Surfactant‐assisted electromembrane extraction coupled with cyclodextrin‐modified capillary electrophoresis was developed for the separation and determination of Tranylcypromine enantiomers in biological samples. This combination would provide a new strategy for selective and sensitive determination of target analytes. The addition of surfactant in the donor solution improved the analyte transport into the lumen of hollow fiber that resulted in an enhancement in the analytes migration into acceptor solution. Optimization of the variables, affecting proposed method, was carried out and best results were achieved with a 175 V potential as driving force of the electromembrane extraction, 2‐nitrophenyloctylether as the supported liquid membrane, donor solution containing 0.2 mM Triton X‐100 with pH 3 and 0.1 M HCl for acceptor solution. Then, the extract was analyzed using cyclodextrin‐modified capillary electrophoresis method for separation of Tranylcypromine enantiomers. The best results were obtained with a phosphate running buffer (100 mM, pH 2.0) containing 7% w/v hydroxypropyl‐α‐cyclodextrin. Under the optimum conditions, a low limit of detection (3.03 ng/mL), good linearity (R2 > 0.9953), and relative standard deviations below 4.0% (n = 5) were obtained. Finally, this procedure was applied to determine the concentration of Tranylcypromine enantiomers in urine samples with satisfactory results.  相似文献

We establish a triple-stacking capillary electrophoresis (CE) separation method to monitor methotrexate (MTX) and its eight metabolites in cerebrospinal fluid (CSF). Three stacking methods with different mechanisms were combined and incorporated into CE separation. Complete stacking and sharp peaks were achieved. Firstly, the optimized buffer (60 mM phosphate containing 15% THF and 100 mM SDS) was filled into the capillary, which was followed by the higher conductivity buffer (100 mM phosphate, 2 psi for 45 s). The analytes extracted from CSF were injected at 2 psi for 99.9 s, which provided long sample zones and pH junction for focusing. Finally, the stacking step was performed by sweeping, and separation was achieved by micellar electrokinetic chromatography. The results of the linear regression equations indicated high linearity (r ≥ 0.9981) over the range of 0.5–7 μM. In intra- and inter-batch results, all data of RSD and RE were below 11%, indicating good precision and accuracy of this method. The LODs (S/N = 3) were 0.1 μM for MTX, 7-hydroxymethotrexate (7-OHMTX) and MTX-polyglutamates (MTX-(Glu) n, n = 2–5), 0.2 μM for MTX-(Glu)6, and 0.3 μM for 2,4-diamino-N 10-methylpteroic acid (DAMPA) and MTX-(Glu)7. Our method was implemented for analysis of MTX and its metabolites in the CSF, and could be used for evaluation of its curative effects of acute lymphoblastic leukemia patients. The data were also confirmed by matrix-assisted laser desorption ionization time-of-flight mass spectrometry. The results showed good coincidence.  相似文献

A simple, rapid and sensitive CE method coupled with electrochemiluminescence (ECL) detection for direct analysis of ibandronate (IBAN) has been developed. Using a buffer solution of 20 mM sodium phosphate (pH 9.0) and a voltage of 13.5 kV, separation of IBAN in a 30‐cm length capillary was achieved in 3 min. ECL detection was performed with an indium tin oxide working electrode bias at 1.6 V (versus a Pt wire reference) in a 200‐mM sodium phosphate buffer (pH 8.0) containing 3.5 mM Ru(bpy)32+ (where bpy=2,2′‐bipyridyl). Derivatization of IBAN prior to CE‐ECL analysis was not needed. Linear correlation (r=0.9992, n=7) between ECL intensity and analyte concentration was obtained in the range of 0.25–50 μM IBAN. The LOD of IBAN in water was 0.08 μM. The developed method was applied to the analysis of IBAN in a drug formulation and human urine sample. SPE using magnetic Fe3O4@Al2O3 nanoparticles as the extraction phase was employed to pretreat the urine sample before CE‐ECL analysis. The linear range was 0.2–12.0 μM IBAN in human urine (r=0.9974, n=6). The LOD of IBAN in urine was 0.06 μM. Total analysis time including sample preparation was <1 h.  相似文献

