Prussian blue nanowire array (PBNWA) was prepared via electrochemical deposition with polycarbonate membrane template for effective modification of glassy carbon electrode. The PBNWA electrode thus obtained was demonstrated to have high-catalytic activity for the electrochemical reduction of hydrogen peroxide in neutral media. This enabled the PBNWA electrode to show rapid response to H2O2 at a low potential of −0.1 V over a wide range of concentrations from 1 × 10−7 M to 5 × 10−2 M with a high sensitivity of 183 μA mM−1 cm−2. Such a low-working potential also substantially improved the selectivity of the PBNWA electrode against most electroactive species such as ascorbic acid and uric acid in physiological media. A detection limit of 5 × 10−8 M was obtained using the PBNWA electrode for H2O2, which compared favorably with most electroanalysis procedures for H2O2. A biosensor toward glucose was then constructed with the PBNWA electrode as the basic electrode by crosslinking glucose oxidase (GOx). The glucose biosensor allowed rapid, selective and sensitive determination of glucose at −0.1 V. The amperometric response exhibited a linear correlation to glucose concentration through an expanded range from 2 × 10−6 M to 1 × 10−2 M, and the response time and detection limit were determined to be 3 s and 1 μM, respectively.  相似文献

The construction of amperometric enzymeless biosensors for phenolic compounds determination, using carbon paste electrode modified with copper phtalocyanine (CuPc) and histidine (His), based on the chemistry of the dopamine β-monooxygenase (DβM) enzyme that catalyzes the hydroxylation of the dopamine and its analogs is shown. The modified carbon paste was evaluated on electrodes constructed in two ways: putting the paste into a cavity of a rotating disk electrode and a platinum slide electrode fixed into a glass tube. The sensor in hydrodynamic conditions presented a linear response range between 30 and 250 μmol l−1, with a sensitivity of 4.6±0.1 nA l μmol−1 cm−2 for catechol, response time of 3 s and lifetime of about 50 days when stored at room temperature. The sensor in static conditions showed a linear response range from 40 to 250 μmol l−1, with a sensitivity of 0.30±0.01 nA l μmol−1 cm−2 for catechol. The sensors presented the following relative response order for dopamine and some analog species: catechol>dopamine>guaiacol>serotonin>phenol.  相似文献

A chemically modified carbon paste electrode with 3,4-tetra pyridinoporphirazinatocobalt(II) (Co(3,4 tppa) was applied to the determination of free cyanide ion. The electrode has a linear range between 1.5 × 10−5 M and 1.0 × 10−2 M with a Nernstian slope of 60 ± 1.5 mV/decade and its detection limit is 9 × 10−6 M. The response time of electrode is 5 min. The proposed electrode was applied successfully for the determination of cyanide in commercially available spring water. Some anions, such as SCN, I, Cl, Br and oxalate that are usually serious interfering species for most of cyanide selective electrodes, did not have any interfering effect for this proposed electrode.  相似文献

A novel and sensitive electrochemical DNA biosensor based on nanoparticles ZrO2 and multi-walled carbon nanotubes (MWNTs) for DNA immobilization and enhanced hybridization detection is described. The MWNTs/nano ZrO2/chitosan-modified glassy carbon electrode (GCE) was fabricated and oligonucleotides were immobilized to the GCE. The hybridization reaction on the electrode was monitored by differential pulse voltammetry (DPV) analysis using electroactive daunomycin as an indicator. Compared with previous DNA sensors with oligonucleotides directly incorporated on carbon electrodes, this carbon nanotube-based assay with its large surface area and good charge-transport characteristics increased DNA attachment quantity and complementary DNA detection sensitivity. The response signal increases linearly with the increase of the logarithm of the target DNA concentration in the range of 1.49 × 10−10 to 9.32 × 10−8 mol L−1 with the detection limit of 7.5 × 10−11 mol L−1 (S/N = 3). The linear regression equation is I = 32.62 + 3.037 log CDNA (mol L−1) with a correlation coefficient value of 0.9842. This is the first application of carbon nanotubes combined with nano ZrO2 to the fabrication of an electrochemical DNA biosensor with a favorable performance for the rapid detection of specific hybridization.  相似文献

