Reaction of 3-R-chromones with dimedone and ammonium acetate leads to 2-(2-hydroxyaryl)-7,7-dimethyl-7,8-dihydroquinolin-5(6H)-ones (R = CF3CO, H), 3-(2-hydroxyaroyl)-7,7-dimethyl-7,8-dihydroquinolin-5(6H)-ones (R = CHO), and 8,8-dimethyl-8H-chromeno[2,3-b]quinoline-10,12(7H,9H)-diones (R = CN) depending on the nature of substituent R.  相似文献

Reaction of dialkyl (2S,3S)- or (2S,3R)-tetrahydro-3-hydroxy-5-oxo-2,3-furandicarboxylates with POCl3 in pyridine followed by diazomethane resulted in the isolation of dialkyl 2S-4-methoxy-5-oxo-2,5-dihydro-2,3-furandicarboxylates, which are analogues of the Quararibea metabolite chiral enolic-γ-lactone (3-hydroxy-4,5-(R)-dimethyl-2(5H)-furanone). An unusual α-hydroxylation of γ-butyrolactone takes place involving POCl3 in pyridine. When the dehydration was facilitated with methanesulfonyl chloride in triethylamine, instead of POCl3, aromatic dialkyl 5-[(methylsulfonyl)oxy]-2,3-furandicarboxylates were obtained.  相似文献

Libraries of skeletally diverse potential bioactive polycyclic/spirocyclic heterocyclic compounds; 2-amino-7,9-dimethyl-5-oxo-4-aryl-4,5,6,7-tetrahydropyrano[2,3-d]pyrazolo[3,4-b]pyridine-3-carbonitrile, 2′-amino-7′,9′-dimethyl-2,5′-dioxo-6′,7′-dihydro-5′H-spiro[indoline-3,4′-pyrano[2,3-d]pyrazolo[3,4-b]pyridine]-3′-carbonitrile, and 5,5′-(arylmethylene)bis(4-hydroxy-1,3-dimethyl-1H-pyrazolo[3,4-b]pyridin-6(7H)-one) have been synthesized through a multi-component reaction using novel heterocyclic active methylene compound 4-hydroxy-1,3-dimethyl-1H-pyrazolo[3,4-b]pyridine-6(7H)-one as one of the building blocks. This protocol can be considered to be an efficient and eco-friendly strategy for diversity oriented synthesis.  相似文献

Isopropyl 2-(1-aryl-4,5-dioxo-2-phenyl-4,5-dihydro-1H-pyrrol-3-yl)-2-oxoacetates reacted with N-substituted 3-amino-5,5-dimethylcyclohex-2-en-1-ones to give the corresponding 1′-substituted (Z)-6′,6′-dimethyl-3-[phenyl(arylamino)methylidene]-6′,7′-dihydro-3H-spiro[furan-2,3′-indole]-2′,4,4′,5(1′H,5′H)-tetraones. The structure of 1′-benzyl-6′,6′-dimethyl-3-[(Z)-phenyl(phenylamino)methylidene]-6′,7′-dihydro-3Hspiro[furan-2,3′-indole]-2′,4,4′,5(1′H,5′H)-tetraone was proved by X-ray analysis.  相似文献

