Silver nanoparticles (Ag NPs) are becoming increasingly popular as antimicrobial agents in consumer goods with consequent risk to environmental health from discharges. Environmentally relevant fate and transport investigations are limited but essential to gain understanding towards bioavailability and toxicology. In this study, monodisperse 15 nm citrate-stabilised Ag NPs were synthesised, characterised and then fractionated by flow field-flow fractionation (FlFFF) at environmentally relevant conditions (pH 5 or 8, presence of natural organic macromolecules (NOM) and presence of sodium or calcium). At low ionic strength, Ag NPs particle size increased as pH increased from 5 to 8. However, changing the ionic strength from 10−3 to 10−2 M Na increased instability of the Ag NPs, and loss of peak at pH 5 but in the presence of humic substance (HS), a reduction in NP size was seen, most likely due to a reduction in the diffuse layer. The presence of Ca2+ ions, at the higher ionic strengths caused complete loss of the solution Ag NPs with or without HS, most likely due to aggregation. At the lower Ca2+ ionic strength the Ag NPs were still unstable, but again, in the presence of HS the NPs were largely dispersed. The presence of HS improved stability of Ag NPs under these conditions by forming a surface coating resulting in both steric and charge stabilisation. This work implies that Ag NPs could have long residence times in aquatic systems in the presence of HS potentially resulting in increased bioavailability.  相似文献

Antifouling biocides used in boat paints were analyzed with a battery of toxicity bioassays to evaluate the toxic effects of these compounds on Vibrio fischeri, Daphnia magna and Selenastrum capricornotum. The antifoulants tested were Irgarol 1051, Kathon 5287, chlorothalonil, diuron, dichlofluanid, 2-thiocyanomethylthiobenzothiazole (TCMTB) and tributyltin (TBT). In most cases, the sensitivity of the organisms towards the toxicants followed the order: S. capricornotum > D. magna > V. fischeri. Toxicity by concentration level had the following order: TBT=Kathon 5287>chlorothalonil>Irgarol 1051>diuron>dichlofluanid>TCMTB for S. capricornotum. For D. magna (48 h test), the toxicity order of compounds was TBT>Kathon 5287>chlorothalonil>TCMTB>dichlofluanid>Irgarol 1051>diuron. For V. fischeri (30 min test), the compound toxicity had the following order: Kathon 5287>TBT>TCMTB>dichlofluanid>Irgarol 1051>chlorothalonil.Degradation products of Irgarol 1051 and diuron were also tested. Degradation product of Irgarol 1051 was found to be less toxic to the crustacean and the microalga but more toxic to the bacterium. Degradation products of diuron were less toxic to the microalga in comparison with the bacterium. For mixtures of compound, toxicities were additive in only 33% of the cases and 21% of mixtures were less toxic than expected based on the sum of concentrations of toxicants (antagonistic effect). Synergistic enhancements of toxicity were observed for a majority (46%) of the mixtures.The average reproducibility of the EC50 and LOEC measurements was 27, 24 and 28%, respectively, in the V. fischeri, S. capricornotum and D. magna bioassays. For single compound, the reproducibility of EC50 was better than ±20% for a vast majority of the measurements with the V. fischeri system, thus agreeing closely with the reported reproducibility values for this relatively well-known assay.  相似文献

Flow field-flow fractionation (Fl-FFF) with off-line electrothermal atomic absorption spectrometry (ETAAS) detection was developed and employed for particle size characterization of Ag NPs stabilized by citrate, pectin, and alginate. Citrate stabilized-Ag NPs were prepared from sodium borohydride reduction of silver nitrate. Sodium citrate was used as the capping agent to stabilize Ag NPs and prevent the aggregation process. Pectin stabilized- and alginate stabilized-Ag NPs were prepared from ascorbic acid reduction of silver nitrate. Pectin and alginate were used as the capping agent for pectin stabilized- and alginate stabilized-Ag NPs, respectively. Three types of Ag NPs were characterized by using FlFFF, zeta potentiometer, and TEM technique. The mean particle sizes of Ag NPs as characterized by FlFFF were 9 nm, 19 nm, and 45 nm for citrate stabilized-, pectin stabilized-, and alginate stabilized-Ag NPs, respectively, in deionized water. Further, FlFFF with ETAAS detection was employed to observe the aggregation of Ag NPs of various types in environmental water in the absence and presence of humic acid. Citrate stabilized-Ag NPs underwent aggregation more rapid than the pectin stabilized- and alginate stabilized-Ag NPs as the latter two types were sterically stabilized. Further, humic acid could prolong the stability of Ag NPs in the environment.  相似文献

