The separation and determination of seven sulphonamides and trimethoprim by micellar electrokinetic capillary chromatography were successfully achieved, employing sodium dodecyl sulphate (SDS) as a micellar phase and tetrabutylammonium bromide as additive. The effects of surfactant and modifier concentrations, pH and applied voltage on the retention behaviour of the solutes and the column efficiency were studied. The migration time of sulponamides increase with increasing SDS concentration and decreasing the applied voltage, but varies only slightly with pH. There is an optimum applied voltage at which a higher theoretical plate number is achieved, in contrast to the sulphonamides, the retention behaviour of trimethoprim gave a more obvious response to changes in the experimental conditions. The determination of three active ingredients in tablets was performed using sulphathiazole as an internal standard with good results. The theoretical plate number ranged between 2.0 x 10(5) and 2.8 x 10(5) with a 50-cm capillary.  相似文献

The separation of fourteen active ingredients used in a cold medicine was investigated by micellar electrokinetic chromatography (EKC) employing bile salts. Basic drugs were also successfully separated by micellar EKC using bile salts with high theoretical plate numbers (2.0 x 10(5)-3.5 x 10(5)) within a relatively short time (ca. 20 min). The separation of these solutes by micellar EKC was not successful using sodium dodecyl sulphate. The effects of micellar concentration, pH and organic modifier content on migration times and selectivity were investigated. This technique was also applied to the determination of several active ingredients combined in commercial preparations by an internal standard method.  相似文献

建立了胶束电动毛细管色谱分离和测定大黄及其制剂三黄片中蒽醌类活性组分的方法.考察了背景电解质pH、表面活性剂浓度、有机改性剂种类和浓度对分离的影响.实验结果表明:在缓冲液pH为9.5、SDS浓度为25mmol/L、乙氰浓度为20%时的优化条件下,大黄及三黄片中蒽醌类活性组分得到基线分离且方法具有较好的重现性.  相似文献

A method of micellar electrokinetic capillary electrophoresis for determining the six flavonoids in Scutellariae Radix (i.e., baicalin, baicalein, wogonin 7-O-glucuronide, wogonin, oroxylin A 7-O-glucuronide, and oroxylin A) has been developed. The buffer solution (pH 7.24) composed of 20 mM sodium dodecyl sulfate (SDS), 20 mM sodium dihydrogen phosphate, and 25 mM sodium borate was found to be the most suitable electrolyte for the separation. The contents of the six flavonoids in crude Scutellariae Radix could easily be determined within about 15 min. On-column UV (254 nm) monitoring allowed the quantitative determination of baicalin. The effects of pH, surfactant concentration, and applied voltage on the migration behavior of the solutes were studied.  相似文献

A micellar electrokinetic capillary electrophoresis (MEKC) method was developed for the determination of three flavonoids: diplacone (DI), mimulone (MI) and apigenin (AP) in the flowers of Paulownia tomentosa (Thunb) Steud. A buffer solution (pH 10.00) of 20 mM sodium borate, 10 mM sodium dodecyl sulfate (SDS) and 5% methanol was found to be the most suitable electrolyte for this separation. Regression equations revealed linear relationships (correlation coefficients: 0.9997 and 0.9998) between the peak area of each compound (DI, MI and AP) and its concentration. The relative standard deviations of migration times and peak areas were < 1.93 and 6.84%, respectively. The effects of pH, surfactant (SDS) concentration and organic modifier (methanol) on migration were also studied. The contents of three flavonoids in the extracts from the flowers of P. tomentosa applying different solvents were successfully determined within 12 min.  相似文献

采用紫外吸收检测,毛细管电泳分离,研究了九种苯胺及其衍生物在毛细管区带电泳(CZE)体系和胶束电动毛细管色谱(MECC)体系中的行为特征。讨论了缓冲溶液的浓度与pH、胶束浓度及混合胶束等在不同体系中对分离组分的影响,发现在CZE体系中,控制分离的主要因素是pKb值;在MECC体系中,控制分离的主要因素是溶质分子中碳原子数。建立了一种分离测定九种苯胺及其衍生物的高效毛细管电泳方法。  相似文献

