In this study, a flow injection analysis (FIA) system using a cartridge of immobilized isocitrate lyase (ICL) and isocitrate dehydrogenase (ICDH) was developed to monitor the concentrations of succinic acid in biotechnological processes. The ICL and ICDH immobilized on VA-Epoxy Biosynth E3-carrier had a good operational lifetime (up to 24 h) and storage stability (up to 30 days). The FIA system with the immobilized ICL/ICDH cartridge was characterized with respect to the factors affecting the activity of the immobilized enzymes, such as pH of carrier solution, temperature, sample matrix, etc. Optimal pH value of the immobilized enzymes was slightly shifted in the alkaline range, i.e. 9.0. Some components such as 10 g l−1 lactose, 3 g l−1 malate and 3 g l−1 oxaloacetate in sample solution had significant activating effects (more than 10%) on the response of the FIA system. But the activity of the immobilized ICL and ICDH was not largely influenced by some components like imidazole (1 mM), sodium azide (10 mM) and semicarbazide (2 g l−1) added to carrier buffer solution. The FIA system with an enzyme cartridge was applied to on-line monitor the concentrations of succinic acid in a continuously stirred reactor and a fermentation process of immobilized Escherichia coli, and showed good sensitivity and reliability of the FIA system developed in this work.  相似文献

A flow injection on-line sorption preconcentration system has been synchronously coupled to an electrothermal atomic absorption spectrometry (ETAAS) system for the selective determination of trace amounts of Sb(III) in water, soil and plant. The determination was achieved by selective complexation and sorption of Sb(III) with [1,5-bis(2-pyridyl)-3-sulphophenyl methylene thiocarbonohydarzide (PSTH) immobilized on an anion-exchange resin (Dowex 1× 8-200)] at a wide range of pH, quantitative elution with 50 μl of 2 M HNO3 and subsequent ETAAS detection. ETAAS determination of the analyte was performed in parallel with the preconcentration of the next sample. Using a preconcentration time of 60 s and a sample loading flow rate of 2.8 ml min−1, an enhancement factor of 12 was obtained in comparison with direct injection of 50 μl aqueous solution, resulting in a sampling frequency of 31 samples h−1. The detection limit (3 s) was 2 μg l−1 and the precision was 3.1% (R.S.D.) for 11 replicate determinations at 10 μg l−1. The accuracy of the proposed method was demonstrated by analyzing one certified sample and different spiked samples.  相似文献

A flow injection analysis (FIA) method using on-line separation and preconcentration with a novel metal scavenger beads, QuadraSil™ TA, has been developed for the ICP-OES determination of traces of palladium. QuadraSil TA contains diethylenetriamine as a functional group on spherical silica beads and shows the highest selectivity for Pd(II) at pH 1 (0.1 mol l−1 hydrochloric acid) solution. An aliquot of the sample solution prepared as 0.1 mol l−1 in hydrochloric acid was passed through the QuadraSil TA column. After washing the column with the carrier solution, the Pd(II) retained on the column was eluted with 0.05 mol l−1 thiourea solution and the eluate was directly introduced into an ICP-OES. The proposed method was successfully applied to the determination of traces of palladium in JSd-2 stream sediment certified reference material [0.019 ± 0.001 μg g−1 (n = 3); provisional value: 0.0212 μg g−1] and SRM 2556 used auto catalyst certified reference material [315 ± 4 μg g−1 (n = 4); certified value: 326 μg g−1]. The detection limit (3σ) of 0.28 ng ml−1 was obtained for 5 ml of sample solution. The sample throughputs for 5 ml and 100 μl of the sample solutions were 10 and 15 h−1, respectively.  相似文献