Acute toxicity of pesticides in water was assessed singly and in mixtures using the responses of the luminescent bacterium Vibrio fischeri (BioTox™), the aquatic invertebrate Daphnia magna (Daphtoxkit™), and the MitoScan™ assay. The latter utilized fragmented mitochondria to enzymatically convert β-nicotinamide adenine dinucleotide (NADH) to its oxidized form, NAD+. The rate of the conversion being sensitive to type and concentration of toxicants. The pesticides tested were Carbofuran (2,3-dihydro-2,2-dimethylbenzofuran-7-yl methylcarbamate), Cyromazine (N-cyclopropyl-1,3,5-triazine-2,4,6-triamine), Fenamiphos (ethyl 4-methylthio-m-tolyl isopropylphosphoramidate), and Formetanate (3-dimethylaminomethyleneiminophenyl methylcarbamate). The toxicity bioassays were characterized in terms of relative sensitivity, reproducibility, range of the linear response, and the ability to reveal synergistic/antagonistic interactions among toxicants. The D. magna assay was the most sensitive and best able to detect toxic interactions of mixtures. Also, unlike the other assays used, the response of the daphnid system was linear over a 10-fold change in pesticide concentration. Relative to the BioTox™, the MitoScan™ was 2- to 11-fold more sensitive for the compounds and mixtures tested. The EC50 reproducibility of all tests was within ±20% coefficient of variation; however, the lowest observable effect concentration (LOEC) were only reproducible to ±35% on average. Cyromazine was the least toxic of the pesticides tested. To test the predictive value of the concept of concentration addition, toxicities of binary and quaternary mixtures of four different pesticides were analyzed. Synergistic/antagonistic responses were most frequently observed in testing with D. magna. Synergistic/antagonistic effects were seen only in 25 and 50% of the cases with the BioTox™ and the MitoScan™ assays, respectively.  相似文献

This article presents recent developments on the use of inorganic nanoparticles (NPs) for environmental remediation in polluted soil, water and gas. The number of publications on these topics has grown exponentially in recent years, especially those focused on wastewater treatment. Among these topics, removal of metals has become the most popular, although some works relate to the use of nanomaterials for the elimination of nutrients (e.g., nitrogen and some persistent organic pollutants). However, this growth has not been accompanied by knowledge about the behavior of NPs once used and released into the environment. In this article, we also comment upon the current situation with respect to NP toxicology (nanotoxicology). A remarkable number of different toxicology tests has been applied to NPs, often making it very difficult to interpret or to generalize the results. We analyze in detail the bioluminescence, Daphnia magna and other tests, and give some preliminary results obtained in our work.  相似文献

Visualization of elemental distributions in thin sections of biological tissue is gaining importance in many disciplines of biological and medical research. Laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) and scanning micro-X-ray fluorescence spectrometry (micro-XRF) are two widely used microanalytical techniques for elemental mapping. This article compares the capabilities of the two techniques for imaging the distribution of selected elements in the model organism Daphnia magna in terms of detection power and spatial resolution. Sections with a thickness of 10 and 20 μm of the fresh water crustacean Daphnia magna were subjected to LA-ICP-MS and micro-XRF analysis. The elemental distributions obtained for Ca, P, S and Zn allow element-to-tissue correlation. LA-ICP-MS and micro-XRF offer similar limits of detection for the elements Ca and P and thus, allow a cross-validation of the imaging results. LA-ICP-MS was particularly sensitive for determining Zn (LOD 20 μg g−1, 15 μm spot size) in Daphnia magna, while the detection power of micro-XRF was insufficient in this context. However, LA-ICP-MS was inadequate for the measurement of the S distributions, which could be better visualized with micro-XRF (LOD 160 μg g−1, 5 s live time). Both techniques are thus complementary in providing an exhaustive chemical profiling of tissue samples.  相似文献

