采用交联法制备了羧基二茂铁功能化Fe3O4纳米粒子(FMC-AFNPs)复合材料,并将该复合纳米材料与多壁碳纳米管(MWNTs)、壳聚糖(CS)及葡萄糖氧化酶(GOD)混合修饰于自制的磁性玻碳基底(MGC)表面,制备了GOD/FMC-AFNPs/MWNTs/CS复合膜生物传感器电极. 实验结果表明,FMC-AFNPs复合材料有效地克服了二茂铁在电极表面的泄漏,且FMC-AFNPs/MWNTs/CS复合膜良好的生物兼容性较大地改善了固定化GOD的生物活性. MWNTs具有良好的导电性和大比表面积,在修饰膜内可作为电子传递“导线”,极大地促进电极的电子传递速率,提高电极的电催化活性和灵敏度. 该电极的葡萄糖检测的线性范围为1.0×10-5 ~ 6.0×10-3 molL-1,检测限为3.2×10-6 mmolL-1(S/N=3),表观米氏常数为5.03×10-3 mmolL-1,且有较好的稳定性和重现性.  相似文献

Novel electroanalytical sensing nanobiocomposite materials are reported. These materials are prepared by mixing multiwalled carbon nanotubes (MWNTs), a Nafion cation exchanger, and glucose oxidase (GOD) in appropriate amounts. The MWNTs are cylindrical with a diameter in the range 40-60 nm and with a length of up to several micrometers, and they provide electrical conductivity. Nafion acts as a polymer backbone to give stable and homogeneous cast thin films. Both MWNTs and Nafion provide negative functionalities to bind to positively charged redox enzymes such as glucose oxidase. The resulting biosensing composite material is inexpensive, reliable, and easy to use. The homogeneity of the MWNT-Nafion-GOD nanobiocomposite films was characterized by atomic force microscopy (AFM). Amperometric transducers fabricated with these materials were characterized electrochemically using cyclic voltammetry and amperometry in the presence of hydrogen peroxide and in the presence of glucose. Their linear response to hydrogen peroxide was demonstrated. The glucose biosensor sensitivity was strongly influenced by the glucose oxidase concentration within the nanobiocomposite film. The optimized glucose biosensor (2.5 mg/mL GOD) displayed a sensitivity of 330 nA/mM, a linear range of up to 2 mM, a detection limit of 4 microM, and a response time of <3 s.  相似文献

A novel amperometric glucose biosensor was fabricated by in situ incorporating glucose oxidase (GOD) within the sol‐gel silica film on a Prussian blue (PB) modified electrode. The method is simple and controllable, which combined the merits of in situ immobilizing biomolecules in sol‐gel silica film by electrochemical method and the synergic catalysis effects of PB and GOD molecules. Scanning electron microscopy (SEM) showed that the GOD/sol‐gel silica film was homogeneous with a large number of three‐dimensional nanopores, which not only enhanced mass transport, but also maintained the active configuration of the enzyme molecule and prevented the leakage of enzyme, therefore improved the stability and sensitivity of the biosensor. The fabricated biosensor showed fast response time (10 s), high sensitivity (26.6 mA cm?2 M?1), long‐term stability, good suppression of interference, and linear range of 0.01 mM–5.8 mM with a low detection limit of 0.94 μM for the detection of glucose. In addition, the biosensor was successfully applied to determine glucose in human serum samples.  相似文献

Platinum nanoparticles (Pt NPs) were deposited onto multi-walled carbon nanotubes (MWNTs) through direct chemical reduction without any other stabilizing agents. Transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and cyclic voltammetry were employed to characterize the morphology of the as-prepared nanocomposite (noted as Pt NPs-MWNTs) and further identify the Pt NPs on the surface of MWNTs. The nanocomposite demonstrated the ability to electrocatalyze the oxidation of hydrogen peroxide and substantially raises the response current. A sensitivity of 591.33 μA mM−1 cm−2 was obtained at Pt NPs-MWNTs modified electrode. Thus, we immobilized glucose oxidase (GOD) as a model enzyme on the nanocomposite-based electrode with a thin layer of Nafion to fabricate a glucose biosensor, which showed sensitive and fast response to glucose. The influence of the GOD loading was investigated and the biosensor with an enzyme loading concentration of 10 mg/mL shows optimal performance for glucose detection, that is, a detection limit of 3 μM and a response time of 3 s, respectively.  相似文献

