Mitochondria are essential intracellular organelles involved in many cellular processes, especially adenosine triphosphate (ATP) production. Since cancer cells require high ATP levels for proliferation, ATP elimination can be a unique target for cancer growth inhibition. We describe a newly developed mitochondria-targeting nucleopeptide (MNP) that sequesters ATP by self-assembling with ATP inside mitochondria. MNP interacts strongly with ATP through electrostatic and hydrogen bonding interactions. MNP exhibits higher binding affinity for ATP (−637.5 kJ mol−1) than for adenosine diphosphate (ADP) (−578.2 kJ mol−1). To improve anticancer efficacy, the small-sized MNP/ADP complex formed large assemblies with ATP inside cancer cell mitochondria. ATP sequestration and formation of large assemblies of the MNP/ADP–ATP complex inside mitochondria caused physical stress by large structures and metabolic disorders in cancer cells, leading to apoptosis. This work illustrates a facile approach to developing cancer therapeutics that relies on molecular assemblies.

Background

Gamma glutamylcyclotransferase (GGCT) has been proved to be involved in various cancers, but the biological function of GGCT in gastric cancer is still largely unknown.

Methods

The expression level of GGCT was evaluated by informatics analyses based on the Oncomine database. GGCT gene was then effectively knocked down via lentivirus mediated short hairpin RNA (shRNA) system. Then a series of functional assays, including MTT, colony formation and flow cytometry analysis were conducted on gastric cancer cells following GGCT knockdown.

Results

We found GGCT is commonly up-regulated in gastric cancer tissues. Furthermore, MTT analysis showed that GGCT depletion significantly inhibited cell proliferation in MGC80-3 and AGS cells. Colony formation assay revealed that depletion of GGCT reduced the colony formation ability in gastric cancer cells. What’s more, cell cycle analysis showed that depletion of GGCT induced gastric cancer cell cycle arrested G2/M phase. More importantly, cell apoptosis analysis further revealed that GGCT inhibition induced early and late cell apoptosis in gastric cancer.

Conclusion

This study suggests GGCT is essential for gastric cancer proliferation and its downregulation may provide a potential anticancer therapy for gastric cancer.

BackgroundNasopharyngeal cancer is a tumor that occurs in the mucous epithelium of the nasopharynx. Due to its rapid growth and early metastatic nature, the successful treatment of nasopharyngeal cancer is highly challenging.ObjectiveHere, we intended to assess the in vitro anticancer property of brassinin against the nasopharyngeal cancer C666-1 cells.MethodologyThe in vitro free radical scavenging property of the brassinin was assessed by various free radical scavenging activities such as FRAP, DPPH, chemiluminescence (CL), and ORAC assays. The cytotoxic level of the brassinin (1–50 µM) against the nasopharyngeal cancer C666-1 cells and normal Vero cells were assessed by the MTT cytotoxicity assay. The levels of TBARS, GSH, and the SOD activity was assessed using kits. The level of ROS generation, MMP, and apoptosis were investigated by the respective fluorescent staining techniques. The flow cytometry analysis was done to scrutinize the cell cycle arrest. The Bax/Bcl-2 level and caspase activities were examined using respective kits.ResultsThe brassinin treatment effectively scavenged the free radicals, which are assessed by the FRAP, DPPH, chemiluminescence (CL), and ORAC assays. The proliferation of brassinin treated C666-1 cells were decreased remarkably, while the same concentration of brassinin did not disturbed the Vero cell viability. The 30 µM of brassinin effectively increased the ROS production, depleted the MMP, and stimulated the apoptosis in the C666-1 cells. The brassinin increased the TBARS and depleted the GSH and SOD in the C666-1 cells. The flow cytometry analysis revealed that the brassinin administration improved the G0/G1 ratio and decreased the proportion of cells with ‘S’ and ‘G2/M’ phase. The Bax, caspase-3 and ?9 were elevated and Bcl-2 level was decreased in the brassinin administered C666-1 cells.ConclusionOur findings discovered that the brassinin has the capacity to prevent the proliferation and stimulate the apoptotic cell death C666‐1 cells via blocking cell cycle and increasing oxidative stress and apoptotic markers. Hence, it can be a talented therapeutic agent to treat the nasopharyngeal cancer in the future.  相似文献