以阴离子表面活性剂十二烷基硫酸钠(SDS)、非离子表面活性剂吐温20( Tween 20)及两者组成的混合胶束体系作为毛细管胶束电动色谱(MECC)的分离介 质,进行4种结构相似的酸性化合物的MECC分离研究,考察了胶束的类型、表面活 性剂的浓度、缓冲溶液的pH值及有机改性剂乙醇对分离的影响。结果表明各因素对 酸性药物的MECC分离有不同的影响规律。SDS胶束体系对溶质的保留值最大, Tween 20体系的保留值最小,二者的分离选择性正好相反,混合胶束体系的分离行 为则介于两者之间;在SDS和Tween 20体系中,表面活性剂浓度增加,溶质的保留 时间均随之递增,混合胶束体系中,总浓度一定,随Tween 20配比的增加,溶质的 保留时间先减少后增加;缓冲溶液的pH值增大,使溶质的分离效果均能变差;有机 改性剂乙醇的加入对容量因子的影响主要与溶质的疏水性有关,并对分离作用机理 进行了探讨。在SDS和Tween 20 MECC体系下,分别进行了实样测定,取得了满意的 结果。  相似文献

 以十二烷基硫酸钠 (SDS)胶束为准固定相 ,考察了咖啡因及其 9种类似物在胶束电动毛细管 (MECC)分离模式下的分离行为。研究了运行缓冲液的 pH值、磷酸盐浓度、SDS浓度、甲醇体积分数、分离电压、分离温度等因素对这 10种化合物的迁移时间和分离效果的影响。结果发现 ,这些因素对上述 10种化合物的分离有显著的影响 ,尤以pH值为最。它不仅影响化合物的迁移时间和分离效率 ,还改变其出峰顺序 ,这与碱性条件下化合物仲胺基上氢的电离有关。优化后的分离条件 :运行缓冲液为 2 0mmol/L磷酸盐 2 0mmol/LSDS(pH 11 0 ) ,分离电压为2 5kV。  相似文献

以十二烷基硫酸钠(SDS)胶束为准固定相,考察了头孢哌酮、头孢哌酮S-异构体、头孢哌酮杂质A及其他未知杂质在胶束电动毛细管色谱(MECC)分离模式下的分离行为。研究了运行缓冲液的pH值、磷酸盐浓度、SDS浓度、甲醇体积分数、分离电压、分离温度等因素对头孢哌酮、S-异构体、头孢哌酮杂质A及其他杂质的迁移时间、分离度以及可分离出的杂质个数的影响。结果发现,这些因素对头孢哌酮与诸杂质间的分离及检测有显著的影响,尤以pH值为最。它不仅影响它们的迁移时间和分离效率,还直接影响头孢哌酮及其杂质峰的检测。优化后的分离条件:运行缓冲液为70 mmol/L磷酸盐-100 mmol/L SDS (pH 6.5),分离电压为15 kV,分离温度为25 ℃。在此条件下,用非涂渍石英毛细管51.0 cm×75 μm(有效长度42.5 cm),压力进样5 kPa×5 s,在254 nm波长下进行检测,可分离出28个杂质,诸杂质彼此间及与头孢哌酮间可得到有效分离。并将该方法成功地用于测定注射用头孢哌酮钠的含量和有关物质,结果令人满意。  相似文献

