Synthesis and structure of 2-(nitromethylene)-2H-1,4-benzothiazin-3(4H)-one

A new representative of benzothiazines, (2Z)-2-(nitromethylene)-2H-1,4-benzothiazin-3(4H)-one, was synthesized, and its structure was studied. According to ¹H NMR, IR, and UV spectral and X-ray diffraction data, the molecules are planar highly conjugated structures that exist in crystal as centrosymmetric dimers; the latter form layers overlapping with each other and held together by strong π-π interaction between the carbonyl groups of neighboring layers.     

Design,synthesis and biological evaluation of 5-(2-(4-(substituted benzo[d]isoxazol-3-yl)piperazin-1-yl)acetyl)indolin-2-one and 5-(2-(4-substitutedpiperazin-1-yl)acetyl)indolin-2-one analogues as novel anti-tubercular agents

A series of thirty-six novel 5-(2-(4-(benzo[d]isoxazol-3-yl)piperazin-1-yl)acetyl)indolin-2-one and 5-(2-(4-substitutedpiperazin-1-yl)acetyl)indolin-2-one analogues were synthesized, characterized and screened for their in vitro anti-tubercular activity against Mycobacterium tuberculosis H37Rv strain. These compounds exhibited minimum inhibitory concentration between 1.56 and 50 μg/mL. Among these derivatives, compounds 10c, 10d, 10j, 10o and 10v (MIC 6.25 μg/mL) displayed moderate activity, while compounds 10e, 10l, 10q, 10w,10x, 12d, 12e and 12i (MIC 3.12 μg/mL) showed good anti-tubercular activity and compounds 10f, 10k, 10p, 10r, 12f, 12j and 12k (MIC 1.56 μg/mL) exhibited excellent anti-tubercular activity. In addition, MTT assay was accomplished on the active analogues of the series against mouse macrophage (RAW 264.7) cells to evaluate the cytotoxic effect of the newly synthesized compounds and selectivity index of the compounds was determined.

One-pot synthesis of 3-(2-cyanophenyl)quinazolin-4(3H)-one

Anthranilonitrile reacting with formic acid at room temperature for three days gave 64% of 3-(2-cyanophenyl)quinazolin-4(3H)-one. Palladium-catalyzed carbonylative annulation reaction of 2-(1-alkynyl)benzenamines: selective synthesis of 3-(halomethylene)indolin-2-ones

A novel and selective palladium-catalyzed carbonylative annulation process for the synthesis of 3-(halomethylene)indolin-2-ones was demonstrated. In the presence of PdX2 and CuX2, 3-(halomethylene)indolin-2-ones were selectively obtained from the carbonylative annulations of 2-(1-alkynyl)benzenamines with CO in moderate to good yields.     

Efficient synthesis of new oxindol-based heterocyclic entities via indolin-2-one derivatives

New series of oxindol-based heterocyclic entities (2–11) have been designed and synthesized using indolin-2-one derivatives as key materials (1a–d). The chemical structures of the new synthesized compounds were characterized by FTIR, ¹HNMR, ¹³CNMR, MS spectroscopy and elemental analyses. Three of the newly synthesized compounds were tested for anticancer activity in the National Cancer Institute (NCI) against human panel breast cancer cell line MCF7, from the in vitro assays compound 6c presented promising anti-cancer activity using Doxorubicin as a reference. Compound 6c could be a lead compound for discovery of new anticancer agent.     

5-(Trimethylstannyl)-2H-pyran-2-one and 3-(Trimethylstannyl)-2H-pyran-2-one: New 2H-Pyran-2-one Synthons

5-(Trimethylstannyl)-2H-pyran-2-one (11) and 3-(trimethylstannyl)-2H-pyran-2-one (30), readily prepared from the corresponding bromo-2H-pyran-2-ones, undergo Pd(0)-catalyzed coupling reactions with a variety of enol triflates to give 5- and 3- substituted 2H-pyran-2-ones, respectively. This reaction is applicable to the enol triflates of 14beta-hydroxy-17-ketosteroids, and therefore may prove useful in convergent syntheses of lucibufagins and bufadienolides.     

超声技术合成1,5-二-(4-羟基苯基)-1,4-戊二烯-3-酮

利用对羟基苯甲醛和丙酮为原料,以氯化氢气体为催化剂,超声波辐射下成功合成了1,5-二-(4-羟基苯基)-1,4-戊二烯3-酮(HPD).与传统方法相比,该方法具有反应时间短,操作简便及产率高等优点,并讨论了影响产率的因素.     

Electrochemical synthesis of 4-(dihydroxyphenylthio)-2H-chromen-2-one derivatives

The 4-(dihydroxyphenylthio)-2H-chromen-2-one derivatives have been synthesized by direct electrochemical oxidation of catechols in the presence of 4-mercaptocoumarin as a nucleophile in water/acetonitrile (50/50) solution, in a one-pot process, at carbon rod electrode, in an undivided cell and in constant current conditions, through an EC mechanism. The products are characterized by spectra data. Besides, the difference in electrochemical oxidation of catechol in the presence of 4-hydroxycoumarin and 4-mercaptocoumarin explained by computational structure, natural bond orbital (NBO) analysis and density functional theory (DFT: B3LYP/6-31G*//B3LYP/6-31G*) based methods, using the GAUSSIAN 98 package of programs.     

Facile synthesis of 3,3-di(heteroaryl)indolin-2-one derivatives catalyzed by ceric ammonium nitrate (CAN) under ultrasound irradiation

Ceric ammonium nitrate efficiently catalyzes the reaction of isatin with indoles under sonic waves to afford symmetrical 3,3-di(indolyl)indolin-2-ones in excellent yields, as well as the reaction of 3-hydroxy-3-indolylindolin-2-ones with indoles, pyrrole to afford the corresponding adducts in excellent yields, which provides an efficient route to the synthesis of symmetrical and unsymmetrical 3,3-di(indolyl)indolin-2-one derivatives, respectively.