Nanosized Prussian blue (PB) particles were synthesized with a chemical reduction method and then the PB nanoparticles were assembled on the surface of multiwall carbon nanotubes modified glassy carbon electrode (PB/MWNTs/GCE). The results showed that the PB/MWNTs nanocomposite exhibits a remarkably improved catalytic activity towards the reduction of hydrogen peroxide. Glucose oxidase (GOD) was immobilized on the PB/MWNTs platform by an electrochemically polymerized o‐phenylenediamine (OPD) film to construct an amperometric glucose biosensor. The biosensor exhibited a wide linear response up to 8 mM with a low detection limit of 12.7 μM (S/N=3). The Michaelis–Menten constant Km and the maximum current imax of the biosensor were 18.0 mM and 4.68 μA, respectively. The selectivity and stability of the biosensor were also investigated.  相似文献

A novel amperometric biosensor for glucose was developed by entrapping glucose oxidase (GOD) in a chitosan composite doped with ferrocene monocarboxylic acid‐aminated silica nanoparticles conjugate (FMC‐ASNPs) and multiwall carbon nanotubes (MWNTs). The entrapped FMC‐ASNPs conjugate performed excellent redox electrochemistry and the presence of MWNTs improved the conductivity of the composite film. This matrix showed a biocompatible microenvironment for retaining the native activity of the entrapped GOD and was in favor of the accessibility of substrate to the active site of GOD, thus the affinity to substrates is improved greatly. Under optimal conditions this biosensor was able to detect glucose with a detection limit of 10 μM (S/N=3) in the linear range of 0.04 to 6.5 mM. The proximity of these three components FMC‐ASNPs, MWNTs and GOD enhanced the electron transfer between the film and electrode. This composite film can be extended to immobilize other enzymes and biomolecules, which will greatly facilitate the development of biosensors and other bioelectrochemical devices.  相似文献

A new type of amperometric glucose biosensor based on silicon dioxide coated magnetic nanoparticle decorated multiwalled carbon nanotubes (Fe3O4@SiO2/MWNTs) on a glassy carbon electrode (GCE) has been developed. MWNTs have been synthesized by catalytic chemical vapour decomposition (CCVD) of acetylene over rare earth (RE) based AB3 alloy hydride catalyst. The as-grown MWNTs have been purified and further functionlized. Functionalized MWNTs have been decorated with magnetic Fe3O4 nanoparticles which have been uniformly coated with biocompatible SiO2 using a simple chemical reduction method. The characterization of magnetic nanoparticle modified MWNTs have been done by X-ray diffraction (XRD), Fourier transform infra red spectroscopy (FT-IR), scanning electron microscope (SEM), transmission electron microscope (TEM), vibrating sample magnetometer (VSM), energy dispersive X-ray analysis (EDX) and UV-vis spectroscopy. Amperometric biosensor has been fabricated by the deposition of glucose oxidase (GOD) over Nafion-solubilized Fe3O4@SiO2/MWNTs electrode. The resultant bioelectrode retains its biocatalytic activity and offers fast and sensitive glucose quantification. The performance of the biosensor has been studied using cyclic voltammetry and amperometry and the results have been discussed. The fabricated glucose biosensor exhibits a linear response from 1 μM to 30 mM with an excellent detection limit of 800 nM indicating the potential applications in food industries.  相似文献