A convenient and rapid synthesis of hitherto unknown 3-aroyl-4-aryl-2-phenylamino-4H-benzo[g]chromene-5,10-diones in high yield from β-aroyl-thioacetanilide, aromatic aldehyde, and 2-hydroxy-1,4-naphthoquinone via InCl3 catalyzed one-pot three-component tandem Knoevenagel condensation–Michael addition–intramolecular cyclization–elimination reaction sequence is disclosed for the first time. This domino protocol has been used to obtain highly substituted pyrano[3,2-c]chromen-5(4H)-ones and 7,7-dimethyl-7,8-dihydro-4H-chromen-5(6H)-ones from N,3-diaryl-3-oxo-propanethioamide, aromatic aldehyde, and 4-hydroxycoumarine or dimedone under mild reaction conditions. A plausible reaction mechanism is proposed. The 4H-pyrano[3,2-c]chromen-5-one and 7,7-dimethyl-7,8-dihydro-4H-chromen-5(6H)-one derivatives possessing 3-(2-chlorobenzoyl)-2-phenylamino-substituents further cyclized under basic conditions to yield penta-cyclic 7,13-diaryl-5,14-dioxa-13-aza-benzo[a]naphthacen-6,8(7H,13H)-dione and tetra-cyclic 6,12-diaryl-3,3-dimethyl-3,4-dihydro-2H-chromeno[2,3-b]quinolin-1,11(6H,12H)-dione, respectively.  相似文献

Methyl 3-aroyl-1-aryl-4,5-dioxo-4,5-dihydro-1H-pyrrole-2-carboxylates reacted with 3-amino-5,5-dimethylcyclohex-2-en-1-one having no substituent on the nitrogen atom to give 3-aroyl-4-arylamino-6′,6′-dimethyl-6′,7′-dihydro-5H-spiro[furan-2,3′-indole]-2′,4′,5′(1′H,5′H)-triones or methyl 12-aroyl-11-aryl-9-hydroxy-5,5-dimethyl-3,10-dioxo-8,11-diazatricyclo[7.2.1.02,7]dodec-2(7)-ene-1-carboxylates. The latter underwent thermal recyclization to 3′-aroyl-1′-aryl-4′-hydroxy-6,6-dimethyl-6,7-dihydrospiro[indole-3,2′-pyrrole]-2,4,5′(1H,1′H,5H)-triones.  相似文献

A novel Schiff base, N-(4-amino-2,3-dimethyl-1-phenyl-3-pyrazolin-5-one)pyridoxaldimine (HL·HCl), was prepared and structurally characterized on the basis of elemental analyses, 1H and 13C NMR, and IR spectral data. The synthesis and characterization of several Cu(II) (1-6) and V(IV) (7) complexes with N-(4-amino-2,3-dimethyl-1-phenyl-3-pyrazolin-5-one)pyridoxaldimine are reported. The composition and structures of the copper(II) and vanadium(IV) complexes were proposed based on elemental analyses, molar conductance, magnetic susceptibility measurements, IR, electronic and EPR spectroscopy. In addition, the structures of the ligand and the complex [CuL(H2O)2]NO3·2.25H2O (1) have been determined by single-crystal X-ray diffraction, showing that the Cu(II) center has a distorted square-pyramidal geometry. The ligand and the complexes were also tested for their in vitro antibacterial activity.  相似文献

Reactions of N-substituted 2-polyfluoroalkyl-4-quinolones and 8-aza-5,7-dimethyl-2-polyfluoroalkylchromones with (trifluoromethyl)trimethylsilane proceed mainly as a 1,4-nucleophilic trifluoromethylation to give N-substituted 2,2-bis(polyfluoroalkyl)-2,3-dihydroquinolin-4(1H)-ones and 5,7-dimethyl-2,2-bis(polyfluoroalkyl)-2,3-dihydro-4H-pyrano[2,3-b]pyridin-4-ones after acid hydrolysis. Similar reaction with 2-trifluoromethyl-4H-thiochromen-4-one proceeds as a 1,2-addition to give 2,4-bis(trifluoromethyl)-4H-thiochromen-4-yl trimethylsilyl ether.  相似文献

