A simple, sensitive, and precise high performance liquid chromatographic method for the analysis of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride, with ultraviolet detection at 210 nm, has been developed, validated, and used for the determination of compounds in commercial pharmaceutical products. The compounds were well separated on a Hypersil BDS C18 reversed-phase column by use of a mobile phase consisting of 0.05 M, 4.70 pH, potassium dihydrogen phosphate buffer - acetonitrile (720:280 v/v) at a flow rate of 1.0 mL min?1. The linearity ranges were 400–4,000 ng mL?1 for pantoprazole, 200–2,000 ng mL?1 for rabeprazole, 400–4,000 ng mL?1 for esomeprazole, 300–3,000 ng mL?1 for domperidone and 500–5,000 ng mL?1 for itopride. Limits of detection (LOD) obtained were: pantoprazole 147.51 ng mL?1, rabeprazole 65.65 ng mL?1, esomeprazole 131.27 ng mL?1, domperidone 98.33 ng mL?1 and itopride 162.35 ng mL?1. The study showed that reversed-phase liquid chromatography is sensitive and selective for the determination of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride using single mobile phase.  相似文献

A sensitive, simple, and accurate method for determination and pharmacokinetic study of ferulic acid and isoferulic acid in rat plasma was developed using a reversed-phase column liquid chromatographic (RP-LC) method with UV detection. Sample preparations were carried out by protein precipitation with the addition of methanol, followed by evaporation to dryness. The resultant residue was then reconstituted in mobile phase and injected into a Kromasil C18 column (250 × 4.6 mm i.d. with 5 μm particle size). The mobile phase was methanol-1% formic acid (33:67, v/v). The calibration plots were linear over the range 5.780–5780 ng·mL?1 for ferulic acid and 1.740–348.0 ng·mL?1 for isoferulic acid. Mean recoveries were 85.1% and 91.1%, respectively. The relative standard deviations (RSDs) of within-day and between-day precision were not above 15% for both of the analytes. The limits of quantification were 5.780 ng·mL?1 for ferulic acid and 1.740 ng·mL?1 for isoferulic acid. This RP-LC method was used successfully in pharmacokinetic studies of ferulic acid and isoferulic acid in rat plasma after intravenous injection of Guanxinning Lyophilizer.  相似文献

A simple, rapid, specific and sensitive method has been developed and validated for simultaneous determination of lopinavir and ritonavir in human plasma. The analytical procedure involves a protein precipitation method using fluoxetine as internal standard Separation was carried out on an Inertsil ODS column using a mobile phase consisting of acetonitrile and 5 mM ammonium acetate buffer. The total run time of analysis was 2.0 min. A linear response function was established for the range of concentrations 50.67–10,008.82 ng mL?1 for lopinavir and 5.066–1,000.693 ng mL?1 for ritonavir. The method was successfully applied to an oral bioequivalence study in humans.  相似文献

A simple, rapid and sensitive HPLC method has been developed and validated for the analysis of letrozole in human plasma. The separation was achieved on a monolithic silica column using acetonitrile–phosphate buffer. A fluorescence detector was used for the quantitation with excitation and emission wavelengths at 230 and 295 nm. The assay enables the measurement of letrozole for therapeutic drug monitoring with a minimum quantification limit (LOQ) of 0.5 ng mL?1. The method involves a simple, one-step extraction procedure with complete recovery. Calibration was linear over the concentration range 0.5–80 ng mL?1. The coefficients of variation for inter-day and intra-day assay were found to be less than 8%.  相似文献

Rhodamine 123 has been frequently used to evaluate the functional activity of P-glycoprotein, assess the related drug interactions, analyze mitochondrial distribution and function, sort functionally distinct cell subpopulations and measure mitochondrial or cellular membrane potential. We developed a sensitive and rapid liquid chromatographic method with fluorescence detection after simple sample preparation procedure for the determination of Rhodamine 123 in P-glycoprotein efflux studies. The mobile phase consisted of methanol and 15 mM dibasic sodium phosphate buffer (pH 6.0) (80:20,v/v), delivered at a rate of 1.0 mL min?1. 15 μL of the samples were injected into a reversed-phase C18 column with a fixed excitation wavelength at 505 nm and altered emission wavelengths. The whole LC analysis was accomplished within 6 min. The established linearity range from 1 to 100 ng mL?1, with the inter-day and intra-day RSD below 7.57 and 4.89% at concentrations of 4.4, 44 and 88 ng mL?1. All the calibration standards and quality controls were prepared in cell lysate and were stable for three freeze-thaw cycles, for 12 h at 4 °C and for 6 h at room temperature. This rapid LC method has been applied to the quantification of Rhodamine 123 in cell lysate obtained from P-glycoprotein efflux study conducted in rat brain capillary endothelial cells.  相似文献

