones and with thiosemicarbazones were shown to afford the corresponding pyrazolyl ethers and 4-substituted 2-(arylidenehydrazino)thiazoles. The product yields are quantitative in all cases and the products do not require purifying workup. Therefore, these reactions are truly solvent-free, sustainable, and no wastes are produced. A diazonium nitrate is quantitatively accessible by gas–solid reaction of the corresponding amine with NO2 gas. It is a useful material for environmental synthesis of azo dyes through solid-state coupling with a variety of coupling compounds, as e.g. β-naphthol, acetoacetanilide, pyrazolones, and barbituric acid.  相似文献

9-Chloro-2,4-(un)substituted acridines (1) on condensation with sulpha- diazine, sulphathiazole, and sulphaacetamide gave condensation products 3a-h. 3-Aryl-4-phenyl-2-imino-4-thiazolines (4) on condensation with 9-chloro-2,4-(un)substituted acridines (1) gave condensation products 5a–5o. Both 3a–3h and 5a–5o were purified by crystallization or by chromatography. Structures assigned to 3a–3h and 5a–5o are supported by correct spectral data. Antiinflammatory and analgesic activity screening of 3a, 3e, 3f and 5a–5c, 5e, 5g, 5i, 5m, 5n were carried out using carrageenin induced paw oedema and phenyl quinone writhing assay. Some of the compounds exhibited interesting antiinflammatory or analgesic activities.  相似文献

Reaction of ethyl or methyl 2-dimethylaminomethylene-3-oxoalkanoates with N-C-N dinucleophiles such as guanidine, acetamidine or benzamidine afforded in high yields the relative esters of 4-substituted 2-amino-, 2-methyl- or 2-phenyl-5-pyrimidinecarboxylic acids, respectively. These esters were hydrolyzed to the corresponding carboxylic acids, which were converted by heating to 4-substituted 2-pyrimidinamines, 2-methyl or 2-phenylpyrimidines, respectively, generally in excellent yields. The 4-unsubstituted ethyl 2-amino-, 2-methyl- and 2-phenyl-5-pyrimidinecarboxylates were obtained in moderate yields by reaction of the above dinucleophiles with ethyl 2,2-diformylacetate. These esters were hydrolyzed and the corresponding acids (with the exception of the 2-methyl derivative) were decarboxylated to give 2-pyrimidinamine and 2-phenylpyrimidine in satisfactory yields.  相似文献

5,6-Unsubstituted 1,3-oxazine-2,4-diones ( 3 ) and 6-unsubstituted 5-methyl-1,3-oxazine-2,4-diones ( 4 ) were prepared by reduction of the corresponding 6-chloro derivatives ( 1 and 2 ). Treatment of 6-chloro-3-methyl-1,3-oxazine-2,4-dione ( 1a ) with sodium azide, sodium cyanide, secondary amines and aniline gave the corresponding 6-substituted compounds ( 7, 9, 10 and 11 ) while the reaction of 1a and 2a,b with primary aliphatic amines such as methylamine and ethylamine caused a ring transformation to pyrimidine ring system giving barbituric acids ( 13a-d ).  相似文献

The synthesis of some substituted ll-methoxy-7-nitro-4H,8H-2,3-dihydro-1,4-diazepino[5,6,7-kl]acridines 4 with more diversified chains at position 4 is described. A convenient method for transformation of these compounds into the corresponding 4-substituted 12-methoxy-4H-2,3-dihydroimidazo[4,5,1-de][1,4]diazepino-[5,6,7-mn]acridine (5) is reported.  相似文献

Dehydrogenative cyclization of the 6-substituted 7-arylidenehydrazinotetrazolo[1,5-b][1,2,4]triazines derived from 6-methyl and 6-phenyl derivatives of 7-hydrazinotetrazolo[1,5-b][1,2,4]triazines and aromatic aldehydes gave the corresponding 6-substituted 9-aryl-[1,2,4]triazolo[4,3-d]tetrazolo-[1,5-b][1,2,4]triazines. The latter compounds were also obtained by an alternative route involving dehydrative cyclization of 6-methyl and 6-phenyl derivatives of 7-chlorotetrazolo[1,5-b][1,2,4]triazines with aromatic hydrazides through the isolable aroylhydrazino intermediates. Also, the triazolotetrazolotriazine rings were accomplished by one-pot cyclization of cyclic amidrazones with aromatic acid chlorides. The ditetrazolo[1,5-b:1′,5′-d][1,2,4]triazine systems were synthesized by cyclization of the former cyclic amidrazones with nitrous acid, or cyclic imidoyl chlorides with sodium azide. The bis-triazolotetrazolotriazine derivatives were synthesized by cyclization of two equivalents of each cyclic imidoyl chloride with acid dihydrazides through the isolable bis-hydrazide products. The antimicrobial activity of representative compounds was studied.  相似文献

