It is of sustainable interest to improve the sensitivity and selectivity of the ionization process, especially for direct analysis of complex samples without matrix separation. Herein, four ambient ionization methods including desorption atmospheric pressure chemical ionization (DAPCI), heat‐assisted desorption atmospheric pressure chemical ionization (heat‐assisted DAPCI), microwave plasma torch (MPT) and internal extractive electrospray ionization (iEESI) were employed for comparative analysis of the navel orange tissue samples by mass spectrometry. The volatile organic compounds (e.g. ethanol, vanillin, leaf alcohol and jasmine lactone) were successfully detected by non‐heat‐assisted DAPCI‐MS, while semi‐volatile organic compounds (e.g. 1‐nonanol and ethyl nonanoate) together with low abundance of non‐volatile organic compounds (e.g. sinensetin and nobiletin) were obtained by heat‐assisted DAPCI‐MS. Typical nonvolatile organic compounds [e.g. 5‐(hydroxymethyl)furfural and glucosan] were sensitively detected with MPT‐MS. Compounds of high polarity (e.g. amino acids, alkaloids and sugars) were easily profiled with iEESI‐MS. Our data showed that more analytes could be detected when more energy was delivered for the desorption ionization purpose; however, heat‐sensitive analytes would not be detected once the energy input exceeded the dissociation barriers of the analytes. We present a new procedure for the determination of oxygenated volatile organic compounds in samples of postoxidative effluents from the production of petroleum bitumens using dispersive liquid–liquid microextraction and gas chromatography with mass spectrometry. The eight extraction parameters were optimized for 43 oxygenated volatile organic compounds. The detection limits obtained ranged from 0.07 to 0.82 μg/mL for most of the analytes, the precision was good (relative standard deviation below 2.91% at the 5 μg/mL level and 4.75% at the limit of quantification), the recoveries for the majority of compounds varied from 70.6 to 118.9%, and the linear range was wide, which demonstrates the usefulness of the procedure. The developed procedure was used for the determination of oxygenated volatile organic compounds in samples of raw postoxidative effluents and in effluents after chemical treatment. In total, 23 compounds at concentration levels from 0.37 to 32.95 μg/mL were identified in real samples. The same samples were also analyzed in the SCAN mode, which resulted in four more phenol derivatives being identified and tentatively determined. The studies demonstrated the need for monitoring volatile organic compounds content in effluents following various treatments due to the formation of secondary oxygenated volatile organic compounds.  相似文献

The analysis of samples contaminated by organic compounds is an important aspect of environmental monitoring. Because of the complex nature of these samples, isolating target organic compounds from their matrices is a major challenge. A new isolation technique, solid phase microextraction, or SPME, has recently been developed in our laboratory. This technique combines the extraction and concentration processes into one step; a fused silica fiber coated with a polymer is used to extract analytes and transfer them into a GC injector for thermal desorption and analysis. It is simple, rapid, inexpensive, completely solvent-free, and easily automated. To minimize matrix interferences in environmental samples, SPME can be used to extract analytes from the headspace above the sample. The combination of headspace sampling with SPME separates volatile and semi-volatile analytes from non-volatile compounds, thus greatly reducing the interferences from non-target compounds. This paper reports the use of headspace SPME to isolate volatile organic compounds from various matrices such as water, sand, clay, and sludge. By use of the technique, benzene, toluene, ethyl-benzene, and xylene isomers (commonly known as BTEX), and volatile chlorinated compounds can be efficiently isolated from various matrices with good precision and low limits of detection. This study has found that the sensitivity of the method can be greatly improved by the addition of salt to water samples, water to soil samples, or by heating. Headspace SPME can also be used to sample semi-volatile compounds, such as PAHs, from complex matrices.  相似文献