An accurate and sensitive reversed-phase high-performance liquid chromatographic method for analysis of sertraline in human serum, using 4-chloro-7-nitrobenzofurazan as pre-column derivatization agent, is described. The drug and an internal standard (azithromycin) were extracted from serum by use of a mixture of diethyl ether and chloroform, and subjected to pre-column derivatization with the reagent. Analysis of the resulting derivatives was performed on a 250 mm × 4.0 mm cyano column with 63:37 (v/v) methanol–sodium phosphate buffer (0.05 M, pH 3.7) containing 2 mL L?1 triethylamine as mobile phase. Detector response was monitored at excitation and emission wavelengths of 470 and 537 nm, respectively. The calibration plot was linear over the concentration range 2–640 ng mL?1. The lower limits of detection and quantification were 0.5 and 2 ng mL?1, respectively. The method was validated for specificity, sensitivity, linearity, precision, accuracy, and stability and shown to be accurate (intra-day and inter-day accuracy from 0.3 to 4.2%) and precise (intra-day and inter-day precision from 2.4 to 15.5%). The drug was detected at concentrations as low as 2 ng mL?1 in 0.5 mL serum and the method described can be easily applied to human single-dose pharmacokinetic studies of sertraline.  相似文献

Abstract

A simple and reliable high-performance liquid chromatographic (HPLC) method was developed for the determination of belotecan in the plasma, urine, and bile samples of rats. Belotecan was analyzed with HPLC using a C18 column with fluorescence detector. A mixture of acetonitrile–0.1 M potassium phosphate buffer at pH 2.4 (25:75, v/v) and 0.2% trifluoroacetic acid was used as the mobile phase. The lower limits of quantitation (LOQ) were 5 ng mL?1 for the plasma and 5 µg mL?1 for the urine and bile samples. The method has been readily applied for the routine pharmacokinetic study of belotecan in small laboratory animals.  相似文献

A rapid, sensitive, and specific high-performance liquid chromatography tandem mass spectrometric method was developed for the simultaneous determination and confirmation of amoxicillin and clavulanic acid in plasma. Plasma sample was subjected to a simple deproteinization with acetonitrile, and then the supernatant was directly diluted by water. Analysis was performed on a Phenomenex Luna C8 reversed-phase column by detection with mass spectrometry in negative ions multiple reaction monitoring mode. A gradient elution program with 0.1% formic acid and acetonitrile was performed at a flow of 0.25 mL min?1. There is good linearity in the range of 0.5–500 ng mL?1 for both amoxicillin and clavulanic acid. The decision limits of amoxicillin and clavulanic acid were 0.06 ng mL?1 and 0.08 ng mL?1 in plasma, respectively, and the detection capabilities of two analytes were below 0.5 ng mL?1. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. The extraction recoveries of amoxicillin and clavulanic acid were between 102% and 115% in plasma at three spiked levels of 0.5, 50, and 500 ng mL?1, with the relative standard deviations less than 15% for each analyte. The developed method was applied to pharmacokinetic studies of amoxicillin and clavulanic acid tablets in healthy beagles.  相似文献

A new LC method has been developed and validated for the direct determination of bupropion and its main metabolite, hydroxybupropion in human plasma. Plasma samples were analyzed after a simple, one step protein precipitation with trichloroacetic acid using a C8 column and mobile phase, consisting of methanol/acetonitrile/phosphate buffer (10 mM, pH 3.0) (40:10:50, v/v/v) and 20 mM 1-heptane sulfonic acid sodium salt with carbamazepine as the internal standard. UV detection was performed at 214 and 254 nm. The method was validated over the concentration range of 60–2,400 and 150–4,700 ng mL?1 for bupropion and hydroxybupropion, respectively. The intra- and inter-day assay variability was less than 15% for the two analytes. Limit of detection values were 24.8 and 63.4 ng mL?1 for bupropion and hydroxybupropion, respectively. The method developed was applied to quantification of bupropion and hydroxybupropion in human plasma.  相似文献

A simple, sensitive and selective LC–MS–MS method has been developed for the simultaneous determination of acyclovir and valacyclovir in human plasma. Acyclovir and valacyclovir in plasma were concentrated by solid phase extraction and chromatographed on a C18 column using a mobile phase of 0.1% formic acid: methanol (30:70% v/v). The method was validated over a linear range of 47–10,255 and 5–1,075 ng mL?1 for acyclovir and valacyclovir respectively. The LOQs were 47.6 and 5.0 ng mL?1. The validated method was applied for the quantitation of acyclovir and valacyclovir from plasma samples in a pharmacokinetic study.  相似文献

