A simple and effective procedure for the enantioselective synthesis of (R)- and (S)-moprolol was described. The key step was the asymmetric synthesis of enantiopure (R)- and (S)-guaifenesin, which were synthesized from enantioenriched (R)-3-chloro-l,2-propanediol and (S)-epichlorohydrin via kinetics of hydrolysis resolution of racemic epichlorohydrin by chiral Salen-Co^Ⅲ complex. The e.e. values of both the optical compounds were above 98%, and the chemical structures of the target compounds were confirmed by ^1H NMR, ^13C NMR, IR, and MS.  相似文献

The title compound,(E)-ethyl 2-(5-(3-methyl-2-butenyloxy)-2-(3-(4-(3-methyl-2-butenyloxy)phenyl) acryloyl) phenoxy)acetate(1),has been synthesized and characterized by FT-IR and 1H-NMR spectroscopy,ESI-MS,and X-ray single-crystal diffraction.FT-IR,1H-NMR and ESI-MS confirm the functional groups,particularly the ethyl groups in the ester moiety,of the compound.The single-crystal X-ray diffraction has revealed a monoclinic structure,space group P21/n with a = 14.6832(14),b = 7.7581(7),c = 23.075(2),β = 101.670(2)o,V = 2574.2(4) ?3,Z = 4,Dc = 1.235 g/cm,μ = 0.085 mm-1,and F(000) = 1024.The skeleton of chalcone in the molecular structure is coplanar.  相似文献

The facile enantioselective synthesis of the di-O-methyl ethers of (-)-agatharesinol (1b), (-)-sugiresionl (2b), (+)-nyasol (3b) and (+)-tetrahydronyasol (4) were achieved in high yield. The absolute configuration of (+)-3a was confirmed via first total synthesis of (+)-3b and (+)-4.  相似文献

A novel molecule tetra-N-alkylation of cyclen (1,4,7,10-tetraazacyclododecane), 1,4,7,10-tetrakis(2-((4-hydroxy)phenoxy)ethyl)-1,4,7,10-tetraazacyclododecane 2, was synthesized and structurally characterized by single-crystal X-ray diffraction. The molecule turned into chiral helical compound crystals grown from EtOH by slow diffusion at room temperature and three of the four hydroquinone groups of the benzene ring formed a g-electron-rich cavity by C-H…π stacking interaction. The crystal belongs to the monoclinic system, space group P21/C with a = 13.9192(9), b = 13.2871(6), c = 22.1894(15)A^°, β = 91.4600(10)°, V = 4102.5(4)A^°3, Z = 4, Dc = 1.219 g/cm^3, C40H52N4O8, Mr = 752.89, F(000) = 1616,μ = 0.088 mm^-1, MoKa radiation (λ = 0.71073), R = 0.0578 and wR = 0.1389 for 5588 observed reflections with I 〉 2σ(I). Moreover, compound 2 was characterized with ^1H NMR, ^13C NMR, IR spectra and MS.  相似文献

The title compound (Z)-1-(3-fluorobenzyl)-5-((3-fluorobenzylimino)(2-hydroxy-4- methoxyphenyl)methyl)pyridin-2(1H)-one (C27H22F2N2O3,Mr = 460.47) was synthesized and crystallized. The crystal belongs to the triclinic system,space group P1 with a = 9.951(6),b = 10.059(6),c = 12.927(7)A,α = 109.828(7),β = 102.304(7),γ = 104.898(7)°,Z = 2,V = 1110.2(11)A^3,Dc =1.377 Mg/m^3,μ(MoKα) = 0.102 mm^-1,F(000) = 480,the final R = 0.0449 and Rw = 0.1250 for 4595 observed reflections (I 〉 2σ(I)). X-ray analysis reveals that both m-fluorobenzyl groups are diorientationally disordered because of the rotation of C–C single bond which was represented as F(1),F(2) and F(1'),F(2'). The diorientational disorder was refined and gave the occupancies of 0.665(4) and 0.335(4) for F(1) and F(1'),0.631(3) and 0.369(3) for F(2) and F(2'). Hydrogen bonds existing in the cell link different components to stabilize the crystal structure. The active data proved that the title compound has good anti-HBV activity.  相似文献

