The thienopyridine derivative 2 , obtained from reaction of acetoacetic ester with 1 in the presence of tin tetrachloride, was treated with triphenylphosphine in hexachloroethane and Et3N to give iminophosphorane 3 . Iminophosphorane 3 reacted with phenyl isocyanate to give carbodiimide 4 , which was further treated with phenols or ethenol to produce 2‐substituted 5,8,9‐trimethyl‐3‐phenyl‐thieno[3′,2′‐5,6]pyrido[4,3‐d]pyrimidin‐4(3H)‐ones 5 in presence of catalytic amount of K2CO3 or EtONa. The structures of compounds 5 were confirmed by 1H NMR, IR, MS, and elemental analysis.  相似文献

The carbodiimides 4 , obtained from reactions of iminophosphorane 3 with aromatic isocyanates, were reacted with secondary amines to give 2‐dialkylamino‐5‐ethyl‐6‐methyl‐thieno[2,3‐d]pyrimidin‐4(3H)‐ones 6 in the presence of catalytic amount of EtONa. Reactions of 4 with phenols or ROH in the presence of the catalytic amount of K2CO3 or RONa gave 2‐aryloxy‐ or 2‐alkoxy‐5‐ethyl‐6‐methyl‐thieno[2,3‐d]pyrimidin‐4(3H)‐ones 6 in satisfactory yields. A general and efficient palladium‐catalyzed direct C3 alkenylation of 4H‐pyrido[1,2‐a]pyrimidin‐4‐ones using AgOAc/O2 as the oxidant has been developed. A variety of 4H‐pyrido[1,2‐a]pyrimidin‐4‐ones were successfully coupled with acrylate esters, styrenes, methylvinylketone, and acrylamide in moderate to excellent yields. The reaction exhibited complete regio‐ and stereoselectivity. This transformation provides an attractive new approach to functionalize 4H‐pyrido[1,2‐a]pyrimidin‐4‐ones.  相似文献

In the presence of N, N′‐dicyclohexylcarbodiimide, 2‐aminopyridine and its derivatives (2) condensed with 2, 2‐di‐hydropolyfluoroalkanoic adds (1) to give the corresponding amides. Subsequent intramolecular Micheal addition‐elimination reactions of the fluorine‐containing amides under basic conditions gave 4‐fluoroalkyl‐2H‐pyrido[1,2‐a]pyrimidin‐2‐ones (3) in good yields.  相似文献

1‐Methyl‐3‐propyl‐1H‐pyrazole‐5‐carboxylic acid ( 3 ) was exclusively brominated at the 4‐position by bromine in the dark. Brominated product 8 was then converted into 1‐methyl‐3‐propyl‐1H‐pyrazole‐5‐car‐boxamide 9 by successive treatment with thionyl chloride and ammonium hydroxide. Carboxamide 9 was treated with various aroyl amides under microwave (MW) irradiation to afford 4‐aroylamino‐1‐methyl‐3‐propyl‐1H‐pyrazole‐5‐carboxamides 10‐22 and 5‐aryl‐1‐methyl‐3‐propyl‐1,6‐dihydro‐1H‐pyrazolo[4,3‐d]pyrimidin‐7‐ones 23‐35. The 1H‐pyrazole‐5‐carboxamides 10‐22 were also converted to pyrimidinones 23‐35 either by conventional heating or by MW irradiation. However, MW irradiation method gives excellent yields in very short time.  相似文献

Alkylation of 2‐hydroxy‐4H‐pyrido[1,2‐a]pyrimidin‐4‐one ( 1 ) was investigated under solid–liquid phase transfer catalysis conditions (PTC), using tetrabutylammonium bromide and potassium carbonate. The reaction with alkyl halides led to the formation of various 2‐alkoxy products, in fair yields. Reaction of compound 1 with epichlorohydrin and chloroacetonitrile, under the same PTC conditions, afforded novel O1,O3‐disubstituted glycerol and oxazolopyridopyrimidone betaine derivatives, respectively. Some 3‐halo‐, 3,3‐dihalo, and/or 2,3‐dihalopyrido[1,2‐a]pyrimidines were also prepared using different halogenating agents at different reaction conditions. © 2007 Wiley Periodicals, Inc. Cyclocondensation of 2,3,3‐trimefhyl‐3H‐indoles 2 with malonates 3 gives 8‐hydroxy‐10,10‐dimefhyl‐10H‐pyrido[1,2‐a]indol‐6‐ones 4 , which were halogenated in position 7, 8 and 9 with sulfuryl chloride, bromine or phosphoroxychloride to give the corresponding halo‐10,10‐dimethyl‐10H‐pyrido[1,2‐a]indoles 5, 6, 7 and 8 . Amination affords the 8‐amino‐10,10‐dimethyl‐10H‐pyrido[1,2‐a]indol‐6‐one 9 . Nitration gives either the 10,10‐dimethyl‐7‐nitro‐10H‐pyrido[1,2‐a]indoles 10 or 10,10‐dimethyl‐7‐hydroxy‐10H‐pyrido[1,2‐a]indoles 11 , depending on the conditions.  相似文献