通过对螺旋型铂铱电极表面进行化学腐蚀和电化学沉积铂纳米粒子实现电极表面的重建和优化,研究了螺旋型铂铱电极在不同腐蚀时间和电沉积时间下的形貌及对过氧化氢(H2O2)的催化活性.对表面重建的工作电极涂覆氧化酶和半透膜,制备出了铂纳米粒子/葡萄糖氧化酶/环氧聚氨酯酶电极,并将其用作葡萄糖传感器的工作电极.传感器计时电流检测结果表明,表面重建后的酶电极传感器对葡萄糖的检测范围扩大为2~45 mmol/L,优于裸铂铱酶电极传感器,电流响应值和灵敏度得到明显提升,同时传感器还具有良好的稳定性和选择性.  相似文献

以中性红为电子媒介体,电聚合于Nafion修饰的玻碳电极表面,以戊二醛作交联剂固定葡萄糖氧化酶,最后覆盖一层Nafion膜防止酶流失,构建一种新型葡萄糖生物传感器.详细探讨了传感器的电化学性能及对葡萄糖的最佳响应条件.结果表明,30℃时,传感器在pH 7.0的PBS中对葡萄糖的线性响应范围为1.0×10-5~5.0×10-3mol.L-1.该传感器制作简单、性能优良,有潜在应用前景.  相似文献

We report on a bienzyme-channeling sensor for sensing glucose without the aid of mediator. It was fabricated by cross-linking horseradish peroxidase (HRP) and glucose oxidase (GOx) on a glassy carbon electrode modified with multiwalled carbon nanotubes (MWNTs). The bienzyme was cross-linked with the MWNTs by glutaraldehyde and bovine serum albumin. The MWNTs were employed to accelerate the electron transfer between immobilized HRP and electrode. Glucose was sensed by amperometric reduction of enzymatically generated H2O2 at an applied voltage of ?50 mV (vs. Ag/AgCl). Factors influencing the preparation and performance of the bienzyme electrode were investigated in detail. The biosensor exhibited a fast and linear response to glucose in the concentration range from 0.4 to 15 mM, with a detection limit of 0.4 mM. The sensor exhibited good selectivity and durability, with a long-term relative standard deviation of <5 %. Analysis of glucose-spiked human serum samples yielded recoveries between 96 and 101 %.

A novel composite was fabricated through dispersing multiwalled carbon nanotubes (MWNTs) in gold nanoparticle (GPs) colloid stabilized by chitosan and ionic liquid (i.e., 1‐butyl‐3‐methylimidazolium tetrafluoroborate, BMIMBF4). Transmission electron microscopy (TEM) experiment showed that the GPs highly dispersed on the MWNTs probably due to the electrostatic interaction among GPs, MWNTs and the imidazolium cation of BMIMBF4. X‐ray photoelectron spectroscopy (XPS) indicated that thus‐formed gold nanostructure was mediated by BMIMBF4. When glucose oxidase (GOD) was immobilized on the composite (MWNTs‐GPs) its ultraviolet‐visible absorption spectrum kept almost unchanged. The immobilized GOD coated glassy carbon electrode (GOD/MWNTs‐GPs/GC) exhibited a pair of well‐defined peaks in 0.10 M pH 7.0 phosphate buffer solution (PBS), with a formal potential of ?0.463 V (vs. SCE). The electrochemical process involved two‐electron transfer. The electron transfer coefficient was ca.0.56 and the electron transfer rate constant was 9.36 s?1. Furthermore, the immobilized GOD presented good catalytic activity to the oxidation of glucose in air‐saturated PBS. The Km and Im values were estimated to be 13.7 μM and 0.619 μA. The GOD/MWNTs‐GPs/GC electrode displayed good stability and reproducibility.  相似文献