A new mixed‐ligand copper(II) complex, [Cu(L)(phen)]⋅MeOH (L = 4‐chloro‐2‐[(2‐hydroxyphenyl)iminomethyl]phenol), was synthesized. It belongs to the orthorhombic crystal system and Cu(II) is five‐coordinated in a seriously distorted square pyramidal geometry. DNA binding experiments confirmed an intercalative mode of interaction of the complex with calf thymus DNA. In a DNA cleavage experiment with the complex, as hydrogen peroxide was involved, oxidative DNA cleavage occurred and double‐stranded breaks even appeared at certain concentration. The strong interaction with bovine serum albumin suggested that the complex might be transported by protein. The complex exhibited more significant cytotoxicity in HeLa cells (IC50 = 0.46 ± 0.01 μM) for 48 h, compared with cisplatin (10.61 ± 0.86 μM). This work indicated that the complex could induce apoptosis in a dose‐dependent manner and was associated with cell cycle arrest to some extent. Being consistent with the results of DNA cleavage experiment, comet assay indicated that the complex induced severe DNA fragmentation. The results showed the production of reactive oxygen species increased with increasing concentration of the complex. The complex was suggested to be capable of promoting HeLa cell apoptosis through an oxidative DNA damage pathway.  相似文献

BackgroundEsophageal cancer is a sixth most cause of cancer-associated mortalities worldwide with increased global prevalence in each year. It has a poor prognosis with 5-year survival rate are less than 10%.ObjectiveThe present study was focused to fabricate the chitosan-sodium alginate-polyethylene glycol-crocin nanocomposites (CSP-Cr-NCs) and evaluate its in vitro anticancer potential against the esophageal cancer KYSE-150 cells.MethodologyThe fabricated CSP-Cr-NCs were characterized using different techniques such as UV–visible spectroscopy, photoluminescence assay, DLS analysis, XRD, SEM and EDX analyses. The antimicrobial study was conducted by well diffusion technique against the S. pneumoniae, K. pneumoniae, E. coli, S. aureus, and C. albicans. The viability of CSP-Cr-NCs treated KYSE-150 and Het-1A cells were assessed by the MTT assay. The ROS production, MMP level, and apoptotic cell death in the CSP-Cr-NCs administered cells were assessed by using different fluorescent staining techniques. The cell migration of CSP-Cr-NCs treated KYSE-150 cells were assessed by wound scratch assay. The levels of TBARS, GSH, and SOD activity in the CSP-Cr-NCs treated KYSE-150 cells were assessed by kits.ResultsThe outcomes from the different characterization analyses witnessed the formation of CSP-Cr-NCs with the 100–210 nm size, tetragonal and agglomerated morphological appearances. The CSP-Cr-NCs effectively repressed the microbial growth. The CSP-Cr-NCs treated KYSE-150 cells were demonstrated the decreased cell viability and IC50 was found at 2.5 µg, furthermore it did not affected the normal Het-1A cells. The formulated CSP-Cr-NCs treatment at 2.5 and 5 µg improved the ROS production, and decreased the MMP status in the KYSE-150 cells. The elevated incidences of apoptotic cells death was found in the CSP-Cr-NCs treated KYSE-150 cells. The CSP-Cr-NCs also inhibited the migration of KYSE-150 cells. The increased TBARS content and decreased GSH and SOD activity was also found in the CSP-Cr-NCs treated KYSE-150 cells.ConclusionOur findings proved that the formulated CSP-Cr-NCs treatment effectively inhibited the esophageal cancer KYSE-150 cell growth via increasing ROS production and apoptotic cell death. Therefore, it could be a promising anticancer candidate in the future for the esophageal cancer treatment.  相似文献

Semiconductor quantum dots (Qdots) have recently been shown to offer significant advantages over conventional fluorescent probes to image and study biological processes. The stability and low toxicity of QDs are well suited for biological applications. Despite this, the potential of Qdots remains limited owing to the inefficiency of existing delivery methods. By conjugating Qdots with small antibody fragments targeting membrane-bound proteins, such as GRP78, we demonstrate here that the Quantum dot- Anti-GRP78 scFv (Qdot-GRP78) retains its immunospecificity and its distribution can be monitored by visualization of multi-color fluorescence imaging both in vitro and in vivo. Moreover we demonstrate here for the first time that Qdot-GRP78 scFv bioconjugates can be efficiently internalized by cancer cells, thus upregulate phophosphate-AKT-ser473 and possess biological anti-tumour activity as shown by inhibition of breast cancer growth in a xenograft model. This suggests that nanocarrier-conjugated scFvs can be used as a therapeutic antibody for cancer treatment.  相似文献