The fabrication and evaluation of pyrolyzed photoresist films (PPF) for microchip capillary electrophoresis (CE) with dual‐electrode electrochemical (EC) detection is described. The sensitivity, linearity, and reproducibility were evaluated using catecholamines and related compounds, including dopamine (DA), 5‐hydroxyindole‐3‐acetic acid (5‐HIAA), ascorbic acid (AA), and catechol. Initial studies with DA show the response of the PPF electrodes to be linear between 25 and 500 μM (r2=0.999) with a limit of detection (LOD) of 5 μM (S/N=3) and sensitivity of 5.8 pA/μM. Selectivity was further enhanced by employing dual‐electrode detection in the series configuration for detection of species exhibiting chemically reversible redox reactions.  相似文献

Quercetin can effectively accumulate at multi-walled carbon nanotubes-paraffin oil paste electrodes (CNTPE) and cause a sensitive anodic peak at around 0.32 V (vs. SCE) in a 0.10 M phosphate buffer solution (pH = 4.0). Under optimized conditions, the anodic peak current is linear to quercetin concentration in the ranges of 2.0 × 10− 9−1.0 × 10− 7 M and 1.0 × 10− 7−2.0 × 10− 5 M, and the regression equations are ip (μA) = 0.0017 + 0.928c (μM, r = 0.999) and ip (μA) = 0.183 + 0.0731c (μM, r = 0.995), respectively. This paste electrode can be regenerated by repetitively cycling in a blank solution for about 2 min. A 1.0 × 10− 6 M quercetin solution is measured for 10 times using the same electrode regenerated after every determination, and the relative standard deviation of the peak current is 1.7%. The method has been applied to the determination of quercetin in hydrolysate product of rutin and the recovery is 99.2–102.6%. In comparison with graphite paste electrode, carbon nanotubes-nujol paste electrode and carbon nanotubes casting film modified glassy carbon electrode, the CNTPE gives higher ratio of signal to background current and better defined voltammetric peak.  相似文献

Fluorinated porous materials, which can provide specific fluorine-fluorine interaction, hold great promise for fluoride analysis. Here, a novel fluorinated covalent-organic polymer was prepared by using 2,4,6-tris(4-aminophenyl)-1,3,5-triazine and 2,3,5,6-tetrafluorotelephtal aldehyde as the precursors and introduced as stationary phase for open-tubular capillary electrochromatography. The as-synthesized fluorinated covalent-organic polymer and the modified capillary column were characterized by infrared spectroscopy, scanning electron microscopy, and energy dispersive X-ray spectrometry. Based on strong hydrophobic interaction and fluorine–fluorine interaction provided by fluorinated covalent-organic polymer coating layer, the modified column showed powerful separation selectivity toward hydrophobic compounds, organic fluorides, and fluorinated pesticides. Additionally, the fluorinated covalent-organic polymer with good porosity and regular shape was uniformly and tightly coated on the capillary inner wall. The obtained highest column efficiency could reach up to 1.2 × 105 plates⋅m−1 for fluorophenol. The loading capacity of the modified column can reach 141 pmol for trifluorotoluene. Besides, the relative standard deviations of retention times for intraday run (n = 5), interday run (n = 3), and between columns (n = 3) were all less than 2.55%. Significantly, this novel fluorinated material-based stationary phase shows great application potential in fluorides analysis.  相似文献

A solid state polyvinyl chloride (PVC) membrane Li+-selective electrode was prepared and used as a detector in a low-dead volume flow through cell for the determination of Li+ in pharmaceutical formulations and human serum samples. The potentiometric performance characteristics of the electrode were calculated under the optimized flow conditions. The electrode had near-Nernstian behavior in the concentration range of 0.1–100 mM (R 2 = 0.9981) with a slope of 61.34 mV decade−1 and detection limit of 0.080 mM. The relative standard deviation of the electrode response for eight replicate measurements of 100, 10, and 1 mM Li+ was 0.43%, 0.45%, and 0.99%, respectively. The designed flow-through cell detector system revealed sampling rates of approximately 70 injections per hour. Flow injection potentiometry (FIP) results obtained for the pharmaceutical formulations were in good harmony with the atomic emission spectrophotometry results. However, the electrode could not be used successfully for the direct analysis of real serum samples in FIP.  相似文献

