A number of 4‐aryloxymethyl‐6‐phenyl‐2H‐pyrano[3,2‐c][1,8]naphthyridin‐5(6H)‐ones ( 4a‐f ) are regioselectively synthesized in 72‐78% yield by the Claisen rearrangement of 4‐(4′‐aryloxybut‐2′‐ynyloxy)‐1‐phenyl‐1,8‐naphthyridin‐2(1H)‐ones ( 3a‐f ) in refluxing chlorobenzene for 4‐6 h. These products are then subjected to a second Claisen rearrangement catalyzed by anhydrous AlCl3 at room temperature for 2 h to give hitherto unreported pentacyclic heterocycles ( 5a‐f ) in 78‐85% yield.  相似文献

A simple and efficient protocol for the synthesis of 5‐aryl‐2‐(2‐substituted‐1,8‐naphthyridin‐3‐yl)‐thiazolo[3,2‐b][1,2,4]triazoles ( 4 ) is achieved by cyclocondensation of 3‐(2‐substituted‐1,8‐naphthyridin‐3‐yl)‐1,2,4‐triazoles ( 3 ) with α‐halogenoketones in anhyd. methanol under microwave irradiation. The products are obtained in good yields and in a state of high purity.  相似文献

In the course of our search for compounds with potential antitumor properties we have undertaken the synthesis of benzo[c][1,8]phenanthroline derivatives. Our project required the preparation of 8,9‐dimethoxy benzo[c][1,8]phenanthrolin‐6‐ones. This was first attempted by the lithiumdiisopropylamide cyclization of N‐(isoquinol‐5‐yl)‐2‐bromo‐4,5‐dimethoxybenzamide. The reaction led to 40% of the unexpected internal Diels‐Alder adduct 3,4‐dimethoxy‐6H‐pyrido[2,3‐i]6,8a‐ethenoindolo[cd]isoquinoline‐2(1H)‐one, which arose from a benzyne intermediate. In a second and more successful approach, the internal biaryl palladium diacetate‐assisted coupling reaction of properly N‐protected N‐(isoquinol‐5‐yl)‐2‐bromo‐4,5‐dimethoxybenzamide was studied. The optimisation of the protecting group necessary for this procedure led to a 64% yield of the target compound starting from N,N‐(isoquinol‐5‐yl)‐bis‐(2‐bromo‐4,5‐dimethoxybenzamide).  相似文献

3‐Nitrosoimidazo[1,2‐a]pyridine, 3‐nitrosoimidazo[1,2‐a]pyrimidine, 3‐nitrosoquinoxaline, 2‐nitroso‐4H‐benzo[b]thiazine, 2‐nitroso‐4H‐benzo[b]oxazine, isoxazoles, isoxazolo[3,4‐d]pyridazines and pyrrolo[3,4‐d]isoxazole‐4,6‐dione were synthesized from 2‐chloro‐2‐(hydroximino)‐1‐(4‐methyl‐2‐phenylthiazol‐5‐yl)ethanone and different reagents. Structures of the newly synthesized compounds were confirmed by elemental analysis and spectral data.  相似文献

The synthesis of a new set of selenadiazoles, 4‐aryl‐5‐(1‐aryl‐2‐methyl‐2‐nitropropyl)‐1,2,3‐selenadiazoles ( 4 ) derived from 2‐[4‐methyl‐4‐nitro‐1,3‐diarylpentylidene]‐1‐hydrazinecarboxamide ( 3 ) has been reported. THF has been found to be the solvent of choice for this reaction. Structural features of 3 and 4 have been analyzed by NMR and X‐ray techniques.  相似文献

A series of N‐carboxymethylacridine‐1,8‐dione derivatives were synthesized by one‐pot reaction of aldehyde, dimedone and glycine in glycol under microwave irradiation without catalyst with excellent yields (78‐92%) and short reaction time (4‐8min). And the reaction was not only suitable for aromatic monoaldehyde, but also aromatic dialdehyde.  相似文献