Treatment of 2‐bromoaryl pyrrole/indol‐2‐yl ketones with cesium carbonate in DMF resulted in the formation of 9H‐pyrrolo[1,2‐a]indol‐9‐ones and 10H‐indolo[1,2‐a]indol‐10‐ones in moderate to excellent isolated yields.  相似文献

Several pyrido[2,3‐e]pyrimidine fused with other rings have been prepared by intramolecular cyclization of 5‐(4‐chlorophenyl)‐2‐hydrazino‐benzo [6,7]cyclohepta‐[1,2‐b]pyrido[2,3‐e]pyrimidine‐4‐one ( 1 ) with acids, carbon disulfide to form triazole derivatives ( 2,4 ), halo‐ketones to give triazine derivative ( 5 ), β‐ketoesters, β‐cyanoesters, and β‐diketones to yield 2‐(1‐pyrazolyl) derivatives ( 7,9,10 ), and aldehydes to form arylhydrazone derivatives ( 11a,b ) which cyclized to form triazoles ( 12a,b ). Also, acyclic N‐nucleosides are prepared by heating under reflux 2‐hydrazino‐benzo[6,7]cyclohepta[1,2‐b]pyrido[2,3‐e] pyrimidin‐4‐one ( 1 ) with xylose and glucose to give the corresponding acyclic N‐nucleosides ( 13a,b ) which are cyclized to afford the corresponding protected tetra and penta–O‐acetate C‐nucleosides ( 14a,b ). Deacetylating of the latter nucleosides afforded the free acyclic C‐nucleosides ( 15a,b ). © 2007 Wiley Periodicals, Inc. A convenient approach to the synthesis of the previously unknown 7H‐furo[3,2‐b]pyran‐7‐ones based on the intramolecular cyclization of carbonyl derivatives of 5‐hydroxy‐2‐methyl‐4H‐pyran‐4‐one has been elaborated. Key intermediates in the synthesis of the target 7H‐furo[3,2‐b]pyran‐7‐ones are 3‐hydroxy‐6‐methyl‐2‐(2‐oxo‐2‐arylethyl)‐4H‐pyran‐4‐ones. They are formed as a result of multicomponent condensation of 5‐hydroxy‐2‐methyl‐4H‐pyran‐4‐one with arylglyoxals and 4‐methoxyaniline.  相似文献

5‐Methoxy and 5‐anilinopyrido[2,3‐d]pyrimidin‐7(8H)‐ones 2a‐2f were obtained by a tandem Michael addition‐cyclization reaction of methanol and anilines with pyrimidinylpropynoate 5. Methoxy derivative 2a was obtained in 62% yield by treatment of 5 with methanol and potassium carbonate. Anilino derivatives 2b‐2f were prepared in 31–71% yields by reacting 5 with the corresponding anilines in refluxing methanol. This methodology accomplishes Michael‐addition and pyridopyrimidinone ring formation in one‐pot and affords the desired products in reasonable yield without chromatography. Propynoate 5 did not react with 4‐cyanoaniline under these conditions. Reaction of 5 with 2‐aminopyridine gave the unexpected arylpyrido[2,3‐d]pyrimidinone 8 in 58% yield and reaction of 5 with imidazole afforded Michael‐adduct 9 in 69% yield. Compounds 2a and 5 were submicromolar inhibitors of epidermal growth factor receptor (EGFR) tyrosine kinase.  相似文献

Sixteen 5‐alkylamino‐3,6‐diaryl‐2‐thioxo‐2,3,6,7‐tetrahydrothiazolo[4,5‐d]pyrimidin‐7‐ones 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h , 4i , 4j , 4k , 4l , 4m , 4n , 4o , 4p were designed and easily synthesized via a tandem aza‐Wittig reaction. The iminophosphorane 2 , obtained from reaction of 1 with triphenylphosphine, hexachloroethane and Et3N, reacted with aromatic isocyanate to give carbodiimide 3 . carbodiimide 3 reacted with alkylamines to provide the title compounds in 45–61% isolated yields in presence of catalytic amount of ethoxide. Useful oxidation reaction of 2‐alkyl(aryl)‐3‐methylthiopyrano[4,3‐c]pyrazol‐4(2H)‐ones, leading to either the corresponding sulfoxides or sulfones, using hydrogen peroxide and acetic acid in 1,2‐dichloroethane, is described.