This study developed a facile, highly sensitive technique for extracting and quantifying barbiturates in serum samples. This method combined ultrasound and surfactant-assisted dispersive liquid–liquid microextraction with poly(ethylene oxide)-mediated stacking in capillary electrophoresis. Factors influencing the extraction and stacking performance, such as the type and volume of extraction solvents, the type and concentration of surfactant, extraction time, salt additives, sample matrix, solution pH, and composition of the background electrolyte, were carefully studied and optimized to achieve the optimal detection sensitivity. Under the optimized extraction (injecting 140 μL C2H4Cl2 into 1 mL of sample with pH 4 (5 mM sodium phosphate containing 0.05 mM Tween 20 and sonication for 1 min) and separation conditions (150 mM tris(hydroxymethyl)aminomethane-borate with pH 8.5 containing 0.5% (m/v) poly(ethylene oxide)), the limits of detection (signal-to-noise ratio = 3) of five barbiturates ranged from 0.20 to 0.33 ng/mL, and the calculated sensitivity improvement ranged from 868- to 1700-fold. The experimental results revealed excellent linearity (R2 > 0.99), with relative standard deviations of 2.1%–3.4% for the migration time and 4.3%–5.7% for the peak area. The recoveries of the spiked serum samples were 97.1% –110.3%. Our proposed approach offers a rapid and practical method for quantifying barbiturates in biological fluids.  相似文献

An anodic stripping voltammetric procedure for the determination of Cu(II) at an in situ-plated stannum film electrode (SnFE) was described. The results indicated that the SnFE had an attractive electroanalytical performance, with two distinct voltammetric stripping signals for copper and stannum, and showed the superior advantage for the determination of copper compared with the bismuth film electrode. Several experimental parameters were optimized. The SnFE exhibited highly linear behavior in the concentration range from 1.0 to 100.0 μg L−1 of Cu(II) (r = 0.994) with the detection limit of 0.61 μg L−1 (S/N = 3), and the relative standard deviation for a solution containing 40.0 μg L−1 Cu(II) was 2.2% (n = 8). The procedure has been successfully applied for the determination of Cu(II) in lake water sample.  相似文献

A new series of xanthone derivatives against the oral human epidermoid carcinoma (KB) cancer cell line is examined to determine the relationship between the structural properties and the biological activity of these compounds—the 3-D quantitative structure–activity relationship (3D-QSAR)—using comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). The best CoMFA and CoMSIA models were obtained using the atom-based alignment of 33 compounds, 22 training compounds and 11 tested compounds, and these give desirable statistics; those for the CoMFA standard model were: r cv2 = 0.691, r 2 = 0.998, S press = 0.178, s = 0.014 and F = 1080.765, while CoMSIA combined steric, electrostatic, hydrophobic and hydrogen-bond acceptor fields: r cv2 = 0.600, r 2 = 0.988, S press = 0.206, s = 0.034 and F = 284.433. The 3D-QSAR models calculated satisfactory test set activities. The 3D-QSAR contour plots correlated strongly with the experimental data for the binding topology. For this reason, these results would be beneficial for predicting affinities with the compounds of interest, and they are advantageous for guiding the design and synthesis of new and more effective anticancer agents.  相似文献

The objective of this research was to test suitability of the Dumas combustion method to completely substitute the Kjeldahl method in routine laboratory determination of crude protein content in cereals and oilseeds. The validation of the method demonstrated that it is able to determine crude protein content in cereals and oilseeds in an efficient and accurate manner, with a detection limit w(N) = 0.006%, quantification limit w(N) = 0.019%, repeatability precision RSD r = 0.41%, intra-laboratory reproducibility precision RSD R = 0.74%, trueness, expressed in terms of bias b = 0.43%, and linear response between (2.36–19.2) mg N. Measurement uncertainty, expressed as relative expanded uncertainty (coverage factor k = 2, confidence level 95%), was calculated from validation data (U rel = 2.24%). In order to examine the relationship between two methods, 15 cereal grain and oilseed samples were analyzed using Dumas and Kjeldahl procedure. The Kjeldahl procedure gave slightly lower w(N) values than the Dumas procedure: w K(N) = 0.9905 w D(N) = 0.0376 (R 2  = 0.9996). Relative standard deviations and results of homogeneity test obtained during analysis of complex cereal products (cereal breakfast and muesli bars) show that the Dumas combustion method may be less suitable for analysis of such samples compared to Kjeldahl method.  相似文献