We present a comparison of two sensitive methods, HPLC with fluorescence detector (HPLC/FLD) and UPLC with electrospray tandem mass spectrometry (UPLC/MS/MS), for the determination of indoleamine neurotransmitters (NTs) and their metabolites in sea lamprey plasma samples. Liquid–liquid extraction (LLE) and solid-phase extraction (SPE) were also tested for recovery and matrix effect. The recoveries of SPE determined by HPLC/FLD and UPLC/MS/MS ranged from 75 to 123% and 78 to 105%, respectively, while the recoveries of LLE ranged from 45 to 73% and 48 to 75%, respectively. SPE combined with HPLC/FLD and UPLC/MS/MS to determine the target analytes in plasma samples were validated of the sensitivity, reproducibility, accuracy and precision. Both methods exhibited excellent linearity in the range of 0.2–50 ng mL−1 for all analytes. The limits of detection (LOD) varied from 0.04 ng mL−1 to 0.13 ng mL−1 for HPLC/FLD method and 0.003 ng mL−1 to 0.02 ng mL−1 for UPLC/MS/MS method. The inter-day accuracy ranged from 82.5 to 127.0% for HPLC/FLD and 93.0 to 113.0% for UPLC/MS/MS. The inter-day precision ranged from 9.9 to 32.3% for HPLC/FLD and 5.4 to 13.2% for UPLC/MS/MS. These results demonstrated that the values obtained by both methods were within the satisfactory range and the UPLC/MS/MS method provided more accurate and precise measurements than HPLC/FLD method. The comparison is of great importance to determine the available detectors, considering the complexity and expensiveness versus quality parameters. These two methods were applied to the analysis of four important indoleamine neurotransmitter analytes (5-hydroxytryptamine, 5-hydroxyindole-3-acetic acid, tryptamine and melatonin) in sea lamprey plasma samples.  相似文献

建立了一种气相色谱(GC)与单光子电离-飞行时间质谱(SPI-TOF MS)联用(GC/SPI-TOF MS)的分析方法。首先,设计了一种双层套管的传输管用于连接GC与SPI-TOF MS,实现了GC与单光子电离离子源的无缝连接。在此基础上,以n-十五烷标准品和苯/甲苯/二甲苯的标准气为对象,对电离源的重要电压参数进行了优化,得到了纯净的分子离子峰,实现了对各类有机物的快速和准确定性。最后,将该方法用于分析柴油中的挥发性与半挥发性有机物,获得了柴油组分的二维GC×SPI-TOF MS谱图。不需要复杂的谱图解析和数据处理,根据谱图中离子的质荷比(m/z)归纳了柴油的主要成分,包括脂肪烃、芳香烃和含量很低的苯并吡咯等含氮化合物;根据色谱的保留时间将柴油中的同分异构体区分开来。结果表明GC/SPI-TOF MS法是一种简单、有效的分析方法,非常适于柴油及复杂环境样品等的分析表征。  相似文献

采用热脱附(TD)结合气相色谱-三重四极杆串联质谱(GC-MS/MS)建立了环境空气中23种挥发性有机物(VOCs)同时检测的分析方法。空气样品通过主动采样的方式富集到装有Tenax-TA填料的热脱附管中,热解吸后在选择反应监测(SRM)模式下用GC-MS/MS进行检测,内标法定量。结果表明,23种VOCs在0.01~1 ng和1~100 ng低、高两个范围内线性关系良好,相关系数(r2)均大于0.99,方法定量限为0.00008~1 μ g/m3。加标水平为2、10和50 ng时,23种VOCs的平均回收率为77%~124%。除了最低加标水平的氯苯,相对标准偏差(RSD, n=6)均小于20%。对市内3个采样点的环境空气进行测定,其中苯、甲苯、乙苯、二甲苯、苯乙烯、1,2,4-三甲基苯和六氯丁二烯均有检出。实验证明,该TD和GC-MS/MS相结合的检测方法具有准确、可靠、灵敏度高等优点,适用于环境空气中VOCs的同时测定。  相似文献