A rapid LC method with UV detection was developed for the quantification of carboxylic acid metabolite of clopidogrel in human plasma. Following a simple protein precipitation using a mixture of methanolic solution of ZnSO4, the analyte and commercially available internal standard were separated using a mobile phase of water–acetonitril (85:15, v/v) adjusted to pH 3.5 on a Chromolith C18 column at a flow rate of 2.5 mL min?1 with a total retention time of 4 min. Linearity was verified over the range of 20–3,000 ng mL?1 where the LOQ was 20 ng mL?1. This method was applied in a pharmacokinetic study.  相似文献

A sensitive, stability-indicating liquid-chromatographic method for analysis of racecadotril in the presence of its degradation products has been developed and validated. Efficient chromatographic separation was achieved on a C18 column with a simple isocratic mobile phase—60:40 methanol–water. Quantification was by photo-diode array (PDA) detection at 220 nm. The linearity of the method was excellent over the range 1–32 μg mL?1. The method was sensitive, with low limits of detection (20 ng mL?1) and quantification (100 ng mL?1). The recovery of the method was consistently good (98.7–100.9%), with low (<1%) intra-day and inter-day relative standard deviation. Robustness studies confirmed that peak area was unaffected by small changes in temperature, and mobile phase composition and flow rate. Both alkaline and acidic hydrolytic degradation were performed in methanolic solution. In alkaline medium the drug was degraded immediately; it was degraded within 90 min in acidic medium. The validated, stability-indicating, method was used for analysis of racecadotril in pharmaceutical dosage form and also to reveal the hydrolytic degradation profile of the racecadotril.  相似文献

A sensitive liquid chromatography?Cmass spectrometry method for the simultaneous determination of sertraline (SER) and its major metabolite norsertraline (NOR) from serum was developed and validated in the context of a pharmacokinetic study in pregnant women. The separations were achieved on a silica column with a non-aqueous polar mobile phase consisting of acetonitrile, methanol and ammonium acetate at a flow rate of 0.5 mL min?1. The concentrations were measured using a single quadruple mass spectroscopic detector supplied with atmospheric pressure ionization electrospray. Sample preparation consisted of a simple liquid?Cliquid procedure. The detector was set in selective ion mode for each compound of interest, 306 m/z for SER and 275 m/z for NOR. Calibration curves were generated by least square linear regression for concentration of 5?C160 ng mL?1 for SER and from 10 to 320 ng mL?1 for NOR. The curves for both compounds of interest were linear, with correlation coefficients r 2 ?? 0.999.  相似文献

A rapid, simple, sensitive and specific liquid chromatography–tandem mass spectrometry method was developed and validated for simultaneous quantification of itopride hydrochloride and domperidone in human plasma. Both drugs were extracted by liquid–liquid extraction with ethyl acetate and saturated borax solution. The chromatographic separation was performed on a reversed-phase C18 column with a mobile phase of water–methanol (2:98, v/v) containing 0.5% formic acid. The protonated analyte was quantitated in positive ionization by multiple reaction monitoring with a mass spectrometer. The assay exhibited linearity over the concentration range of 3.33–500 ng mL?1 for itopride hydrochloride and 3.33–100 ng mL?1 for domperidone in human plasma. The precursor to product ion transitions of m/z 359.1–72.3 and 426.0–147.2 were used to measure itopride hydrochloride and domperidone respectively. The method was found suitable for the analysis of plasma samples collected during phase 1 pharmacokinetics study of itopride HCl 50 mg and domperidone 20 mg in 12 healthy volunteers after single oral doses of the combination drug.  相似文献

A simple and specific analytical method for the quantitative determination of shikimic acid from the methanol extract of the fruits of Illicium species and from various plant samples was developed. The LC–UV separation was achieved by reversed-phase chromatography on a C18 column with potassium dihydrogen phosphate and methanol as the mobile phase. In the LC–MS method, the separation was achieved by a C12 column using water and acetonitrile, both containing 0.1% acetic acid as the mobile phase. The methods were successfully used to study the percentage compositions of shikimic acid present in nine species of Illicium and various other plant samples. The detector response was linear with concentrations of shikimic acid in the range from 1.0–500.0 μg mL?1 by LC–UV and 100–1000 ng mL?1 by LC–MS. Mass spectrometry coupled with electrospray ionization interface is described for the identification of shikimic acid in various plant samples. This method involved the use of the [M-H]? ions of shikimic acid at m/z 173.0455 (calculated mass) in the negative ion mode with extractive ion monitoring.  相似文献