A flow system was coupled to a graphite furnace with a platform coated with tungsten-rhodium permanent chemical modifier for in-line separation and preconcentration of copper by employing a minicolumn loaded with 1-(2-tiazolylazo)-2-naphthol (TAN) immobilized on C18-bonded silica fixed in the tip of the autosampler arm. Elution was made by sampling 35 μl of 0.50 mol l−1 HCl with further delivering into a coated platform. Remarkable improvements in both selectivity and sensitivity were observed. Copper(II) was effectively separated from solutions containing up to 20 g l−1 Na+; 10 g l−1 K+, Ca2+ and Mg2+; 1.0 g l−1 Fe3+ and Zn2+. For a sample flowing at 3.0 ml min−1 and a loading of 60 s, the detection limit was estimated as 5 ng l−1 Cu(II) at the 99.7% confidence level, and an enrichment factor of 33 was calculated. Coefficient of variation was estimated as 4% for a 0.30 μg l−1 copper solution (n=20). The W-Rh permanent chemical modifier was used to improve system stability, analytical performance and atomizer lifetime. More than 1500 firings were carried out with the same atomizer without significant variations in sensitivity and precision. On account of the reagent immobilization, its consumption was lower than 0.2 μg per determination. In addition, TAN purification was unnecessary.  相似文献

A single automatic method for continuous flow determination of β-naphthol based on the enhancement of its native fluorescence once the analyte was transitorily retained on-line on a solid support (QAE A-25 resin) is reported. So, a flow-through optosensor was developed using a flow-injection analysis system with solid phase fluorimetric transduction. KCl (0.15 mol l−1) at pH 12.0 was used as carrier solution. To obtain the optimum fluorescence signal the wavelengths chosen were 245 nm (excitation) and 420 nm (emission). The response of the sensor was directly proportional to the sample volume injected in the studied range 40-1500 μl. Approximately one higher order of magnitude is achieved in sensitivity when 1500 μl are used with respect to the use of 40 μl of sample. The sensor was calibrated for three different injection volumes: 40, 600 and 1500 μl, responding linearly in the measuring range of 2-60, 0.5-15 and 0.2-5 μg l−1 with detection limits of 0.5, 0.09 and 0.05 μg l−1, respectively. The relative standard deviation for ten independent determination is 0.6% (40 μl), 0.9% (600 μl) and 2.3% (1500 μl). A recovery study was performed onto three different spiked water samples at concentration levels from 1 to 2.5 μg l−1 and the recovery percentage from the experimental data ranged between 101±2 and 105±5.  相似文献

The use of a permeation liquid membrane system for the preconcentration and separation of nickel in natural and sea waters and subsequent determination by atomic absorption spectroscopy is presented. 2-Hydroxybenzaldehyde N-ethylthiosemi-carbazone (2-HBET) in toluene is used as the active component of the liquid membrane. A study strategy based on a simplex design has been followed. Several chemical and physical parameters were optimized. Maximum permeation coefficient was obtained at a feed solution pH of 9.4, 0.3 mol l−1 of HNO3 in the stripping solution and 1.66 mmol l−1 of 2-HBTE in toluene as carrier. The precision of the method was 4.7% at 95% significance level and a detection limit of 0.012 μg l−1 of nickel was achieved. The preconcentration procedure showed a linear response within the studied concentration range from 3 to 500 μg l−1 of Ni in the feed solution. The method was validated with different spiked synthetic seawater and certified reference water samples: TMDA-62 and LGC 6016, without matrix interferences and showing good concordance with the certified values, being the relative errors −5.9% and −2.2%, respectively. Under optimal conditions, the average preconcentration yield for real seawater samples was 98 ± 5%, with a nickel preconcentration factor of 20.83 and metal concentrations ranging between 2.8 and 5.4 μg l−1.  相似文献

The existence of dimethylselenium (DMSe) and dimethyldiselenium (DMDSe) in some environmental samples can cause serious interference on Se(IV) determination by hydride generation atomic fluorescence spectrometry (HG-AFS) due to their contribution on HG-response. A flow injection separation and preconcentration system coupled to HG-AFS was therefore developed by on-line coprecipitation in a knotted reactor (KR) for eliminating interference subjected from organoselenium. The sample, spiked with lanthanum nitrate, was merged with an ammonium buffer solution (pH 8.8), which promoted coprecipitation of Se(IV) and quantitative collection by 150 cm PTFE KR. DMSe and DMDSe, however, were unretained and expelled from the KR. An air flow was introduced to remove the residual solution from the KR, then a 1.2 mol l−1 HCl was pumped to dissolve the precipitates and merge with KBH4 solution for HG-AFS detection. The interference of DMSe and DMDSe on the Se(IV) determination by conventional HG-AFS and its elimination by the developed separation and preconcentration system were evaluated. With optimal experimental conditions and with a sample consumption of 12.0 ml, an enhancement factor of 18 was obtained at a sample frequency of 24 h−1. The limit of detection was 0.014 μg l−1 and the precision (R.S.D.) for 11 replicate measurements of 1.0 μg l−1 Se(IV) was 2.5%. The developed method was successfully applied to the determination of inorganic selenium species in a variety of natural water samples.  相似文献