采用层层自组装的方法和异种电荷互相吸引的原理,将Nafion修饰在金电极上固载带正电荷的天青Ⅰ,并利用天青Ⅰ中的氨基固载纳米金,再通过纳米金将酶固定在金电极表面,制成了葡萄糖传感器.采用循环伏安法和交流阻抗法,研究了金电极表面组装各层之后的电化学特征,以及电极对葡萄糖的电化学催化作用. 结果表明,天青Ⅰ不仅可以固定酶和纳米金,而且还可以在酶和电极之间有效地传递电子.在优化的实验条件下,该传感器对葡萄糖响应的线性范围为5.1×10-6 ~4.0×10-3 mol/L,检出限(S/N=3)为1.0 μmol/L.该生物传感器显示出较好的稳定性和抗干扰能力,将其用于人体血清中葡萄糖的测定,结果令人满意.  相似文献

A mediator-free glucose biosensor, termed a “third-generation biosensor,” was fabricated by immobilizing glucose oxidase (GOD) directly onto an oxidized boron-doped diamond (BDD) electrode. The surface of the oxidized BDD electrode possesses carboxyl groups (as shown by Raman spectra) which covalently cross-link with GOD through glutaraldehyde. Glucose was determined in the absence of a mediator used to transfer electrons between the electrode and enzyme. O2 has no effect on the electron transfer. The effects of experimental variables (applied potential, pH and cross-link time) were investigated in order to optimize the analytical performance of the amperometric detection method. The resulting biosensor exhibited fast amperometric response (less than 5 s) to glucose. The biosensor provided a linear response to glucose over the range 6.67×10−5 to 2×10−3 mol/L, with a detection limit of 2.31×10−5 mol/L. The lifetime, reproducibility and measurement repeatability were evaluated and satisfactory results were obtained.  相似文献

Abstract

A new nanocomposite was developed by combination of prussian blue (PB) nanoparticles and multiwalled carbon nanotubes (MWNTs) in the matrix of biopolymer chitosan (CHIT). The PB and MWNTs had a synergistic electrocatalytic effect toward the reduction of hydrogen peroxide. The CHIT/MWNTs/PB nanocomposite‐modified glassy carbon (GC) electrode could amplify the reduction current of hydrogen peroxide by ~35 times compared with that of CHIT/MWNTs/GC electrode and reduce the response time from ~60 s for CHIT/PB/GC to 3 s. Besides, the CHIT/MWNTs/PB nanocomposite‐modified GC electrode could reduce hydrogen peroxide at a much lower applied potential and inhibit the responses of interferents such as ascorbic acid (AA) uric acid (UA) and acetaminophen (AC). With glucose oxidase (GOx) as an enzyme model, a new glucose biosensor was fabricated. The biosensor exhibited excellent sensitivity (the detection limit is down to 2.5 µM), fast response time (less than 5 s), wide linear range (from 4 µM to 2 mM), and good selection.  相似文献

Iron‐phthalocyanines (FePc) are functionalized at multi‐walled carbon nanotubes (MWNTs) to remarkably improve the sensitivity toward hydrogen peroxide. We constructed a highly sensitive and selective glucose sensor on FePc‐MWNTs electrode based on the immobilization of glucose oxidase (GOD) on poly‐o‐aminophenol (POAP)‐electropolymerized electrode surface. SEM images indicate that GOD enzymes trapped in POAP film tend to deposit primarily on the curved tips and evenly disperse along the sidewalls. The resulting GOD@POAP/FePc‐MWNTs biosensor exhibits excellent performance for glucose with a rapid response (less than 8 s), a wide linear range (up to 4.0×10?3 M), low detection limits (2.0×10?7 M with a signal‐to‐noise of 3), a highly reproducible response (RSD of 2.6%), and long‐term stability (120 days). Such characteristics may be attributed to the catalytic activity of FePc and carbon nanotube, permselectivity of POAP film, as well as the large surface area of carbon nanotube materials.  相似文献