The system Pd(OAc)2/BQ/Co(acac)3 (BQ=benzoquinone), in combination with tetrabutylammonium bromide (TBAB) as a surfactant agent and a chelating ligand such as 2,9-dimethyl-1,10-phenanthroline (dmphen) or 2,9-dimethyl-4,7-diphenyl-1,10-phenanthroline (dmdpphen), is an efficient catalyst for the oxidative carbonylation of phenol to diphenyl carbonate (DPC). The best results have been obtained using the system Pd(OAc)2/BQ/Co(acac)3/dmphen=1/30/8/5 (molar ratio) in which [Pd]=10−3 mol l−1 and TBAB/Pd=60/1. This system gives the maximum productivity of 700 mol DPC/mol Pd h at 135°C and under Ptot=60 atm (CO/O2=10/1 molar ratio). The role of each component of the catalytic system is discussed and a catalytic cycle is proposed.  相似文献

A series of novel N-benzylcarboxamide derivatives of bicyclic compounds, 3,4-dihydropyrido[3,2-f][1,4]oxazepin-5(2H)-one and 2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-one, were synthesized by cyclization of N-benzyl-2-chloro-N-(2-hydroxyethyl)- [and -(3-hydroxypropyl)-] nicotinamides, respectively. Atropisomerism was observed in 5-[3,5-bis(trifluoromethyl)benzyl]-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-ones due to steric hindrance of the carboxamide moiety and restriction of its rotation. Cyclization of N-[3,5-bis(trifluoromethyl)benzyl]-2-chloro-N-[(2S)-3-hydroxy-2-methylpropyl]-5-methyl-4-phenylnicotinamide gave (3S)-5-[3,5-bis(trifluoromethyl)benzyl]-3,8-dimethyl-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3b][1,5]oxazocin-6-one, which exists predominantly in the thermodynamically stable aR-conformer in CDCl3. This compound showed excellent NK1-antagonistic activity with IC50 value (in vitro inhibition of [125I]-Bolton-Hunter-substance P binding in human IM-9 cells) of 0.47 nM, which is ca. 200-fold more potent than that of its enantiomer, indicating that the atropisomer chirality affects NK1-receptor recognition.  相似文献

A new, efficient, and environmentally benign protocol for the one-pot, four-component synthesis of 2,2-dimethyl-13-phenyl-2,3-dihydro-1H-indazolo[2,1-b]phthalazine-1,6,11(13H)-trione by condensation of phthalic anhydride, hydrazinium hydroxide, aromatic aldehydes, and dimedone catalyzed by Ce(SO4)2·4H2O as an ecofriendly catalyst with high catalytic activity and reusability at 125 °C under solvent-free conditions is reported. The reaction proceeds to completion within 5-10 min in 71-95% yield. To the best of our knowledge, this new procedure provides the first example of an efficient synthetic method for 2H-indazolo[2,1-b]phthalazine-1,6,11(13H)-trione via a four-component reaction.  相似文献

2,3-Homotroponeiron tricarbonyl, 8-methyl- and 8,8-dimethyl-2,3-homotroponeiron tricarbonyl complexes have been shown to undergo O-protonation in trifluoroacetic (TFA) and 96% sulfuric acids. In the latter acid the O-protonated cations rearrange to give the thermodynamically more stable C-protonated isomers. Cyclooctatrienoneiron tricarbonyl undergoes protonation H2SO4 to give the same cation as was obtained from the protonation of the homotroponeiron tricarbonyl complex in H2SO4. On the basis of reaction in D2SO4, it is suggested that the kinetically preferred site of protonation of the cyclooctatrienone complex is at C(2) one of the coordinated carbon atoms.  相似文献

Nitration of 2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-one gave its 5-nitro derivative which was subjected to alkylation with dimethyl sulfate, diethyl sulfate, and benzyl(dimethyl)phenylammonium chloride. The resulting 1,3-dimethyl-, 1,3-diethyl-, and 1,3-dibenzyl-5-nitro-2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-ones were reduced to the corresponding 1,3-dialkyl-5-amino-2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-ones, and the latter reacted with itaconic acid to produce 1-(1,3-dialkyl-2-oxo-2,3-dihydro-1H-imidazo[4,5-b]pyridin-5-yl)-5-oxopyrrolidine-3-carboxylic acids. 1-(2-Oxo-2,3-dihydro-1H-imidazo[4,5-b]pyridin-5-yl)-5-oxopyrrolidine-3-carboxylic acid was obtained by analogous reaction with 5-amino-2,3-dihydro-1H-imidazo[4,5-b]-pyridin-2-one.  相似文献