Lung cancer is the widespread carcinogenesis in men and the third most familiar cancer in women. It is one of the mostly aggressive human cancers, which is responsible for around 1.4 million deaths per annum and has utmost mortality and incidence with 1.8 million new incidences and 1.6 million new deaths yearly. In this present study, we have evaluated the anticancer potential of zinc oxide nanopartices (ZnONPs) synthesized from a root extract of Euphorbia fischeriana (EF), through the apoptosis signaling markers in A549 lung cancer cells. The synthesized EF-ZnONPs were evaluated through the transmission electron microscope (TEM), Fourier transform infra red (FTIR), UV–visible spectroscopy and dynamic light scattering (DLS) techniques. The EF-ZnONPs were assessed for their cytotoxicity activity towards A549 cells by MTT test. The induction of apoptosis was analysed by the mitochondria membrane potential (MMP), reactive oxygen species (ROS), cell migration and dual staining. Furthermore, pro and anti-apoptotic signaling protein expression was evaluated by western blotting method. We found the bioformulated EF-ZnONPs has a spherical morphology and revealed the existence of diverse bioactive compounds. Also we found the cytotoxic effect of EF-ZnONPs. Apoptosis was activated by the EF-ZnONPs with improved ROS, decreased MMP, inhibited cell migration and altered dual staining was observed. Furthermore, the diminished expression of anti-apoptotic protein Bcl-2 was noted. In this study, we observed the formulation, characterization and anticancer potency of ZnONPs of EF plant extract (EF-ZnONPs) was useful for treatments of lung cancer.  相似文献

Glucosamine, a naturally occurring amino monosaccharide, has been reported to play a role in the regulation of apoptosis more than half century. However the effect of glucosamine on tumor cells and the involved molecular mechanisms have not been thoroughly investigated. Glucosamine enters the hexosamine biosynthetic pathway (HBP) downstream of the rate-limiting step catalyzed by the GFAT (glutamine:fluctose- 6-phosphate amidotransferase), providing UDPGlcNAc substrates for O-linked β-N-acetylglucosamine (O-GlcNAc) protein modification. Considering that O-GlcNAc modification of proteasome subunits inhibits its activity, we examined whether glucosamine induces growth inhibition via affecting proteasomal activity. In the present study, we found glucosamine inhibited proteasomal activity and the proliferation of ALVA41 prostate cancer cells. The inhibition of proteasomal activity results in the accumulation of ubiquitinated proteins, followed by induction of apoptosis. In addition, we demonstrated that glucosamine downregulated proteasome activator PA28γ and overexpression of PA28γ rescued the proteasomal activity and growth inhibition mediated by glucosamine. We further demonstrated that inhibition of O-GlcNAc abrogated PA28γ suppression induced by glucosamine. These findings suggest that glucosamine may inhibit growth of ALVA41 cancer cells through downregulation of PA28γ and inhibition of proteasomal activity via O-GlcNAc modification.  相似文献

Abstract

Valtrate is a principle compound isolated from Valeriana jatamansi Jones, a traditional Chinese folk medicine originally used to treat various nervous disorders. Here, we found that valtrate exhibited significant anti-cancer activity in vitro, especially in human breast cancer cells, while displayed relatively low cytotoxicity to normal human breast epithelial cells (MCF 10A). Valtrate induced cell cycle arrest at G2/M stage and apoptosis in MDA-MB-231 and MCF-7 cells, with reduced expression of p-Akt (Ser 473), cyclin B1 and caspase 8, and increased expression of p21, p-cdc2, cleaved-caspase 3, cleaved-caspase 7 and poly (ADP-ribose) polymerase (PARP). In addition, valtrate inhibited cell migration through down-regulation of MMP-9 and MMP-2 expression. These results demonstrate that valtrate possesses anti-breast cancer activities via cell cycle arrest, apoptosis, and inhibition of cell migration, thus supporting valtrate as a potential antitumor agent.  相似文献