A glucose biosensor using an enzyme-immobilized eggshell membrane and oxygen electrode for glucose determination has been fabricated. Glucose oxidase was covalently immobilized on an eggshell membrane with glutaraldehyde as a cross-linking agent. The glucose biosensor was fabricated by positioning the enzyme-immobilized eggshell membrane on the surface of a dissolved oxygen sensor. The detection scheme was based on the depletion of dissolved oxygen content upon exposure to glucose solution and the decrease in the oxygen level was monitored and related to the glucose concentration. The effect of glutaraldehyde concentration, pH, phosphate buffer concentration and temperature on the response of the glucose biosensor has been studied in detail. Common matrix interferents such as ethanol, d-fructose, citric acid, sodium benzoate, sucrose and l-ascorbic acid did not give significant interference. The resulting sensor exhibited a fast response (100 s), high sensitivity (8.3409 mg L−1 oxygen depletion/mmol L−1 glucose) and good storage stability (85.2% of its initial sensitivity after 4 months). The linear response is 1.0×10−5 to 1.3×10−3 mol L−1 glucose. The glucose content in real samples such as commercial glucose injection preparations and wines was determined, and the results were comparable to the values obtained from a commercial glucose assay kit based on a spectrophotometric method.  相似文献

Biosensors for malic acid and glucose have been developed, using screen-printed electrodes and two different classes of enzymes: NAD(P)+-dependent dehydrogenases and oxidases. The active surface of the electrodes was modified using Meldola Blue (malic acid) and Prussian Blue (glucose) and in this way sensitive, low cost and reliable NAD(P)H and H2O2 probes were obtained. Fixed potential amperometry was used for the detection of substrates in small volumes of sample (50 μl). Immobilization of the enzymes in a polyethylenimine-glutaraldehyde cross-linking membrane allowed sensors to be obtained with sufficient operational stability. The detection limits were of 10−5 M for malic acid and 10−6 M for glucose. The sensors were applied in the analysis of different samples of wine.  相似文献

An amperometric biosensor was prepared by in situ deposition of horseradish peroxidase (HRP) enzyme on a polyaniline (PANI)-doped platinum disk electrode. The PANI film was electrochemically deposited on the electrode at 100 mV s−1/Ag-AgCl. Cyclic voltammetric characterization of the PANI film in 1 M HCl showed two distinct redox peaks, which prove that the PANI film was electroactive and exhibited fast reversible electrochemistry. The surface concentration and film thickness of the adsorbed electroactive species was estimated to be 1.85×10−7 mol cm−2 and approximately 16 nm, respectively. HRP was electrostatically immobilized onto the surface of the PANI film, and voltammetry was used to monitor the electrocatalytic reduction of hydrogen peroxide under diffusion-controlled conditions. Linear responses over the concentration range 2.5×10−4 to 5×10−3 M were observed. Spectroelectrochemistry was used to monitor the changes in UV-vis properties of HRP, before and after the catalysis of H2O2. The biosensor surface morphology was characterized by scanning electron microscopy (SEM) using PANI-doped screen-printed carbon electrodes (SPCEs) in the presence and absence of (i) peroxidase and (ii) peroxide. The SEM images showed clear modifications of the conducting film surface structure when doped with HRP, as well as the effect of hydrogen peroxide on the morphology of biosensor.  相似文献

An amperometric nicotine inhibition biosensor has been substantially simplified and used for determination of nicotine in tobacco sample. Besides the use of single enzyme choline oxidase to replace bienzyme, the use of 1,4-benzoquinone as an electron mediator makes it possible to avoid the use of oxygen or hydrogen peroxide sensor as the internal transducer. Choline oxidase was immobilized on the carbon paste electrode through cross-linking with bovine serum albumin (BSA) by glutaraldehyde. In the presence of choline oxidase and its endogenous cofactor flavin-ademine dinneleotide (FAD), choline was oxidized into betaine while FAD was reduced to FADH2 which subsequently reduced 1,4-benzoquinone into hydroquinone. The later was finally oxidized at a relatively low potential of +450 mV versus saturated calomel electrode (SCE). Nicotine inhibits the activity of enzyme with an effect of decreasing of oxidation current. The experimental conditions were optimized. The electrode has a linear response to choline within 1.25×10−4 to 1.25×10−3 mol l−1. The nicotine measurements were carried out in 0.067 mol l−1phosphate buffer of pH 7.4 at an applied potential of 450 mV versus SCE. The electrode provided a linear response to nicotine over a concentration range of 2.0×10−5 to 9.2×10−4 mol l−1 with a detection limit of 1.0×10−5 mol l−1. The system was applied to the determination of nicotine in tobacco samples.  相似文献