A variety of extraction procedures were evaluated for the extraction of arsenic and other analytes from lobster tissue samples using inductively coupled plasma mass spectrometry (ICP-MS) detection. Soxhlet, room temperature mixing, sonication, microwave assisted, supercritical carbon dioxide and subcritical water extractions were evaluated for a variety of solvent systems and optimum conditions determined using a partially defatted Lobster Hepatopancreas marine certified reference material, TORT-2 (National Research Council of Canada). The solubility trends and solvents into which the analytes extracted gave an indication as to the polar/non-polar nature of the compounds present. Analytes that prefer water are probably more polar or inorganic, while those preferring methanol solutions are less polar or organic in nature. Arsenic, cadmium, cobalt, molybdenum and selenium were probably all present in TORT-2 in both polar inorganic and non-polar organic forms. While TORT-2 may have contained similar amounts of selenium in both forms, the results suggested that more of the arsenic was present as less polar or more organic compounds, and cobalt existed mainly as more polar or inorganic species. Most of the extraction techniques suggested that, although there may be some less polar organic forms present, more of the cadmium was probably present as polar inorganic compounds. Additionally, most techniques indicated that molybdenum was possibly all less polar or more organic in nature. In general, microwave assisted extraction (MAE) yielded comparable or improved recoveries for all of the analytes monitored and usually required less solvent. Additionally, MAE proved to be the mildest, fastest, least complicated and most reproducible extraction technique evaluated. MAE at 75 degrees C for 2 min exposure time yielded quantitative recovery of arsenic from TORT-2. These conditions were evaluated for lobster tissue samples purchased from a local restaurant. Separate evaluation of the lobster meat and organs resulted in quantitative recoveries of arsenic from both tissue samples. The results indicated that the extraction efficiencies might have some dependence upon the extraction technique, extraction conditions, analyte, solvent, and sample matrix.  相似文献

This paper describes the matrix effect during the analysis of ten antibiotic compounds in water by SPE followed by HPLC‐ESI‐MS/MS. The target analytes were tetracycline, oxytetracycline (tetracyclines), sulfathiazole, sulfamethazine, sulfadiazine (sulfonamides), erythromycin‐H2O, roxithromycine, spiramycin (macrolides), ofloxacin, and norfloxacin (quinolones). The matrix effect was examined for internal standards and the target analytes in five different water matrixes, with signal suppression being increased in the order: ultrapure water, tap water, river water, sewage effluent, and sewage influent. A combined application of the internal standards and matrix‐matched extract calibration was shown to be successful in compensating the matrix effect for the analytes. The procedural recovery of the target compounds in sewage effluents and influents was higher than in river water samples, which was further enhanced by sample acidification to pH 2. The validity of the internal standard based matrix‐matched calibration approach was verified by the standard addition method.  相似文献

Known methods of quantitative chromatographic analysis (calibration, external standard, internal standard and standard addition) require the application of sample preparation techniques without significant losses of analytes. If this condition cannot be satisfied, the compensation of these losses should be provided. The modification of known method of quantitative chromatographic analysis (double internal standard), implying the addition of two homologues (previous and following) of target analytes as internal standards into initial samples is considered. This approach permits us to compensate significant losses both analytes and standards at all stages of sample preparation. The advantages of this method are demonstrated on the examples of liquid-liquid extraction, head space analysis (HSA), distillation of volatile compounds with volatile solvents (concentration in condensates) and evaporation of volatile solvents (concentrating in the residues of solvents). In all cases the application of two homologues as internal standards provides accurate results (the typical relative errors are within 1-6%) at the values of a factor of composition distortion of initial samples (K', the definition is suggested) from 0.2 up to 4. These results are in accordance with general relationships between variations in any physicochemical properties of organic compounds within homologous series. The single found exception was the evaporation of volatile solvents (the open phase transition process) when to get the results with relative errors not more then +10% requires the minimal changes in the composition of initial samples (K' values should not be more then approximately 1.5).  相似文献

