For the first time, tetracyclic compounds, namely, furo[2′,3′:3,4]cyclohepta[1,2‐b]indoles were synthesized by recyclization of ortho‐substituted aryldifurylmethanes containing tert‐butyl groups at C5 positions of the furan rings. It was shown that [2‐(benzoylamino)phenyl]bis(5‐tert‐butyl‐2‐furyl)methanes 12 are transformed into tetracycles 15 at room temperature under treatment with POCl3 in benzene solution containing some drops of water. The reaction proceeds via the intermediate formation of 1‐benzoylamino‐3‐(5‐tert‐butyl‐2‐furyl)‐2‐(4,4‐dimethyl‐3‐oxopentyl)indoles 14 which can be isolated from the reaction mixture. The method is very simple but its application is restricted due to side reactions if electron‐releasing groups are present in 12 . On the other hand, the decrease of electron density on furan ring in the starting compounds (for example, the use of [2‐X‐phenyl]difurylmethanes (where X = tosylamino or hydroxy group) prevents cyclization under the studied reaction conditions. From an analysis of nmr spectral data, 1,6,7,12b-tetrahydro-2H,4H-[1,3 ]oxazino[3′, 4′ :1,2]-pyrido[ 3,4-b ]indole is shown to exist in solution at room temperature almost entirely in the cis-fused ring conformation with the nitrogen lone pair bisecting the C4 methylene group whereas under the same conditions 1,2,3,6,7,12b-hexahydro-3-methyl-4H-pyrimido[3′,4′:1,2] pyrido-[3,4-b ]indole exists as an approximately 50:50 equilibrium mixture of the cis and trans-fused ring conformations.  相似文献

Novel conjugated polymers composed of benzo[1,2‐b:4,5‐b′]dithiophene and thieno[3,4‐b]pyrazine or dithieno[3′,2′:3,4;2″,3″:5,6]benzo[1,2‐d]imidazole units are synthesized by Stille polycondensation. The resulting polymers display a longer wavelength absorption and well‐defined redox activities. The effective intramolecular charge‐transfer and energy levels of all polymers are elucidated by computational calculations. Bulk‐heterojunction solar cells based on these polymers as p‐type semiconductors and [6,6]‐phenyl‐C61‐butyric acid methyl ester (PC61BM) as an n‐type semiconductor are fabricated, and their photovoltaic performances are for the first time evaluated. © 2015 Wiley Periodicals, Inc. The failure to obtain the N-(13)alkylrutaecarpines ( 1d,e,f ) by heating rutaecarpine ( 1a ) with neat alkyl halides at 120° is discussed in comparison with the facile reaction with methyl iodide. In contrast, with alkyl halide-potassium carbonate in acetone, the corresponding N-(13)alkyl-rutaeearpines ( 1d-l ) are obtained in good yield. By use of 1,3-diiodopropane and 1,2-dibromo-ethane, this reaction provides a facile route to 12a and 13 which are derivatives of the heretofore unknown indolo[1,2′:3,4]pyrazo[1,2-α]quinazoline and indolo[1,2 :3,4][1,4]diazepino[1,2-α]quinazoline ring systems.  相似文献

Reaction of 1-methyl-3,4-dihydro-β-carboline 1 with methyl glycidate 4 gave the novel tetracyclic conjugated γ-lactam, 2-methylpyrrolo[1′,2′:1,2]pyrido[3,4–6]indol-3-one 5 . The same compound 5 was obtained in higher yield when 1 was treated with methyl 2-methoxyacrylate 7 . Sodium borohydride treatment of 5 resulted in the cleavage of lactam to yield the unexpected alcohols 8 and 9 .  相似文献

The classical Pictet-Spengler reaction of tryptamine with the isomeric N-benzylpiperidones 3a, 3b and N-benzylpyrrolidone 3c yielded the spiro derivatives of 1,2,3,4-tetrahydro-β-carboline 5a, 5b and 5c . Cyclocon-densation of the spirotetrahydrocarboline with chloroacetic chloride and the subsequent reductive debenzylation afforded the new ring systems of trihydrodiazabicyclo[3.m.n]alkano[4′,5′:1,2]pyrido[3,4-b]indoles 8a , 8b , and 8c . The structures of the bicyclic systems 8a, 8b , and 8c were determined by using both, high-resolution 1H and 13C nmr techniques and force field and MNDO calculations.  相似文献

The reaction of the isothiourea derivative 2 with methylaminc or pyrrolidine resulted in guanidines 3a-3b . Using hydrazine under the same conditions the tetrazole derivative 4 was obtained. On reacting 2 with piperidine, morpholine, methylhydrazine, phenylhydrazine, hydroxylamine or sodium hydroxide, cycliza-tion took place leading to the novel 4-cyanimino-1,2,3,4,6,7,12,12b-octahydro-3,12b-ethanopyrim-ido[1′,6′:1,2]pyrido[3,4-b]indole ( 5 ). Some structural aspects of 5 and other model compounds were analysed mainly by 13C nmr spectroscopy.  相似文献