An enzymatic electrochemical biosensor was prepared based on glucose oxidase (GOD) immobilized on single-walled carbon nanotubes/Nile blue (Nb–SWCNTs) nanocomposite cross-linked with glutaraldehyde on graphite electrode. The function of glucose biosensor is based on the electrocatalytic reduction of H2O2 generated by the reaction of the enzyme with glucose molecules. The biosensor was characterized by Fourier transform infrared (FTIR) spectroscopy, emission scanning (FESEM), UV–Vis spectrometry, and voltammetric and amperometric methods. Parameters influencing the performance of the biosensor, namely pH and applied potential, were optimized. Inhibition was carried out for the detection of Hg2+ and Pb2+ species under optimized condition. Inhibition investigations showed that the type of inhibition for both the abov- mentioned heavy metal ions were reversible and competitive.  相似文献

We report herein the development of a novel glucose chemiluminescence (CL) biosensor based on covalent immobilization of glucose oxidase (GOD) in glutaraldehyde-functionalized glass cell and direct coupling of chitosan-induced Au/Ag alloy nanoparticles on it, and how it may be useful for determination of glucose due to CL detection of enzymatically generated H2O2. In addition, the nanoalloy offers excellent catalytic activity toward hydrogen peroxide generation in enzymatic reaction between GOD and glucose and increases stability of covalent-linked enzyme. Chitosan molecules act as both the reducing and stabilizing agents for the preparation of nanoparticles (NPs) and also as a coupling agent between GOD and Au/Ag alloy NPs, which made possible the fabrication of a sensitive, accurate and stable biosensor for glucose. Under the optimum conditions, the biosensor can be used for the determination of glucose in the range of 1.2 × 10–6 to 6.25 × 10–3 M with a detection limit of 5.0 × 10–7 M. The CL biosensor exhibited good storage stability, i.e., 90% of its initial response was retained after 2 months storage at pH 7.0. The present CL biosensor has been used to determine glucose in real serum and urine samples and validated against colorimetric spectrophotometry method.  相似文献

A novel electrochemical platform based on nickel oxide (NiO) nanoparticles and TiO2–graphene (TiO2–Gr) was developed for the direct electrochemistry of glucose oxidase (GOD). The electrochemical behavior of the sensor was studied using cyclic voltammetry and chronoamperometry. The experimental results demonstrated that the nanocomposite well retained the activity of GOD and the modified electrode GOD/NiO/TiO2–Gr/GCE exhibited excellent electrocatalytic activity toward the redox of GOD as evidenced by the significant enhancement of redox peak currents in comparison with bare GCE. The biosensor responded linearly to glucose in the range of 1.0–12.0?mM, with a sensitivity of 4.129?μA?mM?1 and a detection limit of 1.2?×?10?6?M under optimized conditions. The response time of the biosensor was 3?s. In addition, the developed biosensor possessed good reproducibility and stability, and there was negligible interference from other electroactive components.  相似文献

This article describes the investigation of direct electron transfer (DET) between glucose oxidase (GOD) and the electrode materials in an enzyme-catalyzed reaction for the development of improved bioelectrocatalytic system. The GOD pedestal electrochemical reaction takes place by means of DET in a tailored Vulcan carbon paste electrode surfaces with GOD and chitosan (CS), allowing efficient electron transfer between the electrode and enzyme. The key understanding of the stability, biocatalytic activity, selectivity, and redox properties of these enzyme-based glucose biosensors is studied without using any reagents, and the properties are characterized using electrochemical techniques like cyclic voltammogram, amperometry, and electrochemical impedance spectroscopy. Furthermore, the interaction between the enzyme and the electrode surface is studied using ultraviolet–visible (UV–Vis) and Fourier transform infrared (FTIR) spectroscopy. The present glucose biosensor exhibited better linearity, limit of detection (LOD?=?0.37?±?0.02 mol/L) and a Michaelis–Menten constant of 0.40?±?0.01 mol/L. The proposed enzyme electrode exhibited excellent sensitivity, selectivity, reproducibility, and stability. This provides a simple “reagent-less” approach and efficient platform for the direct electrochemistry of GOD and developing novel bioelectrocatalytic systems.  相似文献