The rapid development of nanotechnology and the related production and application of nanosized materials such as engineered nanoparticles (ENP) inevitably lead to the emission of these products into environmental systems. So far, little is known about the occurrence and the behaviour of ENP in environmental aquatic systems. In this contribution, the influence of natural organic matter (NOM) and ionic strength on the stability and the interactions of silver nanoparticles (n-Ag) in aqueous suspensions was investigated using UV–vis spectroscopy and asymmetrical flow field-flow fractionation (AF4) coupled with UV–vis detection and mass spectrometry (ICP-MS). n-Ag particles were synthesized by chemical reduction of AgNO3 with NaBH4 in the liquid phase at different NOM concentrations. It could be observed that the destabilization effect of increasing ionic strength on n-Ag suspensions was significantly decreased in the presence of NOM, leading to a more stable n-Ag particle suspension. The results indicate that this behaviour is due to the adsorption of NOM molecules onto the surface of n-Ag particles (“coating”) and the resulting steric stabilization of the particle suspension. The application of AF4 coupled with highly sensitive detectors turned out to be a powerful method to follow the aggregation of n-Ag particle suspensions at different physical–chemical conditions and to get meaningful information on their chemical composition and particle size distributions. The method described will also open the door to obtain reliable data on the occurrence and the behaviour of other ENP in environmental aquatic systems.  相似文献

重力场流分离作为最简单的一种场流分离技术,常用于分离微米级颗粒。选择两种不同粒径(20 μ m和6 μ m)的聚苯乙烯(PS)颗粒作为样品,通过改变载液中叠氮化钠浓度、混合表面活性剂的比例及载液流速,利用自行设计生产的重力场流分离(gravitational flow field-flow fractionation, GrFFF)仪器,对颗粒混合样品进行分离,得到了相关谱图与数据,考察了这3种因素对分离效果(保留比(R)、塔板高度(H))的影响。结果表明:20 μ m PS颗粒的R值均大于6 μ m PS颗粒的R值,H值均小于6 μ m颗粒的H值;PS颗粒的R值与H值均随着载液中叠氮化钠浓度的增加而增加;但随着载液流速的增加,R值增加,H值减小。该研究为GrFFF系统的开发及应用提供了重要的参考价值。  相似文献

Characterization of the in vivo behavior of nanomaterials aims to optimize their design, to determine their biological effects, and to validate their application. The characteristics of the model organism Caenorhabditis elegans (C. elegans) advocate this 1 mm long nematode as an ideal living system for the primary screening of engineered nanoparticles in a standard synthetic laboratory. This review describes some practicalities and advantages of working with C. elegans that will be of interest for chemists and materials scientists who would like to enter the “worm” community, anticipates some drawbacks, and offers relevant examples of nanoparticle assessment by using C. elegans.  相似文献

The focus of the study is to compare the antibacterial efficacy of silver nanoparticles (AgNPs) fabricated by exploiting biological (a mangrove plant, Rhizophora apiculata) and chemical means (Glucose). The synthesized nanoparticles were characterised using UV-visible absorption spectrophotometry (UV-vis), Fourier transform Infra-red Spectroscopy (FTIR) and Transmission electron microscopy (TEM). Biologically synthesized silver nanoparticles (BAgNPs) were observed at 423 nm with particle sizes of 19-42 nm. The chemically synthesized silver nanoparticles (CAgNPs) showed a maximum peak at 422 nm with particle sizes of 13-19 nm. An obvious superiority of the antibacterial potency of BAgNPs compared to the CAgNPs as denoted by the zone of inhibition (ZoI) was noted when the nanoparticles were treated against seven different Microbial Type Culture Collection (MTCC) strains. The current study therefore elucidates that the synthesized AgNPs were efficient against the bacterial strains tested.  相似文献

A simple gravitational field-flow fractionation (GrFFF) system was used for size separation of micron sized silica particles coated with hydrous iron oxide (geothite). The amount of iron on the particles was monitored either on-line by reverse-flow injection analysis (r-FIA) with chemiluminescence detection using luminol or off-line by electrothermal atomic absorption spectrophotometry (ETAAS). The combination of GrFFF with reverse FIA or with ETAAS has been demonstrated to be a cost-effective tool for size based iron speciation of particles.  相似文献