6‐(2‐Thienyl)‐4‐(trifluoromethyl)‐1H‐pyrazolo[3,4‐b]pyridine‐3‐amine reacted with different active methylene compounds to afford pyridopyrazolopyrimidine derivatives. On the other hand, it reacted with some halo compounds to give the imidazo[1′,2′:1,5]pyrazolo[3,4‐b]pyridine derivatives. Also, it diazotized to give the corresponding diazonium chloride that is coupled with several active methylene compounds to give the corresponding triazine derivatives. Furthermore, compound 3‐amino‐6‐(2(thienyl)‐4‐(trifluoromethyl)thieno[2,3‐b]pyridine‐2‐carbohydrazide reacted with some β‐dicarbonyl compounds and some sulfur‐containing compounds to afford the corresponding pyrazolyl oxadiazolylthieno[2,3‐b]pyridine derivatives.  相似文献

A new bridgehead nitrogen hetero‐ cycle viz. 11‐carboethoxy‐9‐oxo‐pyrimido[3′2′:3,4]‐1,2,4‐triazino[5,6‐b]indole 3 has been synthesized from 3‐azido‐5H‐1,2,4‐triazino[5,6‐b]indole 2 by its reaction with diethyl fumerate. The intermediate 2 was obtained by treating 3‐hydrazino‐5H‐1,2,4‐triazino[5,6‐b]indole with NaNO2 in presence of polyphosphoric acid. A novel series of thieno[2′,3′:4,5]pyrimido[1,2‐b][1,2,4,5]tetrazin‐6‐one derivatives 14 were prepared from the reaction of 3‐amino‐2‐thioxo‐1,2,3,5,6,7‐hexahydro‐4H‐cyclopenta[4,5]thieno[2,3‐d]pyrimidin‐4‐one 3 or its methylthio 4 with hydrazonoyl chlorides 9 . The mechanism of the studied reactions has been discussed and further evidence for the assigned structure of the products is based on alternative synthesis. A single crystal X‐ray analysis of compound 14e has been carried out.  相似文献

The addition of organometallic reagents to the 13b‐position of the indolo[2′,3′:3,4]pyrido[1,2‐b]isoquinoline imminium salt 4 is described. Reaction of 4 with tetraallyl tin in 2‐methoxyethanol gave the allyl adduct 7 in moderate yield. Further elaboration of 7 yielded the pentacyclic benzylidene alcohols 13 and 14 . Structure elucidation of the compounds prepared was achieved by a combination of 1H nmr spec troscopy and X‐ray crystallography.  相似文献

2-Thioxo-1,2-dihydropyridine derivatives 2a, 2b were reacted with methyl iodide to give 2-methylthiopyridines 3a, 3b, which were reacted with hydrazine hydrate to produce 3-aminopyrazolo[5,4-b]pyridines 4a, 4b. A new convenient synthon for heterocyclic chemistry, namely 1H‐pyrazolo[3,4‐b]pyridin‐3‐ylguanidine was successfully prepared by selective guanylation of 1H‐pyrazolo[3,4‐b]pyridin‐3‐amine. A series of 3,4‐dihydropyrido[2′,3′:3,4]pyrazolo[1,5‐a][1,3,5]triazin‐2‐amines was synthesized from 1H‐pyrazolo[3,4‐b]pyridin‐3‐ylguanidine using aldehydes or ketones as one‐carbon inserting reagents. The tautomeric preferences of the products were determined using spectroscopic (e.g., 2D NOESY NMR) and single crystal X‐ray diffraction data.  相似文献

The 3‐amino‐6‐(trifluoromethyl)furo[2,3‐b]pyridine‐2‐carbohydrazide ( 5 ) was prepared from 3‐cyano‐6‐trifluoromethyl‐2(1H)pyridone ( 2 ) in series of steps via selective O‐alkylation, Thorpe–Ziegler cyclization followed by reaction with hydrazine hydrate. The 2‐carbohydrazide ( 5 ) was further reacted with aliphatic acids under different reaction temperatures to form a series of novel N‐acylfuro[2,3‐b]pyridine‐2‐carbohydrazide ( 6 ) and pyrido[3′,2′:4,5]furo[3,2‐d]pyrimidine derivatives ( 7 ). All the compounds 6 and 7 were screened for cytotoxic activity against breast carcinoma MD Anderson‐Metastatic Breast (MDA‐MB) 231 (aggressive) cell lines at 10 µM concentration. Compounds 6a , 6b , and 6c showed promising activity.  相似文献

The synthesis of new pyrido[3′,2′:5,6]thiopyrano[3,2‐b]indol‐5(6H)‐ones was accomplished by the Fischer‐indole cyclization of some 2,3‐dihydro‐3‐phenylhydrazonothiopyrano[2,3‐b]pyridin‐4(4H)‐ones, obtained from the 2,3‐dihydro‐3‐hydroxymethylenethiopyrano[2,3‐b]pyridin‐4(4H)‐one, by the Japp‐Klingemann reaction. 6H‐Pyrido[3′,2′:5,6]thiopyrano[4,3‐b]quinolines were obtained by reaction of 2,3‐dihydrothiopyrano‐[2,3‐b]pyridin‐4(4H)‐ones with o‐aminobenzaldehyde or 5‐substituted isatins. The preparation of some derivatives, functionalized with an alkylamino‐substituted side chain, is also described.  相似文献