2-(1-Iodoethyl)-3,5-dimethyl-1-(4-methylphenylsulfonyl)-2,3-dihydro-1H-indole reacted with pyridine, piperidine, N-alkylpiperidines, and dimethylformamide to give dehydrohalogenation and halogen substitution products whose ratio depended on the reagent structure. Six new copper(II) complexes, CuLCl·H2O (1), CuL(NO3)·2H2O (2), [Cu(L)2] (3), CuL(SCN)·2H2O (4), CuL(ClO4)·2H2O (5) and (CuL)2(SO4)·4H2O (6), where HL = 1-phenyl-2,3-dimethyl-4-(N-2-hydroxy-4-methoxy-benzaldehyde)-3-pyrazolin-5-one, have been synthesized. The characterization of the newly formed compounds was done by 1H NMR, UV-Vis, IR, ESR spectroscopy, elemental analysis and molar electric conductivity. The crystal structure of 1-phenyl-2,3-dimethyl-4-(N-2-hydroxy-4-methoxy-benzaldehyde)-3-pyrazolin-5-one has been determined by X-ray diffraction studies, as well as the crystal structure of one of its copper(II) complexes, [Cu(L)2] (3). The copper atom is coordinated to two nitrogen and two oxygen atoms of the Schiff base ligand. The in vitro antibacterial activity against Klebsiella pneumoniae ATCC 100131, Staphylococcus aureus var. Oxford 6538, Pseudomonas aeruginosa ATCC 9027 and Escherichia coli ATCC 10536 strains was studied and compared with that of free ligand. The anti-microbial activity was dependent on the microbial species tested and the metal salt anion used.  相似文献

Summary Novel mixed-ligand nickel(II) complexes having octahedral stereochemistry and general formula [Ni-(acac)2(L)2]/[Ni(meacac)2(L)2]/[Ni(etacac)2(L)2] (where L=3-methyl-1-phenyl-2-pyrazoline-5-one, 2,3-dimethyl-4-methylaminomethanesulphonate sodium monohydrate-1-phenyl-3-pyrazoline-5-one or 4-amino-2,3-dimethyl-1-phenyl-3-pyrazoline-5-one) have been isolated by reacting [Ni(acac)2(H2O)2]/[Ni(meacac)2(H2O)2]/ [Ni(etacac)2(H2O)2] with L in alcohol. Reaction of 3-Dimethylamino-2,2-dimethyl-2H-azirine with Barbituric Acid The reaction of 3-dimethylamino-2,2-dimethyl-2H-azirine (1) with barbituric acid (4) in dimethyl formamide at room temperature yields a mixture of several compounds. The two main products 5 and 6 have been isolated in 40 and 10% yield, respectively, and their structures established by X-ray analysis. In Schemes 4–6 reaction mechanisms for the formation of 5 and 6 are postulated, the first step beeing either a C- or an N-alkylation of barbituric acid. Reduction of 5 and 6 with NaBH4 in ethanol at room temperature yields 6,6-dimethyl-1,5,6,7-tetrahydro-pyrrolo[2,3-d]pyrimidin-2,4(3H)-dione (7) and 3,3-dimethyl-2,3-dihydro-imidazo[1,2-c]pyrimidin-5,7(1H, 6H)-dione (8) in 38 and 48% yield, respectively. Treatment of 6 with 3N aqueous NaOH at room temperature gives 3,3-dimethyl-imidazo[1,2-c]pyrimidin-2,5,7 (1H, 3H, 6H)-trione (9) in 51% yield (Scheme 3).  相似文献