Phospholipase D (PLD) is a potential therapeutic target against cancer. However, the contribution of PLD inhibition to the antitumor response remains unknown. We developed a potent and selective PLD1 inhibitor based on computer-aided drug design. The inhibitor enhanced apoptosis in colorectal cancer (CRC) cells but not in normal colonic cells, and in vitro cardiotoxicity was not observed. The inhibitor downregulated the Wnt/β-catenin signaling pathway and reduced the migration, invasion, and self-renewal capacity of CRC cells. In cancer, therapeutic engagement of immunogenic cell death (ICD) leads to more effective responses by eliciting the antitumor immunity of T cells. The CRC cells treated with the inhibitor showed hallmarks of ICD, including downregulation of “do not eat-me” signals (CD24, CD47, programmed cell death ligand 1 [PD-L1]), upregulation of “eat-me” signal (calreticulin), release of high-mobility group Box 1, and ATP. PLD1 inhibition subsequently enhanced the phagocytosis of cancer cells by macrophages through the surface expression of costimulatory molecules; as a result, the cancer cells were more susceptible to cytotoxic T-cell-mediated killing. Moreover, PLD1 inhibition attenuated colitis-associated CRC and orthotopically injected tumors, probably by controlling multiple pathways, including Wnt signaling, phagocytosis checkpoints, and immune signaling. Furthermore, combination therapy with a PLD1 inhibitor and an anti-PD-L1 antibody further enhanced tumor regression via immune activation in the tumor environment. Collectively, in this study, PLD1 was identified as a critical regulator of the tumor microenvironment in colorectal cancer, suggesting the potential of PLD1 inhibitors for cancer immunotherapy based on ICD and immune activation. Fluorescent aptamer sensors have shown enormous potential for intracellular imaging of small molecule metabolites. Since metabolites distribute differently at different subcellular locations and their concentrations and locations fluctuate with time, methods are needed for spatiotemporally controlled monitoring of these metabolites. Built upon previous success in temporal control of aptamer-based sensors, we herein report an aptamer sensor containing a photocleavable linker and using DQAsomes to target mitochondria for spatiotemporally controlled monitoring of ATP in the mitochondria of living cells. The photocleavable modification on the DNA ATP aptamer sensor can prevent sensor activation before reaching mitochondria and the sensor can then be activated upon light irradiation. The sensor has a detection limit of 3.7 μM and high selectivity against other nucleotides, allowing detection of ATP concentration fluctuations in mitochondria induced by Ca2+ or oligomycin. This work represents the first successful delivery of a DNA aptamer sensor to mitochondria, providing a new platform for targeted delivery to subcellular organelles for monitoring energy producing processes, as well as mitochondrial dysfunction-related diseases in different cells.

Leaf expansion, comprising cell division and cell enlargement, is controlled by light quality and quantity. The role of UV-B irradiance on leaf cell enlargement has not been determined. We studied the effect of a wide range of UV-B irradiances on the cell-enlargement-driven expansion of Phaseolus vulgaris L., cv. Contender (bush bean) leaf discs. Our growth method allowed separation of the cell enlargement phase of leaf expansion from the cell division phase. In two series of experiments with different types of UV-B screening filters, the effect of increasing levels of UV-B on the area of excised P. vulgaris leaf discs was investigated. One set of experiments utilized polyester (UV-B-absorbing) and cellulose acetate (UV-B-transmitting) filters. The other set utilized UV-B-absorbing and UV-B-transmitting acrylic filters. Regardless of which type of filter was used for screening, high (above summer solstice) levels of supplemental UV-B inhibited cell enlargement in a linear, dose-dependent manner, resulting in smaller leaf discs than treatment with UV-B-absorbing filters. Conversely, low levels of supplemental UV-B enhanced cell enlargement in a linear, dose-dependent manner, resulting in larger leaf discs than did treatment with UV-B-absorbing filters. The results suggest a biphasic response to UV-B, and that there is an optimum UV-B level that results in maximum leaf expansion by cell enlargement.  相似文献

Methyl-beta-cyclodextrin, a cyclic oligosaccharide known for its interaction with the plasma membrane induces several events in cells including cell growth and anti-tumor activity. In this study, we have investigated the possible role of cyclooxygenase 2 (COX-2) in cell growth arrest induced by methyl-beta-cyclodextrin in Raw264.7 macrophage cells. Methyl-beta-cyclodextrin inhibited cell growth and arrested the cell cycle, and this cell cycle arrest reduced the population of cells in the S phase, and concomitantly reduced cyclin A and D expressions. Methyl-beta-cyclodextrin in a dose- and time-dependent manner, also induced COX-2 expression, prostaglandin E(2) (PGE(2)) synthesis, and COX-2 promoter activity. Pretreatment of cells with NS398, a COX-2 specific inhibitor completely blocked PGE(2) synthesis induced by methyl-beta-cyclodextrin, however inhibition on cell proliferation and cell cycle arrest was not effected, suggesting non-association of COX-2 in the cell cycle arrest. These results suggest that methyl-beta-cyclodextrin induced cell growth inhibition and cell cycle arrest in Raw264.7 cells may be mediated by cyclin A and D1 expression.  相似文献