Two on-line SPE-LC-ESI-MS/MS methods have been developed for the rapid determination and confirmation of 18 polar pesticides and nine transformation products (TPs) in water samples. Given the very different physico-chemical characteristics of the analytes, it was not feasible the simultaneous determination of all selected compounds in only one method. Thus, it was necessary to use heptafluorobutyric acid and formic acid in order to obtain good retention in the SPE cartridge for basic and acidic analytes, respectively. The developed analytical methodology based on the direct injection of 2 mL of water sample in the system allowed the quantification of all analytes at the 25 ng/L level (LOQ) with limits of detection normally lower than 5 ng/L. Satisfactory recoveries (70-110%) were obtained for most compounds in ground and surface water samples. Some exceptions were found mainly in surface water, due to the ion suppression produced by the higher amount of matrix interferents in these samples. The acquisition of two MS/MS transitions for each compound allowed the reliable confirmation of positive findings even at the LOQ level. The developed methodology was applied to real ground and surface water samples showing the interest of including TPs in monitoring methods, as several of them were found at concentrations higher than that of parent compounds.  相似文献

Solid-phase extraction (SPE) procedure on octadecylsilica (C18) was developed for accumulation of volatile compounds from grape distillates. The procedure was optimised for final analysis by capillary gas chromatography. At mass concentrations in model solutions ranging from 0.1 to 50 mg/l solid-phase extraction recoveries of all analytes ranged from 69% for 2-phenylethanol to 102% for capric acid, with RSD values from 2 to 9%. SPE recoveries of internal standards to be added in the sample solution prior to extraction, higher alcohols 2-ethyl-1-hexanol and 1-undecanol, were 97 and 93%, respectively, with RSD values of 3%. Detection limits of analyzed compounds in model solutions ranged from 0.011 mg/l for isoamyl acetate to 0.037 mg/l for caproic acid. Method efficiency was tested in relation to acetic acid content, volume fraction of ethanol and possible matrix effects. A significant influence of matrix on SPE efficiency for geraniol, cis-2-hexen-1-ol and cis-3-hexen-1-ol was detected. For the same reason, 2-phenylethanol could not be determined by developed SPE method in samples of grape distillates. The developed solid-phase extraction method was successfully applied to determine the differences in volatile compound content in different grape distillates produced by the distillation of crushed, pressed and fermented grapes.  相似文献

The potential of gas chromatography coupled to tandem mass spectrometry (GC/MS/MS) with a triple quadrupole analyzer (QqQ) has been investigated for the quantification and reliable identification of sixteen polycyclic aromatic hydrocarbons (PAHs) from the EPA priority list in animal and vegetable samples from aquaculture activities, whose fat content ranged from 5 to 100%. Matrices analyzed included fish fillet, fish feed, fish oil and linseed oil. Combining optimized saponification and solid‐phase extraction led to high efficiency in the elimination of interfering compounds, mainly fat, from the extracts. The developed procedure minimized the presence of these interfering compounds in the extracts and provided satisfactory recoveries of PAHs. The excellent sensitivity and selectivity of GC/(QqQ)MS/MS in selected reaction monitoring (SRM) allowed to reach limits of detection at pg/g levels. Two SRM transitions were acquired for each analyte to ensure reliable identification of compounds detected in samples. Confirmation of positive findings was performed by GC coupled to high‐resolution time‐of‐flight mass spectrometry (GC/TOFMS). The accurate mass information provided by GC/TOFMS in full acquisition mode together with its high mass resolution makes it a powerful analytical tool for the unequivocal confirmation of PAHs in the matrices tested. The method developed was applied to the analysis of real‐world samples of each matrix studied with the result of detecting and confirming the majority of analytes at the µg/kg level by both QqQ and TOF mass spectrometers. A fast method was developed for the extraction and analysis of volatile organic compounds, including disinfection by-products (DBPs), with headspace solid-phase microextraction (HS-SPME) and gas chromatography/mass spectrometry (GC/MS) techniques. A GC/time-of-flight (TOF)-MS instrument, which had fast acquisition rates and powerful deconvolution software, was used. Under optimum conditions total runtime was 45s. Volatile organic compounds (VOCs), including purgeable A and B compounds (listed in US Environmental Protection Agency method 624), were identified in standard water samples. Extraction times were 1min for more volatile compounds and 2min for less volatile compounds. The method was applied to the analysis of water samples treated under different disinfection processes and the results were compared with those from a liquid-liquid extraction method.  相似文献