An automated and versatile sequential injection spectrofluorimetric procedure for the simultaneous determination of multicomponent mixtures in micellar medium without prior separation processes is reported. The methodology is based upon the segmentation of a sample slug between two different buffer zones in order to attain both an improvement of sensitivity and residual minimization for the whole species. Resolution of overlapping fluorescence profiles is achieved using a variable angle scanning technique coupled to multivariate least-squares regression (MLR) algorithms at both sample edges.The potentialities of the described methodology are illustrated with the spectrofluorimetric determination of four widespread pesticides with different acid-base properties; viz. carbaryl (CBL) (1-naphthyl-N-methylcarbamate), fuberidazole (FBZ) (2-(2′-furyl)benzimidazole), thiabendazole (TBZ) (2-(4′-thiazolyl)benzimidazole) and warfarin (W) (3-α-acetonylbenzyl)-4-hydroxycoumarin). Detection limits at the 3σ level were 3.9, 0.02, 0.03 and 10 μg l−1 for CBL, FBZ, TBZ and W, respectively at the maximum sensitivity pH. Dynamic ranges of 13-720 μg l−1 CBL, 0.10-14 μg l−1 FBZ, 0.19-60 μg l−1 TBZ and 0.05-5 mg l−1 W were achieved. Relative standard deviations (n=10) were 0.2% for 100 μg l−1 CBL and 2.4 μg l−1 FBZ, 0.7% for 8 μg l−1 TBZ and 1.0% for 1 mg l−1 W. The proposed automated methodology, which handles 17 samples/h, was validated and applied to spiked real water samples with very satisfactory results.  相似文献

A new multisyringe flow injection system for total inorganic selenium determination by hydride generation-atomic fluorescence spectrometry (HGAFS) has been proposed. The flow methodology is based on the simultaneous injection of sample in the acid media (50% HCl), a reducing sodium tetrahydroborate solution (0.18%) and a solution of hydrochloric acid (50%) which are dispensed into a gas-liquid separation cell by using a multisyringe burette coupled with one multiport selection valve. The usage of the time-based injection increases the sample throughput and provides precise known volumes of sample. The hydride of selenium is delivered into the flame of an atomic fluorescence spectrometer by means of an argon flow. A hydrogen flow has been used to support the flame.The technique can be applied over a wide range of concentrations of selenium between 0.1 and 3.5 μg l−1 with good repeatability (relative standard deviation (R.S.D.) values 4.6-7% for 1 μg l−1 of Se). The detection limit of the developed technique (3σb/S) was 0.01 μg l−1. A sample throughput was 28 samples per hour (84 injections). The multisyringe technique has been validated by means of reference solid (sea lettuce) and water (hard drinking water) materials with good agreement with certified values. The analytical features were compared with those obtained by using of the commercial flow injection analysis (FIA) system. The proposed method provides a higher sampling frequency and a significant reduction of reagent and sample consumption in front the flow injection application.  相似文献

A not expensive automatic flow system based on multicommutation and exploiting the liquid-liquid extraction methodology for the determination of lead in plant material is described. The spectrophotometric procedure for lead determination was based on the reaction with dithizone followed by extraction using an organic solvent. The facilities afforded by the multicommutation approach allowed the use of an air stream as carrier, thus contributing to reduce the overall waste generation. The results obtained analysing plant materials compare very well with those obtained employing inductive coupled plasma optical emission spectrometry (ICP OES) at 90% confidence level. Others profitable features such as a linear response range between 50 and 200 μg l−1 Pb (r = 0.999); a sampling rate of 15 determination per hour; a relative standard deviation of 1.8% (n = 12) for a typical sample containing 163 μg l−1 Pb; a detection limit of 12 μg l−1; a reagent consumption of 4.5 mg dithizone; and a waste generation of 225 μl organic solvent per determination were also achieved.  相似文献