A combined gas-phase electron diffraction and quantum chemical (B3LYP/6-311+G**, B3LYP/cc-pvtz, MP2/cc-pvtz) study of molecular structure of 2-nitrobenzenesulfonamide (2-NBSA) was carried out. Quantum chemical calculations showed that 2-NBSA has four conformers, two of which are stabilized by intramolecular hydrogen bond. The latter (with the S–N bond in a close to orthogonal position around the phenyl ring and differing from each other by staggered or eclipsed positions of the N–H and S=O bonds in the SO2NH2 group) presented in a saturated vapor over 2-NBSA at T = 433 (3) K in commensurable amounts. Experimental internuclear distances (Ǻ) for the staggered conformer are (?): r h1(C–H)av. = 1.071(9), r h1(C–C)av. = 1.390(4), r h1(C–S) = 1.789(8), r h1(S=O)av. = 1.427(6), r h1(S–N) = 1.644(6), r h1(N–O)av. = 1.221(4), r h1(C′–N) = 1.487(8), r h1(N–H)av. = 1.014. Calculations at B3LYP/cc-pvtz level were performed to determine the structure and the energies of the transition states between conformers. It was shown that the conformer structures of free molecule differ from those of a molecule stabilized by intermolecular hydrogen bonds in a crystal. Influence of a substituent X (X = –CH3, –NO2) on conformational features of the ortho-substituted benzenesulfonamide was established.  相似文献

The burning rate of AN–HTPB-based propellant catalysed with chromium salt has been studied using conventional strand burner under the various pressure range, i.e. from atmospheric pressure to 6.897 MPa and verified with Piobert law, i.e. r = aP n . At atmospheric pressure, the burning rate AN–HTPB propellant was being accelerated with the chromium-based catalysts used. In case of lead chromate-catalysed system, burning rate was observed 2.655 times higher than burning rate (r = 0.200 mm s−1) of virgin AN–HTPB propellant sample. However, the Copper chromate-catalysed propellant burned with slower rate (r = 0.160 mm s−1) than the virgin AN–HTPB propellant sample. The burning rate of all catalysed propellant samples are found to be the pressure sensitive and accelerated higher with rise of pressure. The highest burning rate (r = 2.422 mm s−1) was recorded with ammonium dichromate and lowest (r = 1.40 mm s−1) with lead chromate-catalysed propellant sample with the rise of pressure up to 6.897 MPa at different pressures. A linear relationship was observed between the burning rate and pressure rise which followed the Piobert law, i.e. r = aP n . The pressure index (n) values of AN–HTPB-based samples were calculated higher when catalysed with ammonium dichromate, Copper Chromate, Cr2O3, Potassium dichromate (n = 0.525, 0.555, 0.429, and 0.408 respectively) and lower (n = 0.226) with lead chromate compared to virgin sample (n = 0.405). Higher value indicates the positive effect on accelerating the burning rate with catalyst at higher pressure ranges.  相似文献

A novel and fully automated sequential injection analysis manifold coupled to a capillary electrophoresis apparatus with amperometric detection, is described. The sequential injection manifold was isolated from the high voltage by inserting an air plug into the circuit. Small buffer reservoirs were used to avoid the need to pump fresh buffer to the interface during the electrophoretic separation. No decoupling device was used to mitigate the interference from the high voltage electric field, instead the potential shift induced by the separation voltage, was accounted for. The new hydrodynamic injection method presented is based on the overpressure created in the circuit when a pinch valve is closed for a predetermined time. The injection method yields RSD values of peak height and area below 2.55 and 1.82%, respectively, at different durations of valve closure (n = 5). The capillary and working electrode alignment was achieved by adapting a commercial available capillary union. When the electrode was replaced, the alignment method proved to be very reliable, yielding RSD values of peak height and area lower than 2.64 and 2.08%, respectively (n = 8). Using this system with a gold microelectrode, dopamine, and epinephrine could be quantified within the concentration range of 1–500 μM and detected at a concentration of 0.3 μM. The methods here presented could be applied for the development of new capillary electrophoresis systems with amperometric detection and/or to the design of fully automated systems for online process monitoring purposes.  相似文献