The feasibility of a newly synthesized Rh(III) complex, Rh[(trpy)(bpy)Cl](PF6)2, as a novel ionophore for the preparation of anion-selective polymeric membrane electrodes was tested. The ionophore exhibited anti-Hofmeister behavior with enhanced potentiometric selectivity toward thiocyanate ion compared to other anions. The influence of some experimental parameters such as membrane composition, nature and amount of plasticizer and additive and concentration of internal solution on the potential response of the SCN sensor were investigated. The electrode exhibits a Nernstian response for SCN over a wide concentration range (1.0 × 10−5 to 1.0 × 10−1 M) with a slope −58.7 ± 0.5 mV per decade and a detection limit of 4.0 × 10−6 M (0.23 ppm). It could be used in a pH range of 3.0-8.0 and has a fast response time of about 15 s. The proposed sensor was used for the determination of thiocyanate ions in real samples such as urine and saliva of smokers and nonsmokers and, as an indicator electrode, in potentiometric titrations of SCN ion.  相似文献

The aim of this work was the synthesis and characterization of new modified sol-gel carbon composite electrodes and their application to the determination of trace mercury species with positive charge. Two types of modified electrodes were synthesized, sol-gel and sol-gel-PVSA carbon composite electrodes. In the last ones, poly(vinylsulfonic acid) (PVSA) was used as a functional polymer entrapped within the sol-gel material due to its cationic exchange properties. In a first stage, parameters affecting both, the sol-gel process and the electrode preparation were optimized. In a second stage, usefulness of developed electrodes applied to the determination of cationic mercury species was evaluated, optimizing the activation, preconcentration, measurement and regeneration steps. Developed electrodes showed very favourable electroanalytical properties for their use as amperometric sensors, such as small size, low cost, simple fabrication and handling, renewability and reusability. By means of an easy and low-cost methodology, satisfactory experimental results were obtained in Hg2+ determination. In this sense, developed analytical methodology showed adequate response times, linear concentration range up to three orders of magnitude (from 5.0 × 10−8 to 5.0 × 10−5 M) and detection limits of 1.5 × 10−8 M (3.0 μg L−1). These results suggest that the incorporation of different receptor molecules at the sol-gel carbon composite material in combination with a selected electrochemical reaction could improve the detection limit achieved and obtain electrochemical sensors adapted to the determination of different species of mercury and other heavy metals.  相似文献

Simple and sensitive DNA sensors have been developed on a base on graphite screen-printed electrodes modified with DNA and enzymes. Cholinesterase and peroxidase immobilized by treatment with glutaraldehyde were used for the detection of human DNA antibodies of systemic lupus erythematosus and bronchial asthma patients. The amperometric signal was measured at +680 mV versus Ag/AgCl for DNA-cholinesterase sensor and −150 mV for DNA-peroxidase sensor 5 min after the injection of acethylthiocholine and hydroquinone, respectively. The addition of serum samples results in the sharp decrease of the signal due to the formation of DNA-antibody adducts followed by the suppression of the access of substrate to the enzyme active site. Sulfonamide medicines suppress the DNA-antibody interaction due to the competitive binding along DNA minor grooves. DNA sensor labeled with peroxidase showed the linear calibration range of 5×10−9 to 7×10−5 mol l−1 of sulfamethoxazole and of 5×10−8 to 1×10−4 mol l−1 of sulfathiazole.  相似文献

A new biosensor for detection of phenols, based on tyrosinase immobilization with alumina sol-gel on Sonogel-Carbon transducer, has been developed. The electrode was prepared using high energy ultrasounds directly applied to the precursors. The alumina sol-gel provided a microenvironment for retaining the native structure and activity of the entrapped enzyme and a very low mass transport barrier to the enzyme substrates. Phenols are oxidized by tyrosinase biosensor to form a detectable product, which was determined at −300 mV vs. Ag/AgCl reference electrode. For phenol, the sensor exhibited a fast response which resulted from the porous structure and high enzyme loading of the sol-gel matrix. The linear range was from 5 × 10−7 M to 3 × 10−5 M, with a detection limit of 3 × 10−7 M. The stability of the biosensor was also evaluated.  相似文献