Summary.  The synthesis of the title compounds through variations of the Gewald reaction is presented. Knoevenagel condensation of methylketone derivatives with methyl cyanoacetate and subsequent treatment of the α,β-unsaturated nitriles with sulfur and amine resulted in the corresponding 2-aminothiophenes 5 or isomers 9 and 10. Reaction of methylketone derivatives bearing a leaving group at the methyl group under modified Gewald conditions selectively led to the formation of 4-substituted 2-aminothiophenes 9a and 12. Aryl 2-[(2-imidazolyl)ethyl or 3-(2-imidazolyl)propyl]ketones were ketalized by glycerol or 3-mercapto-1,2-propanediol in boiling benzene in the presence of 4-toluenesulfonic acid to provide the title compounds. The aryl substituents are 4-chloro-, 4-bromo-, 4-fluoro-, or 2,4-dichlorophenyl. While aryl (2-imidazolyl)methyl ketones condensed with glycerol to form cis- and trans-{2-aryl-2-[(2-imidazolyl)methyl]-4-(hydroxymethyl)}-1,3-dioxolanes, related condensations with 3-mercapto-1,2-propanediol, under similar, or even more stringent reaction conditions, produced no 1,3-oxathiolane analogs, with the starting ketones being recovered. Separation and structure determination of these racemic cis and trans isomeric products are described. The structure of these stereoisomers was established by means of 1H and 13C nmr correlation and nOe experiments. Selective methylation of the N-unsubstituted 2-imidazolyl alcohols with one equivalent sodium hydride and methyl iodide provided the corresponding N-methyl alcohols in excellent yields. With excess benzoyl chloride, N-unsubstituted 2-imidazolyl alcohols were initially converted to O, N-dibenzoates from which the N-benzoyl group was easily cleaved by ammonium hydroxide in ethanol to provide benzoate esters.  相似文献

Summary. Several 2-substituted 4-amino-2-azabicyclo [3.2.2]nonane derivatives were synthesized. The new compounds were investigated for their activities against the causative organism of East African sleeping sickness, Trypanosoma b. rhodesiense, and a protozoan parasite which causes Malaria tropica, Plasmodium falciparum K1, a strain which is resistant to chloroquine and pyrimethamine. The results are compared to the activities of 2-unsubstituted 4-amino-2-azabicyclo[3.2.2]nonanes.  相似文献

Aminomethylation of thiourea with aqueous formaldehyde and simple amino acids (glycine, β-alanine, γ-aminobutyric acid) have resulted in the formation of (4-thioxo-1,3,5-triazinan-1-yl)-substituted acetic, propionic, and butyric acids, respectively. By alkylation of these compounds corresponding S-methyl and S-ethyl iodides were obtained, and by the action of tert-butylamine, the corresponding salts. The same salts were obtained by the reaction of amine exchange between 5-tert-butyl-1,3,5-triazinan-2-thione and these amino acids in water. As a result of neutralization of S-methyl iodides with tert-butylamine in 2-propanol or aqueous 2-propanol zwitterionic [4-(methyl-sulfanyl)-3,6-dihydro-1,3,5-triazin-1(2H)-yl] derivatives of these acids were isolated. From aqueous solutions of S-methyl iodides and tert-butylamine ion associates of the corresponding zwitter-ions and tert-butylammonium iodide have crystallized. The same associates have formed at treating S-methyl iodides with tert-butylamine or diethylamine in the absence of a solvent.  相似文献

Quinolizine compounds 1 and 2 or their monocyclic tautomers 3 and 4 have been synthesized using 2-pyri-dineacetic acid derivatives 6a, b, A, B and ethoxymethylenemalonic acid derivatives 7a, b, c in base catalyzed or thermic reaction. In the 6-unsubstituted series, both the 4-oxo and 4-imino derivatives could have been obtained, in the 6-substituted series, however the 4-oxo ones only, whereas instead of the 4-imino derivatives, their monocyclic tautomers 3, 4 have been isolated. In the 6-unsubstituted series, the primarily formed 4-imino compounds have been rearranged into 4-oxo ones under stronger conditions. The structure of the isolated compounds have been proved by ultraviolet, infrared and 1H nmr spectra, that of 3B=C by X-ray analysis as well.  相似文献

A number of tertiary aza-9-fluorenols were obtained from 4- and 2-azafluofenones and were subjected to dehydration in order to synthesize the previously unknown 9-methyleneazafluorenes. The corresponding 9-methyleneazafluorenes and their polymers were obtained simultaneously in relatively stable form from both pyridine-ring-substituted and -unsubstituted 4-aza-9-fluorenols and from 1,3-diphenyl-2-aza-9-fluorenol. On the basis of an analysis of the mass spectrum of the polymer obtained from 9-methylene-4-azafluorene it was concluded that it is evidently isolated in the form of two dimers — dispiro [bis (4-aza-9-fluorene)-1, 3-cyclobutane] and the analogous product of dispiro addition with a 1,2-substituted cyclobutane ring. Condensation of the corresponding azafluorenes with benzaldehyde gave their 9-benzylidene derivatives in the form of geometrical isomers, the structures of which were established on the basis of the PMR spectra.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 9, pp. 1234–1240, September, 1978.  相似文献