Traditional matrix does not allow matrix-assisted laser desorption/ionization mass spectrometry(MALDI MS) to analyze volatile compounds,because volatile analytes may vaporize during the sample preparation process or in the high vacuum circumstance of ion source.Herein,we reported a Co and N doped porous carbon material(Co-NC) which were synthesized by pyrolysis of a Schiff base coordination compound.Co-NC could simultaneously act as adsorbent of volatile compounds and as matrix of MALDI MS,to provide the capability of MALDI MS to analyze volatile compounds.As adsorbent,Co-NC could stro ngly adsorb and enrich the volatile compounds in perfume and herbs,and hold them even in the high vacuum circumstance.On the other hand,Co-NC could absorb the energy of the laser,and then transfer the energy to the analyte for desorption and ionization of analyte in both negative and positive ionization modes.Additionally,the background interferences were avoided in the low-mass region(<500 Da) when using Co-NC as matrix,overcoming the challenges of MALDI MS analysis of small molecule compounds.In summary,Co-NC as matrix tremendously extended the application of MALDI MS.  相似文献

An ultra performance liquid chromatography electrospray tandem mass spectrometry (UPLC/MS/MS) method was developed and validated for the determination of 17 antibiotics in natural waters in one single extraction and chromatographic procedure. Gradient separation conditions were optimised for 17 compounds belonging to five different antibiotic groups: quinolones (oxolinic acid, nalidixic acid, pipemidic acid, flumequine), fluoroquinolones (enoxacin, ciprofloxacin, norfloxacin, ofloxacin, enrofloxacin, sarafloxacin, danofloxacin, difloxacin, lomefloxacin), sulphonamides (sulphamethoxazole, sulphamethazine), nitro-imidazole (ornidazole) and diaminopyrimidine (trimethoprim). The separation of all compounds, obtained using a 1.7 μm particle size column (100 mm?×?2.1 mm), was achieved within 10 min time. Water samples were adjusted to pH 7 and extracted using Oasis hydrophilic–lipophilic balance (HLB) solid phase extraction cartridges. After elution with methanol and concentration, extracts were injected in a C18 column (Acquity UPLC BEH C18) and detected by tandem mass spectrometry. Average recovery from 100 ng L?1 fortified samples was higher than 70% for most of the compounds, with relative standard deviations below 20%. Performances of the method (recoveries, detection limit, quantification limit and relative standard deviation) and matrix effects were studied, and results obtained showed that method was suitable for routine analysis of antibiotics in surface water. Samples analysis from Seine River (France) confirmed the interest of antibiotic contamination evaluation in that area.

建立了同时测定调味品中糠醛、5-甲基糠醛和5-羟甲基糠醛的气相色谱-串联质谱法(GC-MS/MS)。对样品的前处理方法及GC-MS/MS分析条件进行优化,样品经乙酸乙酯萃取,GC-MS/MS多反应监测模式(MRM)进行测定。3种糠醛类物质在0.001~20 mg/L范围内均呈良好的线性关系,相关系数为0.999 0~0.999 5;检出限(S/N=3)均为0.005 mg/kg,加标回收率为86%~95%。该法简便、快速、溶剂用量少,可消除调味品中复杂基质的干扰,结果准确可靠、灵敏度高,适用于调味品中3种糠醛类物质的同时测定。  相似文献