In this work, Amberlite XAD-2 resin functionalized with 4,5-dihydroxy-1,3-benzenedisulfonic acid was synthesized, characterized and applied as a new packing material for an on-line system to nickel preconcentration. The method is based on the sorption of Ni(II) ions in a minicolumn containing the synthesized resin, posterior desorption using an acid solution and measurement of the nickel by spectrophotometry (PAR method). The optimization of the system was performed using factorial design and Doehlert matrix considering five variables: eluent concentration, PAR solution pH, sample flow rate, PAR solution concentration and sample pH. Signals were measured as peak height by using an instrument software. Using the experimental conditions defined in the optimization, the method allowed nickel determination with achieved sampling rate of 25 samples per hour, detection limit (3 s) of 2 μg l−1 and precision (assessed as the relative standard deviation) of 8.2-2.6%, for nickel solutions of 10.0-200.0 μg l−1 concentration, respectively. The experimental enrichment factor of the proposed system was 46, for 120 s preconcentration time. The proposed procedure was applied for nickel determination in food samples. Recoveries of spike additions (5 or 10 μg g−1) to food samples were quantitative (94-110%).  相似文献

In the present paper, an on-line system for preconcentration and determination of zinc by Flame Atomic Absorption Spectrometry (FAAS) is proposed. It is based in the sorption of zinc(II) ions on a minicolumn packed with polyurethane foam loaded with 2-[2′-(6-methyl-benzothiazolylazo)]-4-bromophenol (Me-BTABr) reagent. Chemical and flow variables as pH effect, sample flow rate and eluent concentration were optimized using univariate methodology. The results demonstrated that zinc can determinate using the sample pH in the range of 6.5-9.2, sample flow rate of 6.0 ml min−1, and the elution step using 0.10 mol l−1 hydrochloric acid solution at flow rate of 5.5 ml min−1. In these conditions, an enrichment factor of 23 and a sampling rate of 48 samples per hour were achieved. The detection limit (DL, 3σ) as IUPAC recommendation was 0.37 μg l−1 and the precision (assessed as the relative standard deviation, R.S.D.) reached values of 5.9-1.8% in zinc solutions of 1.0-10.0 μg l−1 concentration, respectively. The method was successfully applied to the determination of trace amounts of zinc in natural water samples from Salvador (Brazil).  相似文献

A time-based multisyringe flow injection procedure with spectrofluorimetric detection is proposed in this paper for the determination of aluminium in drinking water. The flow methodology is based on the simultaneous or sequential injection of sample and chelating reagent (viz. 8-hydroxyquinoline-5-sulphonic acid) plugs using a multicommutation approach so that three successive injections may be performed with a sole displacement of the piston driver bar of the burette. Thus, an injection throughput as high as 154 h−1 is achieved by sampling a 182 μl sample zone. In order to enhance the luminescence, the reaction is carried out in micellar medium using hexadecyltrimethylammonium chloride as surfactant. The influence of geometric and hydrodynamic variables as well as several parameters such as multicommutation timing, ligand and surfactant concentration and reagent pH was assessed.Under the selected working conditions, a linear dynamic range from 10 to 500 μg l−1 Al(III), a 3σ detection limit of 0.5 μg l−1 and a coefficient of variation of 0.6% at the 30 μg l−1 level were obtained. The analytical features were compared with those reported in previous flow injection and sequential injection methods. The multisyringe technique was successfully applied to the determination of aluminium in drinking water at low mineralisation levels, validating the results by inductively coupled plasma atomic emission spectrometry.  相似文献