Methyl‐3‐(2‐chloroquinolin‐3‐yl)acrylates 5a‐i on reaction with methyl amine in acetonitrile yielded methyl‐3‐[2‐(methylamino)quinolin‐3‐yl]acrylates 6a‐i . When, these were followed by the reaction with the Vilsmeier reagent, they afforded methyl benzo[b][1,8]naphthyridin‐3‐carboxylate 7a‐i in good yields.  相似文献

Tricyclic dihydropyridines like ZM244085 are potential KATP channel openers. In this study 3‐cyanophenyl ring of ZM244085 was replaced with imidazolyl ring. So, 9‐[1‐benzyl‐5‐(alkylsulfonyl)‐1H‐2‐imidazolyl]perhydro‐1,8‐acridinediones ( 5d‐f ) were synthesized from 2‐alkylsulfonyl‐1‐benzyl‐5‐formylimidazole ( 4d‐f ) and cyclohexane‐1,3‐dione according to classical Hantzch synthesis as potential potassium channel modulators.  相似文献

The dehydrogenations with iron(III) chloride, potassium permanganate, and for the first time, CAN, tris‐[trinitrato‐cerium(IV)]‐paraperiodate (TTNCPP), and (diacetoxyiodo)benzene (IBDA) are investigated. Hydrogen peroxide, sodium periodate, potassium permanganate, and ammonium cerium(IV) nitrate (CAN) oxidation of chiral benzothiazolines, O‐ and O,N‐acylated derivatives of the condensates of D‐galactose with 2‐aminothiophenols or L‐cysteine, are also reported.  相似文献

Short pathways are described for the synthesis of a representative example of each of the 7,8‐dihydro‐and 1,2,3,4‐tetrahydro‐1,6‐naphthyridine‐5(6H)‐one ring systems from simple pyridine precursors. An attempted synthesis of the related 4,6‐dihydro‐1,6‐naphthyridin‐5(1H)‐one ring system from a common intermediate was unsuccessful.  相似文献

A simple one‐pot procedure for ring enlargement of α‐chloromethyl N‐containing heterocycles has been developed. By reaction of chloromethyl tetrahydroisoquinoline and its thieno analog with benzyl or allyl bromide under basic conditions, ring expansion and N‐substitution were achieved simultaneously. The key to the transformation was proposed to involve the formation of aziridinium salt and subsequent bond breaking between the nitrogen and tertiary carbon atoms.  相似文献

Reaction of 6‐methyl‐4‐oxo‐4H‐[1]‐benzopyran‐3‐carboxaldehyde 1 with 5‐amino‐3‐methyl‐1‐phenylpyrazole 2 in alcoholic reaction media in the presence of 4‐toluenesulfonic acid as catalyst afforded 5‐(2‐hydroxy‐5‐methylbenzoyl)‐3‐methyl‐1‐phenyl‐1H‐pyrazolo[3,4‐b]pyridine 3 and 2‐methoxy‐6‐methyl‐3‐(3‐methyl‐1‐phenylpyrazol‐5‐ylaminomethylene)chroman‐4‐one 7 . 3‐Alkyl/aryl‐3‐amino‐1H,3H‐quinoline‐2,4‐diones react with alkyl/aryl isocyanates to give novel 3‐alkyl/aryl‐3‐ureido‐1H,3H‐quinoline‐2,4‐diones or 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones. In some cases, a mixture of both products was obtained and separated by fractional crystallization. All compounds were characterized by their 1H, 13C, ir and ms data and some of them also by 15N nmr data.  相似文献

A number of substituted 4H,5H,6H‐thiazolo[3,2‐a][1,5]benzodiazepinium salts 2a‐h, 5, 9 , which are based on the novel thiazolobenzodiazepine system, were prepared by condensation‐cyclization of 1,5‐benzodiazepine‐2‐thiones 1a‐f, h, 4 with α‐haloketones, as well as with α‐bromoacetaldehyde diethyl acetal. The structure and stereochemistry of the ring system obtained were investigated by 1H and 13C nmr spectroscopy: the additional heterocyclic nucleus was found to appreciably influence the conformational mobility of the heptatomic ring. Upon treatment of salt 2d with alkali the presence of the base enamine structure in solution has been postulated.  相似文献