Comparative studies of the potentiometric behavior of three mercapto compounds [2-((5-mercapto-1,3,4-thiadiazol-2-ylimino)methyl)phenol] (MTMP), [5-(2-methoxy benzylidene amino)-1,3,4-thiadiazole-2-thiol] (MBYT) and [5-(pyridin-2-ylmethyleneamino)-1,3,4-thiadiazole-2-thiol] (PYTT) self-assembled on gold nanoparticles (GNPs) as ionophores in carbon paste electrodes (CPEs) have been made. These mercapto thiadiazole compounds were self-assembled onto gold nanoparticles and then incorporated within carbon paste electrode. The self-assembled ionophores exhibit a high selectivity for copper ion (Cu2+), in which the sulfur and nitrogen atoms in their structure play a role as the effective coordination donor site for the copper ion. These carbon paste electrodes were applied as indicator electrodes for potentiometric determination of copper ions. The sensor based on PYTT exhibits the working concentration range of 4.0 × 10−9 to 7.0 × 10−2 M and a Nernstian slope of 28.7 ± 0.3 mV decade−1 of copper activity. The detection limit of electrode was 1.0 × 10−9 M and potential response was pH independent across the range of 3.0-6.5. It exhibited a quick response time of <5 s and could be used for a period of 45 days. The ion selectivity of this electrode for Cu2+ was over 104 times that for other metal cations. The application of prepared sensors has been demonstrated for the determination of copper ions in spiked water and natural water samples.  相似文献

In this paper, a polydopamine (PDA) film is electropolymerized on the surface of bilayer lipid membrane (BLM) which is immobilized with horseradish peroxidase (HRP). The coverage of the PDA film on HRP/BLM electrode is monitored by electrochemical impedance spectroscopy (EIS). The electrocatalytic reduction of H2O2 at the PDA/HRP/BLM electrode is studied by means of cyclic voltammetry (CV). The biosensor has a fast response to H2O2 of less than 5 s and an excellent linear relationship is obtained in the concentration range from 2.5 × 10−7 to 3.1 × 10−3 mol L−1, with a detection limit of 1.0 × 10−7 mol L−1 (S/N = 3). The response current of BLM/HRP/PDA biosensor retains 84% of its original response after being stored in 0.1 mol L−1 pH 7.0 PBS at 4 °C for 3 weeks. The selectivity, repeatability, and storage stability of PDA/HRP/BLM biosensor are greatly enhanced by the coverage of polydopamine film on BLM.  相似文献

A Prussian Blue (PB)/polyaniline (PANI)/multi-walled carbon nanotubes (MWNTs) composite film was fabricated by step-by-step electrodeposition on glassy carbon electrode (GCE). The electrode prepared exhibits enhanced electrocatalytic behavior and good stability for detection of H2O2 at an applied potential of 0.0 V. The effects of MWNTs thickness, electrodeposition time of PANI and rotating rate on the current response of the composite modified electrode toward H2O2 were optimized to obtain the maximal sensitivity. A linear range from 8 × 10−9 to 5 × 10−6 M for H2O2 detection has been observed at the PB/PANI/MWNTs modified GCE with a correlation coefficient of 0.997. The detection limit is 5 × 10−9 M on signal-to-noise ratio of 3. To the best of our knowledge, this is the lowest detection limit for H2O2 detection. The electrode also shows high sensitivity (526.43 μA μM−1 cm−2) for H2O2 detection which is more than three orders of magnitude higher than the reported.  相似文献