A sensitive and selective method was developed for the determination of traces of manganese in urine using on-line electrochemical preconcentration followed by flame atomic absorption spectrometry detection. A home made flow-through polypropylene cell (4.5 cm long × 0.8 cm diameter filled with glass marbles) with an effective inner volume of 0.5 ml containing a working and a counter electrode, both of glassy carbon and a Pt pseudo reference electrode was located in a flow injection manifold specially designed for the purpose of this work. The manganese was deposited from buffer solution of NH3/NH4Cl at pH 9.00 through an oxidizing process at a current of 400 mA during 7 min. A flow of HCl 0.1 mol l−1 at 4 ml min−1 through the cell, chemically dissolved the deposit. A small portion (15 μl) of the concentrate was introduced in a continuously flowing system by means of a timing device and was then carried to the detector for the manganese quantification. All electrochemical and spectroscopic variables as well as possible interferences in both systems were systematically studied. The relative standard deviations for ten consecutive measurements of manganese solutions of 2.0 and 20 μg l−1 were of 2.3 and 1.5%, respectively, while for a sample processed five times was less then 5%. The accuracy of the developed procedure was evaluated by adding known amounts of manganese standard to urine samples and following the whole procedure. Recoveries within the range 97.2-102.8% were obtained. To further prove the accuracy, a Seronorm Trace Elements in Urine, Batch 403125 sample with a reported concentration of 13 μg Mn l−1 was also analyzed. The experimental value obtained was of 12.7 ± 0.1 μg l−1, which does not differ significantly from the reported amount (p < 0.05). A preconcentration factor of 40, a linear range between 0.015 and 60 μg l−1 and a limit of detection of 15 ng l−1 permitted the determination of manganese in real urine samples from non-exposed subjects in the range 0.5-2.8 μg l−1.  相似文献

A selective matrix removal/separation/enrichment method, utilizing a microcolumn of a chelating resin with SH functional groups (Duolite GT-73), was proposed for the determination of Sb(III) in waters by segmented flow injection-hydride generation atomic absorption spectrometry (SFI-HGAAS). The resin was selective to Sb(III) at almost all pH and acidity values employed, whereas Sb(V) was not retained at all and could be determined after a pre-reduction step with l-cysteine. Spike recoveries were tested at various concentration levels in different water types and were found to vary between 85 and 118%. Accuracy of the proposed methodology was checked by analyzing a standard reference material and a good correlation was found between the determined (13.3 ± 1.1 μg l−1) and the certified value (13.79 ± 0.42 μg l−1). The method was applied to several bottled drinking water samples for antimony determination with and without preconcentration and none of the samples were found to contain antimony above the permissible level (5 μg l−1). The characteristic concentration (the concentration of the analyte corresponding to an absorbance of 0.0044) was 0.55 μg l−1 and the 3 s limit of detection (LOD) based on five times preconcentration was 0.06 μg l−1. The applicability of the microcolumn separation/preconcentration/matrix removal method for flow injection systems was also demonstrated.  相似文献

In the present work, thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS) was employed for Cu and Zn determination in bovine milk and fruit juice samples without any pretreatment. TS-FF-AAS system was optimized and a sample volume of 300 μl was injected into the carrier stream (0.014 mol l−1 HNO3 at a flow rate of 0.4 ml min−1), and it was introduced into a hot Ni tube. The detection limits obtained for Cu and Zn in aqueous solution were 2.2 and 0.91 μg l−1, respectively, and 3.2 μg l−1 for Cu in a medium containing water-soluble tertiary amines. The relative standard deviations varied from 2.7 to 4.2% (n=12). Sample preparation was carried out by simple dilution in water or in water-soluble tertiary amines medium. Accuracy was checked by performing addition-recovery experiments as well as by using reference materials (whole milk powder, non-fat milk powder, and infant formula). Recoveries varied from 97.7 to 105.3% for Cu and Zn. All results obtained for reference materials were in agreement with certified values at a 95% confidence level.  相似文献