A microwave-assisted extraction used to extract trace triorganotin from aquatic organisms and a sensitive analytical method for the determination of ultratrace triorganotin (namely trimethyltin, triethyltin, tripropyltin and tributyltin) with capillary electrophoresis-inductively coupled plasma mass spectrometry were firstly described in this study. The extraction method is simple, effective and can be used to extract trace triorganotin in aquatic organisms within several min. The analytical method has a much lower detection limit of 0.2-0.7 ng Sn/mL for triorganotin compounds, and can be used to determine trace triorganotin in aquatic organisms directly without any derivatization and preconcentration. Using above methods, we have successfully determined trimethyltin, triethyltin, tripropyltin and tributyltin in dried Mya arenaria Linnaeus and Corbicula fluminea within 17 min with a recovery of 93-104% and a RSD (relative standard deviation, n = 6) of 2-5%. Our results showed that dried M. arenaria Linnaeus contained an extremely high tributyltin of 5.1 μg Sn/g dried weight, indicating that it may be a good biomarker for the organotin pollution in ocean.  相似文献

Nickel ferrite nanoparticles have several possible applications as cathode materials for rechargeable batteries, named “lithium-ion” batteries. In this study, NiFe2O4 was prepared by microwave induced thermohydrolysis. The obtained nanoparticles were characterized by scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), BET method, transmission electron microscopy (TEM) and small angle X-ray scattering (SAXS). All the results show that the microwave one-step flash synthesis leads in a very short time to NiFe2O4 nanoparticles with elementary particles size close to 4-5 nm, and high specific surfaces (close to 240 m2/g). Thus, microwave heating appears as an efficient source of energy to produce quickly nanoparticles with complex composition as ferrite.  相似文献

Quantitative structure–activity relationship (QSAR) models for predicting acute toxicity to Daphnia magna are often associated with poor performances, urging the need for improvement to meet REACH requirements. The aim of this study was to evaluate the accuracy, stability and reliability of a previously published QSAR model by means of further external validation and to optimize its performance by means of extension to new data as well as a consensus approach. The previously published model was validated with a large set of new molecules and then compared with ChemProp model, from which most of the validation data were taken. Results showed better performance of the proposed model in terms of accuracy and percentage of molecules outside the applicability domain. The model was re-calibrated on all the available data to confirm the efficacy of the similarity-based approach. The extended dataset was also used to develop a novel model based on the same similarity approach but using binary fingerprints to describe the chemical structures. The fingerprint-based model gave lower regression statistics, but also less unpredicted compounds. Eventually, consensus modelling was successfully used to enhance the accuracy of the predictions and to halve the percentage of molecules outside the applicability domain.  相似文献

The principal objective of this work was to develop and demonstrate a new methodology for silver nanoparticle (AgNP) detection and characterization based on asymmetric-flow field flow fractionation (A4F) coupled on-line to multiple detectors and using stable isotopes of Ag. This analytical approach opens the door to address many relevant scientific challenges concerning the transport and fate of nanomaterials in natural systems. We show that A4F must be optimized in order to effectively fractionate AgNPs and larger colloidal Ag particles. With the optimized method one can accurately determine the size, stability and optical properties of AgNPs and their agglomerates under variable conditions. In this investigation, we couple A4F to optical absorbance (UV–vis spectrometer) and scattering detectors (static and dynamic) and to an inductively coupled plasma mass spectrometer. With this combination of detection modes it is possible to determine the mass isotopic signature of AgNPs as a function of their size and optical properties, providing specificity necessary for tracing and differentiating labeled AgNPs from their naturally occurring or anthropogenic analogs. The methodology was then applied to standard estuarine sediment by doping the suspension with a known quantity of isotopically enriched 109AgNPs stabilized by natural organic matter (standard humic and fulvic acids). The mass signature of the isotopically enriched AgNPs was recorded as a function of the measured particle size. We observed that AgNPs interact with different particulate components of the sediment, and also self-associate to form agglomerates in this model estuarine system. This work should have substantial ramifications for research concerning the environmental and biological fate of AgNPs.  相似文献