An in‐house flow‐injection capillary electrophoresis with capacitively coupled contactless conductivity detection method was developed for the direct measurement of colistin in pharmaceutical samples. The flow injection and capillary electrophoresis systems are connected by an acrylic interface. Capillary electrophoresis separation is achieved within 2 min using a background electrolyte solution of 5 mM 2‐morpholinoethanesulfonic acid and 5 mM histidine (pH 6). The flow‐injection section allows for convenient filling of the capillary and sample introduction without the use of a pressure/vacuum manifold. Capacitively coupled contactless conductivity detection is employed since colistin has no chromophore but is cationic at pH 6. Calibration curve is linear from 20 to 150 mg/L, with a correlation coefficient (r2) of 0.997. The limit of quantitation is 20 mg/L. The developed method provides precision, simplicity, and short analysis time.  相似文献

A magnetic mesoporous carbon material (i.e., mesoporous iron oxide/C, mesoFe/C) is synthesized for protein immobilization, using glucose oxidase (GOx) as model. Transmission electron microscopy images show that mesoFe/C has highly ordered porous structure with uniform pore size, and iron oxide nanoparticles are dispersed along the wall of carbon. After adsorption of GOx, the GOx-mesoFe/C composite is separated with magnet. The immobilized GOx remains its natural structure according to the reflection–absorption infrared spectra. When the GOx-mesoFe/C composite is coated on a Pt electrode surface, the GOx gives a couple of quasireversible voltammetric peaks at −0.5 V (vs. saturated calomel electrode) due to the redox of FAD/FADH2. The electron-transfer rate constant (k s) is ca. 0.49 s−1. The modified electrode presents remarkably amperometric response to glucose at 0.6 V. The response time (t 95%) is less than 6 s; the response current is linear to glucose concentration in the range of 0.2–10 mM with a sensitivity of 27 μA mM−1 cm−2. The detection limit is 0.08 mM (S/N = 3). The apparent Michaelis–Menten constant (K mapp) of the enzyme reaction is ca. 6.6 mM, indicating that the GOx immobilized with mesoFe/C has high affinity to the substrate.  相似文献

A rapid, sensitive, and selective liquid chromatography with tandem mass spectrometry method was developed and fully validated for the simultaneous quantification of arotinolol and amlodipine in rat plasma. Two internal standards were introduced with metoprolol as the internal standard of arotinolol and (S)‐amlodipine‐d4 as the internal standard of amlodipine. The analytes were isolated from 50.0 μL plasma samples by a simple protein precipitation using acetonitrile. The chromatographic separation was achieved in 5 min on a C18 column. The mobile phase consisted of phase A 5% methanol and phase B 95% methanol (both containing 0.5% formic acid and 5 mM ammonium acetate) and was delivered in gradient elution at 0.300 mL/min. Quantification was performed in multiple reaction monitoring mode with the transition m/z 372.1 → 316.1 for arotinolol, m/z 268.2 → 116.2 for metoprolol, m/z 409.1 → 238.1 for amlodipine and m/z 413.1 → 238.1 for (S)‐amlodipine‐d4. Linearity was obtained over the range of 0.200–40.0 ng/mL for arotinolol (r= 0.9988) and 0.500–100 ng/mL for amlodipine (r= 0.9985) in rat plasma. The validated data have met the acceptance criteria in FDA guideline. This method was successfully applied to a pharmacokinetic interaction study in rats, and the results indicated that there was no significant drug–drug interaction between arotinolol and amlodipine.  相似文献

In this communication, an amperometric glucose biosensor based on MnO2/MWNTs electrode was reported. MnO2 was homogeneously coated on vertically aligned MWNTs by electrodeposition. The MnO2/MWNTs electrode displayed high electrocatalytic activity towards the oxidation of glucose in alkaline solution, showing about 0.30 V negative shift in peak potential with oxidation starting at ca. −0.20 V (vs. 3 M KCl–Ag/AgCl) as compared with bare MWNTs electrode. At an applied potential of +0.30 V, the MnO2/MWNTs electrode gives a linear dependence (R = 0.995) in the glucose concentration up to 28 mM with a sensitivity of 33.19 μA mM−1. Meanwhile, the MnO2/MWNTs electrode is also highly resistant toward poisoning by chloride ions. In addition, interference from the oxidation of common interfering species such as ascorbic acid, dopamine, and uric acid is effectively avoided. The MnO2/MWNTs electrode allows highly sensitive, low-potential, stable, and fast amperometric sensing of glucose, which is promising for the development of nonenzymatic glucose sensor.  相似文献