This paper reports the development of a new strategy for low-level determination of copper in water samples by using a flow-injection system coupled to solid-phase extraction (SPE) using flame atomic absorption spectrometry (F AAS) as detector. In order to preconcentrate copper from samples, a minicolumn packed with a styrene-divinylbenzene resin functionalized with (S)-2-[hydroxy-bis-(4-vinyl-phenyl)-methyl]-pyrrolidine-1-carboxylic acid ethyl ester was used and the synthesis procedure is described. System operation is based on the on-line retention of Cu(II) ions at pH 9.0 ± 0.2 in a such minicolumn with posterior analyte elution with 2 mol l−1 HCl directly to the F AAS nebulizer. The influence of several chemical (sample pH, buffer concentration, HCl eluent concentration and effect of the ionic strength) and flow (sample and eluent flow rates and preconcentration time) variables that could affect the performance of this system were investigated as well as the possible interferents. At optimized conditions, for 2 min of preconcentration time (13.2 ml of sample volume), the system achieved a detection limit of 1.1 μg l−1, a R.S.D. 1% at 20 μg g l−1 and an analytical throughput of 25 h−1, whereas for 4 min of preconcentration time (26.4 ml of sample volume), a detection limit of 0.93 μg l−1, a R.S.D. 5.3% at 5 μg l−1 and a sampling frequency of 13 h−1 were reported.  相似文献

This paper deals on the determination of Strychnine, a potent and dangerous pesticide and the analytical procedure is based on the photo-induced chemiluminescence of the pesticide by means of the Multicommutation continuous-flow methodology. Small segments of the pesticide solution were sequentially alternated with segments of the solution for adjusting the suitable medium for the photodegradation. The required time of UV irradiation was obtained by stopped-flow during 150 s; then, the resulting solution formed alternated segments with the oxidizing solution containing 5 × 10−3 mol l−1 Ce(IV) in 0.6 mol l−1 nitric acid. The calibration range, from 2 μg l−1 to 50 mg l−1, resulted in a linear behaviour over the range 25 μg l−1 to 20 mg l−1 and fitting the equation: I = 4706x + 624 with a correlation coefficient of 0.9955. The limit of detection was 2 μg l−1 and the sample throughput 15 h−1. After testing the influence of a large series of potential interferents, the method was applied to different kinds of samples.  相似文献

A novel method for the determination of ethanol in tequila based on the immobilized enzyme fluorescence capillary analysis (IE-EFCA) has been proposed. Alcohol dehydrogenase (ADH) was immobilized in inner surface of a capillary and an immobilized enzyme capillary bioreactor (IE-ECBR) was formed. After nicotinamide adenine dinucleotide (NAD+) as an oxidizer is mixed with alcohol sample solution, it was sucked into the IE-ECBR. The fluorescence intensity of the mixed solution in the IE-ECBR was detected at λex = 350 nm and λem = 459 nm. The experimental conditions are as follows: The reaction time is 20 min; temperature is 40 °C; the concentrations of phosphate buffer solution (pH 7.5) and NAD+ are 0.1 mol L−1 and 5 mmol L−1, respectively; immobilization concentration of ADH is 10 U L−1. The determination range of ethanol is 2.0-15.0 g L−1 (F = 10.44C + 6.6002, r > 0.9958); its detection limit is 1.11 g L−1; and relative standard deviation is 1.9%. IE-EFCA method is applicable for the determination of the samples containing alcohol in medicine, industry and environment.  相似文献

An automated system to perform liquid-liquid extraction is proposed, in which the effective mixture (the intimate contact) between the aqueous phase and the organic phase, as well as the separation of the phases, are carried out in a micro-batch glass extraction chamber. Sample, reagents and organic solvent are introduced into the glass extraction chamber by a peristaltic pump using air as carrier. The detection of the extracted species from the aqueous phase is made in a small volume (120-150 μl) of isobutyl methyl ketone (MIBK). The system allows enrichment factors of 2-10-fold. The proposed automatic system was evaluated for Cu(II) extraction based on complex formation between copper(II) and 1-(2′-pyridylazo)naphthol (PAN) in MIBK. When a volumetric ration of 2:1 (aqueous:organic) was implemented, copper was detected in the concentration range of 100-1600 μg l−1 (r = 0.9995) with a relative standard deviation of 2% (200 μg l−1, n = 5) and a detection limit of 20 μg l−1. The analytical curve was linear over the concentration range 25-500 μg l−1 (r = 0.9994) when a volumetric ratio of 10:1 was employed.