This paper proposes a multiple headspace solid-phase microextraction (MHS-SPME) method coupled to gas chromatography-tandem mass spectrometry detection (GC/MS/MS) for the simultaneous determination of 2,4,6-trichloroanisole, 2,3,4,6-tetrachloroanisole, pentachloroanisole, 2,4,6-tribromoanisole, 4-ethylphenol, 4-ethylguaiacol, 4-vinylphenol and 4-vinylguaiacol in wines. These compounds are involved in the presence of "cork taint" and Brett character in wines. The MHS-SPME method is a modification of SPME developed for quantitative analysis that avoids possible matrix effects based on an exhaustive analyte extraction from the sample. After demonstrating the existence of matrix effect in the analysis of the target compounds by HS-SPME with a divinylbenzene/Carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fibre, the MHS-SPME method was developed and validated. The proposed method showed satisfactory linearity, precision and detection limits, all below the odour detection thresholds of the compounds in wine matrices. Good recoveries were observed for all compounds, always above 90%, and the repeatability obtained was considered acceptable, ranging between 2 and 11%. After checking the applicability of the method by comparing the results recorded with those obtained with the standard addition method, the method was applied successfully to the analysis of wine samples. To our knowledge, this is the first time that MHS-SPME combined with GC/MS/MS has been applied to simultaneously determine haloanisoles and volatile phenols in wine.  相似文献

Pressurized liquid extractions were performed on eight sediments in order to investigate if a modified US EPA method (100 °C, 100 bar, n-heptane/acetone (1:1), 2 × 5 min) provided exhaustive extractions of polychlorinated biphenyls (PCBs) from sediment, and to study if the extractability of PCBs from the different sediments was affected by characteristics of the sediment. The recovery from the eight native sediments, contaminated in nature, was between 96.4% and 98.9%, as an average of the recoveries from 10 PCB congeners. Hundred percent recovery was defined as the sum of two consecutive extractions (2 × 5 min each) at the stated conditions. The recoveries of the individual congeners were above 94%, except for one congener in one sediment, which had a recovery of 92%. When the recoveries and different characteristics of the sediments were compared, no correlation appeared between recoveries and sediment PCB concentration, total organic carbon (TOC), soot carbon (SC) or amorphous carbon (AC). The fact that carbon did not influence the extractions was somewhat surprising, since previous experiments have indicated a connection. Instead, statistically significant (p < 0.05) correlations were observed for water content and carbon/nitrogen (C/N) ratio. The decrease in recoveries with decreased water content was attributed to less access of the solvent to the analytes due to less matrix swelling. The lowered recoveries with increased C/N ratio can indicate that a difference in structure of the organic matter exists, which influences the binding strength between the analytes and the matrix. The difference in structure can possibly be explained by different origin of the organic matter or by aging effects. Overall the method was found to be exhaustive and the excellent recoveries show that sediment characteristics do not influence the extractions markedly.  相似文献

A method was developed for screening crops for a range of pesticide residues by liquid chromatography/tandem mass spectrometry (LC/MS/MS). A complete set of LC, electrospray ionization (ESI), and tandem MS acquisition parameters was established for the determination of 108 analytes; these parameters were used for the simultaneous acquisition of 98 analytes in the positive ESI mode and 10 analytes in an additional MS/MS method in the negative ESI mode. The entire procedure involves extraction of residues with methanol-water and partition into dichloromethane. The utility of the method is demonstrated by the analysis of crops of 5 matrix types (water-containing, acidic, dry, sugar-containing, and fatty). Of 108 pesticides/metabolites tested, 104 showed sufficient stability in most matrixes for determination by LC/MS/MS. These analytes belong to 20 chemical classes, which demonstrate the general applicability of the method for multiclass analysis. By using matrix-matched standards, 67 compounds could be determined in most matrixes with recoveries of 70-120% and a relative standard deviation of < or = 25% at the 0.01 mg/kg level.  相似文献