考察了利用安大略假丝酵母(Candida ontarioensis)静息细胞不对称催化2-氯-1-(3-氯苯基)乙酮合成(R)-2-氯-1-(3-氯苯基)乙醇的转化反应条件.结果表明,当底物浓度为10g/L时,在最适转化条件下反应72h,产物的ee值和产率分别达到99.9%和99.0%.采用4g/L十六烷基三甲基溴化铵对Candida ontarioensis细胞于4℃通透性处理20min后,全细胞的酶活提高2倍以上.当底物浓度提高为30g/L,转化24h后,产物的ee和产率分别达到99.9%和97.5%.该研究为高效制备(R)-2-氯-1-(3-氯苯基)乙醇提供了可行途径,并为生物催化合成芳基手性醇类手性中间体提供了理论指导。  相似文献

A total of 82 fungal isolates were screened for their ability to reduce 3,5-bis(trifluoromethyl)-acetophenone 1. Penicillium expansum was found to successfully reduce ketone 1 in the submerged culture. A second screening was performed on the active P. expansum strains identified by the first screening. A number of strains of P. expansum were found to produce (R)-3,5-bistrifluoromethylphenyl ethanol 2 with over 99% enantiomeric excess (ee) from ketone 1. The most productive strain was identified as P. expansum EBK-9, and this strain was selected for further experiments. The total production of 2 was carried out by P. expansum EBK-9 in a laboratory-scale bioreactor employing optimized conditions as determined by our experiments. P. expansum EBK-9 gave 2 with ee >99% and 76% yield. On a large scale, a total of 3.35 g/L of 2 was produced from 1 after 56 h.  相似文献

首次成功实现了光学纯(R)-1-(2-萘基)乙胺的高效酶法动力学拆分制备,考察了脂肪酶种类、溶剂、酰基供体、底物浓度、反应温度等对拆分效果的影响,发现新型酰基供体——正戊酸对氯苯酯能够很好地抑制非酶促自催化酰胺化效应.在甲苯溶剂中,底物浓度300 mmol/L,40℃条件下,采用该供体在脂肪酶Novozym 435催化下,动力学拆分反应8 h转化率达到理论最佳值50%,eep>99%.  相似文献

以红酵母静息细胞催化苯乙酮不对称还原合成(S)-α-苯乙醇为模型反应, 研究了在反应体系中加入吸附树脂对反应的影响. 结果表明, 水相中较高浓度(>50 mmol/L)的苯乙酮对红酵母细胞具有显著毒性; 加入适量的吸附树脂可以降低水相中苯乙酮的浓度从而减小对细胞的毒害作用. 考察了3种不同极性的吸附树脂(HZ-801, HZ-806和HZ-816)对底物和产物的吸附能力, 发现在3种树脂中疏水性最强的HZ-816树脂对反应底物和产物具有相对较高的吸附能力. 进一步研究结果表明, 当底物浓度为100 mmol/L, 细胞湿重为500 g/L时, 与不加树脂的对照实验相比, 加入100 g/L HZ-816树脂后, 使得48 h的反应转化率从54%提高到91%, 与此同时细胞的死亡率从99%下降为10%. 含有产物的树脂经乙醇解吸后即可循环使用.  相似文献

An α-diimine-stabilized Al-Al-bonded compound [L(2-)Al(II)-Al(II)L(2-)] (L = [{(2,6-iPr(2)C(6)H(3))NC(Me)}(2)]; 1) consists of dianionic α-diimine ligands and sub-valent Al(2+) ions and thus could potentially behave as a multielectron reductant. The reactions of compound 1 with azo-compounds afforded phenylimido-bridged products [L(-)Al(III)(μ(2)-NPh)(μ(2)-NAr)Al(III)L(-)] (2-4). During the reaction, the dianionic ligands and Al(2+) ions were oxidized into monoanions and Al(3+), respectively, whilst the [NAr](2-) imides were produced by the four-electron reductive cleavage of the N=N double bond. Upon further reduction by Na, the monoanionic ligands in compound 2 were reduced to the dianion to give [(L(2-))(2)Al(III)(2)(μ(2)-NPh)(2)Na(2)(thf)(4)] (5). Interestingly, when asymmetric azo-compounds were used, the asymmetric adducts were isolated as the only products (compounds 3 and 4). DFT calculations indicated that the reaction was quite feasible in the singlet electronic state, but the final product with the triplet-state monoanionic ligands could result from an exothermic singlet-to-triplet conversion during the reaction process.  相似文献