In this work, a capillary electrophoresis (CE) method for the determination of a group of eleven triazine compounds by micellar electrokinetic capillary chromatography (MEKC) with diode array detection was developed. The eleven herbicides studied were: desethylatrazin-2-hydroxy (DEA), simazine, prometon, atrazine, simetryn, ametryn, propazine, prometryn, trietazine, terbutylazine, and terbutryn The separation of these compounds was optimised as a function of buffer concentration and pH, concentration of sodium dodecyl sulphate (SDS) and voltage applied. To increase the selectivity of the separation and the resolution of the solutes, different organic solvents were tested as buffer additives, obtaining the best results when 1-propanol was used. The optimised buffer (24 mM of sodium borate, 18 mM of disodium hydrogen phosphate, 25 mM of SDS, pH 9.5, and 5% of 1-propanol) provides the best separation in terms of resolution and migration time. This method allowed the determination of these compounds at concentrations of 0.05 μg l−1 in ground water samples pretreated using solid-phase extraction (SPE).  相似文献

A micellar electrokinetic chromatography method is presented for the determination of the partition coefficient for the distribution of nonpolar and moderately polar solutes between the micelle and the aqueous phases in aqueous micellar solutions. In comparison with the literature the method is, theoretically and experimentally, the most straightforward for this application. An equation is derived for the determination of the partition coefficient in a coated fused silica capillary, with neglible electroosmotic flow, from simple measurements of the migration times of solutes and a marker for micelle migration. The advantages and limitations of this method are discussed. The method is tested by using sodium dodecylsulfate (SDS) surfactant and naphthalene, benzene, toluene, and phenol solutes. Micellar electrokinetic chromatography in coated columns isideally suited for the separation of hydrophobic solutes in aqueous samples.  相似文献

The influence of solubilized solutes on the micellization and critical micelle concentration (CMC) of sodium dodecyl sulfate (SDS) were investigated by means of capillary electrophoresis (CE). Three different structural types of test solutes, including chloropyridines. chlorophenols and cephalosporins with different binding strength to SDS micelles, were selected in this study. The variations of the effective electrophoretic mobility of these solutes as a function of SDS concentration in the premicellar and micellar regions were analyzed. Interestingly, the results indicate that, in the presence of these solubilized solutes, the micellization of SDS may occur over a range of SDS concentration, with the aggregate size increasing over this range. Depending on the nature of solubilized solutes and the extent of the interactions between solubilized solutes and SDS micelles, the CMC value of SDS may vary significantly. The incorporation of solubilized solutes into SDS micelles to form mixed micelles is proposed to interpret the migration behavior of solubilized solutes in CE.  相似文献

Summary A group of flavonoids of special interest to wine quality were separated by micellar electrokinetic chromatography (MEKC) with diode array detection. Their separation was optimized as a function of the buffer concentration and pH, the concentration of sodium dodecyl sulfate (SDS) and the applied voltage. Selectivity and resolution of the solutes were increased by adding organic solvents to the separation buffer, the best results being obtained at lower concentrations. An optimized buffer with 5% methanol provided optimum separation with regard to efficiency, resolution and migration time. The optimized method was applied to the determination of these compounds in wine samples.  相似文献

We present a new method for homocysteine quantitation in human plasma based on in-capillary reaction of homocysteine with 2,2′-dipyridyl disulfide. Homocysteine is in this so-called thiol-exchange reaction quantitatively transformed in mixed disulfide concomitantly with formation of an equimolar amount of 2-thiopyridone that is further separated by micellar electrokinetic chromatography and determined specifically at 343 nm. The concentration of homocysteine is thus estimated indirectly from the result of 2-thiopyridone determination. The linear detection range for concentration versus peak area for the assay was from 0.03–3 mM (correlation coefficient 0.994) with a detection limit of 6 μM and a limit of quantitation 20 μM. The inter-day reproducibility of the peak area and the migration time were 1.37% and 0.05%, respectively. The method is simple, relatively rapid and can be easily automated. Moreover the common capillary electrophoresis apparatus with a UV detector can be used to distinguish between normal and pathological hyperhomocysteinemia plasma samples.  相似文献