Transarterial chemoembolization (TACE) is the first-line treatment for unresectable intermediate-stage hepatocellular carcinoma (HCC). It is of high clinical significance to explore the synergistic effect of TACE with antiangiogenic inhibitors and the molecular mechanisms involved. This study determined that glucose, but not other analyzed nutrients, offered significant protection against cell death induced by sorafenib, as indicated by glucose deprivation sensitizing cells to sorafenib-induced cell death. Next, this synergistic effect was found to be specific to sorafenib, not to lenvatinib or the chemotherapeutic drugs cisplatin and doxorubicin. Mechanistically, sorafenib-induced mitophagy, as indicated by PINK1 accumulation, increased the phospho-poly-ubiquitination modification, accelerated mitochondrial membrane protein and mitochondrial DNA degradation, and increased the amount of mitochondrion-localized mKeima-Red engulfed by lysosomes. Among several E3 ubiquitin ligases tested, SIAH1 was found to be essential for inducing mitophagy; that is, SIAH1 silencing markedly repressed mitophagy and sensitized cells to sorafenib-induced death. Notably, the combined treatment of glucose restriction and sorafenib abolished ATP generation and mitophagy, which led to a high cell death rate. Oligomycin and antimycin, inhibitors of electron transport chain complexes, mimicked the synergistic effect of sorafenib with glucose restriction to promote cell death mediated via mitophagy inhibition. Finally, inhibition of the glucose transporter by canagliflozin (a clinically available drug used for type-II diabetes) effectively synergized with sorafenib to induce HCC cell death in vitro and to inhibit xenograft tumor growth in vivo. This study demonstrates that simultaneous treatment with sorafenib and glucose restriction is an effective approach to treat HCC, suggesting a promising combination strategy such as transarterial sorafenib-embolization (TASE) for the treatment of unresectable HCC.Subject terms: Liver cancer, Mitophagy, Apoptosis  相似文献

Parthenolide, the principal component of sesquiterpene lactones present in medical plants such as feverfew (Tanacetum parthenium), has been reported to have anti-tumor activity. In this study, we evaluated the therapeutic potential of parthenolide against bladder cancer and its mechanism of action. Treatment of bladder cancer cells with parthenolide resulted in a significant decrease in cell viability. Parthenolide induced apoptosis through the modulation of Bcl-2 family proteins and poly (ADP-ribose) polymerase degradation. Treatment with parthenolide led to G1 phase cell cycle arrest in 5637 cells by modulation of cyclin D1 and phosphorylated cyclin-dependent kinase 2. Parthenolide also inhibited the invasive ability of bladder cancer cells. These findings suggest that parthenolide could be a novel therapeutic agent for treatment of bladder cancer.  相似文献

Trichosanthes dioica seed extract was loaded on a QA-cellulose column and the unbound fraction with the chitinase activity was run on SDS-PAGE. Multiple bands were observed and were separated by a Sephadex G-50 column. The combination of the 6 and 33 kDa masses supported the degradation of chitinase as purified earlier. Only the 33 kDa fraction contained sugar and showed chitinase activity. The chitinase was also isolated by using a chitin column. At 200 µg/ml protein concentration, the chitinase inhibited 49.1 %, 48.8 % and 38.12 % of Ehrlich ascites carcinoma, HCT-116 and MCF-7 cells growth, respectively, in a dose-dependent manner. Exactly, 46 % and 82 % EAC cell growth inhibition were observed after treating the EAC cells bearing Swiss albino mice with the chitinase at the doses of 1.0 and 2.0 mg/Kg/day respectively. EAC, HCT-116 and MCF-7 cells growth inhibitions were due to the induction of apoptosis. ROS was accumulated in HCT-116 and MCF-7 cells. After treatment of HCT-116 cells, the expression level of p53 and TNFα genes increased and PARP gene decreased. On the other hand, elevated expression was observed for PARP, MAPK, NFκB, FAS, FADD, and Caspase-8 genes in MCF-7 cells. The induction of apoptosis in HCT-116 was further confirmed by caspase protein expression. The chitinase causes ‘S’ cell cycle arrest in MCF-7 and HCT-116 cells. T. dioica seed chitinase inhibited EAC, HCT-116 and MCF-7 cells by inducing apoptosis in vitro and EAC in vivo in mice. These promising results indicated that T. dioica seed chitinase can be an anticancer agent.  相似文献

A series of benzofuran-isatin conjugates 6a-l and 7a,b tethered by various alkyl linkers were synthesized and evaluated for their VEGFR-2 inhibitory activity and in vitro activity against a panel of cancer cell lines. Seven of them were comparable with or better than Sunitinib against all tested cancer cells, demonstrating benzofuran-isatin conjugates were potential anticancer candidates. The mechanism study revealed that VEGFR-2 was at least one of the targets for this kind of conjugates. The structure-activity relationship demonstrated that the carbon spacer between benzofuran and isatin moieties, substituents on the C-2 position of benzofuran moiety, and substituents on C-3 as well as C-5 position of isatin motif influenced the anticancer activity significantly, and the enriched structure-activity relationship may provide an insight for rational design of more effective conjugates.  相似文献