3-(4-Chlorophenyl)-1-phenylprop-2-en-1-one reacted with 3-amino-N-phenyl-3-thioxopropanamide under basic conditions in ethanol to give a mixture of 3-(4-chlorophenyl)-2-cyano-5-oxo-N,5-diphenylpentanamide and 3-(4-chlorophenyl)-1,5-diphenylpentane-1,5-dione. The structure of the former was determined by X-ray analysis. The reaction of the same compounds in DMSO in the presence of sodium hydroxide produced 4-(4-chlorophenyl)-N,6-diphenyl-2-thioxo-1,2,3,4-tetrahydropyridine-3-carboxamide.  相似文献

Lactobacillus kefir was used as the whole cell biocatalyst for the asymmetric reduction of ethyl 4-chloro acetoacetate 1 to the chiral synthon ethyl (S)-4-chloro-3-hydroxybutanoate 2. Ketoester 1 was obtained as micro-droplets, without the use of an organic solvent as substrate reservoir. 2 (1.2 M) was produced using 2-propanol as co-substrate with a final yield of 97% within 14 h. A high space-time yield and a high specific product capacity of 85.7 mmol/L h and of 24 mmol/gDCW were measured. The enantiomeric excess of the (S)-alcohol 2 was 99.5%.  相似文献

 采用近平滑假丝酵母细胞用于催化乙酰基三甲基硅烷不对称还原反应, 可高选择性地生成 (R)-1-三甲基硅乙醇. 结果表明, 固定化于海藻酸钙的细胞催化该反应的产物收率比游离细胞的高. 不同辅底物对该反应的影响显著, 以葡萄糖为辅底物时, 反应的初速率较快, 产物收率较高. 该反应的最适条件为: 辅底物 (葡萄糖) 浓度 110 mmol/L, 振荡速度 180 r/min, 缓冲液 pH 值 6.0, 反应温度 30 oC, 底物浓度 20 mmol/L. 在此反应条件下反应的初速率、产物收率和产物的 ee 值分别为 11.4 μmol/h, 96.5% 和 99.9%.  相似文献

(S)-1-(2,6-二氯-3-氟苯基)乙醇(2)是合成抗癌药物克唑替尼的关键手性前体。本文以1-(2,6-二氯-3-氟苯基)乙酮为起始原料,利用二异松莰基氯化硼[(-)-Ipc2BCl]不对称还原制得光学纯的2;并将中间体2经Mitsunobu反应、还原、溴代、 Suzuki偶联及脱除Boc保护合成克唑替尼,其结构1H NMR,13C NMR和HR-MS(ESI)确证。对关键中间体2的合成条件进行了优化,并其对反应机理进行了推测。  相似文献

构建了羰基还原酶CR2重组酶体系,并优化了相关的酶促催化反应条件.通过在催化体系中添加辅酶NADP+(0.1 mmol/L)和辅底物葡萄糖(120 g/L),在30℃及p H=8.0的条件下反应4 h,CR2重组酶体系不对称还原N,N-二甲基-3-酮-3-(2-噻吩)-1-丙胺(DKTP,10 g/L),合成了高光学纯度(S)-N,N-二甲基-3-羟基-3-(2-噻吩)-1-丙胺[(S)-DHTP,e.e.值99.9%],产率为62%.在酶促催化过程中,由于辅酶循环生成葡萄糖酸导致反应体系p H值下降而影响催化效率.通过调控反应体系p H值,(S)-DHTP的产率提高到68%.不同浓度底物的反应过程表明底物对CR2酶促反应具有抑制作用,且在10 g/L底物浓度下反应的时空产率可达1.3 g·L-1·h-1.  相似文献