A group of phenolic compounds including phenolic aldehydes, acids and flavonoids are separated by micellar electrokinetic chromatography (MECC). The influence of buffer (concentration and pH), concentration of sodium dodecylsulphate (SDS) and applied voltage were studied. To increase the selectivity of the separation and the resolution of the solutes organic solvents are added to the separation buffer, the best results were obtained when methanol was used at lower percentages. An optimized buffer (150 mM boric acid (pH 8.5)-50 mM SDS-5% methanol) provides the optimum separation with regard to resolution and migration time. This method was applied to the determination of these compounds in wine samples with good results.  相似文献

A micellar electrokinetic capillary chromatography (MECC) method has been developed for the determination of the four isoflavones, i.e. biochanin A, formononetin, genstein and daidzein in red clover (Trifolium Pratense L.). The effect of running buffer pH and concentration were investigated. An electrolyte composed of 30 mm borate, 20 mm sodium dodecyl sulfate (SDS) and 4 mg/mL HP-beta-CD containing 5% (v/v) ethanol at pH 10.1 provides a satisfactory separation for all the analytes. The applied voltage was 25 kV, and the capillary temperature was kept constant at 25 degrees C with a UV detection at 254 nm. The relative standard deviations (RSD) of the migration time and peak area were less than 1.73 and 3.94% (intra-day), and 2.29 and 4.38% (inter-day), respectively, under the optimized separation conditions. Regression equations revealed a good linear relationship between the peak area of each compound and its concentration. The contents of the four compounds in red clover were successfully determined with satisfactory repeatability and recovery.  相似文献

The migration behavior of cationic solutes and influences of the interactions of cationic solutes with sodium dodecyl sulfate (SDS) on the formation of micelles and its critical micelle concentration (CMC) were investigated by capillary electrophoresis at neutral pH. Catecholamines and structurally related compounds, including epinephrine, norepinephrine, dopamine, norephedrine, and tyramine, which involve different extents of hydrophobic, ionic and hydrogen-bonding interactions with SDS surfactant, are selected as cationic solutes. The dependence of the effective electrophoretic mobility of cationic solutes on the concentration of surfactant monomers in the premicellar region provides direct evidence of the formation of ion-pairs between cationic solutes and anionic dodecyl sulfate monomers. Three different approaches, based on the variations of either the effective electrophoretic mobility or the retention factor as a function of surfactant concentration in the premicellar and micellar regions, and the linear relationship between the retention factor and the product of a distribution coefficient and the phase ratio, were considered to determine the CMC value of SDS micelles. The suitability of the methods used for the determination of the CMC of SDS with these cationic solutes was discussed. Depending on the structures of cationic solutes and electrophoretic conditions, the CMC value of SDS determined varies in a wide concentration range. The results indicate that, in addition to hydrophobic interaction, both ionic and hydrogen-bonding interactions have pronounced effects on the formation of SDS micelles. Ionic interaction between cationic solutes and SDS surfactant stabilizes the SDS micelles, whereas hydrogen-bonding interactions weakens the solubilization of the attractive ionic interaction. The elevation of the CMC of SDS depends heavily on hydrogen-bonding interactions between cationic solutes and SDS surfactant. Thus, the CMC value of SDS is remarkably elevated with catecholamines, such as epinephrine and norepinephrine, as compared with norephedrine. In addition, the effect of methanol content in the sample solution of these cationic solutes on the CMC of SDS was also examined.  相似文献

A simple and rapid method for the simultaneous determination of six ephedrine alkaloids (ephedrine, pseudoephedrine, norephedrine, norpseudoephedrine, methylephedrine and methylpseudoephedrine) in Ephedrae herba by capillary electrophoresis was developed. A buffer solution that contained 0.005 M barium hydroxide and 0.02 M isoleucine and adjusted to pH 10.0 with ammonia solution was found to be the most suitable electrolyte for this separation. The contents of the six alkaloids in the crude drug of Ephedrae herba could be easily determined.  相似文献