A selective, sensitive, and accurate method has been developed and validated for the quantification of tangeretin in rat plasma. The application of LC-electrospray-ion trap mass spectrometry in full scan and multiple reactions monitoring modes were investigated. Following solid phase extraction using a hydrophilic–lipophilic balance cartridge, the analytes were separated on a C18 column using an isocratic mobile phase composed of acetonitrile/water (50:50, v/v) containing 0.3% formic acid. In full scan mode, the LOQ was 2 ng mL?1. The standard calibration curve was linear (R 2 = 0.9999) over the concentration range 2–200 ng mL?1. The precision over the concentration range was within 15% (RSD) and the accuracy was ranged from 86 to 115%. In multiple reaction monitoring mode, the LOQ was 1 ng mL?1 and the standard calibration curve was linear (R 2 = 0.9976) over the concentration range 1–100 ng mL?1 with a precision of 12% and accuracy rangeing from 91 to 113%.  相似文献

A simple, sensitive, precise and accurate reversed phase liquid chromatographic method has been developed for the simultaneous estimation of atorvastatin (AT) calcium, ramipril (RA) and aspirin (AS) from capsule dosage form. The method was developed using a Phenomenex Luna C18 (250 mm, 4.6 mm i.d., 5 µm) column with a mobile phase consisting of 0.1%, orthophosphoric acid buffer:acetonitrile:methanol (45:50:5 v/v/v), pH 3.3, at a flow rate of 1 mL min?1. Detection was carried out with ultra-violet detection at 210 nm. The retention times were about 12.19, 2.35, and 3.95 min for AT calcium, RA and AS, respectively. The developed method was validated for linearity, accuracy, precision, limit of detection, limit of quantitation and robustness. The linearity ranges were 1–6 µg mL?1 for AT calcium, 0.5–3 µg mL?1 for RA and 7.5–45 µg mL?1 for AS with mean recoveries of 100.59 ± 0.68, 100.62 ± 0.83 and 100.49 ± 0.73% for AT calcium, RA and AS, respectively. Limit of detection obtained were 29.85 ng mL?1 for AT calcium, 4.71 ng mL?1 for RA and 85.13 ng mL?1 for AS. Impurity of salicylic acid was found in capsule dosage form at the retention time of about 4.84 min. The proposed method can be used for the estimation of these drugs in combined dosage forms.  相似文献

A simple, rapid and sensitive liquid chromatography/electrospray tandem mass spectrometry quantitative detection method, using amantadine as internal standard, was developed for the simultaneous analysis of paracetamol, pseudoephedrine and chlorpheniramine concentrations. Analytes were extracted from plasma samples by liquid–liquid extraction with n-hexane–dichloromethane–2-propanol (2:1:0.1, v/v), separated on a C18 reversed-phase column with 0.1% formic acid–methanol (40:60, v/v) and detected by electrospray ionization mass spectrometry in positive multiple reaction monitoring mode. Calibration curves for plasma were linear over the concentration range 10–10,000 ng mL?1 of paracetamol, 2–2,000 ng mL?1 of pseudoephedrine and 0.2–200 ng mL?1 of chlorpheniramine. The method has a lower limit of quantitation of 10 ng mL?1 for paracetamol, 2.0 ng mL?1 for pseudoephedrine and 0.2 ng mL?1 for chlorpheniramine. Recoveries, precision and accuracy results indicate that the method was reliable within the analytical range, and the use of the internal standard was very effective for reproducibility by LC-MS-MS. This method is feasible for the evaluation of pharmacokinetic profiles of a novel multicomponent sustained release formulation containing 325 mg of paracetamol, 30 mg of pseudoephedrine hydrochloride and 2 mg of chlorpheniramine maleate. It is the first time the pharmacokinetic evaluation of a novel sustained-action formulation containing paracetamol, pseudoephedrine and chlorpheniramine has been elucidated in vivo using LC-MS-MS.  相似文献

A reversed-phase high-performance liquid chromatographic method coupled with photodiode-array detection (HPLC–PAD) has been developed and validated for simultaneous quantitation of dl-tetrahydropalmatine (dl-THP) and protopine (PTP), two active constituents of the herbal medicine Rhizoma Corydalis, in rabbit plasma. The pharmacokinetics of dl-THP and PTP were researched in rabbits after oral administration of extracts of Rhizoma Corydalis. Optimum separation was achieved on a reversed-phase column with methanol–phosphate buffer (pH 7.75, 65:35, v/v) as mobile phase. The experimental results showed the intra-day and inter-day precision and accuracy of the method were satisfactory for simultaneous determination of dl-THP and PTP at low, middle, and high concentrations. The lower limits of quantitation were 2.05 ng mL?1 for dl-THP and 2.10 ng mL?1 for PTP. Plasma concentration–time data for dl-THP were best fitted by the two-compartment linear pharmacokinetic model whereas data for PTP were best fitted by the one-compartment model.  相似文献