We report a low cost selective analytical method based on inner filter effect (IFE) for citrate-silver nanoparticle (cit-AgNP) detection, in which fluorescent amine-derivatized carbon dots (a-CDs) act as the donor and aggregated cit-AgNPs as the energy receptor. Carbon dots (CDs) were chemically modified with ethylenediamine (EDA) moieties via amidic linkage displaying an emission band at 440 nm. The presence of cit-AgNPs produces a remarkably quenching of a-CD fluorescence via IFE, since the free amine groups at CD surface induce the aggregation of cit-AgNPs accompany by a red-shifting of their characteristic plasmon absorption wavelength, which resulted in “turn-on” of the IFE-decreased in CD fluorescence. The proposed method, which involves the use of chelating agents for removal of metal ions interferences, exhibits a good linear correlation for detection of cit-AgNPs from 1.23 × 10−5 to 6.19 × 10−5 mol L−1, with limits of detection (LOD) and quantification (LOQ) of 5.17 × 10−6 and 1.72 × 10−5 mol L−1, respectively. This method demonstrates to be efficient and selective for the determination of cit-AgNPs in complex matrices such as cosmetic creams and reveals many advantages such as low cost, reusability, high sensitivity and non time-consuming compared with other traditional methods.  相似文献

A method based on matrix solid-phase dispersion (MSPD) has been developed for the determination of 12 isoflavones in Trifolium pratense L. Dried leaf samples were blended with C18, placed in small columns and isoflavones extracted with dichloromethane–methanol. Analyses were performed by high performance liquid chromatography with diode array detection (HPLC-DAD) with 2-methoxyflavone as internal standard. Several dispersants, eluents and clean-up steps were tested during the optimization of the process in order to obtain the best selectivity and yields. Mean recoveries ranged from 70% to 119%, with relative standard deviations <18%. The limits of detection were between 0.006 mg/l for biochanin A and 0.108 mg/l for daidzin. The performance of the optimized method in real samples was compared with a conventional method based in solid–liquid extraction (SLE).  相似文献

Microcalorimetry and measurement of culture turbidity using a Bioscreen C Analyzer System were applied to study the toxic effect of phenanthrene on Cunninghamella elegans IM 1785/21Gp spore germination. The results of C. elegans spore incubation in Bioscreen C microbiology reader showed the inhibition of spore germination by 70% (with 25 mg l−1 of phenanthrene) and total inhibition of the fungus growth with a higher content of the xenobiotic (50-100 mg l−1). The microcalorimetric technique showed to be useful for the estimation of metabolic activity of C. elegans spores in growth medium up to xenobiotic concentrations of 90 mg l−1. These data corresponded with the microscopic observations. The obtained results showed that the microcalorimetry method could be a valuable supplement in the study on the mechanism of PAHs detoxification by fungi.  相似文献

The application of nanoscale materials and structures, usually ranging from 1 to 100 nanometers (nm), is an emerging area of nanoscience and nanotechnology. Nanomaterials may provide solutions to technological and environmental challenges in the areas of solar energy conversion, catalysis, medicine, and water-treatment. The development of techniques for the controlled synthesis of nanoparticles of well-defined size, shape and composition, to be used in the biomedical field and areas such as optics and electronics, has become a big challenge. Development of reliable and eco-friendly processes for synthesis of metallic nanoparticles is an important step in the field of application of nanotechnology. One of the options to achieve this objective is to use ‘natural factories’ such as biological systems. This study reports the optimal conditions for maximum synthesis of silver nanoparticles (AgNPs) through reduction of Ag+ ions by the culture supernatant of Escherichia coli. The synthesized silver nanoparticles were purified by using sucrose density gradient centrifugation. The purified sample was further characterized by UV–vis spectra, fluorescence spectroscopy and TEM. The purified solution yielded the maximum absorbance peak at 420 nm and the TEM characterization showed a uniform distribution of nanoparticles, with an average size of 50 nm. X-ray diffraction (XRD) spectrum of the silver nanoparticles exhibited 2θ values corresponding to the silver nanocrystal. The size-distribution of nanoparticles was determined using a particle-size analyzer and the average particle size was found to be 50 nm. This study also demonstrates that particle size could be controlled by varying the parameters such as temperature, pH and concentration of AgNO3.  相似文献