A reagentless signal-on electrochemiluminescence (ECL) biosensor for DNA hybridization detection was developed based on the quenching effect of ferrocene (Fc) on intrinsic cathodic ECL at thin oxide covered glassy carbon (C/CxO1−x) electrodes. To construct the DNA biosensor, molecular beacon (MB) modified with ferrocene (3′-Fc) was attached to a C/CxO1−x electrode via the covalent bound between labeled amino (5′-NH2) and surface functional groups. It was found that the immobilization of the probe on the electrode surface mainly depended on the fraction of surface carbonyl moiety. When a complementary target DNA (cDNA) was present, the stem-loop of MB on the electrode was converted into a linear double-helix configuration due to hybridization, resulting in the moving away of Fc from the electrode surface, and the restoring of the cathodic ECL signal. The restoration of the ECL intensity was linearly changed with the logarithm of cDNA concentration in the range of 1.0 × 10−11 to 7.0 × 10−8 M, and the detection limit was ca. 5.0 pM (S/N = 3). Additionally, single-base mismatched DNA can be effectively discriminated from the cDNA. The great advantage of the biosensor lies in its simplicity and cost-effective with ECL generated from the electrode itself, and no adscititious luminophore is required.  相似文献

A novel amperometric biosensor utilizing two enzymes, glucose oxidase (GOD) and horseradish peroxidase (HRP), was developed for the cathodic detection of glucose. The glucose biosensor was constructed by electrochemical formation of a polypyrrole (PPy) membrane in the presence of GOD on the surface of a HRP-modified sol-gel derived-mediated ceramic carbon electrode. Ferrocenecarboxylic acid (FCA) was used as mediator to transfer electron between enzyme and electrode. In the hetero-bilayer configuration of electrode, all enzymes were well immobilized in electrode matrices and showed favorable enzymatic activities. The amperometric detection of glucose was carried out at +0.16 V (versus saturated calomel reference electrode (SCE)) in 0.1 M phosphate buffer solution (pH 6.9) with a linear response range between 8.0×10−5 and 1.3×10−3 M glucose. The biosensor showed a good suppression of interference in the amperometric detection.  相似文献

A new dodecyl benzene sulfonate (DBS) ion-selective electrode based on polyaniline is reported. The films of polyaniline doped with DBS were prepared electrochemically on platinum electrodes in the solution containing 1.0×10−3 M aniline and 7.0×10−3 M DBS. The optimum potentiometric response was obtained for prepared polymeric film by passing electricity of 7.5 C cm−2. The electrode exhibits an excellent Nernstian slope of −59.1±0.3 mV per decade for DBS ion over a wide concentration range (5.0×10−6 to 4.1×10−3 M) with a low detection limit (1.0×10−6 M). The proposed electrode revealed good sensitivities for DBS ion over a wide variety of other anions and can be used in the wide pH range of 5-10. It shows good stability, good reproducibility, wide range of pH independency and fast response (<20 s) without using internal solution. This electrode could be used for the determination of DBS in the real samples.  相似文献

This paper describes a potentiometric method for determination of l-histidine (l-his) in aqueous media, using a carbon paste electrode modified with tetra-3,4-pyridinoporphirazinatocopper(II) (Cu (3,4tppa)). The electrode exhibits linear response to the logarithm of the concentration of l-histidine from 2.4 × 10−5 to 1.0 × 10−2 M, with a response slope of −49.5 ± 1 mV and response time of about 1.5 min. The detection limit according to IUPAC recommendation was 2.0 × 10−5 M. The proposed electrode shows a good selectivity for l-his over a wide variety of anions. This chemically modified carbon paste electrode was successfully used for the determination of l-his in a synthetic serum and RANDOX control serum solutions.  相似文献

A new amperometric biosensor for hydrogen peroxide was developed based on cross-linking horseradish peroxidase (HRP) by glutaraldehyde with multiwall carbon nanotubes/chitosan (MWNTs/chitosan) composite film coated on a glassy carbon electrode. MWNTs were firstly dissolved in a chitosan solution. Then the morphology of MWNTs/chitosan composite film was characterized by field-emission scanning electron microscopy. The results showed that MWNTs were well soluble in chitosan and robust films could be formed on the surface. HRP was cross-linked by glutaraldehyde with MWNTs/chitosan film to prepare a hydrogen peroxide biosensor. The enzyme electrode exhibited excellent electrocatalytic activity and rapid response for H2O2 in the absence of a mediator. The linear range of detection towards H2O2 (applied potential: −0.2 V) was from 1.67 × 10−5 to 7.40 × 10−4 M with correction coefficient of 0.998. The biosensor had good repeatability and stability for the determination of H2O2. There were no interferences from ascorbic acid, glucose, citrate acid and lactic acid.  相似文献
